This study is aimed at investigating the effects of dietary supplementation with Artemisia ordosica crude polysaccharides (AOCP) on lactation performance, antioxidant status, and immune status of lactating donkeys and analyzing rectal microbiomes and serum metabolomes. Fourteen lactating Dezhou donkeys with similar age (6.16 ± 0.67 yr of BW ± SD), weight (250.06 ± 25.18 kg), DIM (39.11 ± 7.42 d), and average parity of 3 were randomly allocated into 2 treatments: a control group (CON, basal diet) and an AOCP group (AOCP, basal diet with 1.0 g/kg DM AOCP). Ten weeks were allotted for the experiment, 2 wk for adaptation, and 8 wk for collecting data and samples. The results showed that supplementation of donkey diets with AOCP increased lactation performance, including DMI, milking yield, estimated milk yield, solids-corrected milk, ECM, milk fat yield, milk protein yield, milk lactose yield, milk TS yield, and milk SNF yield. The digestibility of DM, CP, ADF, and NDF was increased in the AOCP group compared with the CON group. The AOCP group increased the concentrations of IgA, IgG, and IgM, the activities of the superoxide dismutase, catalase, and total antioxidant capacity in the serum. Artemisia ordosica crude polysaccharides decreased the concentrations of tumor necrosis factor-α, nitric oxide, reactive oxygen species, and malondialdehyde in the serum. Compared with the CON group, AOCP increased propionate, butyrate, isovalerate, and total VFA concentrations in rectal feces (P < 0.05). The addition of AOCP to increased diversity (Shannon index) and altered structure of the rectal microflora. As a result of AOCP supplementation, there has been a significant improvement in the colonization of beneficial bacteria, including Lactobacillus, Unclassified_f_Prevotellacea, Ruminococcus, and Fibrobacter genera. In contrast, a decrease in the colonization of the Clostridium_sensu_stricto_1 bacterial genus and other pathogenic bacteria was observed. Meanwhile, metabolomics analysis found that AOCP supplementation upregulated metabolites l-tyrosine content while downregulating 9(S)-HODE, choline, sucrose, lysophosphatidylcholine (LysoPC) (18:0), LysoPC (18:1(9Z)), and LysoPC (20:2(11Z,14Z)) concentrations. These altered metabolites were involved in the PPAR signaling pathway, prolactin signaling pathway, glycerophospholipid metabolism, carbohydrate digestion and absorption, and tyrosine metabolism pathways, which were mainly related to antioxidant capacity, immune responses, and protein metabolism in the lactating donkeys. As a consequence of feeding AOCP diets, beneficial bacteria were abundant, and antioxidant and protein metabolism-related pathways were enriched, which may enhance lactation performance in donkeys. Therefore, supplementing AOCP diets is a desirable dietary strategy to improve donkey health and lactation performance.

Objective: To verify the efficacy and safety of daily oral minodronate in postmenopausal women with established osteoporosis. Methods: In this randomized, double-blinded, placebo-controlled trial, 262 postmenopausal women were enrolled. Patients were randomized to receive daily oral minodronate 1 mg with supplements of 500 mg calcium and 200 U vitamin D3 (n=130) or placebo (n=132) with daily supplements of 500 mg calcium and 200 U vitamin D3, for 48 weeks. The primary endpoint was the average bone mineral density (BMD) change in the lumbar vertebrae 48 weeks post-treatment. Secondary outcome measures was the incidence of vertebral fractures. Safety assessments included the rate of adverse events. Results: At the end of 48 weeks treatment, the average BMD change rate from baseline were: full analysis set results: (3.52±4.82)% in the minodronate group and (2.00±5.74)% in the placebo group; per-protocol set results: (3.99±5.05)% in the minodronate group and (2.07±6.20)% in the placebo group; the differences were all significant (all P<0.05). Vertebral fracture occured in 3 patients (2.3%, 3/132) in the placebo group, and 1 case (0.8%, 1/130) in the minodronate group (P>0.05). The incidence of adverse events was 71.5% (93/130) in the minodronate group and 78.0% (103/132) in the placebo group (P>0.05). Conclusion: Minodronate is effective and safe in the treatment of postmenopausal osteoporosis without severe side effects.

