We examined the effects of a supplement of plant polyphenols extracts of green tea, capsicum, and fenugreek, and electrolytes ([Na+, K+]; AXT, Axion ThermoPlus, CCPA, France] during summer heat load on production, welfare, and oxidative stress proteins in adipose tissue (AT) of dairy cows. A total of 42 multiparous mid-lactation cows were divided into 3 groups during summer, and were fed for 2 wk either a standard milking cow diet (CTL, n = 14) or diets supplemented with 100 g/d of AXT (100AXT, n = 14), or 150 g/d of AXT (150AXT, n = 14), while being cooled 5 times a day. Then, half of the cows from each dietary treatment were cooled (CL) or not cooled (NCL) for 2 wk, after which the cooled and uncooled groups were switched for additional 2 wk. Cows were milked 3 times a day, and milk composition was analyzed at the end of each 2-wk period. Vaginal temperature (VT) was measured for 3 consecutive days in each period. Biopsies of subcutaneous AT were taken from 10 NCL cows (5 each of CTL and 150AXT) at the end of the period and examined by liquid chromatography-tandem mass spectrometry proteomics analysis. Data were analyzed with PROC MIXED of SAS (version 9.2, SAS Institute Inc.). The model included the effects of dietary treatment, cooling regimen, period, and their interactions. Protein and mRNA abundances and proteomic data (P ≤ 0.05 and fold change [FC] ± 1.5) were analyzed by t-test. Milk yields and 4% fat-corrected milk (FCM) were higher in 100AXT than in CTL; milk components were not different. Dry matter intake (DMI) was higher in 100AXT than in CTL. The effect of cooling and the interactions of period × cooling were significant for DMI, 4% FCM, energy-corrected milk, and milk/DMI. The proportion of time that VT was >39°C was lower in 100AXT and in 150AXT than in CTL. Daily rumination time was greater in 150AXT than in CTL, and lying time was greater in 100AXT and 150AXT than in CTL. Proteomics of AT demonstrated that 150AXT had increased abundances of peroxidasin (FC = 1.6), microsomal glutathione S-transferase 2 (FC = 2.5), and heme oxygenase 1 (FC = 3.6) compared with CTL. Top enriched canonical pathways included acute phase response signaling, Nrf2-mediated oxidative stress response, and lipopolysaccharide (LPS)/IL-1-mediated inhibition of RXR function. Immunoblots of AT showed a higher abundance of the transient receptor potential vanilloid 1 and of LPS binding protein in AT of 150AXT compared with CTL. Supplementation of AXT increased DMI, milk, and 4% FCM, lowered VT, improved welfare indices, and enriched the AT with Nrf2-oxidative stress response and acute phase response proteins in heat-stressed dairy cows.

Pregnancy toxemia is the most frequent metabolic disorder of ewes in late pregnancy. Although propylene glycol (PG) and glycerol (GLY) are common glucogenic supplements for treating pregnancy toxemia in ewes, the relative benefit of these 2 supplements is not entirely clear. Therefore, the objectives of the present study were to determine the changes during 24 h in key blood metabolites and insulin in response to PG or GLY drenching in prolific ewes. To this end, 36 multiparous late-pregnant Afec-Assaf ewes (∼132.4 d pregnant) bearing 2 to 4 fetuses, divided into 2 blocks (18 ewes in each block), with a blood ß-hydroxybutyrate (BHB) concentration of 0.5 to 1.6 mmol/L were included. Ewes were divided into 3 groups (12 ewes each; 6 ewes in each experimental day), according to their BHB levels, expected litter size, body weight, and body condition score, and were drenched with the following: (1) control group (CTL), 55 mL of water; (2) PG, 106 mL of PG (100% PG, 448 calories); or (3) GLY, 108 mL of Koforin 80 (80% GL; 448 calories). Blood samples were taken before drenching and every hour after drenching for 24 h. Plasma concentration of glucose, BHB, nonesterified fatty acids, lactate, glycerol, and insulin were determined. Because there were no effects of treatments after 12 h in the first block, the data were analyzed for 12 h after drenching rather than 24 h. The plasma glucose concentration during the first 5 h after drenching was the highest in the GLY, BHB concentration was the lowest in the PG, and the nonesterified fatty acid levels were lower in the PG compared with the CTL ewes during the first 5 h after drenching. However, glucose concentration was higher in the PG ewes at 9, 11, and 12 h after drenching than in CTL or GLY ewes. The mean lactate concentration in plasma for 12 h was 2.5- and 1.9-fold higher in the PG compared with the CTL and GLY ewes, respectively, and except at 11 h after drenching, it was significantly higher at each time point. The insulin concentration was higher in the GLY than in both other groups at 2 to 5 h after drenching. These results suggest that during the first few hours after drenching the effect of PG was more effective in reducing the BHB concentration, whereas the GLY effect was more effective in enhancing glucose concentration. The increased concentration in lactate following PG treatment suggests that the PG contribution to gluconeogenesis is mediated through its metabolism to lactate. In contrast, the lack of an effect on lactate, and the faster increase in blood glucose in response to GLY suggest that GLY has a more advanced entry point to gluconeogenesis, which influences the immediate response in enhancing the glucose blood concentration.

