Background: In a context of long-term highly intensive grazing in grassland ecosystems, a better understanding of how quickly belowground biodiversity responds to grazing is required, especially for soil microbial diversity. Methods: In this study, we conducted a grazing experiment which included the CK (no grazing with a fenced enclosure undisturbed by livestock), light and heavy grazing treatments in a desert steppe in Inner Mongolia, China. Microbial diversity and soil chemical properties (i.e., pH value, organic carbon, inorganic nitrogen (IN, NH4+-N and NO3--N), total carbon, nitrogen, phosphorus, and available phosphorus content) both in rhizosphere and non-rhizosphere soils were analyzed to explore the responses of microbial diversity to grazing intensity and the underlying mechanisms. Results: The results showed that heavy grazing only deceased bacterial diversity in the non-rhizosphere soil, but had no any significant effects on fungal diversity regardless of rhizosphere or non-rhizosphere soils. Bacterial diversity in the rhizosphere soil was higher than that of non-rhizosphere soil only in the heavy grazing treatment. Also, heavy grazing significantly increased soil pH value but deceased NH4+-N and available phosphorus in the non-rhizosphere soil. Spearman correlation analysis showed that soil pH value was significantly negatively correlated with the bacterial diversity in the non-rhizosphere soil. Combined, our results suggest that heavy grazing decreased soil bacterial diversity in the non-rhizosphere soil by increasing soil pH value, which may be due to the accumulation of dung and urine from livestock. Our results highlight that soil pH value may be the main factor driving soil microbial diversity in grazing ecosystems, and these results can provide scientific basis for grassland management and ecological restoration in arid and semi-arid area.

BACKGROUND: The term "vascular cognitive impairment" (VCI) describes various cognitive conditions that include vascular elements. It increases the risk of morbidity and mortality in the elderly population and is the most common cognitive impairment associated with cerebrovascular disease. Understanding the etiology of VCI may aid in identifying approaches to target its possible therapy for the condition. Treatment of VCI has focused on vascular risk factors. There are no authorized conventional therapies available right now. The medications used to treat VCI are solely approved for symptomatic relief and are not intended to prevent or slow the development of VCI. PURPOSE: The function of Chinese medicine in treating VCI has not yet been thoroughly examined. This review evaluates the preclinical and limited clinical evidence to comprehend the "multi-component, multi-target, multi-pathway" mechanism of Traditional Chinese medicine (TCM). It investigates the various multi-omics approaches in the search for the pathological mechanisms of VCI, as well as the new research strategies, in the hopes of supplying supportive evidence for the clinical treatment of VCI. METHODS: This review used the Preferred Reporting Items for Preferred reporting items for systematic reviews and meta-analyses (PRISMA) statements. Using integrated bioinformatics and network pharmacology approaches, a thorough evaluation and analysis of 25 preclinical studies published up to July 1, 2023, were conducted to shed light on the mechanisms of TCM for vascular cognitive impairment. The studies for the systematic review were located using the following databases: PubMed, Web of Science, Scopus, Cochrane, and ScienceDirect. RESULTS: We discovered that the multi-omics analysis approach would hasten the discovery of the role of TCM in the treatment of VCI. It will explore components, compounds, targets, and pathways, slowing the progression of VCI from the perspective of inhibiting oxidative stress, stifling neuroinflammation, increasing cerebral blood flow, and inhibiting iron deposition by a variety of molecular mechanisms, which have significant implications for the treatment of VCI. CONCLUSION: TCM is a valuable tool for developing dementia therapies, and further research is needed to determine how TCM components may affect the operation of the neurovascular unit. There are still some limitations, although several research have offered invaluable resources for searching for possible anti-dementia medicines and treatments. To gain new insights into the molecular mechanisms that precisely modulate the key molecules at different levels during pharmacological interventions-a prerequisite for comprehending the mechanism of action and determining the potential therapeutic value of the drugs-further research should employ more standardized experimental methods as well as more sophisticated science and technology. Given the results of this review, we advocate integrating chemical and biological component analysis approaches in future research on VCI to provide a more full and objective assessment of the standard of TCM. With the help of bioinformatics, a multi-omics analysis approach will hasten the discovery of the role of TCM in the treatment of VCI, which has significant implications for the treatment of VCI.

