RESUMO

Hyperbaric oxygen (HBO) therapy is of clinical utility in patients with transient cerebral ischemia. The investigatory study was to identify the potential regulatory mechanism of HBO treatment on neuronal injury and neurological function recovery in rats with intracerebral hemorrhage (ICH). Firstly, the rat model of ICH was established by collagenase, and the experimental rats were treated with HBO at 2.5 absolute atmospheres for 60 min each time. Next, lentivirus interfering with microRNA (miR)-204-5p or chloride channel protein 3 (CLCN3) expression was injected via the tail vein. Afterward, neurological function assessment was conducted, serum S100ß and NSE contents were detected by enzymer-linked immunosorbent assay, and pathological conditions of brain tissue were observed by hematoxylin-eosin staining. Terminal deoxynucleotidyl transferase mediated dUTP nick end labeling staining was used to detect neuronal apoptosis. The results showed that HBO alleviated neuronal injury and neurological function recovery in ICH rats and reduced serum S100ß and NSE content (all P<0.05). At the same time, overexpressing miR-204-5p or depleting CLCN3 further promoted the therapeutic effect of HBO on ICH rats (all P<0.05), while silencing miR-204-5p or elevating CLCN3 did oppositely (all P<0.05). In conclusion, HBO alleviates neuronal injury and neurological function recovery in ICH rats by silencing miR-204-5p-targeted CLCN3.

The passion fruit peel (PFP) is the by-product of juice processing and is rich in phenolic compounds and dietary fibers. As the high ADF content in PFP (34.20%), we proceeded to treat PFP with cellulase. The ADF decreased to 16.70% after enzymatic processing, and we supposed that enzymolytic passion fruit peel (EPF) should have a greater growth performance than PFP to broilers. Two trials were conducted to evaluate the effects of dietary PFP or EPF supplementation on growth performance, serum biochemical indices, meat quality, and cecal short-chain fatty acids, microbiota, and metabolites in broilers. In Exp. 1, 180 1-day-old Sanhuang broilers (male, 36.17 ± 2.47 g) were randomly allocated into 3 treatments, with 6 replicates in each treatment. The 3 experimental diets included 1 basal diet (control) and 2 PFP-added diets supplemented with 1 and 2% PFP, respectively. The trial lasted for 42 d. In Exp. 2, 144 Sanhuang broilers (male, 112-day-old, 1.62 ± 0.21 kg) were randomly allocated to 3 treatments. Each treatment was distributed among 6 pens, and each pen contained 8 broilers. The 3 treatment diets included: a control diet, a positive control diet supplementing 75 mg/kg chlortetracycline, and the experimental diet supplementing 3% EPF. The trial lasted for 56 d. Results showed that dietary 1 and 2% PFP addition did not affect growth performance in Exp. 1, and the 3% EPF supplementation had a negative effect on ADFI (P < 0.05) in Exp. 2. A decreased serum triglyceride (P < 0.05) in broilers was observed in Exp. 1. Broilers fed EPF had a higher glutathione peroxidase (GSH-Px) (P < 0.05), and lower levels of tumor necrosis factor-α (TNF-α) (P < 0.05) and glucose (P < 0.05) in Exp. 2. We also found that broilers from PFP or EPF-treated treatments had an increased butyrate content and higher microbial diversity in the cecum. The effects of antioxidation, anti-inflammatory function, and elevated SCFAs were confirmed after the microbe and untargeted metabolomic analysis. Dietary EPF supplementation significantly increased the SCFA-generating bacteria, anti-inflammatory-related bacteria, the antioxidant-related and anti-inflammatory-related metabolites. Moreover, dietary 3% EPF addition positively affects the biosynthesis of phenylpropanoids, which strongly correlate with the antioxidant and anti-inflammatory properties. In conclusion, the proper addition level did not affect the growth performance, and the PFP and EPF could improve the antioxidation state, anti-inflammatory activity, and intestinal functions of Sanhuang broilers to some extent.