The two most popular rumen-protected fatty acid supplements in dairy cow rations are calcium salts of palm oil fatty acid calcium salts of palm oil fatty acid (CSFA) and prilled saturated fatty acids (SFAs). The objectives of this study were to determine the effects of supplementing SFA in the form of triglycerides (TSFA), as compared to CSFA, on yields, efficiency and diet digestibility in high-yielding dairy cows. Twenty-eight (14 cows in each group) multiparous cows were fed a basal diet supplemented (on DM basis) with either 12 g/kg TSFA (~350 g/cow per day - contained 980 g/kg fat; 882.3 g/kg SFAs) or 14 g/kg CSFA (~440 g/cow per day - contained 800 g/kg fat; 566.4 g/kg SFAs). The supplement amounts in the diet were balanced according to fat content. Rumen samples were taken for measurements of ammonia and volatile fatty acids concentrations, and fecal samples were taken for digestibility measurements. The CSFA cows produced 3% higher milk yields (47.6 v. 46.2 kg/day; P < 0.0001) and 4.7% higher 4% fat-corrected milk (FCM; 44.7 v. 42.7 kg/day; P = 0.02) than the TSFA cows. No difference in milk-fat content was observed, but milk-protein content was higher in the TSFA than CSFA cows. Yields of fat and protein were similar, but lactose yields were higher in TSFA cows. There were no differences in dry matter intake or efficiency calculations between groups. The ruminal ammonia concentrations were similar between groups, whereas acetate concentrations and acetate : propionate ratio were greater for CSFA than TSFA cows. The apparent total-tract digestibility of dry (P < 0.0007) and organic matters (P < 0.0003), fat (P < 0.0001), NDF and ADF (P = 0.02) were lower in the TSFA v. CSFA cows. In conclusion, the CSFA-supplemented cows produced 3% higher milk and 4.7% higher 4% FCM than the TSFA cows. However, TSFA supplementation did not depress milk-protein content. The apparent total-tract digestibility was lower for all dietary components in the TSFA cows, which was probably due to the effects of both degree of saturation and triglyceride form of the TSFA supplement. Considering that diets were balanced according to the fat content of the supplements, the lower yields of milk and FCM observed in the TSFA than CSFA cows were likely due to the lower digestibility of the fat and other nutrients in the TSFA cows, which might have negatively influenced the dietary energy content.