Ancient acupuncture and moxibustion prescriptions are one of the sources for evidence-based acupuncture research, representing the techniques features in treatment. This paper examines and analyses the literature of ancient acupuncture and moxibustion prescriptions for headache. It was found that repeated adaption and private changes of ancient acupuncture prescriptions was common. It was also found that the literature which reported the adaption of the indications of acupoints and the annotation of acupuncture and moxibustion in the form of poetry was often confused with the clinical acupuncture prescriptions. It is essential to trace the literature sources of ancient acupuncture prescriptions and to confirm the earliest documents extant for their authenticity and inheritance. In this sense, they are the two steps for review of the literature of ancient acupuncture prescriptions.

Guanidinoacetic acid (GAA) can improve the growth performance of bulls. This study investigated the influences of GAA addition on growth, nutrient digestion, ruminal fermentation and serum metabolites in bulls. Forty-eight Angus bulls were randomly allocated to experimental treatments, that is, control, low-GAA (LGAA), medium-GAA (MGAA) and high-GAA (HGAA), with GAA supplementation at 0, 0.3, 0.6 and 0.9 g/kg DM, respectively. Bulls were fed a basal diet containing 500 g/kg DM concentrate and 500 g/kg DM roughage. The experimental period was 104 days, with 14 days for adaptation and 90 days for data collection. Bulls in the MGAA and HGAA groups had higher DM intake and average daily gain than bulls in the LGAA and control groups. The feed conversion ratio was lowest in MGAA and highest in the control. Bulls receiving 0.9 g/kg DM GAA addition had higher digestibility of DM, organic matter, NDF and ADF than bulls in other groups. The digestibility of CP was higher for HGAA than for LGAA and control. The ruminal pH was lower for MGAA, and the total volatile fatty acid concentration was greater for MGAA and HGAA than for the control. The acetate proportion and acetate-to-propionate ratio were lower for MGAA than for LGAA and control. The propionate proportion was higher for MGAA than for control. Bulls receiving GAA addition showed decreased ruminal ammonia N. Bulls in MGAA and HGAA had higher cellobiase, pectinase and protease activities and Butyrivibrio fibrisolvens, Prevotella ruminicola and Ruminobacter amylophilus populations than bulls in LGAA and control. However, the total protozoan population was lower for MGAA and HGAA than for LGAA and control. The total bacterial and Ruminococcus flavefaciens populations increased with GAA addition. The blood level of creatine was higher for HGAA, and the activity of l-arginine glycine amidine transferase was lower for MGAA and HGAA, than for control. The blood activity of guanidine acetate N-methyltransferase and the level of folate decreased in the GAA addition groups. The results indicated that dietary addition of 0.6 or 0.9 g/kg DM GAA improved growth performance, nutrient digestion and ruminal fermentation in bulls.

The poem "white rabbits pounding herbs and toads pilling" was recorded in Han Yuefu(Folk) poetry, and the corresponding images were also appeared on the pictorial stones of the Han Dynasty. Some scholars held the opinion that the herbs the white rabbits pounded was "toad pills" , and the "pill" was made of toad venom or the other components of toads.By analyzing literature, the images and unearthed objects, this paper concludes that the meaning of "white rabbits pounding herbs and toads pilling" is that white rabbits and toads work together to make the elixir pills.The word "pill" in the poem is a verb rather than a noun.Toads, like white rabbits, are the agent of actions. They are not the raw materials for elixir pills.

In Gu Kong Lun(Discourse on Bone Hollows), a chapter from Suwen(, Plain Questions): When the governor vessel(Du Mai) generates a disease, treat the governor vessel. Treat it above the bone. In severe cases treat at the camp below the navel(,,). This sentence illuminated acupoint selection methods and treatment thoughts for governor vessel disease, the comprehension of physicians in ancient different dynasties have large divergence. This paper digs and presents its meaning through investigating the following ancient documents: governor vessel disease, acupuncture point indications of governor vessel, relative therapy methods, expert annotation, etc. Then this paper draws a conclusion that "above the bone" means above the vertebral column, where is the circulation area of governor vessel, "under the umbilicus" is the part from umbilicus to pubis symphysis center. This sentence not only indicates the treatment part when governor vessel generates disease, but also emphasizes the therapy ideas that provides different treatment method according to severity degree of disease.