This study was conducted to investigate the effects of citric acid (CA) supplementation on growth performance, intestinal morphology, intestinal microbiota, and blood parameters of geese from 1 to 28 d of age and evaluate the optimum additional level of citric acid. A total of 180 one-day-old male goslings were randomly allotted to 5 treatment groups of 36 birds with 6 replications. The control group was fed a basal diet, and the other groups were fed the basal diet supplemented with 0.25, 0.50, 1.00, and 2.00% of citric acid, respectively. The results showed that goslings fed the diet supplemented with 1.00% CA had higher final body weight (FBW) and average daily gain (ADG) than other groups (P < 0.05). The CA supplementation at 0.25 to 1.00% improved the morphology of duodenum or jejunum (P < 0.05). The jejunal content pH value was significantly reduced with the addition of CA compared with the control group (P < 0.05). As citric acid levels increased, the IgA concentrations in plasma increased and then decreased, and the goslings fed 1.00% CA supplementation had the highest IgA concentrations (P < 0.05). The supplementation of 1.00% and 2.00% CA in diet significantly reduced the malondialdehyde (MDA) concentration in plasma (P < 0.05). No significant difference was found on some indices related to liver function in plasma (P > 0.05), while creatinine significantly increased by the 2.00% CA supplementation (P < 0.05). Besides, the higher Coliform level in cecal content and worse intestinal morphology were observed when CA supplementation was up to 2.00%. Hence, the dietary CA supplementation (especially 1.00%) in goslings improves the growth performance, intestinal morphology, immunity and antioxidant, while excessive CA addition may cause negative effects. According to the quadratic polynomial model, the addition of CA in diet for obtaining maximum average daily feed intake (ADFI) should be 1.09% (10.9 g/kg diet) for goslings from 1 to 28 d of age.

Objective: To investigate the safety and efficacy of the Weitan Waifu patch on the postsurgical gastroparesis syndrome (PGS) of gastrointestinal cancer. Methods: The multi-center, double-blind, randomized controlled trial was conducted with superiority design. Patients with PGS of gastrointestinal cancer diagnosed in 4 AAA hospitals and the abdominal symptom manifested as cold syndrome by Chinese local syndrome differentiation were recruited. These patients were randomly divided into two groups according to 1∶1 proportion. Placebo or Weitan Waifu patch was applied in control group or intervention group, respectively, based on the basic treatments, including nutrition support, gastrointestinal decompression, promoting gastric dynamics medicine.Two acupuncture points (Zhongwan and Shenque) were stuck with placebo in control group or patch in treatment group. The intervention course was 14 days or reached the effective standard. Results: From July 15, 2013 to Jun 3, 2015, 128 participants were recruited and 120 eligible cases were included in the full analysis set (FAS), and 60 cases in each group. 88 cases were included in the per-protocol set (PPS), including 45 cases in the treatment group and 43 cases in the control group. In the FAS, the clinical effective rate in the treatment group was 68.3%, significantly superior than 41.7% of the control group (P=0.003). The medium time of effective therapy in the treatment group was 8 days, significantly shorter than 10 days in the control group (P=0.017). In the FAS, 3 adverse events occurred in the treatment group, including mild to moderate decrustation, pruritus and nausea. The incidence rate of adverse events was 5.0% (3/60) and these symptoms were spontaneously remitted after drug withdrawal. No severe adverse events were observed in the control group. There was no significant difference between these two groups (P=0.244). Conclusion: Weitan Waifu patch is a safely and effectively therapeutic method for patients with PGS (cold syndrome) of gastroenterological cancer. Trial registration: International Standard Randomized Controlled Trial Number Register, ISRCTN18291857.

The insecticidal and repellent activities of the essential oil extracted from Zingiber purpureum Roscoe rhizomes were evaluated against Tribolium castaneum (Herbst) and Lasioderma serricorne (L.) adults. During our screening program for agrochemicals from Chinese medicinal herbs and wild plants, the essential oil of Z. purpureum rhizomes was found to possess strong contact toxicity against T. castaneum and L. serricorne adults, with LD50 values of 39.0 and 16.3 µg per adult, respectively, and also showed strong fumigant toxicity against the two grain storage insects with LC50 values of 13.6 and 9.3 mg/liter of air, respectively. The essential oil obtained by hydrodistillation was investigated by gas chromatography-mass spectrometry. The main components of the essential oil were identified to be sabinene (48.1%), terpinen-4-ol (25.1%), and γ-terpinene (6.7%), followed by α-terpinene (4.3%), ß-thujene (3.4%), and α-phellandrene (2.7%). Sabinene, terpinen-4-ol, and γ-terpinene were separated and purified by silica gel column chromatography and preparative thin-layer chromatography. Terpinen-4-ol showed the strongest contact toxicity against T. castaneum and L. serricorne (LD50=19.7 and 5.4 µg per adult, respectively) and also the strongest fumigant toxicity against T. castaneum and L. serricorne (LC50=3.7 and 1.3 mg/liter of air, respectively). Otherwise, sabinene and terpinen-4-ol were strongly repellent against T. castaneum as well as the essential oil, while γ-terpinene exhibited weaker repellency against T. castaneum compared with the positive control, DEET (N, N-diethyl-3-methylbenzamide). Moreover, only the essential oil exhibited strong repellency against L. serricorne, the three compounds exhibited weaker repellency against L. serricorne relative to DEET.

