ETHNOPHARMACOLOGICAL RELEVANCE: PolyphyllinVI (PPⅥ) is the main bioactive component of Chonglou which is a traditional Chinese herbal with various effects, including antitumor, anti-inflammatory, and analgesia. AIM OF THE STUDY: This study aimed to investigate the properties and mechanisms of the analgesia of PPⅥ by using neuropathic pain (NPP) mice. MATERIALS AND METHODS: The potential targets and mechanisms of PPⅥ in alleviating NPP were excavated based on the network pharmacology. Subsequently, the construction of a spared nerve injury (SNI) mice model was used to evaluate the effect of PPⅥ on NPP and the expression of the P2X3 receptor. We identified the signaling pathways of PPⅥ analgesia by RNA sequencing. RESULTS: The results of network pharmacology showed that BCL2, CASP3, JUN, STAT3, and TNF were the key targets of the analgesic effect of PPⅥ. PPⅥ increased the MWT and TWL of SNI mice and decreased the level of P2X3 receptors in the dorsal root ganglion (DRG) and spinal cord (SC). Additionally, PPⅥ reduced the release of pro-inflammatory mediators (TNF-α, IL-1ß, and IL-6) in the DRG, SC, and serum. Based on the KEGG enrichment of differentially expressed genes (DEGs) identified by RNA-Seq, PPVI may relieve NPP by regulating the AMPK/NF-κB signaling pathway. Western blotting results showed that the AMPK signaling pathway was activated, followed by inhibition of the NF-κB signaling pathway. CONCLUSION: PPⅥ increased the MWT and TWL of SNI mice maybe by inhibiting the expression of the P2X3 receptor and the release of inflammatory mediators. The properties of the analgesia of PPⅥ may be based on the AMPK/NF-κB pathway.

Objective: Inflammatory pain is one of the most common diseases in daily life and clinic. In this work, we analysed bioactive components of the traditional Chinese medicine Chonglou and studied mechanisms of their analgesic effects. Material and methods: Molecular docking technology and U373 cells overexpressing P2X3 receptors combined with the cell membrane immobilized chromatography were used to screen possible CL bioactive molecules interacting with the P2X3 receptor. Moreover, we investigated the analgesic and anti-inflammatory effects of Polyphyllin VI (PPIV), in mice with chronic neuroinflammatory pain induced by CFA (complete Freund's adjuvant). Results: The results of cell membrane immobilized chromatography and molecular docking showed that PPVI was one of the effective compounds of Chonglou. In mice with CFA-induced chronic neuroinflammatory pain, PPVI decreased the thermal paw withdrawal latency and mechanical paw withdrawal threshold and diminished foot edema. Additionally, in mice with CFA-induced chronic neuroinflammatory pain, PPIV reduced the expression of the pro-inflammatory factors IL-1, IL-6, TNF-α, and downregulated the expression of P2X3 receptors in the dorsal root ganglion and spinal cord. Conclusion: Our work identifies PPVI as a potential analgesic component in the Chonglou extract. We demonstrated that PPVI reduces pain by inhibiting inflammation and normalizing P2X3 receptor expression in the dorsal root ganglion and spinal cord.

It is essential to mitigate gaseous emissions that result from poultry and livestock production to increase industry sustainability. Odorous volatile organic compounds (VOCs), ammonia (NH3), hydrogen sulfide (H2S), and greenhouse gases (GHGs) have detrimental effects on the quality of life in rural communities, the environment, and climate. This study's objective was to evaluate the photocatalytic UV treatment of gaseous emissions of odor, odorous VOCs, NH3, and other gases (GHGs, O3-sometimes considered as by-products of UV treatment) from stored swine manure on a pilot-scale. The manure emissions were treated in fast-moving air using a mobile lab equipped with UV-A and UV-C lights and TiO2-based photocatalyst. Treated gas airflow (0.25-0.76 m3∙s-1) simulates output from a small ventilation fan in a barn. Through controlling the light intensity and airflow, UV dose was tested for techno-economic analyses. The treatment effectiveness depended on the UV dose and wavelength. Under UV-A (367 nm) photocatalysis, the percent reduction of targeted gases was up to (i) 63% of odor, (ii) 51%, 51%, 53%, 67%, and 32% of acetic acid, propanoic acid, butanoic acid, p-cresol, and indole, respectively, (iii) 14% of nitrous oxide (N2O), (iv) 100% of O3, and 26% generation of CO2. Under UV-C (185 + 254 nm) photocatalysis, the percent reductions of target gases were up to (i) 54% and 47% for p-cresol and indole, respectively, (ii) 25% of N2O, (iii) 71% of CH4, and 46% and 139% generation of CO2 and O3, respectively. The results proved that the UV technology was sufficiently effective in treating odorous gases, and the mobile lab was ready for farm-scale trials. The UV technology can be considered for the scaled-up treatment of emissions and air quality improvement inside livestock barns. Results from this study are needed to inform the experimental design for future on-farm research with UV-A and UV-C.