Inspired by the bifunctional phototherapy agents (PTAs), constructing compact PTAs with efficient photothermal therapy (PTT) and photodynamic therapy (PDT) effects in the near-infrared (NIR-II) biowindow is crucial for high therapeutic efficacy. Herein, none-layered germanium (Ge) is transformed to layered Ge/germanium phosphide (Ge/GeP) structure, and a novel two-dimensional sheet-like compact S-scheme Ge/GeP in-plane heterostructure with a large extinction coefficient of 15.66 L/g cm-1 at 1,064 nm is designed and demonstrated. In addition to the outstanding photothermal effects, biocompatibility and degradability, type I and type II PDT effects are activated by a single laser. Furthermore, enhanced reactive oxygen species generation under longer wavelength NIR laser irradiation is achieved, and production of singlet oxygen and superoxide radical upon 1,064 nm laser irradiation is more than double that under 660 nm laser irradiation. The S-scheme charge transfer mechanism between Ge and GeP, is demonstrated by photo-irradiated Kelvin probe force microscopy and electron spin resonance analysis. Thus, the obtained S-scheme Ge/GeP in-plane heterostructure shows synergistic therapeutic effects of PTT/PDT both in vitro and in vivo in the NIR-II biowindow and the novel nanoplatform with excellent properties has large clinical potential.

Conductive hydrogels have shown great potential in wound healing and skin tissue engineering, owing to their electroactive, mechanical, and chemical properties. However, it still remains as a challenge to incorporate other functions into conductive hydrogels, such as antibacterial ability, controllable drug release, and biodegradability. In this study, a black phosphorus-based conductive hydrogel (HA-DA@BP) is prepared by an amidation reaction coupled with a coordination of Fe3+ -catechol. The hydrogel could be changed from the sol phase to the gel phase under electrical stimulus (ES). The results show that BP could be released under slight acidity, which is cell compatible but could achieve synergistic electrical antibacterial action and promote wound healing. This study proves that BP is a strong candidate for electroactive materials and provides a new insight for the development of BP-based biomedical materials in skin tissue engineering.

Background: Single-cell RNA sequencing (scRNA-Seq) provides new perspectives and ideas to investigate the interactions between different cell types and organisms. By integrating scRNA-seq with new computational frameworks or specific technologies, better Alzheimer's disease (AD) treatments may be developed. Methods: The single-cell sequencing dataset GSE158234 was obtained from the GEO database. Preprocessing, quality control, dimensionality-reducing clustering, and annotation to identify cell types were performed on it. RNA-seq profiling dataset GSE238013 was used to determine the components of specific cell subpopulations in diverse samples. A set of genes included in the OMIM, Genecards, CTD, and DisGeNET databases were selected as highly plausible AD-related genes. Then, ROC curves were created to predict the diagnostic value using the significantly expressed genes in the KO group as hub genes. The genes mentioned above were mapped to the Coremine Medical database to forecast prospective therapeutic Chinese medicines, and a "Chinese medicine-ingredient-target" network was constructed to screen for potential therapeutic targets. The last step was to undertake Mendelian randomization research to determine the causal link between the critical gene IL1B and AD in the genome-wide association study. Results: Using the scRNA-seq dataset, five unique cell clusters were discovered. These clusters were further subdivided into four distinct cell types using marker genes. The KO group showed a more substantial differential subgroup of macrophages than the WT group. By using the available datasets and PPI network analysis, 54 common genes were discovered. Four clusters were identified using the MCODE approach, and correlation analysis showed that seven genes in those four clusters had a significantly negative correlation with macrophages. Six genes in four sets had a significantly positive correlation. Five genes had different levels of expression in the WT and KO groups. The String database was used to identify the regulatory relationships between the four genes (IL10, CX3CR1, IL1B, and IL6) that were finally selected as AD hub genes. Screening identified potential traditional Chinese medicine to intervene in the transformation process of AD, including Radix Salviae, ginseng, Ganoderma, licorice, Coptidis Rhizoma, and Scutellariae Radix, in addition to promising therapeutic targets, such as PTGS1, PTGS2, and RXRA. Finally, it was shown that IL1B directly correlated with immune cell infiltration in AD. In inverse variance weighting, we found that IL1B was associated with a higher risk of AD, with an OR of 1.003 (95% CI = 1.001-1.006, p = 0.038). Conclusion: Our research combined network pharmacology and the scRNA-seq computational framework to uncover pertinent hub genes and prospective traditional Chinese medicine potential therapeutic targets for AD. These discoveries may aid in understanding the molecular processes behind AD genes and the development of novel medications to treat the condition.