This study was conducted to investigate the effect of two distinct enzyme preparations on nutrients' digestibility and growth performance of growing pigs fed diets based on corn, soya bean meal and Chinese double-low rapeseed meal (DLRM). The two enzyme preparations were Enzyme R, a preparation extracted from fermentation of a non-GMO fungus Penicillum funiculosum, developed for multi-grain and multi-animal species; and Enzyme P, a xylanase preparation from Trichoderma longibrachiatum, for pigs fed corn-based diets only. Both enzymes were tested at 0, 0.25 and 0.50 g/kg feed using 70 crossbred male pigs (Large Yorkshire x Landrace) in five dietary treatments and seven replicates in each treatment, for growth period from 27 to 68 kg live weight in 49 days. Results showed that the supplementation of both enzymes (1) increased total-tract digestibility of dietary energy from 77.5% (control) to 81.4% (Enzyme R, p < 0.05) and 81.9% (Enzyme P, p < 0.05); of neutral detergent fibre from 41.0% (control) to 57.8% (Enzyme R, p < 0.05) and 60.0% (Enzyme P, p < 0.05); (2) improved average daily gain from 786 g (control) to 829 g (Enzyme R, p < 0.05) and 846 g (Enzyme P, p < 0.05); and numerical increases in feed intake from 1.96 kg/pig/day (control) to 2.01 (Enzyme R) and 2.00 (p > 0.05) and feed conversion ratio from 2.50 (control) to 2.42 (Enzyme R) and 2.36 (Enzyme P, p < 0.05); (3) there was a dose response but no significant differences were observed in enzyme efficacy between the two enzyme preparations. The present study demonstrated beneficial effects of applying xylanase-based enzymes to improve feeding values of pig diets based on corn, soya bean meal and DLRM.

Two taraxastane triterpenoids, i.e. taraxast-20-ene-3beta,30-diol (1) and 20alpha,21alpha-epoxy-taraxastane-3beta,22alpha-diol (2), as well as four known triterpenes taraxast-20(30)ene-3beta,21alpha-diol (3), taraxast-20(30)-ene-3beta-ol (4), taraxast-20-ene-3beta-ol (5) and taraxastane-3beta,20alpha-diol (6) were isolated from the Chinese medicinal plant Saussurea petrovii. Their structures were elucidated by spectroscopic methods. These compounds, especially 1 and 2, exhibit significant antitumor and antibacterial activity.

AIM: To study whether Cladonia furcata polysaccharide (CFP-1) might induce apoptosis in human leukemia K562 cells. METHODS: Inhibition of proliferation was measured by MTT assay. Morphological assessment of apoptosis was performed with fluorescence microscope and electron microscope. DNA fragmentation was visualized by agarose gel electrophoresis. The amount of apoptosis cells was measured by flow cytometry. RESULTS: CFP-1 (50 - 800 mg/L) inhibited K562 cell proliferation in a concentration-dependent manner. After incubation of K562 cells with CFP-1 300 mg/L for 5 d, morphological changes of typical apoptosis were observed and agarose gel electrophoresis of DNA revealed "ladder" pattern. CONCLUSION: CFP-1 induced apoptosis in human leukemia K562 cells.

Using hypothalamus as material and lambda gt10 as vector, chicken hypothalamus cDNA library has been constructed. The results showed that the titration of the cDNA library we constructed was 3.8 x 10(6), and the fragment length of inserted DNA ranged mainly from 1.0-3.0 kb. The ESTs was sequenced and primarily analyzed for ten clones randomly selected from above library. After analysis with BLASTn and FASTA3.1, it was found that four ESTs developed matched homologous sequence in chicken, four also had similar sequences in human and other species, have other two sequences were function-unknown gene. The sequences of ten ESTs we studied have been loaded into GenBank.

