OBJECTIVE: To observe the histological and ultrastructural changes of the otopoint "Stomach" (MA-IC) area in chronic gastritis rabbits so as to provide a foundation for auricular acupoint diagnosis and treatment. METHODS: A total of 20 New Zealand rabbits (half male and half female) were randomly and equally divided into control and model groups. The chronic gastritis model was established by gavage of 5% sodium salicylate each day for 5 weeks, while the rabbits of the control group received gavage of clear water at the same volume. Morphological changes of the tissues of gastric mucosa, otopoint "Stomach" and auricular control point were observed under light microscope after staining with hematoxylin-eosin (H.E.), and given scores (0-3 points) according to the levels of inflammatory cells. The ultrastructural changes of the otopoint "Stomach" tissue were observed under transmission electronic microscope (TEM). RESULTS: H.E. staining revealed smoothness of the gastric smooth muscle with no or a few of inflammatory cells in the control group, and appearance of gastric mucosal hemorrhage and erosion, slightly disordered epithelial glands and infiltration of a large number of lymphocytes in the mucosal layer with some clustered lymphocyte aggregation foci in the model group. The pathological score of gastric mucous in the model group was significantly higher than that in the control group (P<0.01). Under light microscope, no obvious changes were observed in the skin of the otopoint "Stomach" of the control group and the control point of the model group, whereas hyperplasia and abscission in the epidermic cuticle and spinous layer and basal layer, dermal tissue and inflammatory cell infiltration were observed in the otopoint "Stomach" of the model group. Results of TEM observation showed no significant changes in the ultrastructure of the otopoint "Stomach" in the control group, and swollen and vacuolated epidermal keratinocyte mitochondria, reduced keratin filament aggregation, widened local cell space, unclear desmosome structure, activation of the dermal fibroblasts, and an increase of the myelinated nerve mitochondria in the "Stomach" region in the model group. CONCLUSION: The otopoint "Stomach" tissue has structural damage and hyperplasia in chronic gastritis rabbits, suggesting a special correlation between the otopoint "Stomach" and gastric tissue, hence, providing a morphological basis for otopoint diagnosis and treatment.

Iron overload remains a concern in myelodysplastic syndrome (MDS) patients especially those requiring recurrent blood transfusions. Whether iron chelating therapy (ICT) is beneficial to the long-term survival of myelodysplastic syndrome is still a controversial issue. Therefore, we conducted a systematic review and meta-analysis to clarify the relationship between ICT and long-term survival in patients with MDS. A total of 14 studies involving 7242 participants were identified; the outcomes revealed that for patients with MDS, ICT resulted in a lower risk of mortality compared to those with no ICT (HR 0.57; 95% CI 0.44-0.70; P < 0.001); what is more, ICT led to a lower risk of leukemia transformation (HR 0.70; 95% CI 0.52-0.93; P = 0.016). Results of subgroup analyses based on adequate ICT or any ICT, low/int-1 IPSS or unclassified IPSS and study types indicated that the ICT had a beneficial role in all these groups of patients.

In this paper, a novel high throughput screening method for antioxidants was described. The screening process was completed on a 5 cm × 5 cm silica gel 60 plate. Luminol was used as the fluorogenic substrate while hydrogen peroxide employed to excite the chemiluminescence. A dark spot was observed due to quenching effect of antioxidant. A linear model of the integral area of the spot as a function of analyte amount was fit by using vitamin C as positive control. Excellent linearity (R2 = 0.9978) was obtained in the range of 2.31-23.1 µg. The variation coefficients of intra- and inter-chip precisions were below 5% (n = 10) and 10% (n = 6) respectively. The new method was validated by comparing the analysis result of six flavonoids with that from a sequential injection method. The Pearson correlation coefficient was up to 0.98. A set of traditionally used herbal medicines was screened, and Rhei Radix et Rhizoma showed the strongest signal. The major bioactive ingredients were further assigned by flow injection and MS analyses. These results reveal the prospects of the proposed method to supply a promising tool in vitro for high throughput screening and activity evaluation of antioxidants in a fast, low-cost and reliable manner.