Reducing milk production during early lactation might be of interest to improve the energy balance (EB) of high-yielding dairy cows. Therefore, the objective of this study was to determine how reducing the milking frequency (MF) of high-yielding dairy cows from thrice to twice a day during the first 30 d in milk (DIM) affects yields, intake, efficiency, metabolic status, and carryover effects. To this end, 42 multiparous cows were divided into 2 groups according to their previous lactation performance, parity, and body weight. The control cows were milked 3 times a day (3ML) and the treated cows were milked twice a day (2ML) until 30 DIM and then both groups were milked 3 times a day. Milk samples were taken twice a week from 2 or 3 consecutive milkings until 45 DIM for analysis of milk solids, and both groups were followed until 100 DIM to determine the carryover effects of MF until 30 DIM. Individual dry matter intake (DMI), milk yield, and body weight were recorded daily. Blood samples were taken 3 times weekly from 14 d prepartum until 45 DIM. Milk yield during the first 30 DIM was 8.6% higher (49.3 and 45.4 kg/d, respectively), milk fat percentage was lower (3.96 and 4.27%, respectively), and the yields of all milk solids were higher in the 3ML cows than in the 2ML cows. Dry matter intake and 4% fat-corrected milk were similar between groups. The EB during the first 30 DIM was lower in the 3ML cows than in the 2ML cows, and milk yield, but not 4% fat-corrected milk yield, per unit of DMI was higher in the 3ML cows. No differences were observed between groups from 31 to 100 DIM in milk yield (∼56.3 kg/d for both groups), milk solids yield, DMI, or milk/DMI; however, fat percentage was lower and EB was higher in the 3ML cows. Blood glucose concentrations between 0 and 30 DIM were lower and ß-hydroxybutyrate concentrations were higher in the 3ML cows than in the 2ML cows, but nonesterified fatty acids concentrations were lower, which may be attributed to the lower clearance frequency of nonesterified fatty acids from the blood stream in the 2ML cows. A lower proportion of the 3ML cows (10%) ovulated ≤15 DIM compared with the 2ML cows (40%), with no beneficial effects on preovulatory follicle characteristics. Reducing the MF from thrice to twice a day during the first 30 DIM improved EB and metabolic status, with only minor effects on production.

Mammals can synthesize all of the fatty acids (FA) necessary for proper health and functioning with the exception of FA in the n-3 (omega-3) and n-6 (omega-6) families of polyunsaturated fatty acids (PUFA), which should be supplied in the diet. The PUFA are the predominant type of lipid in dairy cattle diets; however, common feedstuffs are rich in n-6 FA, whereas the supply of n-3 FA in the intensive dairy industry is mainly limited to flaxseed and fish oils. The n-3 FA are involved in many biological systems and processes, and therefore their dietary supplementation is of special interest in dairy cattle. Furthermore, because milk, milk products, and meat are among the most important and widely used components in traditional and modern human diets, enrichment of these food products with n-3 FA is of special importance. The purpose of this review is to provide a comprehensive description of different aspects and outcomes involved in dietary n-3 FA supplementation in dairy cattle. I provide an inclusive review of the effects of n-3 FA on milk and milk solids and the FA profile in milk fat upon feeding a variety of flaxseed products or fish oil. Selective uptake of n-3 FA has been demonstrated in the ovary compartments, as well as in bull sperm and in the unborn calf through the placenta. Incorporation of these unique FA into the reproductive system influences many processes and exerts some positive effects on fertility. In addition, beneficial effects of feeding n-3 FA on the reproductive system of females and males can be achieved with supplementation of α-linolenic acid from flaxseed or from eicosapentaenoic and docosahexaenoic acids from fish oil. This work provides a broad perspective and demonstrates the importance and potential of n-3 FA dietary supplementation in dairy cattle on the animal itself, as well as its secondary effects, which are associated with human nutrition and health.

The objectives of this study were to determine the differential incorporation of various omega-3 (n-3) fatty acids (FAs) supplemented to dairy cows into ovarian compartments and assess the effects on IVF. Forty-two 256-day pregnant cows were supplemented with encapsulated fats, in treatments designated as i) SFA - saturated fat at 240 and 560 g/day per cow, prepartum and post partum (PP) respectively; ii) FLX - flaxseed oil at 300 and 700 g/day per cow prepartum and PP respectively; and iii) FO - fish oil at 300 and 700 g/day per cow prepartum and PP respectively. Commencing at 60 days in lactation, ovum pickup (OPU) was performed twice weekly (20 sessions; five cows per group) and in vitro maturation and IVF were conducted. The proportion of α-linolenic acid (ALA) was greater in follicular fluid (FF), granulosa cells, and cumulus-oocyte complexes (COCs) of FLX cows than in other groups (P<0.001). The proportion of docosahexaenoic acid (DHA) was 6.7 times as great in FF of FO as in other groups (P<0.001); docosapentaenoic acid n-3 and DHA were detected in COCs of FO but not in others. The follicle number during OPU was higher in FLX and FO than in SFA (P<0.05), and the oocyte cleavage rate was higher in FLX and FO than in SFA (P<0.01). Also, the percentage of oocytes that developed to blastocysts tended to be higher in both n-3 groups than in SFA (P<0.1). In conclusion, both dietary n-3 FAs similarly improved folliculogenesis and IVF performance; therefore, ALA-rich botanical n-3 seems to be a satisfactory approach to improve oocyte quality.

