RESUMO

Antibiotic resistance is a major issue in animal industries and antibiotic-free alternatives are needed to treat infectious diseases and improve performance of pigs. Plant extracts have been suggested as a potential solution. The present study was conducted to investigate the effects of Astragalus polysaccharides (Aps) and ginseng polysaccharide (Gps) on growth performance, intestinal morphology, immune function, volatile fatty acids (VFAs), and microfloral community in weaned piglets. A total of 180 weaned piglets were randomly divided into three treatment groups during a 28-days feeding experiment, including a basal diet (Con), basal diet supplemented with 800 mg/kg Aps (Aps), and basal diet supplemented with 800 mg/kg Gps (Gps). Results showed that both Aps and Gps increased body weight, average daily gain and feed conversion rate, and reduced the rate of diarrhea. Gps also decreased aspartate aminotransferase compared to the Con piglets after 14 days. No significant effects on alanine aminotransferase were observed. Both Aps and Gps piglets exhibited higher serum immunoglobulin M levels after 14 and 28 days, and also decreased jejunal crypt depth, increased jejunal villus length and villus height/crypt depth ratio, and increased expression of toll-like receptor 4, myeloid differentiation primary response 88, nuclear factor-kappa B proteins in the jejunum. Aps and Gps piglets also had higher concentrations of acetic acid, isobutyric acid, and butyrate in their colon. Data of high-throughput sequencing revealed that Aps and Gps affected bacterial quantity and diversity in the colon. Species richness and evenness were higher in both Aps and Gps piglets than the control piglets. Aps and Gps piglets also had a higher relative abundance of Lachnospiraceae and Anaerostipes, and the Aps piglets had a higher relative abundance of Lactobacillus gasseri and L. amylovorus. Therefore, dietary supplementation with Aps and Gps could be beneficial for optimizing the performance of industry pigs and reducing dependence on antibiotics. Furthermore, Plant polysaccharides play a great role in promoting the sustainable development of animal husbandry.

In order to investigate heterosis on a molecular basis, suppression subtractive hybridization was used to analyze the differences in gene expression between porcine F1 hybrids Landrace x Yorkshire and their female parents Yorkshire. From two specific subtractive cDNA libraries, the clones screened out by reverse Northern high-density blots screening were chosen to clone full-length cDNA by RACE. An expression-upregulated gene for Yorkshire skeletal muscle, designated as HUMMLC2B, was identified. Porcine HUMMLC2B contains an open reading frame (ORF) encoding 169 amino acids residues with 59 and 115 nucleotides in the 5' and 3' untranslated regions (UTRs), respectively. In the porcine genome, it contains seven exons separated by six introns. High allelic variations and four SINEs were detected in it. Comparison of derived amino acid sequence of HUMMLC2B with database sequences revealed highly conserved 12 amino acid residues in a putative calcium-binding region. RT-PCR analysis showed a tissue-specific pattern of expression in skeletal muscle and a similar level of expression during skeletal muscle development. The possible role of HUMMLC2B and its relation to porcine heterosis are discussed.

Bacterial pathogens are often involved in contact lens-related adverse responses. This study aimed to find antimicrobial peptides and proteins that effectively eradicate or inhibit ocular bacteria. The antimicrobials were screened against gram-negative and gram-positive bacteria originating from ocular sources. The viability of these ocular bacteria was measured after exposure to the peptides and proteins. Two conditions were used to grow bacteria, low nutrient phosphate-buffered saline and high nutrient tryptone soya broth. Samples were taken at different times up to 48 h. In low nutrient conditions, protamine was found to be the most effective against all strains. Melittin was very effectve against all strains except Serratia and one Pseudomonas isolate which were partially affected. In high nutrient condition, only melittin was effective in killing Staphylococcus aureus. Protamine and the combination of protamine and melittin had the greatest effect in eradicating the bacteria tested in low nutrient condition. Protamine alone and its combination with melittin may have potential therapeutic agents for ocular infections in an era of emerging antibiotic resistance.

OBJECTIVE: To assess the effect of Xinmaitong (XMT) capsule in treating ischemical cardiac disease. METHODS: Sixty coronary heart disease (CHD) patients with myocardial ischemia were divided randomly into two groups. XMT group (30 cases) was treated with XMT plus western medicine, and control group (30 cases) with western medicine alone. The changes of the scores for clinical symptoms, the total ischemia burden (TIB), the plasma endothelin (ET), the nitric oxide (NO), the superoxide dismutase (SOD), and the malonyldialdeyde (MDA) levels were observed before and after treatment. RESULTS: After treatment with XMT the scores of clinical symptoms, TIB, ET and MDA levels were significantly decreased (P < 0.01), the levels of NO and SOD were significantly increased (P < 0.01) in the XMT group, comparing with the control group these changes were statistically different (P < 0.01). CONCLUSIONS: XMT capsule can act against myocardial ischemia effectively, one of the mechanisms of which is protecting the function of vascular endothelium and resisting lipid peroxidation injury. The effects of adding XMT capsule on conventional treatment of western medicine were better than those of using western medicine alone.