Poultry farmers are producing eggs, meat, and feathers with increased efficiency and lower carbon footprint. Technologies to address concerns about the indoor air quality inside barns and the gaseous emissions from farms to the atmosphere continue to be among industry priorities. We have been developing and scaling up a UV air treatment that has the potential to reduce odor and other gases on the farm scale. In our recent laboratory-scale study, the use of UV-A (a less toxic ultraviolet light, a.k.a. "black light") and a special TiO2-based photocatalyst reduced concentrations of several important air pollutants (NH3, CO2, N2O, O3) without impact on H2S and CH4. Therefore, the objectives of this research were to (1) scale up the UV treatment to pilot scale, (2) evaluate the mitigation of odor and odorous volatile organic compounds (VOCs), and (3) complete preliminary economic analyses. A pilot-scale experiment was conducted under commercial poultry barn conditions to evaluate photocatalyst coatings on surfaces subjected to UV light under field conditions. In this study, the reactor was constructed to support interchangeable wall panels and installed on a poultry farm. The effects of a photocatalyst's presence (photocatalysis and photolysis), UV intensity (LED and fluorescent), and treatment time were studied in the pilot-scale experiments inside a poultry barn. The results of the pilot-scale experiments were consistent with the laboratory-scale one: the percent reduction under photocatalysis was generally higher than photolysis. In addition, the percent reduction of target gases at a high light intensity and long treatment time was higher. The percent reduction of NH3 was 5-9%. There was no impact on H2S, CH4, and CO2 under any experimental conditions. N2O and O3 concentrations were reduced at 6-12% and 87-100% by both photolysis and photocatalysis. In addition, concentrations of several VOCs responsible for livestock odor were reduced from 26 to 62% and increased with treatment time and light intensity. The odor was reduced by 18%. Photolysis treatment reduced concentrations of N2O, VOCs, and O3, only. The initial economic analysis has shown that LEDs are more efficient than fluorescent lights. Further scale-up and research at farm scale are warranted.

The analysis of free fatty acids (FFAs) in edible oils can provide important information for quality control and oil authentication. Herein, we report the comprehensive profiling of FFAs in edible oils via magnetic dispersive extraction combined with comprehensive two-dimensional gas chromatography-mass spectrometry (GC × GC-MS). A magnetic extractant was designed for dispersive extraction of FFAs. The extraction conditions were carefully optimized. To assess the extraction method, we first use the method for analysis of 7 targeted FFAs. The limits of detection range from 5.6 to 25.8 ng g-1, and the recoveries in oil samples are 81%-107%. We then performed comprehensive profiling of untargeted FFAs in oil by combining the extraction method with GC × GC-MS. A total of 64 FFAs were identified positively or putatively. The proposed method can provide FFA fingerprint data to guide the processing, storage and authentication of edible oils.

BACKGROUND: Men who have sex with men (MSM) are disproportionately affected by HIV and other STIs worldwide. Rectal douching, which is commonly used by MSM in preparation for anal sex, may increase the risk of HIV and other STIs by injuring the rectal mucosa. Results from individual studies reporting associations between rectal douching and HIV and other STIs among MSM are inconsistent. We performed a systematic review and meta-analysis to estimate the association between rectal douching and HIV and other STIs among MSM. METHODS: We searched PubMed, Embase, Scopus and Web of Science for studies published from January 1970 to November 2018. Studies that reported ORs and 95% CIs of associations between rectal douching and infection with HIV/STIs, or reported enough data to calculate these estimates, were included. We assessed risk of bias using the Newcastle-Ottawa Scale. ORs were pooled using a random effects model. RESULTS: Twenty-eight eligible studies were identified in our review, of which 24 (20 398 participants) were included in the meta-analysis. Rectal douching was associated with increased odds of infection with HIV (OR 2.80, 95% CI 2.32 to 3.39), and any STI other than HIV (including hepatitis B virus (HBV), hepatitis C virus (HCV), chlamydia, gonorrhoea, syphilis and human papillomavirus) (OR 2.46, 95% CI 1.95 to 3.11) among MSM. For specific STIs, douching was associated with increased odds of viral hepatitis (HBV, HCV) (OR 3.29, 95% CI 2.79 to 3.87), and chlamydia or gonorrhoea (OR 3.25, 95% CI 2.02 to 5.23). These associations remained significant in studies that adjusted for potential confounders. CONCLUSION: Rectal douching may put MSM at increased risk for infection with HIV and other STIs. Longitudinal studies are needed to clarify this association, and health education materials should inform men of the potential for increased risk of infection with rectal douching.