Antioxidative effects of the main polyphenolic components extracted from green tea leaves, i.e. (-)-epicatechin (EC), (-)-epigallocatechin (EGC), (-)-epicatechin gallate (ECG), (-)-epigallocatechin gallate (EGCG) and gallic acid (GA), against free radical initiated peroxidation of human low density lipoprotein (LDL) were studied. The peroxidation was initiated either thermally by a water-soluble initiator 2,2'-azobis(2-amidinopropane hydrochloride) (AAPH), or photochemically by a triplet sensitizer benzophenone (BP). The reaction kinetics was monitored by the uptake of oxygen and the depletion of alpha-tocopherol (TOH) presented in the native LDL. Kinetic analysis of the antioxidation process demonstrates that these green tea polyphenols are effective antioxidants against both AAPH-initiated and BP-photosensitized LDL peroxidation. The antioxidative action of the green tea polyphenols includes trapping the initiating and/or propagating peroxyl radicals with the activity sequence EC>EGCG>ECG>EGC>GA for the AAPH initiated peroxidation, and reducing the alpha-tocopheroxyl radical to regenerate alpha-tocopherol with the activity sequence of ECG>EC>EGCG>EGC>GA and ECG>EGCG>GA>EC>EGC for the AAPH-initiated and BP-photosensitized peroxidations respectively.

A supercritical fluid extraction (SFE) method was developed for the extraction of paeonol in Paeonia suffruticosa Andr. and its patent preparations. A systematic method was used to optimize five variables (temperature, pressure, modifier concentration, static extracting time and CO2 dynamic extracting volume) of SFE. Sample extracts were analyzed by wide-bore capillary gas chromatography (CGC). The method showed good linearity (gamma = 0.9999) in the range of 0.1-1.6 mg.ml-1 of paeonol. The average recovery was 97.8% with RSD = 2.35% (n = 3) for Paeonia suffruticosa andr., 100.3% with RSD = 1.89% (n = 3) for its patent preparations.

OBJECTIVE: To explore the effect of Xuanfa Moyuan principle ([symbol: see text], XFMYP) on arthritic mice. METHODS: The type II collagen induced arthritis in DBA mice was taken as Bizheng ([symbol: see text]) animal model. The Dayuanyin ([symbol: see text], DYY) was taken as the representative prescription for XFMYP. The arthritis incidence and arthritis index were tested by scoring system. The anti-C II antibody IgG and its subsets, IgM rheumatoid factor, interferon gamma and interleukin 10 produced by lymph node cells were tested by ELISA method. RESULTS: XFMYP could delay the onset day of arthritis, decrease the arthritic incidence and index, decrease anti-II IgG2a, IgG2b, IgG1 level and the IgG2a + IgG2b/IgG1 ratio in serum, decrease the production of interferon gamma by lymphocytes and increase the production of interleukin 10. CONCLUSIONS: XFMYP could inhibit the anti-C II immune response, and balance the anti-C II Th1 and Th2 type immune responses.

The whole plants of Ajuga lupulina afforded five compounds, including three new clerodane diterpenes, lupulins A-C (1-3), whose structures were elucidated by spectral methods. Among these compounds, lupulins A (1) and B (2) as well as the acid hydrolysate (5) of lupulin D (4) showed antibacterial activities against Staphylococcus aureus, Pseudomonas aeruginosa, and Escherichia coli.

RESUMO

Two new flavones, 4',6,7-trihydroxy-3',5'-dimethoxy-flavone (2) and 5',5- dihydroxy-3',4',8-trimethoxyflavone (3) were isolated from Artemisia giraldii and their structures were identified by spectroscopic methods. These two new flavones showed antibiotic activity against Staphylococcus aureus, Sarcina lutea, Escherichia coli, Pseudomonas aeruginosa, Proteus sp. Aspergillus flavus, and Trichoderma viride.

An HPLC method using unmodified silica with reversed-phase eluents was developed for the determination of DL-tetrahydropalmatine in Corydalis Yanhusuo W. T. Wang and its set prescription preparations. The mobile phase was a mixture of methanol and 2 mmol/L phosphate buffer (75:25, v/v). The result was calculated by external standardization. The method is simple, rapid and precise, and could also be used for the separation and determination of alkaloids in other crude drugs and preparations.

Human blood samples were investigated in vitro to observe the antithrombotic effect of water extract of Gynostemma pentaphyllum (GP). The results showed that GP could inhibit significantly the platelet aggregation induced by ADP and compound agonists (P < 0.05), accelerate obviously the disaggregation (P < 0.05) and inhibit effectively the experimental thrombosis (P < 0.05). The delayed effects of GP on KPTT, PT, TT, AT, RVV-RT, RVV-CT suggested that this drug could decrease the activity of multiple coagulation factors. And it showed that GP could accelerate the erythrocyte electrophoresis rate. This study revealed that GP is an antithrombotic agent affecting the links of thrombotic chain which is worthwhile to be studied further.