A general feature of unconventional superconductors is the existence of a superconducting dome in the phase diagram. Here we report a series of discrete superconducting phases in the simplest iron-based superconductor, FeSe thin flakes, by continuously tuning the carrier concentration through the intercalation of Li and Na ions with a solid ionic gating technique. Such discrete superconducting phases are robust against the substitution of 20% S for Se, but they are vulnerable to the substitution of 2% Cu for Fe, highlighting the importance of the iron site being intact. The superconducting phase diagram for FeSe derivatives is given, which is distinct from that of other unconventional superconductors.

Ammonia in poultry houses not only affects worker health but also induces a variety of poultry diseases. Alpha-lipoic acid (LA) is an effective antioxidant that protects cells against oxidative injury during various toxic and pathological processes. This study was designed to evaluate the mitigating effects of LA supplementation on ammonia stress and hepatic proteome changes in broilers. Male broilers (22 d old) were allocated to 3 groups: (1) a control group without ammonia stress (CTRL); (2) exposure to 70 ppm ammonia (AM); and (3) exposure to 70 ppm ammonia and dietary administration of 300 mg/kg LA (AM+LA). Ammonia exposure significantly decreased broiler growth performance and plasma glutathione peroxidase activity (P < 0.05), and increased plasma malondialdehyde content and glutamic-pyruvic transaminase activity (P < 0.05). These negative effects were eliminated by LA supplementation. Comparative proteomic analyses revealed 291 differentially expressed proteins in the AM group compared to the CTRL and AM+LA groups. A total of 30 proteins were differentially expressed between the AM/CTRL and (AM+LA)/AM groups. The addition of LA restored 24 of these proteins to control levels; these proteins were mainly related to transcription regulation, detoxification, protein translation and degradation, and immune and stress responses. The differentially expressed proteins included the high mobility group box (HMGB) and glutathione S-transferase (GST), which is closely related to immune response and oxidative stress, and collagens, which are implicated in liver injury. The addition of LA to broiler diet may reduce ammonia toxicity by maintaining the antioxidant system, xenobiotic metabolism, and metabolic pathways.

Objective: To biologically evaluate the three-dimensional(3D) printed co-poly lactic acid/glycolic acid/tri-calcium phosphate(PLGA/TCP) scaffold which could be used for repairing oral and maxillofacial bone defects, and to provide experimental evidence for its further research and clinical application. Methods: PLGA/TCP scaffolds were fabricated using low temperature rapid prototyping technique. Micro-CT and scanning electron microscope(SEM) were used to characterize the surface morphology. MC3T3-E1 cells were seeded onto the scaffold and stained with the rhodamine phalloidin and calcein acetomethoxy. After that, confocal laser scanning microscope was exploited to observe the features and viability of the cells. Moreover, the cells were co-cultured with the extract of PLGA/TCP and complete medium, respectively. The proliferation capability of the cells was assessed by the cell counting kit-8 (CCK-8) on the 1st, 2nd, and 3rd day. The PLGA/TCP scaffolds incorporated with recombinant human bone morphogenetic protein-2(rhBMP-2) of 0, 30, 60 µg(i.e. blank control group, low-dose group and high-dose group) were implanted into the latissimus dorsi muscle of the rats, and 6 weeks later, the samples were harvested to estimate the volume and pattern of new bone. Results: The 3D printed PLGA/TCP scaffold possessed a regular and well-defined porous stereo-structure with porosity of (73±3)%. Micro-CT and SEM showed that pore size were (379±32) and (453±29) µm respectively, and distance between layers were (452± 24) and (415±25) µm, and cylinder diameter were (342±24) and (350±28) µm. It also exhibited excellent cell adhesion and growth ability on the exterior and inner surface through rhodamine phalloidin and calcein acetomethoxy staining. The CCK-8 test demonstrated that the absorbance value of extract group on the 1st and 2nd day(0.51±0.08 and 0.63±0.09) were significantly higher than those in the blank control group(0.39± 0.05 and 0.53±0.05)(P<0.05), while there was no significant difference between the extract group(0.67±0.06) and the blank control group(0.68±0.04)(P>0.05) on the 3rd day. For in vivo test, there was obvious ectopic new bone formation on the PLGA/TCP scaffold incorporated with rhBMP-2, and this was demonstrated using the histological examination and micro-CT. The bone formation in the low-dose group was similar to the shape of the pre-implanted 3D printed scaffold, while much diversity was revealed in the high-dose group duo to over osteogenesis which was validated by the examinations of gross observation, histology and micro-CT. Conclusions: Customized PLGA/TCP scaffolds can be manufactured by 3D printing technique. The scaffold showed an excellent biocompatibility and ectopic osteogenesis when incorporated with rhBMP-2. However, further research is needed to validate it's effect on repairment of the oral and maxillofacial bone defects.

