RESUMO

To explore the anti-tumor effects of Radix Astragali on osteosarcoma and its mechanism. We analyzed the PPI network of Radix Astragali's potential targets for treating osteosarcoma and got the hub targets. We used KM curves to screen hub targets that could prolong sarcoma patients' survival time. We performed GO and KEGG enrichment analysis of Radix Astragali's potential targets and predicted Radix Astragali's molecular mechanism and function in treating osteosarcoma. The binding process between the hub targets, which could prolong sarcoma patients' survival time, and Radix Astragali was simulated through molecular docking. PPI network analysis of potential therapeutic targets discriminated 25 hub targets. The KM curves of the hub targets showed there were 13 hub targets that were effective in improving the 5-year survival rate of sarcoma patients. GO and KEGG enrichment demonstrated that Radix Astragali regulates multiple signaling pathways of osteosarcoma. Molecular docking results indicated that Radix Astragali could bind freely to the hub target, which could prolong the sarcoma patient's survival time. Radix Astragali act on osteosarcoma by regulating a signaling network formed by hub targets connecting multiple signaling pathways. Radix Astragali has the potential to become a drug for treating osteosarcoma and prolonging the sarcoma patient's survival time.

Osteosarcoma (OS) is a malignant bone tumor of mesenchymal origin. Angelica dahurica is a typical traditional Chinese herb. Angelica dahurica is used in the treatment of a variety of tumors. However, the studies of Angelica dahurica for OS have not been reported. To investigate Angelica dahurica's potential mechanism of action in the treatment of OS, we used network pharmacology and molecular docking methods in this study. Of which the network pharmacology includes the collection of active ingredients of Angelica dahurica, the collection of predicted targets of Angelica dahurica and predicted targets of OS, the analysis of therapeutic targets of Angelica dahurica, gene ontology (GO) enrichment, and Kyoto encyclopedia of genes and genomes (KEGG) enrichment. The Venn plot performance showed that there were 225 predicted targets of Angelica dahurica for the treatment of OS. The therapeutic targets enrichment analysis results showed that Angelica dahurica treated OS through multiple targets and pathways. Angelica dahurica could affect OS's proliferation, apoptosis, migration, infiltration, and angiogenesis through a signaling network formed by pivotal genes crosstalking numerous signaling pathways. In addition, molecular docking results showed that sen-byakangelicol, beta-sitosterol, and Prangenin, have a relatively high potential to become a treatment for patients with OS and improve 5-year survival in OS patients. We used network pharmacology and molecular docking methods to predict the active ingredients and significant targets of Angelica dahurica for the treatment of OS and, to a certain extent, elucidated the potential molecular mechanism of Angelica dahurica in the treatment of OS. This study provided a theoretical basis for Angelica dahurica in the treatment of OS.

The higher extraction efficiency of analytes is crucial for developing immunoassays with high accuracy. Here, we evaluated the extraction efficiency of neonicotinoids in tea samples in terms of grinding degrees, extraction solvents types and contents. Fragments for fresh tea leaves (1 g, 5-10 mm2) or tea powder (1 g, 35 mesh) for commercial tea was extracted with 100 % methanol. The extraction (1 mL) was diluted 10-fold with buffer solution, and then submitted to gold nanoparticles-based lateral flow immunoassay. This optimal extraction protocol exhibited a higher extraction efficiency (72.4-99.3 %) for the positive neonicotinoids samples. The cut-off values of lateral flow immunoassay were 0.325 or 0.65 µg/g, 0.3 or 0.45 µg/g, 0.3 or 0.45 µg/g, 0.03 or 0.06 µg/g for thiamethoxami, clothianidin, acetamiprid and midacloprid in fresh tea leaves and commercial tea. In summary, this proposed lateral flow immunoassay can be used as the point-of-needs analysis method for four neonicotinoids in tea.

Osteosarcoma (OS) is a malignant bone tumor of mesenchymal origin. Tripterygii Wilfordii (TW) is a traditional Chinese medicine widely used for its anti-inflammatory and immunomodulatory effects. Various components of TW have been shown to have antitumor effects, however, no systematic study has been conducted to prove the anti-OS effects of TW. This study aimed to investigate the effects of TW on OS and its mechanism based on network pharmacology and molecular docking. The web pharmacology section includes the gathering of the active components of TW, the collection of predicted targets of TW and OS-related targets, the analysis of therapeutic targets of TW, the enrichment of gene ontology (GO), and the enrichment of Kyoto Encyclopedia of Genes and Genomes (KEGG). The Veen diagram showed 451 targets for OS treatment in TW. The therapeutic target enrichment analysis results showed that TW treated OS via multiple targets and pathways. TW can affect OS proliferation, apoptosis, migration, infiltration, and angiogenesis through a signaling network formed by hub genes that cascade through numerous signaling pathways. In addition, molecular docking results showed that triptolide, kaempferol, and 5,8-Dihydroxy-7-(4-hydroxy-5-methyl-coumarin-3)-coumarin have relatively high potential to become drugs for patients with OS and improve the 5-year survival rate of patients with OS. Network pharmacology and molecular docking suggest that TW affects the biological behavior of OS through multiple pathways involving multiple targets, such as proliferation, apoptosis, migration, and infiltration. Upregulation of the cellular tumor antigen p53 (TP53) gene and downregulation of peroxisome proliferator-activated receptor gamma (PPARG) and signal transducer and activator of transcription 1-alpha/beta (STAT1) genes can prolong the survival time of patients with OS. Triptolide, kaempferol, and 5,8-Dihydroxy-7-(4-hydroxy-5 methyl-coumarin-3)-coumarin have a relatively high potential to become a treatment for patients with OS and improve 5-year survival of OS patients.