Genotype-by-environment interaction (G-by-E) is a common but potentially problematic phenomenon in plant breeding. In this study, we investigated the genotypic performance and two measures of plasticity on a phenotypic and genetic level by assessing 234 maize doubled haploid lines from six populations for 15 traits in seven macro-environments with a focus on varying soil phosphorus levels. It was found intergenic regions contributed the most to the variation of phenotypic linear plasticity. For 15 traits, 124 and 31 quantitative trait loci (QTL) were identified for genotypic performance and phenotypic plasticity, respectively. Further, some genes associated with phosphorus use efficiency, such as Zm00001eb117170, Zm00001eb258520, and Zm00001eb265410, encode small ubiquitin-like modifier E3 ligase were identified. By significantly testing the main effect and G-by-E effect, 38 main QTL and 17 interaction QTL were identified, respectively, in which MQTL38 contained the gene Zm00001eb374120, and its effect was related to phosphorus concentration in the soil, the lower the concentration, the greater the effect. Differences in the size and sign of the QTL effect in multiple environments could account for G-by-E. At last, the superiority of G-by-E in genomic selection was observed. In summary, our findings will provide theoretical guidance for breeding P-efficient and broadly adaptable varieties.

This study aimed to investigate the impact of the replanting of basswood Ganoderma lingzhi on the soil bacterial community and reveal the obstacle phenomenon of replanting basswood G. lingzhi. In this study, the soil bacterial community of wild 20 cm (N0a) and 40 cm deep soil (N0b), cultivated once (N1a, N1b) and twice (N2a, N2b), were investigated by Illumina MiSeq sequencing. The predominant bacterial phyla at the phylum classification level were Acidobacteria, Chloracidobacteria, Nitrospira, Spartobacteria, Gemmatimonadetes, Acidobacteria-6. Still, only the relative abundance of Chloracidobacteria and Acidobacteriia increased after two years of replanting of basswood G. lingzhi. At the genus level, the dominant genus included many unclassified bacteria. Among the known genera, the best genus was DA101, which showed a decreasing trend after two years of replanting. Network analysis showed that more connections of bacterial communities were observed in soil samples of the group "a," indicating that the replanting of basswood G. lingzhi can affect the relationship between soil bacterial communities at depths. The phylogenetic investigation of communities by reconstruction of unobserved states (PICRUSt) analysis showed that the gene metabolism function of soil bacteria was quite different after one year of replanting basswood G. lingzhi. The replanting of basswood G. lingzhi changed the composition and function of the soil bacterial community, and also affected the bacterial community diversity in the soil at different depths.

Phosphorus (P) deficiency is an important challenge the world faces while having to increase crop yields. It is therefore necessary to select maize (Zea may L.) genotypes with high phosphorus use efficiency (PUE). Here, we extensively analyzed the biomass, grain yield, and PUE-related traits of 359 maize inbred lines grown under both low-P and normal-P conditions. A significant decrease in grain yield per plant and biomass, an increase in PUE under low-P condition, as well as significant correlations between the two treatments were observed. In a genome-wide association study, 49, 53, and 48 candidate genes were identified for eleven traits under low-P, normal-P conditions, and in low-P tolerance index (phenotype under low-P divided by phenotype under normal-P condition) datasets, respectively. Several gene ontology pathways were enriched for the genes identified under low-P condition. In addition, seven key genes related to phosphate transporter or stress response were molecularly characterized. Further analyses uncovered the favorable haplotype for several core genes, which is less prevalent in modern lines but often enriched in a specific subpopulation. Collectively, our research provides progress in the genetic dissection and molecular characterization of PUE in maize.