UNLABELLED: We investigated the effect of calcitonin (CT) on lumbar intervertebral disk degeneration (LIDD) in rats with ovariectomy-induced osteopenia. CT protected ovariectomized rats from LIDD by, at least in part, modifying extracellular matrix metabolism of the disks and preserving the microarchitecture and biomechanical properties of adjacent vertebrae. INTRODUCTION: The present study aimed to investigate the effect of CT on lumbar vertebral bone mineral density and intervertebral disk degeneration in ovariectomized (OVX) rats. METHODS: We first subjected 50 3-month-old female rats to either OVX (n = 30) or sham (n = 20). Twelve weeks later, ten OVX and ten sham rats were necropsied. The remaining OVX rats began to receive either saline vehicle (OVX + V, n = 10), or salmon CT (OVX + CT, 16 IU/kg/2 days, n = 10). After 12 weeks of treatment, necropsy was conducted and bone mineral density was determined in L3-4 and L5-6 vertebrae. The microstructure and biomechanical properties of L3 vertebrae were detected by micro-computed tomography and compression test, respectively. L5-6 was also used to measure intervertebral disk height and observe intervertebral disk histological changes by Van Gieson staining and histological scores, as well as immunohistochemistry (IHC) analysis of matrix metalloprotease (MMP)-1, MMP-13, and collagen II expression. RESULTS: At 12 weeks post-OVX, OVX rats had lower BV/TV and Tb.N and higher intervertebral disk histological score than sham rats. After 24 weeks, OVX + CT rats had higher BMD, BV/TV, Tb.N, and bone biomechanical strength values than OVX + V rats. Histological analysis showed OVX + CT rats had significantly lower disk degeneration scores than OVX + V rats. IHC analysis revealed CT treatment decreased expression of MMP-1 and MMP-13 and increased expression of collagen II compared with OVX + V rats. CONCLUSIONS: Our data demonstrate that CT-treated OVX rats display less intervertebral disk degeneration and favorable changes in intervertebral disk metabolism, associated with higher trabecular bone mass, better trabecular microarchitecture, and better biomechanical strength when compared to vehicle-treated OVX rats.

To investigate the effect of humidity and ammonia on the antioxidative capacities and meat qualities of broilers, 192 broilers were divided into 2 groups: high (H, 70 ppm) and low (L, 30 ppm) ammonia concentration. These groups were divided into 30% (Treatment humidity, T) and 60% (Control humidity, C) humidity, giving 4 treatments: C+L, C+H, T+L, and T+H. Blood and muscle antioxidative capacities and meat quality were measured. In the H group, body weight (BW), average daily feed intake (ADFI), average daily weight gain (ADG), blood and muscle antioxidative capacities, and postmortem pectoral muscle a* of broilers were significantly decreased (P < 0.05), and pectoral muscle thiobarbituric acid reactive substance (TBARS) contents and drip losses, postmortem pectoral muscle b* (P < 0.05) and L* (P = 0.054), and pectoral muscle shear forces (P = 0.075) increased. In the T condition, BW, ADFI, pectoral muscle superoxide dismutase (SOD) and glutathione peroxidase (GSH-Px) activities, and pectoral muscle L* decreased (P = 0.053), and pectoral muscle shear forces and TBARS contents increased (P < 0.05). In the T+H group, BW, ADFI, ADG, blood antioxidative capacities, pectoral muscle SOD and GSH-Px activities, and postmortem pectoral muscle a* were significantly lower than those of the C+L group, but postmortem pectoral muscle TBARS contents and pectoral muscle drip losses and shear forces significantly increased (P < 0.05). These results revealed that T+H could significantly reduce growth performance, antioxidative capacities, and meat quality of broilers; T intensified these negative effects.