During diet-induced milk fat depression (MFD), the short and medium-chain fatty acids (SMCFA), which are synthesized de novo in the mammary gland, are reduced to a much greater extent than the long-chain fatty acids (LCFA) that originate from the circulation. Our hypothesis was that increased availability of SMCFA might rescue conjugated linoleic acid (CLA)-induced MFD in lactating dairy cows. To test that hypothesis, 4 rumen-fistulated lactating Holstein cows (128 ± 23 d in milk) were used in a 4 × 4 Latin square design with 3-wk experimental periods. Treatments were applied during the last 2 wk of each period and included 3× daily abomasal infusion of a total of (1) 230 g/d of LCFA (blend of 59% cocoa butter, 36% olive oil, and 5% palm oil); (2) 420 g/d of butterfat (BF); (3) 230 g/d of LCFA with 27 g/d of CLA (LC-CLA), containing 10 g/d of trans-10,cis-12 CLA; and (4) 420 g/d of butterfat with 27 g/d of CLA (BF-CLA). Butterfat provided 50% of C16 (115 g/d) and similar amounts of C18 FA as found in LCFA, such that the difference between the BF and LCFA treatments was 190 g/d of SMCFA. No treatment effects were observed for DMI or milk yield. Milk fat content was reduced by 41 and 32%, whereas milk fat yield was reduced by 41 and 38% with LC-CLA and BF-CLA, respectively, compared with their respective controls. Abomasal infusion of CLA reduced de novo synthesized fatty acid (DNFA; SMCFA and 50% C16:0) concentration, whereas DNFA tended to be greater with BF infusion. An interaction was observed between SMCFA and CLA as the increased availability of SMCFA reduced stearoyl-CoA-desaturase-1 gene expression, whereas it tended to reduce lipoprotein lipase (LPL), 1-acylglycerol-3-phosphate O-acyltransferase 6 (AGPAT-6), sterol regulatory element-binding protein cleavage-activating protein (SCAP), and peroxisome proliferator-activated receptor γ (PPAR-γ) gene expression in the presence of CLA. The mRNA expression of genes involved in de novo fatty acid synthesis [acetyl-coenzyme A carboxylase α (ACACA) and fatty acid synthase (FASN)], fatty acid uptake (LPL), and triglyceride synthesis [AGPAT-6 and diacylglycerol O-acyltransferase 1 (DGAT-1)] along with protein abundance of the ACC and FASN were reduced with CLA. However, the increased availability of SMCFA had no effect on lipogenic gene expression except for LPL, whose expression was increased with BF infusion. The nutritional manipulation by increasing the intestinal availability of SMCFA was not sufficient to rescue CLA-induced MFD.