BACKGROUND/AIMS: In humans, chronic iron excess may induce hepatic fibrosis and/or hepatocellular carcinoma. This work was undertaken to investigate hepatic iron overload outcome in iron-overloaded mice. METHODS: BALB/cJ male mice received supplements of 0, 0.5, 1.5 and 3% carbonyl-iron for 2, 4, 8 and 12 months. Histological staining, immunohistochemistry using ferritin antibodies and electron microscopic studies were performed on liver. Liver iron concentration was measured biochemically. Mitotic index and hepatocyte nuclear size were evaluated on Feulgen-stained slides. RESULTS: Liver iron concentration was increased, reaching 13 times control value after 12 months in 3% iron-overloaded mice, and iron was found predominantly in hepatocytes, with a porto-centrolobular decreasing gradient. Neither hepatic fibrosis nor hepatocellular carcinoma was found. Perls' stain positive inclusions containing ferritin were found within hepatocyte nuclei in 3%-overloaded mice. Electron microscopy disclosed that inclusions consisted of ferritin particle aggregates without a limiting membrane. Mice overloaded with 3% iron for 12 months showed larger hepatocyte nuclei than control mice and a mitotic index increase with presence of abnormal tripolar mitotic figures. In addition, some iron-free hepatocytes were observed. CONCLUSIONS: Carbonyl-iron supplementation produces significant iron overload in mice but does not result in liver fibrosis or hepatocellular carcinoma after 12 months. However, nuclear changes were produced in hepatocytes, and occasional iron-free hepatocytes were observed: these may represent preneoplastic changes caused by iron overload.

RESUMO

This paper summarizes the resources of anticarcinogenic medical fungi, and the information of distribution, eco-environment. It outlines the present statuts of the exploitation and utilization of some species of common anticarcinogenic medical fungi such as Ganoderma lucidum, Coriolus versicolor, Polyporus frondosus etc. The anticarcinogenic mechanism of medical fungi is discussed. Suggestions on how to development and utilize rationally these resources are offered as well.

The amino acid glutamate is one of the major neurotoxins in the pathogenesis of neuronal death after ischemia or trauma. Microdialysis studies in both man and animal have shown elevated extracellular levels after primary lesions. Monitoring of cerebral tissue oxygenation (p(ti)O2) has been used in recent years to detect and prevent episodes of low cerebral oxygenation, e.g. after trauma or subarachnoid hemorrhage. Intraoperative monitoring of p(ti)O2 combined with microdialysis in the peritumoral edema has been chosen to study the responses of glutamate and oxygen levels during resection. In 7/9 patients p(ti)O2 was below "critical" 10 mm Hg. Elevating inspiratory oxygen concentration to 100% led to an increase of p(ti)O2 by 2.5-4 fold and a decrease of glutamate and aspartate by 50-80%. A close correlation between p(ti)O2 and microdialysis glutamate levels was not clearly shown due to frequent intraoperative manipulations.

RESUMO

Studies on the chemical constituents of Phyllanthus urinaria and its antiviral activity against hepatitis B virus were completed. Eleven compounds have been isolated. Two of them are new compounds methyl ester dehydrochebulic acid and methyl brevifolin carboxylate. Antiviral experiments on HBsAg in vitro and liver damage caused by CCl4 have shown that. Phyllanthus urinaria possesses antiviral activities against HBV.

RESUMO

The frontal cortex is recognized as the highest adaptive control center of the human brain. The principle of the "frontalization" of human brain function offers new possibilities for brain research in space. There is evolutionary and experimental evidence indicating the validity of the principle, including it's role in nervous response to gravitational stimulation. The gravitational field is considered here as one of the more constant and comprehensive factors acting on brain evolution, which has undergone some successive crucial steps: "encephalization", "corticalization", "lateralization" and "frontalization". The dominating effects of electrical responses from the frontal cortex have been discovered 1) in experiments under gravitational stimulus; and 2) in processes potentially relating to gravitational adaptation, such as memory and learning, sensory information processing, motor programing, and brain state control. A brain research experiment during space flight is suggested to test the role of the frontal cortex in space adaptation and it's potentiality in brain control.