Biomimetic membrane materials have been widely explored and developed for drug loading and tissue engineering applications due to their excellent biocompatibility and abundant reaction sites. However, novel cytomembrane mimics have been lacking for a long time. In this study, black phosphorus (BP) was used as the foundation for a new generation of promising cytomembrane mimics due to its multiple similarities to cytomembranes. Inspired by the dual function of endotoxins on membranes, we prepared a BP-based cytomembrane mimic with controllable antibacterial ability via electrostatic interaction between BP and [1-pentyl-1-quaternary ammonium-3-vinyl-imidazole]Br ([PQVI]Br). The release of PQVI could be manipulated in different conditions by adjusting the electrostatic force, thereby achieving controllable antibacterial ability. This report confirms the possibility of using BP as a new material to mimic cytomembranes and provides a new concept of controllable antibacterial action based on endotoxins.

OBJECTIVES: The present study aimed to clarify the effects of Gypenosides on myocardial ischemia-reperfusion injury. Using rat H9c2 cardiomyocytes as the research object, the model of cardiomyocyte hypoxia and reoxygenation was established to observe the protective effects of Gypenosides on myocardial ischemia-reperfusion injury, revealing the key targets and possible mechanisms for Gypenosides to exert myocardial protection. MATERIAL AND METHODS: A model of cardiomyocyte hypoxia and reoxygenation was prepared. The activity of cardiomyocytes was detected by CCK-8 method. The cardiomyocyte injury was evaluated by LDH assay. The cardiomyocyte apoptosis rate was detected by flow cytometry. The mitochondrial membrane potential of cardiomyocytes was detected by JC-1 staining. Western blot was used to detect the expression of MPTP downstream apoptotic pathways and MPTP opening-related regulatory factors. RESULTS: The cell survival rate of each Gypenosides pretreatment group was significantly higher than that of the hypoxia-reoxygenation group, indicating that Gypenosides could inhibit cell apoptosis and the decrease of mitochondrial membrane potential of hypoxia-reoxygenation cells. The expressions Cytochrome C, APAFl, Caspase-9, and Caspase-3 proteins were significantly lower than those of the hypoxia-reoxygenation group, the expression of Bax was significantly lower than those of the hypoxia-reoxygenation group, while the expression of Bcl2 was significantly higher than those of the hypoxia-reoxygenation group. CONCLUSION: Gypenosides can effectively reduce myocardial ischemia-reperfusion injury in rats. By regulating Bax and Bcl2, Gypenosides can inhibit MPTP opening and the activation of downstream apoptotic pathways, thereby reducing myocardial ischemia-reperfusion injury.

Phosphorus (P) is an essential macronutrient for plants, but also a limited resource worldwide. Strict regulations for fertilizer applications in the European Union are a consequence of the negative environmental effects in case of improper use. Maize is typically grown with the application of P starter fertilizer, which, however, might be reduced or even omitted if suitable varieties were available. This study was performed with the 20 commercially most important maize hybrids in Germany evaluated in multi-location field trials with the aim to investigate the potential to breed for high-performing maize hybrids under reduced P starter fertilizer. At the core location, three starter fertilizers with either phosphate (triple superphosphate, TSP), ammonium nitrate (calcium ammonium nitrate, CAN), or a combination of ammonium and phosphate (diammonium phosphate, DAP) were evaluated relative to a control and traits from youth development to grain yield were assessed. Significant differences were mainly observed for the DAP starter fertilizer, which was also reflected in a yield increase of on average +0.67 t/ha (+5.34%) compared to the control. Correlations among the investigated traits varied with starter fertilizer, but the general trends remained. As expected, grain yield was negatively correlated with grain P concentration, likely due to a dilution effect. Importantly, the genotype-by-starter fertilizer interaction was always non-significant in the multi-location analysis. This indicates that best performing genotypes can be identified irrespective of the starter fertilizer. Taken together, our results provide valuable insights regarding the potential to reduce starter fertilizers in maize cultivation as well as for breeding maize for P efficiency under well-supplied conditions.