The objectives of the present study were to evaluate the transfer efficiency of α-linolenic acid (ALA) from the abomasum into milk fat, its interaction with milk fat content and yield, and the relationship between ALA and C16:0 in milk fat. Three rumen-fistulated multiparous Holstein cows at midlactation were used in a 3×3 Latin square design. Treatments consisted of abomasal infusion of (1) 110 mL of water/d (control), (2) 110 mL of flaxseed oil/d (low flaxseed oil, LFO), and (3) 220 mL of flaxseed oil/d (high flaxseed oil, HFO). Experimental periods were continued for 2 wk and fat supplements were infused abomasally during the last 7 d of each period. Average dry matter intake and milk yield were not affected by oil infusion. Milk fat and lactose content tended to be greater with flaxseed infusion compared with the control. Plasma ALA was 2.9- and 4.0-fold greater with LFO and HFO, respectively. The apparent transfer efficiency of ALA to milk was 44.8 and 45.7% with LFO and HFO, respectively. The C16:0 content in milk fat was decreased by 3.59 and 5.25 percentage units, whereas the ALA content was increased by 1.68 and 3.09 percentage units with LFO and HFO, respectively. Similarly, C18:2n-6 was increased by 0.95 and 1.31 percentage units with LFA and HFO, respectively, without changes in other fatty acids (FA). Total polyunsaturated FA was 4.4 and 2.7% lower in the HFO and LFO, respectively, than in the control. Furthermore, C16:0 content in the milk fat was reduced to a greater extent than the increase in ALA content, as a 1.68 and 3.09 percentage unit increase occurred in ALA compared with a 3.6 and 5.25 percentage unit decrease in C16:0 for LFO and HFO, respectively, such that a negative correlation existed between ALA and C16:0 (r=-0.72). In conclusion, abomasal infusion of flaxseed oil dramatically increased the ALA content in plasma and milk fat. Because the replacement of C16:0 with ALA and C18:2n-6 occurred without changes in other FA presumed to be synthesized de novo in the mammary gland, this suggests that the preformed C16:0 was replaced, rather than being caused, by an overall suppression of de novo FA synthesis in the mammary gland.

Ruminal wall metabolism was studied in nonlactating heifers by altering the carbohydrate (CHO) digestion site between rumen and intestine. The CHO digestion site was estimated from in situ and total-tract digestibility of control (CONT) diets and diets supplemented with corn (CRN), barley (BARL), or soy hulls (SOYH). Ruminal epithelial metabolism regulating gene expression, morphology, and nutrient delivery was assessed from a combination of rumen volatile fatty acid (VFA) concentration, biopsies for papilla morphology, and expression of putative metabolic regulatory genes encoding enzymes that facilitate VFA utilization. Digestible dry matter and CHO intake were 25 and 45% higher, respectively, in the supplemented diets than in CONT diets. Fiber supplementation increased the intestinal and decreased ruminal CHO digestion. Ruminal nonfiber CHO digestibility was 10% lower in CRN than with the high rumen-degradable supplement. The CONT heifers had lowest total ruminal VFA and highest acetate concentration relative to the other treatments. Total VFA concentration in BARL and CRN diets tended to be higher than in SOYH. The SOYH diet tended to reduce papilla dimension relative to CRN and BARL. The CRN diet tended to increase papilla surface area relative to BARL and SOYH. Gene expression of propionyl-coenzyme A carboxylase was higher in CRN and BARL than in SOYH diets, and tended to be higher in CRN than in BARL and SOYH diets. Lactate dehydrogenase and butyryl coenzyme A synthase gene transcripts tended to be higher in CONT than in the supplemented treatments. Thus, rumen epithelial expression of genes involved in VFA metabolism and ruminal wall-structure development are influenced by other regulatory mechanism that is not directly affected by local signals. The in situ methods used are a useful tool for differentiating ruminal from extraruminal nutrient supply.

The passage of long-chain fatty acids (FA) through the placenta in ruminants is limited. However, essential long-chain polyunsaturated FA, and especially n-3 FA, are crucial for normal development of the bovine fetus; therefore, uptake of these FA by the embryo must occur during pregnancy. The objectives of the present study were to examine the effects of enrichment of dam plasma with various n-3 FA during late gestation on newborn calf plasma FA composition. Twenty-seven multiparous cows at 256 d of pregnancy were divided into 3 groups and fed encapsulated fats as follows: 1) control: supplemented at 240 g/d per cow with saturated FA; 2) flaxseed oil (FLX): supplemented at 300 g/d per cow with fat that provided 56.1g/d per cow of α-linolenic acid (ALA) from flaxseed oil; and 3) fish oil (FO): supplemented at 300 g/d per cow with fat that provided 5.8 and 4.3g/d per cow eicosapentaenoic acid (EPA) and docosahexaenoic acid (DHA) from fish oil, respectively. Blood samples were taken from dams twice weekly and from calves immediately after calving before first colostrum. The FA composition in plasma was determined in dams at the last sample before parturition, on average 2d before calving. Feeding cows with FLX resulted in a 2.6-fold increase in the proportion of ALA in dam plasma as compared with the control. The proportion of EPA in cow plasma was not different between groups; however, the percentage of docosapentaenoic acid (DPA) was 1.4 and 2 times higher, respectively, in cows fed FLX and FO than in the controls. In addition, the plasma proportion of DHA was 15 times higher in FO cows than in controls. In calves, no differences between groups were observed in the plasma proportions of ALA and EPA; however, the proportion of DHA was 1.9 times higher in the FO calves than in controls. Across treatments, data showed no correlation between the proportions of ALA, EPA, and DPA in dam and calf plasma; however, positive correlation was demonstrated between dams and calves in DHA proportion (r=0.55). In conclusion, the distinct plasma FA profile in newborn calves compared with dams was apparently due to low permeability of the bovine placenta to polyunsaturated FA. Enriching late-gestation dairy cows with n-3 FA increased the proportion of DHA, but not ALA, in newborn calf plasma, probably because of the essentiality of DHA to fetal development.