BACKGROUND: The increase in the levels of reactive oxygen species (ROS) in acute myeloid leukemia (AML) patients has been previously described; thus, it is important to regulate ROS levels in AML. METHODS: Flow cytometry were used to assess the in vitro effect of compound kushen injection (CKI). Quantitative proteomics were used to analyse the mechanism. The AML patient-derived xenograft (PDX) model were used to evaluate the in vivo effect of CKI. RESULTS: We found that intracellular ROS levels in AML cells were decreased, the antioxidant capacity were increased when treated with CKI. CKI inhibited the proliferation of AML cells and enhanced the cytotoxicity of AML cells, which has few toxic effects on haematopoietic stem cells (HSCs) and T cells. At the single-cell level, individual AML cells died gradually by CKI treatment on optofluidic chips. CKI promoted apoptosis and arrested cell cycle at G1/G0 phase in U937 cells. Furthermore, higher peroxiredoxin-3 (Prdx3) expression levels were identified in CKI-treated U937 cells through quantitative proteomics detection. Mechanically, the expression of Prdx3 and peroxiredoxin-2 (Prdx2) was up-regulated in CKI-treated AML cells, while thioredoxin 1 (Trx1) was reduced. Laser confocal microscopy showed that the proteins Prdx2 could be Interacted with Trx1 by CKI treatment. In vivo, the survival was longer and the disease was partially alleviated by decreased CD45+ immunophenotyping in peripheral blood in the CKI-treated group in the AML PDX model. CONCLUSIONS: Antioxidant CKI possess better clinical application against AML through the Prdxs/ROS/Trx1 signalling pathway.

RESUMO

One new pentacyclic triterpenoid, 3ß-carboxylicfilic-4(23)-ene (1), and three known pentacyclic triterpenoids, adian-5-en-3α-ol (2), fernenol (3), and fern-7-en-3ß-ol (4) were isolated from the petroleum ether phase of the ethanolic extract of Ainsliaea yunnanensis Franch. Their structures were established by spectroscopic methods including 1-D and 2-D NMR, and MS experiments. Compounds 1, 2, 3, and 4 showed significant selective cytotoxicity against human acute monocytic leukemia cell line (THP-1) with IC50 values of 5.12 µM, 1.78 µM, 1.74 µM, and 1.75 µΜ, respectively. Compound 1 also showed an anti-inflammatory effect through the inhibition of the activity of NF-κB by blocking the nuclear translocation of p65.

We assessed the clinical effectiveness and safety of CKI (compound Kushen injection) plus standard induction chemotherapy for treating adult acute leukemia (AL). We randomly assigned 332 patients with newly diagnosed AL to control (n = 165, receiving DA (daunorubicin and cytarabine) or hyper-CVAD (fractionated cyclophosphamide, doxorubicin, vincristine, and dexamethasone)) or treatment (n = 167, receiving CKI and DA or hyper-CVAD) groups. Posttreatment, treatment group CD3+, CD4+, CD4+/CD8+, natural killer (NK) cell, and immunoglobulin (IgG, IgA, and IgM) levels were significantly higher than those of the control group (p < 0.05), and CD8+ levels were lower in the treatment group than in the control group (p < 0.05). Treatment group interleukin- (IL-) 4 and IL-10 levels were significantly higher compared to the control posttreatment (both p < 0.05) as were complete remission, overall response, and quality of life (QoL) improvement rates (p < 0.05). The control group had more incidences of grade 3/4 hematologic and nonhematologic toxicity (p < 0.05). Responses to induction chemotherapy, QoL improvement, and adverse events incidence between control group patients with acute myeloid leukemia and acute lymphocytic leukemia were not significantly different. CKI plus standard induction chemotherapy is effective and safe for treating AL, possibly by increasing immunologic function.

By fully optimizing the separation and analytical conditions, a rapid-resolution LC (RRLC, 1.8 microm particle size) coupled with TOF-MS method was developed and validated for multiple compounds analysis of traditional Chinese medicinal compound preparations (TCMCPs). As a typical example, determination of Compound Danshen preparations (CDPs, mainly comprising Salvia miltiorrhiza and Panax notoginseng), was improved on three groups of bioactive constituents (phenolic acids, diterpenoids, and saponins). LODs down to 1.2 pg, permitting very good separation of 20 analytes and 3 internal standards (ISs) in 17 min, represented an approximate five-fold reduction of time compared to conventional HPLC. Unequivocal identification of target compounds in CDPs was based on accurate mass measurement of ions, and comparison of retention performance with available standards. Quantitation was carried out using the narrow window extracted ion chromatograms (XICs) of each compound with an interference-free baseline (using a +/-0.02 Da window) yielding good linearity of response (r(2) >0.9940), and excellent precision of retention time (t(R)) both interday (RSD, 0.01-0.19%) and intraday (RSD, 0.02-1.08%). The acceptable recoveries obtained were in the range of 77.69-113.8%. The results demonstrated the robustness and applicability of RRLC-TOF-MS for multicomponents analysis in TCMCPs with diverse chemical compositions and properties.