The effects of clay mineral bentonite on the growth process of submerged macrophyte V. spiralis and sediment microenvironment were investigated in the study for the first time, aiming to determine whether it is suitable for application in the field of ecological restoration. The growth index, and physiological and biochemical index of V. spiralis in the experiments were measured once a month, and the changes of rhizosphere microorganisms and physicochemical properties of sediments were also studied at the same time. The results demonstrated that bentonite can effectively promote the growth of V. spiralis. The treatment groups of RB1/1 and MB1/5 (the mass ratios of bentonite to sediment were 1/1 and 1/5, respectively.) showed the best V. spiralis growth promotion rates which were 18.78%, and 11.79%, respectively. The highest microbial diversity and abundance existed in group of RB10 (the mass ratio of sediment to bentonite was 10/1), in which the OTUs, Shannon, Chao and Ace were 1521.0, 5.20, 1712.26, and 1686.31, respectively. Bentonite was conducive to the propagation of rhizosphere microorganisms, and further changed the physical and chemical properties of the sediment microenvironment. The nutrient elements dissolved from bentonite may be one of the main reasons that promoted the growth of V. spiralis. The purpose of this result is to prove that bentonite can be further applied as sediment improver and growing media in ecological restoration projects in eutrophic shallow lakes.

Sediment phosphorus (P) is the main source of endogenous P for lake eutrophication. An in-situ combined technology for determination the removal effect of sediment P in all fractions was first developed using the novel modified maifanite (MMF) and submerged macrophytes in this study. MMF was synthesized using an acidification process (2.5 mol/L H2SO4) and then a calcination (400 °C) method. The morphology and structure of MMF were characterized by XRD, SEM, XPS, and BET. We tested the removal effects of sediment P by MMF and submerged macrophytes in combination and separately. The results demonstrated that the synergistic removal capacity of sediment P using MMF coupled with submerged macrophytes was higher than the sum of them applied separately. MMF could promote the submerged macrophytes growth and enhance the adsorption of extra P on MMF through root oxygenation and nutrient allocation. The microcosm experiment results showed that sediment from fMMF+V. spiralis exhibited the most microbial diversity and abundance among the sediment. The combination of MMF and submerged macrophytes increased the Firmicutes abundance and decreased the Bacteroidetes. These results indicated that adsorption-biological technology can be regarded as a novel and competitive technology to the endogenous pollution control in eutrophic shallow lakes.

In this study, two types of artificial floating islands (AFIs), group A (consists of 1# and 2# traditional AFIs with plant and soil) and group B (consists of 4# and 5# new-type AFIs with plant, substrate, and with luffa sponge and corncob hanging at the bottom), were constructed, respectively. The removal effects and degradation mechanisms of luffa sponge and corncob in group B were compared and investigated. Plant height, root growth, and packing degradation of the two types of AFIs were studied. Temperature, dissolved oxygen (DO), and pH on the decontamination effects of AFI were discussed. The results showed that group A and group B AFIs showed great significant differences in removal of CODCr, TN, NO3--N, NH4+-N, and TP (p < 0.05). The TP removal of group B was 92.8 ± 0.6%, and the TN removal and NO3--N removal were significantly higher than that of group A, which was 90.3 ± 0.8% and 96.0 ± 2.2%, respectively; The addition of luffa sponge and corncob could enhance the biodegradability of sewage and the nitrogen and phosphorus removal efficiency of group B. The plant growth height of group B planted with Lythrum salicaria was 2.36 times higher than that of group A. The effect of temperature on TP was significantly greater than that of TN, and both groups of AFIs presented continuous improvement capacities of TN and TP removal when the temperature was above 15 °C. Group B was observed with a lower pH range of 6.69~7.12, which was more suitable for denitrification than group A. The release of carbon source of 5#-corncob AFI was 2.51 times higher than 4#-luffa sponge AFI at the end of the experiment.

Floating treatment wetlands (FTWs) and biofilm carriers are widely used in water purification. The objective of the present work was to explore whether and to what extent an FTW integrated with plants and biofilm carriers (FTW-I) could enhance the nutrient removal efficiency. Significantly higher removal rates of ammonia nitrogen (85.2 %), total phosphorus (82.7 %), and orthophosphate (82.5 %) were observed in the FTW-I treatment relative to the FTW with plants (FTW-P; 80.0, 78.5, and 77.6 %, respectively) and the FTW with biofilm carriers (FTW-B; 56.7, 12.9, and 13.4 %, respectively) (p < 0.05). The mass balance results indicated that plant uptake was the main pathway for N and P removal (accounting for 58.1 and 91.4 %, respectively) in FTW-I, in which only 1.2 % of the N and 5.7 % of the P was deposited on the bottom of the tank. In addition, the plants translocated 43.9 and 80.2 % of the N and P in the water and 83.5 and 88.3 % of the absorbed N and P, respectively, into their aboveground tissues. The combination of an FTW and biofilm carriers can improve the efficiency of water purification, and nutrients can be rapidly removed from the system by harvesting the aboveground plant tissues.