The objective of this study was to examine the incorporation of dietary n-3 fatty acids (FAs) into ovarian compartments and the effects on hormonal and behavioral patterns around estrus. Multiparous 256-day pregnant cows were fed either a standard diet both prepartum and postpartum (PP) (control; n=22) or supplemented with extruded flaxseed (E-FLAX) providing C18:3n-3 at 172.2 and 402.5  g/day per cow prepartum and PP respectively (n=22). The estrous cycle was synchronized, and at day 7 of the cycle, the cows were injected with prostaglandin F(2)(α) (PGF(2)(α)) and then subjected to 5 days of intensive examination. Compared with those in the control, in the E-FLAX group, the interval from PGF(2)(α) injection to behavioral estrus peak tended to be longer (3.6  h; P<0.1), that to estradiol (E(2)) peak was 6.5  h longer (P<0.03), and that to LH peak tended to be longer (5.3  h; P<0.07). The durations of behavioral estrus and E(2) surge were longer, and the area under the E(2) curve was greater in the E-FLAX cows. Afterward, 7-8 days following behavioral estrus, follicular fluids (FFs) from >7  mm follicles were aspirated. The proportions of n-3 FA increased in plasma, FF, and granulosa cells in the E-FLAX group. The concentrations of PGE(2) in the E(2)-active follicles tended to be lower in the E-FLAX cows (P<0.06). In conclusion, several modifications in hormonal and behavioral estrus patterns were demonstrated in cows fed n-3 FA, which might be attributed to alterations in membrane FA composition and partly mediated by lower PGE(2) synthesis.

The objective of this study was to determine the effects of feeding an increased amount of extruded flaxseed with high proportions of n-3 fatty acids (FA) to transition dairy cows on performance, energy balance, and FA composition in plasma, adipose tissue, and milk fat. Multiparous Israeli-Holstein dry cows (n = 44) at 256 d of pregnancy were assigned to 2 treatments: (1) control cows were fed prepartum a dry-cow diet and postpartum a lactating-cow diet that consisted of 5.8% ether extracts; and (2) extruded flaxseed (EF) cows were supplemented prepartum with 1 kg of extruded flaxseed (7.9% dry matter)/cow per d, and postpartum were fed a diet containing 9.2% of the same supplement. The EF supplement was fed until 100 d in milk. On average, each pre- and postpartum EF cow consumed 160.9 and 376.2g of C18:3n-3/d, respectively. Postpartum dry matter intake was 3.8% higher in the EF cows. Milk production was 6.4% higher and fat content was 0.4% U lower in the EF group than in the controls, with no differences in fat and protein yields. Energy balance in the EF cows was more positive than in the controls; however, no differences were observed in concentrations of nonesterified fatty acids and glucose in plasma. Compared with controls, EF cows had greater proportions of C18:3n-3 in plasma and adipose tissue. The proportion of n-3 FA in milk fat was 3.7-fold higher in the EF cows, and the n-6:n-3 ratio was decreased from 8.3 in controls to 2.3 in the EF cows. Within-group tests revealed that the C18:3n-3 content in milk fat in the EF cows was negatively correlated with milk fat percentage (r = -0.91) and yield (r = -0.89). However, no decrease in de novo synthesis of less than 16-carbon FA was found in the EF group, whereas C16:0 yields were markedly decreased. It appears that the enrichment of C18:3n-3 in milk fat was limited to approximately 2%, and the potential for increasing this n-3 FA in milk is higher for cows with lower milk fat contents. In conclusion, feeding increased amounts of C18:3n-3 during the transition period enhanced dry matter intake postpartum, increased milk production, decreased milk fat content, and improved energy balance. Increased amounts of EF considerably influenced the FA profile of plasma, adipose tissue, and milk fat. However, the extent of C18:3n-3 enrichment in milk fat was limited and was negatively correlated with milk fat content and yield.