Antibiotic resistance poses severe health threats throughout the world. Exploring new antibiotics is widely recognized as an effective strategy to counter antibiotic resistance, but new antibiotics will eventually lead to further antibiotic resistance when new drugs are misused or overused. An alternative tactic may be antibacterial regulation on demand. Here, we show experimentally and theoretically that unstable black phosphorus nanosheets (BPNs) can function as antibacterial agents without causing antibiotic resistance. This antibacterial strategy relies on an unprecedented synergism: The BPNs use reactive oxygen species, are not toxic towards nonbacterial cells within a wide range of BPN concentration (0.01-2.0 mg mL-1 ), and are chemically degradable on demand. BPNs thus offer a promising approach to fighting bacterial infections without causing antibiotic resistance. We believe this proposed strategy offers new insights into instability-guided antibacterial therapy in clinical applications and indicates a new direction for fighting antibiotic resistance.

Liver-targeted drug delivery improves the efficacy of anti-liver cancer agents and reduces systemic toxicity by limiting the bioavailability of these drugs to within tumors. Liver targeting reagents with galactose residues, which selectively combine to asialoglyco protein receptors, have previously been used to improve liposome-encapsulated drug accumulation within liver cells. They lead to a reduction in liver cancer cell growth and have been used to cure certain hepatic diseases. In the present study, curcumol, which is the primary active component of Chinese traditional medicine Rhizoma zedoariae, was encapsulated in galactosylated-liposomes to enhance its anti-liver cancer efficacy. Galactosylated-liposomes and normal liposomes were labeled with propidium iodide. Galactosylated-liposomes with increasing concentrations of galactosylated-stearate (Gal-s) had a notably increased level of uptake in HepG2 cells (hepatoblastoma) compared with SGC-7901 (gastric cancer) and A549 (non-small cell lung cancer) cells. When the percentage of Gal-s reached 20%, liposome uptake plateaued. In the in vitro anti-liver cancer experiment, the anti-liver cancer efficacy of galactosylated-curcumol-liposomes increased significantly more compared with normal curcumol liposomes and free curcumol as indicated by cell survival rate and lactate dehydrogenase release rate. Collectively, these results demonstrate that galactosylated-liposomes are able to enhance the in vitro liver-targeting effect and anti-liver cancer efficacy of curcumol.

Alkylated PAHs (APAHs) have been shown to be more toxic and persistent than their non-alkylated parent compounds. However, little is known about the extent of soil contamination by these pollutants. To help understand agricultural soil pollution by these compounds at a regional scale, a total of 18 methylated PAHs (MPAHs, a major class of APAHs) in 243 soil samples were analyzed. These soil samples were collected from 11 sites in the Yangtze River Delta (YRD) region, a representative fast developing area in China. The total concentration of MPAHs (∑18MPAHs) ranged from 5.5 to 696.2 ng/g dry soil, with methylnaphthalenes (M-NAPs) and methylphenanthrenes (M-PHEs) accounting for more than 70% of the compositional profile. Relatively high concentrations of ∑18MPAHs were found in Jiaxing and Huzhou areas of Zhejiang province, as well as on the border between the cities of Wuxi and Suzhou. Different MPAH groups showed dissimilar spatial distribution patterns. The spatial distribution of lower molecular weight MPAHs was related to agricultural straw burning and emissions/depositions from industrial activities, whereas that of higher molecular weight MPAHs was much more a function of the total organic carbon (TOC) content of soil. Although coal, biomass (crop straw and wood), and petroleum combustion were identified to be the major emission sources for most of the sampling sites, the areas with relatively severe pollution with ∑18MPAHs resulted from the localized hotspots of petroleum leakage. Isomeric MPAHs with methyl group substituted at 2- (ß) position exhibited significantly higher concentrations than those substituted at 1- (α) position. Results of this work help to understand soil pollution by MPAHs, and are useful for designing effective strategies for pollution control so as to ensure food safety in areas with fast economic growth.