Atherosclerosis is a major cause of cardiovascular disease caused by high cholesterol. Reduced intestinal cholesterol absorption has been shown to exert strong cholesterol-lowering and antiatherogenic effects. Previously, we reported that curcumin reduced cholesterol absorption in high-fat diet-fed hamster by downregulating the intestinal expression of Niemann-Pick C1-like 1. Here, we tested the hypothesis that supplementation with curcumin can also reduce intestinal cholesterol absorption in high-fat diet-fed apolipoprotein E knockout (ApoE-/-) mice and prevent atherosclerosis development. ApoE-/- mice were fed a high-fat diet supplemented with or without curcumin (0.1% w/w) for 16 weeks. Aortic sinus sections revealed that curcumin supplementation reduced the extent of atherosclerotic lesions by 45%. Curcumin treatment also reduced cholesterol accumulation in the aortas by 56% and lowered plasma total cholesterol and low-density lipoprotein cholesterol levels. Moreover, the antiatherogenic and cholesterol-lowering effects of curcumin coincided with a significant decrease in intestinal cholesterol absorption. It was reduced by nearly 51%, and the decreased cholesterol absorption was modulated by inhibiting the intestinal expression of Niemann-Pick C1-like 1, predominantly in the duodenal and jejunal segments of the small intestine. These findings support the hypothesis that curcumin supplementation reduces intestinal cholesterol absorption and prevents atherosclerosis in high-fat diet-fed ApoE-/- mice. Curcumin affords a potent antiatherogenic action by inhibiting intestinal cholesterol absorption in the mouse.

Motor imagery (MI)-based brain-computer interfaces (BCIs) have been widely used for rehabilitation of motor abilities and prosthesis control for patients with motor impairments. However, MI-BCI performance exhibits a wide variability across subjects, and the underlying neural mechanism remains unclear. Several studies have demonstrated that both the fronto-parietal attention network (FPAN) and MI are involved in high-level cognitive processes that are crucial for the control of BCIs. Therefore, we hypothesized that the FPAN may play an important role in MI-BCI performance. In our study, we recorded multi-modal datasets consisting of MI electroencephalography (EEG) signals, T1-weighted structural and resting-state functional MRI data for each subject. MI-BCI performance was evaluated using the common spatial pattern to extract the MI features from EEG signals. One cortical structural feature (cortical thickness (CT)) and two measurements (degree centrality (DC) and eigenvector centrality (EC)) of node centrality were derived from the structural and functional MRI data, respectively. Based on the information extracted from the EEG and MRI, a correlation analysis was used to elucidate the relationships between the FPAN and MI-BCI performance. Our results show that the DC of the right ventral intraparietal sulcus, the EC and CT of the left inferior parietal lobe, and the CT of the right dorsolateral prefrontal cortex were significantly associated with MI-BCI performance. Moreover, the receiver operating characteristic analysis and machine learning classification revealed that the EC and CT of the left IPL could effectively predict the low-aptitude BCI users from the high-aptitude BCI users with 83.3% accuracy. Those findings consistently reveal that the individuals who have efficient FPAN would perform better on MI-BCI. Our findings may deepen the understanding of individual variability in MI-BCI performance, and also may provide a new biomarker to predict individual MI-BCI performance.

Common spatial pattern (CSP) is one of the most popular and effective feature extraction methods for motor imagery-based brain-computer interface (BCI), but the inherent drawback of CSP is that the estimation of the covariance matrices is sensitive to noise. In this work, local temporal correlation (LTC) information was introduced to further improve the covariance matrices estimation (LTCCSP). Compared to the Euclidean distance used in a previous CSP variant named local temporal CSP (LTCSP), the correlation may be a more reasonable metric to measure the similarity of activated spatial patterns existing in motor imagery period. Numerical comparisons among CSP, LTCSP, and LTCCSP were quantitatively conducted on the simulated datasets by adding outliers to Dataset IVa of BCI Competition III and Dataset IIa of BCI Competition IV, respectively. Results showed that LTCCSP achieves the highest average classification accuracies in all the outliers occurrence frequencies. The application of the three methods to the EEG dataset recorded in our laboratory also demonstrated that LTCCSP achieves the highest average accuracy. The above results consistently indicate that LTCCSP would be a promising method for practical motor imagery BCI application.
