Objective: This study aims to investigate the impact of patient-centered health education on individuals with type 2 diabetes coexisting with hyperlipidemia. Methods: A cohort of 80 patients with type 2 diabetes and hyperlipidemia attending our hospital from February 2022 to August 2022 were randomly assigned to either the health education group or the control group. While the control group received routine health education, the health education group received additional patient-centered health education. Subsequently, we compared blood glucose and lipid levels, negative emotions, quality of life, and the incidence of unhealthy eating or overweight between the two groups post-education. Results: Following the health education intervention, the health education group exhibited superior improvements in blood glucose and lipid levels compared to the control group. Moreover, there was a significant decrease in SAS and SDS scores and a notable increase in quality of life compared to the control group. The health education group also demonstrated a lower incidence of unhealthy eating or overweight. Conclusions: Patient-centered health education for individuals with type 2 diabetes and hyperlipidemia proves effective in enhancing glucose and lipid metabolism, mitigating negative emotions, improving quality of life, and reducing unhealthy habits.

Dissolved organic matter (DOM) in petroleum refinery wastewater is an extremely complex mixture. A better understanding of chemical compositions of DOM at the molecular level is necessary for the design and optimization of wastewater treatment processes. In this study, two largely different DOM samples, one from a petroleum refinery wastewater and the other from the Suwannee river water, were characterized by Fourier transform ion cyclotron resonance mass spectrometry (FT-ICR MS) coupled with positive-/negative-ion electrospray ionization (ESI), and positive-ion atmospheric pressure photoionization (APPI). For wastewater DOM, a total of 6226 molecular formulae were assigned in the three ionization modes. However, only 1182 molecular formulae were common in all three mass spectra, indicating that the techniques were highly complementary in the types of molecules they ionize. Acid Ox (x = 1-9) and basic N1Ox (x = 0-2) classes were dominant in the wastewater DOM detected in negative-ion and positive-ion ESI mode, respectively. And the wastewater DOM contains considerable amounts of polycyclic aromatic hydrocarbons that did not respond to ESI but can be ionized selectively by APPI. Compared with riverine DOM, the refinery wastewater DOM has a higher molecular complexity and is more enriched in hydrocarbon, and nitrogen- and sulfur-containing compounds. The results show that the major components of refinery wastewater DOM were distinctive from those of the natural organic matter. Though not quantitative, the results obtained by various ionization techniques were found to be complementary, and are helpful to our understanding of the selectivity of different ionization techniques as well as the molecular compositions of DOM.

Tea polyphenols (TP) is the main functional substances in tea. It has been reported that TP can modulate the composition of gut microbes in the human body, in addition, after the bio-transformation by intestinal flora, the metabolites of TP also have positive effects on the health of the host. Lots of researches have shown that TP have possible therapeutic effect against high fat diet induced obesity, which is closely related to the gut flora of the host. Therefore, this review focused on the interactions of TP with intestinal microbiota and their implication for cellular signal conditioning mechanism that will enable us to better study the two-way effects of TP and intestinal microbiota on host health improvement. PRACTICAL APPLICATIONS: TP have been widely concerned for their health care properties. As the functional food components, TP have strong antioxidant and physiological activities for human body. A better understanding on the interactions of TP with intestinal microbiota and their implication for cellular signal conditioning mechanism will lead us to better evaluate the contribution of the microbial metabolites of TP, as well as the regulation of intestinal bacterial diversity and abundance for host health.

BACKGROUND: The protective effect of melatonin against bone metabolism imbalance in osteoporosis (OP) induced by drugs such as retinoic acid (RA) is unclear. The aim of this study was to explore the role of melatonin in bone destruction based on a mouse model. METHODS: RA-induced OP model mice were established. To assess the effect of melatonin on these mice, micro-CT was used to characterize the trabecular structure of normal mice and those treated with RA (model), RA + low-dose melatonin (Mlt-L), RA + high-dose melatonin (Mlt-H), and RA + alendronate sodium (positive control). The shape of the trabecular bone, the length and diameter of the femoral head and the height and diameter of vertebra(L1) of each group were also measured and the number of osteoclasts was determined by Tartrate-resistant acid phosphatase (TRACP) staining. Meanwhile, the expression of alkaline phosphatase (ALP) was evaluated by immunohistochemistry assays. The differences between groups in terms of liver and kidney oxidation-related indexes and serum and urinary indicators related to bone metabolism were also analyzed. Furthermore, qRT-PCR and western blotting were used to evaluate the effect of melatonin on osteogenic and osteoclastic differentiation in MC3T3-E1 and RAW264.7 cells, respectively. RESULTS: RA induction led to a decrease in the amount and density of trabecular bone, a decrease in the length and diameter of the femur and height, diameter of the vertebra (L1), a decrease in bone mass and density and the expression of ALP, and an increase in the number of osteoclasts. Melatonin treatment alleviated these effects induced by RA, increasing the amount of trabecular bone in OP mice, improving the microstructure of the femur and vertebra(L1) and increasing bone mass bone density and the expression of ALP, as well as decreasing the number of osteoclasts. Additionally, blood and urinary bone metabolism-related indicators showed that melatonin promoted bone formation and inhibited bone resorption. Determination of oxidant and antioxidant biomarkers in the livers and kidneys of the mice revealed that melatonin promoted the antioxidant level and suppressed the level of oxidant molecules in these organs. In vitro, RA promoted osteoclasts and inhibit osteogenesis by increasing oxidative stress levels in the RAW264.7 and MC3T3-E1 cells, but melatonin reversed this effect. Melatonin may, therefore, play a role in the ERK/SMAD and NF-κB pathways. CONCLUSIONS: Melatonin can alleviate bone loss in RA-induced OP model mice, repair the trabecular microstructure, and promote bone formation. These effects may be related to reducing oxidation levels in vivo and vitro through the ERK/SMAD and NF-κB pathways.

In this study, a multifunctional graphene based nanohybrid, termed as GN/Fe3O4/PF127, is engineered via a facile one-pot process consisting of simultaneous reduction of graphene oxide/Fe3O4 and subsequent assembly of Pluronic F127 (PF127) onto graphene nanosheets (GNs). The unique aromatic and planar structure of GNs allows the attachment of multiple functional components including MRI contrast agent (Fe3O4 nanoparticles) and an aromatic anticancer drug like doxorubicin (DOX), as well as PF127 coating which imparts physiological dispersivity and stability to the nanohybrid. The successful assembly process is revealed by TEM observation, size and FITR monitoring. In contrast with the primitive graphene or its oxide derivative, the resulting GN/Fe3O4/PF127 nanohybrids have shown high biological dispersion and MRI effect for diagnosis due to the incorporation of superparamagnetic Fe3O4 nanoparticles without evident cytotoxicity. Moreover, the GN/Fe3O4/PF127 nanohybrid exhibits a photothermal effect due to the considerable optical absorption in the near-infrared region of GNs. The GN/Fe3O4/PF127 nanohybrid could be a further platform for chemophototherapy assisted by the therapeutic DOX. Cellular toxicity assays indicated that the DOX-loaded GN/Fe3O4/PF127 nanohybrid showed a remarkable cytotoxicity to HeLa cells and the cytotoxic effect was intensified when subjected to photoirradiation. Confocal laser scanning microscopy (CLSM) and flow cytometric analysis (FCAS) revealed that the nanohybrid could be easily uptaken into HeLa cells.