Root system architecture (RSA) plays an important role in phosphorus (P) acquisition, but enhancing P use efficiency (PUE) in maize via genetic manipulation of RSA has not yet been reported. Here, using a maize recombinant inbred line (RIL) population, we investigated the genetic relationships between PUE and RSA, and developed P-efficient lines by selection of quantitative trait loci (QTLs) that coincide for both traits. In low-P (LP) fields, P uptake efficiency (PupE) was more closely correlated with PUE (r = 0.48-0.54), and RSA in hydroponics was significantly related to PupE (r = 0.25-0.30) but not to P utilization efficiency (PutE). QTL analysis detected a chromosome region where two QTLs for PUE, three for PupE and three for RSA were assigned into two QTL clusters, Cl-bin3.04a and Cl-bin3.04b. These QTLs had favorable effects from alleles derived from the large-rooted and high-PupE parent. Marker-assisted selection (MAS) identified nine advanced backcross-derived lines carrying Cl-bin3.04a or Cl-bin3.04b that displayed mean increases of 22%-26% in PUE in LP fields. Furthermore, a line L224 pyramiding Cl-bin3.04a and Cl-bin3.04b showed enhanced PupE, relying mainly on changes in root morphology, rather than root physiology, under both hydroponic and field conditions. These results highlight the physiological and genetic contributions of RSA to maize PupE, and provide a successful study case of developing P-efficient crops through QTL-based selection.

Heat-labile enterotoxin (LT) of enterotoxigenic Escherichia coli (ETEC) is one of the major virulence factors for causing diarrhea in piglets, and LT is a strong immunogen. Thus, LT represents an important target for development of vaccines and diagnostic tests. In this study, bioinformatic tools were used to predict six antigenic B cell epitopes in the B subunit of LT protein (LTB) of ETEC strains. Then, seven antigenic B cell epitopes of LTB were identified by polyclonal antisera (polyclonal antibody (PAb)) using a set of LTB-derived peptides expressed as maltose-binding protein (MBP) fusion protein. In addition, one LTB-specific monoclonal antibody (MAb) was generated and defined its corresponding epitope as mentioned above. This MAb was able to specifically bind with native LT toxin and has no cross-reaction with LT-II (type II heat-labile enterotoxin), Stx1 (Shiga toxin I), Stx2 (Shiga toxin II), STa (heat-stable enterotoxin I), and STb (heat-stable enterotoxin II) toxins. Further, this MAb was able to interrupt LT toxin specific binding to GM1 receptor, indicating that the corresponding epitope is the specific binding region to GM1 receptor. Moreover, in vitro and in vivo assay showed that the MAb was able to neutralize the native LT toxin. Diarrheal suckling pigs challenged with LT-positive ETEC strain recovered when an enema with this purified MAb was administered. This study will provide the foundation for further studies about the interaction between LT toxin and GM1 receptor and about the developing of epitope-based vaccines and specific therapeutic agent.

Folic acid and methylenetetrahydrofolate reductase (MTHFR) may affect the development of human cancer. However, few studies have evaluated folate intake and MTHFR in susceptibility to and prognosis of patients with ovarian cancer. We conducted a prospective case-control study in 215 ovarian cancer patients and 218 controls (all Chinese) between Jan. 2004 and Jan. 2007. MTHFR C677T genotyping was done by PCR-RFLP. All patients were followed up until Dec. 2010. We found a 2.43-fold increased risk of ovarian cancer among MTHFR 677TT carriers, and a decreased risk of ovarian cancer in individuals with high folate intake (OR = 0.54, 95% CI = 0.32-0.94). Cox regression survival analysis showed that among the ovarian cancer patients, those carrying the 677TT genotype had a higher risk of death (HR = 2.17, 95% CI = 1.20-4.79), while high folate intake was associated with a lower risk of death (HR = 0.43, 95% CI = 0.33-0.88). Moreover, MTHFR 677CC carriers with higher folate intake showed a lower risk of death from ovarian cancer (HR = 0.32, 95% CI = 0.27-0.82). In summary, high folate intake may lessen susceptibility and improve the prognosis of ovarian cancer patients, while the MTHFR 677TT genotype appears to increase ovarian cancer risk and worsen its prognosis in a Chinese population.

