RESUMO

Cellulose nanofibers (CNFs) aerogel was prepared via simple covalent crosslinking and freeze-drying method. The porous cellulose aerogel possessed high specific surface area and high metal-chelating capacity, which showed fast adsorption kinetics and high adsorption capacity (440.60 mg g-1) in static uranium adsorption process. In the dynamic filtration system, the maximum adsorption capacity reached 194 mg g-1 with the initial concentration of 10 mg L-1. In addition, the CNFs aerogel possessed excellent selectivity and good regeneration ability for uranium adsorption. The integrated analyses of attenuated total reflection Fourier transform infrared (ATR-FTIR), X-Ray photoelectron spectroscopy (XPS) and extended X-ray absorption fine structure (EXAFS) suggested that the predominant UO22+ species formed inner-sphere surface complexes with two active carboxyl groups in the coordination model. This strategy may provide a sustainable route for development of efficient biomass-based adsorbents for selective uranium removal from aqueous solution.

Uranium extraction from seawater is considered as an efficient strategy to meet the increasing demands of uranium. Amidoxime has been reported as one of the most efficient groups for uranium affinity. Herein, amidoximated cellulose fibers were synthesized by grafting polyacrylonitrile (PAN) onto cellulose fibers followed by amidoxime modification. The amidoximated cellulose fibers showed maximum adsorption capacity of 52.88 mg g-1 (pH = 5.0), and its static adsorption process was well fitted with Langmuir model and Pseudo-second-order kinetics. The adsorption mechanism was attributed to the chelating reaction between uranyl complexes and amidoximated cellulose fibers. The prepared fibers were further fabricated into nonwoven membrane for dynamic adsorption, and the breakthrough curves were well fitted to Dose-Response model. The amidoximated cellulose fiber membrane showed a good adsorption capacity of 1.22 mg g-1 at pH 8.0 after filtrating 10.0 L simulated seawater, demonstrating promising efficient engineering materials for uranium extraction from seawater.

A fiber-network chitosan film with three-dimensional interconnected structure was prepared in an alkali/urea solution and regenerated from an ethanol/water coagulation solution. The surface morphology and structure were characterized by scanning electron microscopy (SEM), X-ray diffraction (XRD), N2 adsorption-desorption, attenuated total reflection Fourier transform infrared spectroscopy (ATR-FTIR) and X-ray photoelectron spectroscopy (XPS). Batch adsorption for uranium U(VI) was conducted to investigate the effects of pH, contact time and initial uranium concentration on adsorption capacity. The adsorption of CS-80% was in good agreement with the pseudo-second-order kinetic model and Langmuir isotherm model. The three-dimensional interconnected structure provided more active sites and favored the diffusion of uranium solute, and therefore enhanced the adsorption capacity. The maximum adsorption capacity at pH 5 was 196.735 mg/g. The adsorption mechanism was attributed to chelation and coordination of uranium with -NH2 and -OH groups on chitosan molecules.

The exploration of the relationship between zeolite composition and adsorption performance favored to facilitate its better application in removal of the hazardous substances from water. The adsorption capacity of rhodamine B (RB) onto Beta zeolite from aqueous solution was reported. The relationship between SiO2/Al2O3 ratio and adsorption capacity of Beta zeolite for RB was explored. The structure and physical properties of Beta zeolites with various SiO2/Al2O3 ratios were determined by XRD, FTIR, TEM, BET, UV-vis and so on characterizations. The adsorption behavior of rhodamine B onto Beta zeolite matched to Langmuir adsorption isotherm and more suitable description for the adsorption kinetics was a pseudo-second-order reaction model. The maximum adsorption capacity of the as-prepared Beta zeolite with SiO2/Al2O3 = 18.4 was up to 27.97mg/g.

Nicotinic acid (NA) has recently been shown to inhibit inflammatory response in cardiovascular disease. Sirtuin1 (SIRT1), a NAD(+)-dependent class III histone deacetylase, participates in the regulation of cellular inflammation. We hypothesized that dietary supplementation of NA could attenuate vascular inflammation via modulation of SIRT1 pathway. New Zealand White rabbits received chow or chow supplemented with 0.6% (wt/wt) NA for 2 weeks. Acute vascular inflammation was induced in the animals by placing a non-occlusive silastic collar around the left common carotid artery. At 24 h after collar implantation, the collar-induced production of C-reactive protein and monocyte chemotactic protein-1 was significantly suppressed in the NA-supplemented animals. Meanwhile, NA also decreased the expression of cluster of differentiation 40 (CD40) and CD40 ligand, but up-regulated SIRT1 expression, both in rabbits and in lipopolysaccharide-stimulated endothelial cells. Moreover, knockdown of SIRT1 reversed the inhibitory effect of NA on CD40 expression. Further study revealed that NA also decreased the expression of CD40 partly through mammalian target of rapamycin. These results indicate that NA protects against vascular inflammation via the SIRT1/CD40-dependent signaling pathway.

