Coking industry has been considered as important source of volatile organic compounds (VOCs) emissions. However, few studies have emphasized the occurrence and adverse effects of VOCs from coking wastewater treatment processes. In this research, pollution profiles of both air and water phase VOCs in a typical coking wastewater treatment plant were investigated in terms of distribution characteristics, air-water exchange, ozone formation potential (OFP) and associated human health risks. Thirty VOCs were detected in the air phase, in which benzene and naphthalene were found to be the major VOCs with total contribution of 87.81 %. Nineteen VOCs were detected in the water phase, in which benzene, naphthalene and toluene contribute most to total VOCs with total contribution of 75.1 %. The regulating tank (RT) was the major source of VOCs, and the emission rate of total VOCs from all unites was 2711.03 g/d with annual emission of 0.99 t. The emission factor was estimated to be 1.36 g VOCs/m3 wastewater. The air-water exchange was assessed using the Fugacity model, and water-to-air volatilization was predominant based on the net flux of air-water exchange. OFP evaluated by emission factor indicated that the total OFP in RT was the highest (1.52 g O3/m3 wastewater), and toluene contributed 41.8 % of the total OFP, followed by naphthalene accounting for 38.7 % The total carcinogenic risks were in the range of 8.60 × 10-6 to 2.18 × 10-3, in which the RT exceeded the significant risk threshold (>1 × 10-4). The non-carcinogenic risks of hazard quotient value in RT also exceeded the risk threshold (>1), and naphthalene was the major contributor accounting for 79.02 %. These results not only provided comprehensive knowledge on pollution profiles and environmental risks of VOCs during coking wastewater treatment processes, but also facilitated the implement of VOCs regulation and occupational health protection strategies in coking industries.

Natural microalgae (NA, cyanobacteria) collected from Taihu Lake (Jiangsu, China) were used for biofuel production through pyrolysis. The microalgae were de-ashed via pretreatment with deionized water and hydrochloric acid, and the samples obtained were noted as 0 M, 0.1 M, 1 M, 2 M, 4 M, 6 M, 8 M, respectively, according to the concentration of hydrochloric acid used in the pretreatment. Pyrolysis experiments were carried out at 500 °C for 2 h. The products were examined by various techniques to identify the influence of the ash on the pyrolysis behavior. The results showed that the ash inhibited the thermal transformation of microalgae. The 2 mol/L hydrochloric acid performed the best in removing ash and the liquid yield increased from 34.4% (NA) to 40.5% (2 M). Metal-oxides (mainly CaO, MgO, Al2O3) in ash promoted the reaction of hexadecanoic acid and NH3 to produce more hexadecanamide, which was further dehydrated to hexadecanenitrile. After acid pretreatment, significant improvement in the selectivity of hexadecanoic acid was observed, ranging from 22.4% (NA) to 58.8% (4 M). The hydrocarbon compounds in the liquid product increased from 12.90% (NA) to 26.67% (2 M). Furthermore, the acid pretreatment enhanced the content of C9-C16 compounds and the HHV values of bio-oil. For natural microalgae, the de-ashing pretreatment before pyrolysis was essential for improving the biocrude yield and quality, as well as the biomass conversion efficiency.

Longchun 30, a new wheat variety, was used to investigate seedling growth, element absorption, and antioxidant response under 150 mM NaCl and tea polyphenols (TP) (25 and 100 mg L-1) treatments alone or in combination, thus revealing TP-alleviating mechanism on the salt damage to plants. 150 mM NaCl stress alone inhibited the seedling growth, increased sodium content and reactive oxygen species (ROS) accumulation, but reduced potassium (K) and calcium (Ca) levels at different culture times, thus resulting in the oxidative damage to the leaves. Even though 25 or 100 mg L-1 TP treatment alone led to the significant increases of O2·- and H2O2 generation, TP-treated leaves exhibited the reduction of relative electrical conductivity and no change of malondialdehyde content. Moreover, high TP concentration alone stimulated the seedling growth. In addition, the activities and gene expression of superoxide dismutase, catalase, and peroxidase (POD) as well as diamine oxidase and polyamine oxidase were changed to different degrees due to NaCl or TP treatment alone. Further study showed that the presence of 25 or 100 mg L-1 TP promoted the growth, increased K+ and Ca2+ contents, and reduced O2·- and H2O2 accumulation in salt-stressed wheat seedlings. Taken together, salinity-inhibitory effect on the growth of wheat seedlings might be associated with salt-induced imbalance of element content and the increase of oxidative damage resulting from ROS accumulation, while the application of TP effectively alleviated salinity-inhibitory effect on the seedling growth and improved the tolerance of wheat seedlings to salt environment, which might be associated with the increases of K+ and Ca2+ contents as well as the reduction of oxidative damage in the leaves of wheat seedlings under NaCl and TP treatment in combination.