Multiparous Israeli-Holstein cows (n=42) averaging 158 d in milk and 621 kg of body weight were used to investigate the effects of energy source (fat or concentrate) on production, rumination time, energy balance, metabolic heat production, and efficiency under hot and humid conditions. Cows were assigned to 3 diet treatment groups: a lactating-cow ration (1.75 Mcal of net energy for lactation/kg of dry matter (DM; control); supplemented with 0.825 kg/d per cow of ground corn grain (2.7% of diet; HG); or supplemented with 0.300 kg/d per cow of calcium salts of fatty acids (1.5% of diet; HF). Milk production, body weight, and rumination time were recorded daily. Rectal temperature and respiratory rate (RR) were measured weekly. Mean daily maximum ambient temperature, relative humidity, and temperature-humidity index were 31.5 degrees C, 86.6%, and 76.8, respectively. Dry matter intake was lower in HF and HG cows than in controls. Average daily rumination time was lowest in the HG group: 393.0, 377.7, and 390.8 min/d for control, HG, and HF cows, respectively. Milk production was higher in the control group than in the HG group; milk fat content was 0.38 units higher and fat yield was 11% greater in HF cows than in HG cows. Fat-corrected milk yield was higher in HF cows than in HG cows, but not higher than in controls. Energy balance in HF cows was lower than in both other groups, and nonesterified fatty acid concentrations in plasma were increased by fat supplementation. Rectal temperature was highest in HF cows, and RR was higher in the HF cows than in the controls. Metabolic heat production was similar in HG and HF cows and lower than in controls. Body weight gain of the HG cows tended to be higher than that of the others. Efficiency of conversion of DM or energy intake to milk and fat-corrected milk was higher in HF cows than in both other groups; however, when taking the energy retention in body mass into account, no differences in energy utilization were observed between HF and HG groups. A principal component analysis revealed that rumination time played a pivotal role in the deleterious effect of heat stress in dairy cows. In conclusion, increasing the energy density in diets of heat-stressed mid-lactation cows over 1.75 Mcal/kg of DM was not effective in enhancing production. Both dietary treatments were effective in reducing metabolic heat production, but the changes were not reflected in rectal temperature and RR. Fat supplementation increased metabolic efficiency; however, whereas HF cows prioritized milk production, HG cows channeled energy for body mass deposition.

Our objectives were to determine the effects of rapid growth rate during the preweaning period and prepubertal protein supplementation on long-term growth pattern and milk production during the first lactation. Forty-six Israeli Holstein heifer calves were fed either milk replacer (MR) or whole milk (WM) from 4 to 60 d age. Calves had free access to WM or MR for 30 min twice daily and free-choice water and starter mix for the entire day. From weaning until 150 d of age, all heifers were fed the same ration. At 150 d of age the heifers were divided into 2 subgroups, with one subgroup supplemented with an additional 2% protein until 320 d of age. Thereafter, all heifers were housed and fed together until calving. Another cluster of 20 heifers was raised on MR and WM treatments and 3 animals from each nursery treatment were slaughtered at 60 d and 10 mo age to determine effects of nursery treatment on organ and adipose tissue mass. Prior to weaning, the MR heifers consumed 0.12 kg/d more DM than the WM heifers, but metabolizable energy intake was not different. Body weight at weaning and average daily gain during the preweaning period were 3.1 kg and 0.074 kg/d higher, respectively, in the WM treatment than in the MR treatment, with no differences in other measurements. Nursery feeding treatment and added protein had no effect on growth rate in the prepubertal period, but the postweaning difference in BW between the WM and MR heifers remained throughout the entire rearing period. The age at first insemination was 23 d earlier and age at pregnancy and first calving was numerically lower for the WM heifers than for the MR heifers. Adipose tissue weights at weaning were doubled in the WM calves. First-lactation milk production and 4% fat-corrected milk were 10.3 and 7.1% higher, respectively, for WM heifers than for MR heifers, whereas prepubertal added protein tended to increase milk yield. In conclusion, preweaning WM at high feeding rates appears to have long-term effects that are beneficial to future milk production. The positive long-term effects of feeding WM on first-lactation milk production were independent of their effects on skeletal growth. Enhanced milk production observed with WM treatment may be related to the milk supply, paracrine or endocrine effects of fat tissues on mammary parenchyma, or a combination of both factors.