OBJECTIVE. To determine the prevalence of psychiatric morbidity, identify correlates of psychiatric morbidities, and evaluate the effectiveness of the 12-item General Health Questionnaire as a screening tool for psychiatric morbidity in Chinese patients infected with chronic hepatitis B. METHODS. This cross-sectional study was conducted in a local public specialist outpatient infectious disease clinic from October 2008 to June 2009, in which a total of 160 patients were randomly selected. Psychiatric diagnoses were established by using the Chinese-Bilingual Structured Clinical Interview for the DSM-IV (Axis I Disorders). Scores for the General Health Questionnaire were compared against the psychiatric diagnoses. RESULTS. Among 149 patients, the respective point prevalence of overall psychiatric disorders, depressive disorders, and anxiety disorders was 32%, 16%, and 14%. A family history of psychiatric disorder and absence of knowledge of mode of hepatitis B virus transmission were identified as being associated with current psychiatric disorders, current depressive disorders, and current anxiety disorders. Absence of a confidant was an independent factor for current depressive disorders and use of herbal medicine in the previous month was an independent factor for current anxiety disorders. CONCLUSIONS. Psychiatric disorders are common in Chinese patients with chronic hepatitis B. Identifying associated factors and using the General Health Questionnaire as a screening tool are useful for identifying patients with psychiatric disorders in an infectious disease clinic.

A total of 50 weaning pigs (16 d of age; 4.72 +/- 0.23 kg of BW) were selected to investigate the effect of dietary chito-oligosaccharide (COS) supplementation on growth performance, fecal shedding of Escherichia coli and Lactobacillus, apparent digestibility, and small intestinal morphology. Pigs housed in individual metabolic cages were assigned randomly to 5 treatments (n = 10), including 1 basal diet (control), 3 diets with COS supplementation (100, 200, and 400 mg/kg), and 1 diet with chlortetracycline (CTC) supplementation (80 mg/kg). Fresh fecal samples were collected to evaluate shedding of E. coli and Lactobacillus on d 0, 7, 14, and 21 postweaning. Fresh fecal samples collected from each cage from d 19 to 21 were stored frozen for determination of apparent total tract digestibility. On d 21, all pigs were killed to collect the middle sections of the duodenum, jejunum, and ileum for determination of mucosa morphology. Supplementation of COS at 100 and 200 mg/kg and supplementation of CTC improved (P < 0.05) overall ADG, ADFI, and G:F in comparison with the control. Supplementation of COS at 200 mg/kg as well as supplementation of CTC increased (P < 0.05) apparent total tract digestibility of DM, GE, CP, crude fat, Ca, and P, whereas COS at 100 mg/kg increased (P < 0.05) the digestibility of DM, Ca, and P in comparison with the control diet. Pigs receiving diets supplemented with COS or CTC had a decreased (P < 0.05) incidence of diarrhea and decreased diarrhea scores compared with control pigs. Fecal samples from pigs receiving diets supplemented with COS had greater (P < 0.05) Lactobacillus counts than those from control pigs and pigs receiving diets supplemented with CTC on d 14 and 21. However, supplementation of COS at 200 mg/kg and supplementation of CTC decreased (P < 0.05) E. coli counts in the feces on d 21 compared with the control diet. Dietary supplementation of COS at 200 mg/kg and of CTC increased (P < 0.05) the villus height and villus:crypt ratio at the ileum and jejunum, and COS at 100 mg/kg also increased (P < 0.05) the villus height in the ileum compared with the control diet. The current results indicated that dietary supplementation of COS at 100 and 200 mg/kg enhanced growth performance by increasing apparent digestibility, decreasing the incidence of diarrhea, and improving small intestinal morphology.