PURPOSE: Pancreatic neuroendocrine tumors (pNETs) are uncommon malignancies noted for their propensity to metastasize and comparatively favorable prognosis. Although both the treatment options and clinical outcomes have improved in the past decades, most patients will die of metastatic disease. New systemic therapies are needed. EXPERIMENTAL DESIGN: Tissues were obtained from 43 patients with well-differentiated pNETs undergoing surgery. Gene expression was compared between primary tumors versus liver and lymph node metastases using RNA-Seq. Genes that were selectively elevated at only one metastatic site were filtered out to reduce tissue-specific effects. Ingenuity pathway analysis (IPA) and the Connectivity Map (CMap) identified drugs likely to antagonize metastasis-specific targets. The biological activity of top identified agents was tested in vitro using two pNET cell lines (BON-1 and QGP-1). RESULTS: A total of 902 genes were differentially expressed in pNET metastases compared with primary tumors, 626 of which remained in the common metastatic profile after filtering. Analysis with IPA and CMap revealed altered activity of factors involved in survival and proliferation, and identified drugs targeting those pathways, including inhibitors of mTOR, PI3K, MEK, TOP2A, protein kinase C, NF-kB, cyclin-dependent kinase, and histone deacetylase. Inhibitors of MEK and TOP2A were consistently the most active compounds. CONCLUSIONS: We employed a complementary bioinformatics approach to identify novel therapeutics for pNETs by analyzing gene expression in metastatic tumors. The potential utility of these drugs was confirmed by in vitro cytotoxicity assays, suggesting drugs targeting MEK and TOP2A may be highly efficacious against metastatic pNETs. This is a promising strategy for discovering more effective treatments for patients with pNETs.

Chromium (Cr) contamination of soil and water has become a severe threat to human health. In this study, a series of experiments were conducted to examine the ameliorative effects of Cr toxicity, by exogenous 100 µM sodium sulfate. Our team has examined the plant growth, Cr content, chlorophyll, antioxidant index and soluble protein content, before and after the addition of sodium sulfate. The results showed that the addition of sulfur (S) can reduce the enrichment of Cr and the content of malonyldialdehyde (MDA) under Cr stress. After addition of S in the culture solution, the biomass and roots length of Arabidopsis thaliana increased under Cr stress. Furthermore, the content of chlorophyll, superoxide dismutase (SOD), peroxidase (POD), catalase (CAT), glutathione (GSH), and soluble protein increased with the addition of sulfur. Transmission electron microscope observation point to that the chloroplasts can be damaged in leaf. All data demonstrate that S supplementation should help to alleviate the negative effects caused by both Cr(III) and Cr(VI) on Arabidopsis thaliana.

ETHNOPHARMACOLOGICAL RELEVANCE: Enterovirus 71 (EV71) has a propensity to cause hand-foot-and-mouth disease (HFMD) epidemics associated with neurological sequelae. Unfortunately, no drugs are currently available for the clinical treatment of EV71 infections. Sophoridine (SRI) is one of the most abundant alkaloids in Sophora flavescens Aiton (Leguminosae), which has been used to treat fever, throat inflammation, cancer, and other diseases. MATERIALS AND METHODS: In this study, we found that SRI inhibits EV71 infection in Vero cells. To study the antiviral activity of SRI, Vero cells were divided into 3 treatment groups based on the timing of SRI dosing: prior to viral adsorption (Group A), during viral adsorption (Group B), and after viral adsorption (Group C). We further revealed the antiviral activity of SRI with the attachment assay and the penetration assay. For Group A, 50% viability of Vero cells was observed at a SRI concentration of 61.39 µg/mL, whereas for Groups B, 50% viability was observed at SRI concentrations of 196.86 µg/mL. Furthermore, 29.7% cell viability was observed even at a SRI concentration of 1000 µg/mL in Groups C. The results show that SRI was highly effective against EV71 when Vero cells were pretreated with SRI for 2 h (Group A). Further researches indicate SRI was highly effective at inhibiting EV71 attachment when the SRI concentrations over 250 µg/mL (P < 0.001). CONCLUSIONS: We have shown that Vero cell viability increases when SRI is administered prior to viral adsorption. This suggests that SRI has the considerable potential as an antiviral for EV71 disease prevention.