Hydroxysafflor yellow A (HSYA), the main active natural constituent extracted from Carthamus tinctorius L., has been widely used for the treatment of cerebrovascular and cardiovascular diseases. The aim of this study is to explore the effect of HSYA on alcohol-induced liver injury and the underlying mechanism. Male Sprague-Dawley rats were used to establish the liver injury model induced by alcohol. HSYA treatment ameliorated serum biochemical indicators by reducing the levels of alanine aminotransferase (ALT), aspartate aminotransferase (AST), hyaluronan (HA), laminin (LN), and type III precollagen (III-C) in rats. HSYA efficiently increased the activity and messenger RNA (mRNA) of superoxide dismutase (SOD) and glutathione peroxidase (GPx) in rat liver tissue compared with those of model group, which was obviously reduced by alcohol. HSYA also apparently decreased the levels of reactive oxygen species (ROS) and malondialdehyde (MDA) in rat liver tissue compared with those of model group, which was obviously enhanced by alcohol. Histological studies demonstrated that HSYA substantially reduced the number of macro- and micro-vesicular steatosis, suppressed hepatic fibrogenesis and shrunk ballooning degeneration areas, ameliorated the severity of liver damage induced by long-term drinking, and finally improved the liver architecture. In addition, immunohistochemistry study indicated that the activation of transforming growth factor β1 (TGF-β1) stimulated by alcohol in rat liver tissue was significantly blocked by HSYA. Collectively, these data demonstrated that HSYA can effectively protect the liver of rats from long-term alcohol injury, which relates with the enhanced antioxidant capacity of liver tissues and inhibition of TGF-β1 expression

UNLABELLED: Although high-dose cyclophosphamide seems to achieve durable complete remission, there are still concerns about its too much early toxicity. So, we designed a clinical study to investigate the effects of high-dose cyclophosphamide/ATG combined with cord blood infusion as first-line therapy for patients with severe aplastic anemia. PATIENTS AND METHOD: Between January 2003 and September 2007, we treated 16 treatment-naive patients with severe aplastic anemia with cord blood infusion after high-dose cyclophosphamide (50 mg/kg/d × 2) and rabbit antithymocyte globulin (3 mg/kg/d × 5) therapy. RESULTS: Although only one patient had durable full donor engraftment, 14 of the enrolled 16 patients had rapid autologous hematopoietic recovery. The median recovery time for neutrophils and platelets was only 23 and 37 d after infusion of cord blood. Of the 15 responding patients, all patients achieved treatment-free remission: nine patients met the criteria for a complete remission; six patients achieved a partial remission. CONCLUSION: Infusion of cord blood after high-dose cyclophosphamide/ATG resulted in a rapid autologous hematologic recovery and a high response rate in patients with treatment-naive patients with severe aplastic anemia. These promising results merit further investigation and confirmation on a larger number of patients.

BACKGROUND AND AIM: Curcumin, an active component derived from dietary spice turmeric (Curcuma longa), has been demonstrated antihyperglycemic, antiinflammatory and hypocholesterolemic activities in obesity and diabetes. These effects are associated with decreased level of circulating free fatty acids (FFA), however the mechanism has not yet been elucidated. The flux of FFA and glycerol from adipose tissue to the blood stream primarily depends on the lipolysis of triacylglycerols in the adipocytes. Adipocyte lipolysis is physiologically stimulated by catecholamine hormones. Tumor necrosis factor-α (TNFα) stimulates chronic lipolysis in obesity and type 2 diabetes. In this study, we examined the role of curcumin in inhibiting lipolytic action upon various stimulations in 3T3-L1 adipocytes. METHODS: Glycerol release from TNFα or isoproterenol-stimulated 3T3-L1 adipocytes in the absence or presence of curcumin was determined using a colorimetric assay (GPO-Trinder). Western blotting was used to investigate the TNFα-induced phosphorylation of MAPK and perilipin expression. Fatcake and cytosolic fractions were prepared to examine the isoproterenol-stimulated hormone-sensitive lipase translocation. RESULTS: Treatment with curcumin attenuated TNFα-mediated lipolysis by suppressing phosphorylation of extracellular signal-related kinase 1/2 (ERK1/2) and reversing the downregulation of perilipin protein in TNFα-stimulated adipocytes (p<0.05). The acute lipolytic response to adrenergic stimulation of isoproterenol was also restricted by curcumin (10-20 µM, p<0.05), which was compatible with reduced perilipin phosphorylation(29%, p<0.05) and hormone-sensitive lipase translocation(20%, p<0.05). CONCLUSIONS: This study provides evidence that curcumin acts on adipocytes to suppress the lipolysis response to TNFα and catecholamines. The antilipolytic effect could be a cellular basis for curcumin decreasing plasma FFA levels and improving insulin sensitivity.