Extracts of a perennial and poisonous weed, Stellera chamaejasme L. (Thymelaeaceae) were bioassayed to determine their acaricidal activities against Tetranychus viennensis Zacher (Acari: Tetranychidae) in the laboratory. Extracts had both contact and systemic toxicity to the mite. After liquid chromatography and thin layer chromatography, the extract was further concentrated, separated, and bioassayed. This study is the first to report the acaricidal property of S. chamaejasme and its potential as a botanical pest control agent.

AIM: An efficient, precise, and sensitive method for identifying Atractylodes plants has been established and will contribute significantly to quality control and scientific analysis in Chinese traditional medicine. METHODS: Twenty primers were applied for setting up the RAPD (randomly amplified polymorphic DNA) markers of Atractylodes plants, Atractylodes lancea DC (A lancea DC), Atractylodes japonica Koidz (A japonica K), and Atractylodes ovata DC (A ovata DC). The primer OPF03, OPF05, and OPF14 could discriminate them successfully. The results were also able to apply on the Chinese formulations with Atractylodes purchased from local markets. RESULTS: RAPD was used to investigate phylogenetic relationships among and within closely related species. RAPD analysis reflects heritable changes in the nucleotides sequence in both the coding and noncoding regions, because it is conducted directly from the DNA level. This work first conducted RAPD analysis of Atractylodes plants to establish their RAPD makers. CONCLUSION: The RAPD markers could be applied extensively in the Chinese herbal formulations.

OBJECTIVE: To select high taxol content cell lines of Taxus yunnanensis. METHOD: Choosing different cell aggregates according to their color, texture, growth rate and secretion of colorful substances, culturing them separately, and further analyzing their growth rates and taxolcontents. RESULT AND CONCLUSION: Cell lines with higher taxol contents could be obtained by careful selection; those with darker color, lower growth rate and higher ratio of dry cell weight vs. fresh cell weight usually has higher taxol contents.

RESUMO

130 hospital-discharged elderly patients received our comprehensive assessment by using a Chinese Minimum Data Set-Home Care (MDS-HC). Our case manager developed and implemented care plans with reference to the computer-generated Clients Assessment Protocols. Results showed that the MDS-HC was sensitive to identify elderly persons' holistic needs, and helpful in formulating all-inclusive care plans.

1. A high throughput screening (HTS) method for the evaluation of the seven major human hepatic CYP isoform activities was developed on a 96-well format, with automation. The method utilized pooled human liver microsomes and seven probe substrates, generic conditions for incubation, reaction termination and metabolite extraction with solid phase extraction (SPE) plates. Metabolites from the seven reactions were pooled and quantified using a generic liquid chromatography and tandem mass spectrometry (LCMS/MS) method. 2. The HTS method was validated based on Km values obtained, which were in agreement with literature data. 3. The isoform inhibition profiles of ketoconazole, quinidine, sulfaphenazole, tranylcypromine, alpha-naphthoflavone, and 4-methylpyrazole against CYPs 3A4, 2D6, 2C9, 2A6 land 2C19), 1A2 and 2E1, respectively, were obtained by this HTS method. Graphically obtained IC50 values are in agreement with literature reported values. 4. The HTS method represents a significant efficiency and selectivity improvement over traditional methods, and can be used for CYP inhibition assay and can be extended for liver activity profiling.

RESUMO

A new C18 dibenzocyclooctadiene lignan, kadsumarin A (1) was isolated from Kadsura matsudai Hayata and Schizandra arisanensis Hayata. The anti-HBeAg test revealed that kadsumarin A had activity at a concentration of 40 micrograms/ml (= 90.1 microM). Its structural elucidation by spectral analysis was discussed in this note.