Xijiang River is an important drinking water source in Guangxi Province, China. Along the Xijiang River and surrounding tributary, the pollution profile of three important groups of semi-volatile organic compounds, including polycyclic aromatic hydrocarbons (PAHs), organochlorine pesticides (OCPs) and phthalate esters (PAEs), was analyzed. Relatively low levels of PAHs (64-3.7 × 102 ng L-1) and OCPs (16-70 ng L-1), but high levels of PAEs (7.9 × 102-6.8 × 103 ng L-1) occurred in the water. Comparatively, low levels of OCPs (39-1.8 × 102 ng g-1) and PAEs (21-81 ng g-1), but high levels of PAHs (41-1.1 × 103 ng g-1) were found in sediment. Principal component analyses for source identification indicated petroleum-derived residues or coal and biomass combustion, and vehicular emission was the main sources for PAHs. The OCPs sources of each category were almost independent, whereas the new input of HCHs and p,p'-DDTs probably existed in some areas. PAEs were mainly originated from personal care products of urban sewage, plastic and other industrial sources. Ecological risk through the risk quotient analysis indicated a small or significant potential adverse effect on fish, daphnia and green algae. Nevertheless, the integrated risk of all pollutants should be taken into account in future study.

Previous studies have shown that Lycium barbarum polysaccharides (LBPs) serve an important role in antioxidant activity to protect the cells and tissues. However, the specific mechanism of LBPs in the prevention of endometrial damage remains to be elucidated. Using morphological observation, cell proliferation assay, the detection of superoxide dismutase (SOD) activity and the content of malondialdehyde (MDA) in cell culture supernatant fluid, the detection by western blot analysis and reverse transcription-quantitative polymerase chain reaction of the mRNA and protein expression levels of caspase­3 and Bcl­2 in endometrial stromal cells (ESCs), it was demonstrated that, in vitro, hydrogen peroxide (H2O2)-induced death of ESCs, increased the content of MDA and decreased the activity of SOD, and decreased the expression of Bcl-2 and increased the expression of caspase­3. LBPs can inhibit H2O2­induced cell death of ESCs, decrease the content of MDA in ESCs and increase the activity of SOD, as well as increasing the expression of Bcl­2 and decreasing the expression levels of caspase­3. These findings suggested that LBPs can inhibit H2O2­induced apoptosis of EECs and that LBPs are able to offer a significant protection against oxidative stress to ESCs.

The sustainable intensification of African agriculture is gaining momentum with the compelling need to increase food and agricultural production. In Southern Africa, smallholder farming systems are predominately maize-based and subject to erratic climatic conditions. Farmer crop and soil management decisions are influenced by a plethora of complex factors such as market access resource availability, social relations, environment, and various messages on sustainable farming practices. Such factors pose barriers to increasing sustainable intensification in Africa. This paper characterizes smallholder farming practices in Central Malawi, at Africa Research in Sustainable Intensification for the Next Generation (Africa RISING) project sites. We present findings from a survey of 324 farmers, located within four Africa RISING sites selected in a stratified random manner to represent (1) low agricultural potential (high evapotranspiration, variable rainfall), (2) medium agricultural potential (two sites), and (3) high agricultural potential (well-distributed rainfall). Soil fertility was low overall, and certain farming practices appeared to limit the sustainability of agricultural production. Nearly half of farmers did not value legume residues as a high nutrient value resource for soil amelioration, as legume residues were removed (17.9%) or burned (21.4%). Conversely, maize residues were rarely removed (4.5%) or burned (10.4%). We found that farmers do not allocate soil amendment resources to legume fields (zero instances of mineral fertilizer or manure application to legumes compared to 88 and 22% of maize systems, respectively). Policy makers in Malawi have led initiatives to intensify agricultural systems through subsidizing farmer access to mineral fertilizer as well as maize hybrid seed, and only rarely to improved legume seed. In this survey, farmers allocate mineral fertilizer to maize systems and not legume systems. There is urgent need to invest in education on sustainable reinvestment in natural resources through complementary practices, such as maximization of biological nitrogen fixation through improved legume agronomy and better organic resource and crop residue management. Recent efforts by Malawi agricultural services to promote doubled-up legumes as a sustainable intensification technology are encouraging, but benefits will not accrue unless equal attention is given to an extension campaign on management of organic resources such as crop residues.