Epistatic interactions may contribute substantially to the hybrid performance of sugar beet. The main goal of our study was to dissect the genetic basis of eight important physiological and agronomic traits using two different biometrical models for joint linkage association mapping. A total of 197 genotypes of an elite breeding population were evaluated in multi-location trials and fingerprinted with 194 SNP markers. Two different statistical models were used for the genome-wide scan for marker-trait associations: Model A, which corrects for the genetic background with markers as cofactors and Model B, which additionally models a population effect. Based on the extent of linkage disequilibrium in the parental population, we estimated that for a genome-wide scan at least 100 equally spaced markers are necessary. We mapped across the eight traits 39 QTL for Model A and 22 for Model B. Only 11% of the total number of QTL were identified based on Models A and B, which indicates that both models are complementary. Epistasis was detected only for two out of the eight traits, and contributed only to a minor extent to the genotypic variance. This low relevance of epistasis implies that in sugar beet breeding the prediction of performance of three-way hybrids is feasible with high accuracy based on the means of their single crosses.

Seven gaseous polycyclic aromatic hydrocarbons (PAHs) were determined in the indoor air of 38 folk houses in Beijing during the heating and non-heating seasons, by the passive air samplers. The total average concentration of the gaseous PAHs was around 100 ng/m3, and the dominant components were those with 2 or 3 rings. The inhalation exposure of PAHs by the residents was calculated based on the concentrations of PAHs acquired, average house-staying time and respiratory rates at different ages. The calculated results indicated that, the potential total exposure rates of the seven gaseous PAHs for the adults (including senior people) during the heating and non-heating periods were 66 ng/h and 58 ng/h, respectively; while those for juvenile people were 56 ng/h and 50 ng/h, respectively.

OBJECTIVE: To screen, clone and identify the cDNA fragments of human endometrial carcinoma-related genes,and explore the molecular mechanism of endometrial carcinogenesis. METHODS: Pure endometrial glandular epithelial cells and endometrial carcinoma cells were obtained by laser capture microdissection (LCM). RNA from these cells was isolated, and differentially expressed gene fragments that were specialy relevant to endometrial carcingenesis were identified by using fluorescence differential display reverse transcription polymerase chain reaction (FDD-PCR). The selected fragments were cloned, sequenced and verified by reverse Northern blot analysis, and positive fragments were BLAST analysed and compared with those in Genbank. RESULTS: 38 differential fragments were isolated, 3 of which were expressed more abundantly in normal endometrium and 35 were highly expressed in endometrial carcinoma. 10 fragments were recoverd, cloned and sequenced, confirmed by reverse Northern blot analysis, among which 6 fragments were positive. BLAST analysis showed that T1.1 was homologous to cyclin-dependent protein kinase 7 (CDK7, 99%); L1.9 was homologous to protein phosphatase 1 regulatory (inhibitor) subunit 12A (PPP1R12A, 99%); L1.21 and L1.22 were homologous to cellular repressor of E1A-stimulated genes 1 (CREG, 100%); L1.25 and L1.26 were homologous to solute carrier family 39 (zinc transporter) member 10 (SLC39A10, >98%). CONCLUSION: Gene fragments related to endometrial carcinoma have been obtained by applying LCM and FDD-PCR. To our knowledge it is the first time that the correlation between CDK7, PPP1R12A, CREG, SLC39A10 and endometrial carcinoma is discovered at mRNA level, and their role in molecular mechanism of cancinogenesis is discussed. CDK7, CREG, SLC39A10 as new candidate oncogene and PPP1R12A as new candidate anti-oncogene are worthy of being further investigated in the future.