The pollution levels of typical semivolatile organic compounds (SVOCs) consisting of 15 polycyclic aromatic hydrocarbons (PAHs), 20 organic chlorinated pesticides (OCPs), and 15 phthalate esters (PAEs) were investigated in small rivers running through the flourishing cities in Pearl River Delta region, China. The concentrations of ∑15PAHs were 2.0-48 ng/L and 29-1.2 × 10(3) ng/g in the water and sediment samples, respectively. The ∑20OCPs were 6.6-57 ng/L and 9.3-6.0 × 10(2) ng/g in the water and sediment samples, respectively. The concentrations of ∑15PAEs were much higher both in the water and sediments. The partition process of the detected SVOCs between the water and sediment did not reach the equilibrium state at most of the sites when sampling. The combustion of petroleum products and coal was the major source of the detected PAHs. The OCPs were mainly historical residue, whereas the new inputs of dichlorodiphenyltrichloroethane (DDT), chlordane, and endosulfan were possible at several sites. The industrial and domestic sewage were the major source for the PAEs; storm water runoff accelerated the input of PAEs. No chronic risk of the SVOCs was identified by a health risk assessment through daily water consumption, except for the ∑20OCPs that might cause cancer at several sites. Nevertheless, the integrated health risk of the SVOCs should not be neglected and need intensive investigations.

OBJECTIVE: To observe the clinical efficacy of treating middle-aged and aged patients with knee osteoarthritis (KOA) of yang-deficiency induced cold-damp syndrome (YDICDS) by ozone combined Fugui Gutong Granule (FGG). METHODS: Using a prospective, randomized controlled clinical trial, 200 KOA patients of YDICDS were randomly assigned to four groups. i.e., the control group (Group A), the Chinese medicine treatment group (Group B), the ozone group (Group C), and the Chinese-r medicine treatment plus ozone group (Group D).Patients in Group A took Voltaren Tablet. Those in Group B took FGG. Those in Group C received ozone injection (10 -18 mL) from knee joint cavity at 25 mg/L, once weekly for 4 weeks in total. Those in Group D received injection from knee joint cavity and took FGG. The therapeutic course for all was one month. The efficacy was assessed using visual analogue scale (VAS) and Western Ontario MacMaster University Osteoarthritis index (WOMAC). RESULTS: The VAS score was obviously lower in Group D than in Group B and Group A at 24 h and 1 week (P <0. 05). After one month of treatment, the VAS score was obviously lower in Group D than in Group A, B, and C (P < 0.05). After treatment the total integral of WOMAC was 25.34 +/- 2.12 in Group D, obviously lower than that in Group A (44.72 +/- 6.57), Group B (40.58 +/- 5.98), and Group C (38.53 +/- 5. 13), showing statistical difference (P <0.05). The pain score, the joint stiffness score, the score for daily activities were lower in Group D than in Group A (P <0.05). The cured and markedly effective rate was 76.0% in Group D, higher than that of Group A (25. 0%), Group B (25. 0%), and Group C (43.8%), respectively (P < 0.05). CONCLUSION: Ozone combined FGG had advantages in alleviating joint pain, and improving joint stiffness and daily activities of middle-aged and aged patients with KOA of YDICDS.

OBJECTIVE: To explore the expressions of B lymphocyte activating factor (BAFF) in the serum and peripheral blood B cells (PBBCs) of BXSB lupus nephritis mice, and to investigate the efficacy of Langchuangping Granule (LG). METHODS: Eighteen 11-week-old male BXSB lupus mice were randomly divided into three groups, i.e., the lupus control group, the hormone treatment group, and the LG treatment group, 6 in each group. Besides, another 6 C57BL/6 male mice were recruited as the normal control group. The mice were given with normal sodium (10 mL/d), methylprednisolone (at the daily dose of 5 mg/kg), LG (at the daily dose of 4 g/kg), and the normal saline (10 mL/d) respectively by gastrogavage for 4 weeks. The urine protein, ds-DNA, and body weight were determined. The serum soluble BAFF (sBAFF), the expressions and changes of BAFF-mRNA in the PBBCs were detected using ELISA and RT-PCR respectively. The activity index (AI) and 24 h urine albumin excretion quantitation of renal pathological activities were observed. The correlation between ds-DNA and sBAFF were analyzed. RESULTS: The level of sBAFF in serum, the BAFF mRNA level in PBBCs, 24 h urinary albumin excretion, and serum ds-DNA content increased more obviously in lupus mice than in the normal mice. After being treated by methylprednisolone or LG, the sBAFF and BAFF mRNA expressions decreased more obviously than before treatment, showing statistical difference (P<0.05). But there was no statistical difference in the sBAFF level or the BAFF mRNA expression (P>0.05). There was positive correlation between sBAFF and AI (r=0.8098, P<0.01), 24 h urinary albumin excretion (r=0.8220, P<0.01), and ds-DNA (r=0.8535, P<0.01). CONCLUSIONS: BAFF plays an important role in the occurrence and development of lupus nephritis. It can be used in monitoring the disease progress and predicting its recurrence. It is one of ideal targets for treating lupus nephritis. LG could attenuate the renal injury via suppressing BAFF level. It is worth further clinical application.

OBJECTIVE: To study whether Langchuangping granule (LG) could exert its renal protection by down-regulating monocyte chemoattractant protein-1 (MCP-1) via suppressing nuclear factor kappa B (NF-kappaB) signaling pathway in BXSB lupus nephritis (LN) mice. Methods Eighteen male 11-week-old BXSB LN mice were randomly divided into three groups, i.e., the model group, the hormone group, and the Chinese medicine group, 6 in each. They were administered by gastrogavage with normal saline, methylprednisolone, and LG, respectively. Another six C57BL/6 male mice of the same age was taken as the normal control group, which was administered with normal saline by gastrogavage. All mice were treated once daily, for 4 successive weeks. The 24-h urine protein was determined. The mRNA and protein expressions of MCP-1 in the renal tissue were detected using RT-PCR and Western blot. The expression of NF-kappaB p65 in the renal tissue was detected using immunohistochemical assay. Activity index (AI) of the renal tissue was counted using PAS stain. The content of ds-DNA antibody was detected using ELISA. The correlations of the aforesaid indices were analyzed. RESULTS: The 24-h urine protein level, serum ds-DNA antibody content, protein and mRNA expressions of MCP-1, NF-kappaB p65 expression level, and AI count were obviously higher in the model group than in the normal control group (P < 0.01). The aforesaid indices all obviously decreased after medication in the Chinese medicine group and the hormone group (P < 0.05). MCP-1 protein expression level was positively correlated with MCP-1 mRNA, NF-kappaB p65, AI, 24-h urine protein, and ds-DNA antibody of all LN mice (r= 0.984, 0.936, 0.887, 0.698, 0.679, all P < 0.01). CONCLUSIONS: LG possibly played renal protection by down-regulating NF-kappaB-mediated MCP-1 expression levels. MCP-1 played important roles in the occurrence and development of LN, being one of ideal targets for LN treatment.

The feasibility study of the application of the photoelectrocatalytic decontamination of high saline produced water containing refractory organic pollutants was investigated in the slurry photoelectrocatalytic reactor with nanometer TiO2 particle prepared with sol-gel method using the acetic acid as hydrolytic catalyst. The efficiency of the photoelectrocatalytic decontamination of produced water was determined with both COD removal from the tested wastewater and the decrease of mutagenic activity evaluated by Ames tests. The experimental results showed that the photoelectrocatalysis is a quite efficient process for decontaminating the produced water, although there are high concentration of salt existed in oilfield wastewater. We found that the COD removal efficiencies by photoelectrocatalytic process are much higher than that of by photocatalytic or electrochemical oxidation individually in the photoelectrocatalytic reactor. The COD removal can be substantially improved by the added H2O2 and the generation of active chlorine from high concentration chlorides in the wastewater. The effects of various operating conditions, such as initial COD concentration, applied cell voltage, catalyst amount and initial pH value of solution, on the photoelectrocatalytic efficiencies, is also investigated in detail. The results showed that when the raw produced wastewater was diluted in a 1:1 (v/v) ratio, there is a highest COD removal efficiency. And the photoelectrocatalytic degradation of organic pollutants in saline water is much favored in acidic solution than that in neutral and/or alkaline solution.

Molecular genetic analysis and insect bioassay of transgenic indica rice 'Zhuxian B' plants carrying snowdrop lectin gene (gna) and soybean trypsin inhibitor gene (sbti) were investigated in detail. PCR, 'dot' blot and PCR-Southern blot analysis showed that both transgenes had been incorporated into the rice genome and transmitted up to R3 progeny in most lines tested. Some transgenic lines exhibited Mendelian segregation, but the other showed either 1:1 (positive: negative for the transgenes) or other aberrant segregation patterns. The segregation patterns of gna gene crossed between R2 and R3 progeny. In half of transgenic R3 lines, gna and sbti transgenes co-segregated. Two independent homozygous lines expressing double transgenes were identified in R3 progeny. Southern blot analysis demonstrated that the copy numbers of integrated gna and sbti transgenes varied from one to ten in different lines. Insect bioassay data showed that most transgenic plants had better resistance to both Nilaparvata lugens (Stahl) and Cnaphalocrocis medinalis (Guenee) than wild-type plants. The insect resistance of transgenic lines increased with the increase in transgene positive ratio in most of the transgenic lines. In all, we obtained nine lines of R3 transgenic plants, including one pure line, which had better resistance to both N lugens and C medinalis than wild-type plants.