The objectives were to determine the incorporation of dietary encapsulated fats differing in n-6:n-3 ratio into milk fat, plasma, and various ovarian compartments and to examine the effects on ovarian follicular status, preovulatory follicle characteristics, and oocyte quality. Twenty-four multiparous Israeli Holstein cows, averaging 114 d in milk, were assigned to 1 of 3 treatment groups: 1) control (n=7), in which cows were fed a lactating cow diet; 2) E-FLAX (n=8), in which cows were fed a lactating cow diet that consisted of 1kg/d of encapsulated fat (3.8% of dry matter) containing 40.8% flaxseed oil, providing 242.2g of C18:3n-3 (low n-6:n-3 ratio); or 3) E-SUN (n=9), in which cows were fed a lactating cow diet that consisted of 1kg/d of encapsulated fat (3.8% of dry matter) containing 40.8% sunflower oil, providing 260.0g of C18:2n-6 (high n-6:n-3 ratio). Ovaries were monitored by ultrasonography for follicular status, and after synchronization, follicles >7mm were aspirated and evaluated. Ovum pickup was performed (19 sessions for the control and E-FLAX groups and 11 for the E-SUN group), and in vitro maturation and oocyte fertilization were conducted. The E-FLAX treatment increased the proportions of C18:3n-3 (5.8 fold), C20:5n-3, and C22:5n-3 (approximately 4-fold) in milk fat as compared with the other 2 treatments. The proportion of C18:3n-3 fatty acid in plasma increased dramatically with the E-FLAX treatment, from 1.43 and 1.49% in the control and E-SUN groups, respectively, to 7.98% in the E-FLAX group. Consequently, the n-6:n-3 ratio in plasma was reduced from approximately 42 in the control and E-SUN groups to 6.74 in the E-FLAX group. Proportions of C18:3n-3 in follicular fluid and granulosa cells were approximately 5-fold higher in the E-FLAX group than in the other 2 groups. The percentage of C18:2n-6 in cumulus-oocyte complexes of cows in the E-SUN group was 54% higher than that in the E-FLAX group and was 2.4-fold higher than that in the control group; the proportion of C18:3n-3 in the E-FLAX group was 4.73% and was not detected in the other groups. The average numbers of 2- to 5-mm follicles on d 5 and 9 of the cycle were higher in the E-FLAX group than in the E-SUN group, whereas the average numbers of follicles > or =10mm on d 5, 9, and 13 were higher in the E-SUN group than in the other 2 groups. The estrous cycles of the cows were synchronized and PGF(2alpha) was injected on d 16 to 17 of the cycle. The interval from PGF(2alpha) injection to behavioral estrus was longer in the E-FLAX group than in the E-SUN group, and the beginning of the luteal phase of the subsequent cycle was delayed. Concentrations of estradiol in follicular fluid of the preovulatory follicles were higher in the E-SUN group than in the E-FLAX group. The number of follicles aspirated by ovum pickup was higher in the E-FLAX group than in the control group, and the cleavage rate in the E-FLAX group was higher than in the control group, but not the E-SUN group. In conclusion, dietary n-3 fatty acids influenced the follicular status and increased the cleavage rate of oocytes as compared with those of control cows. These findings could be related to modifications of the fatty acid composition in plasma and ovarian compartments in response to dietary supplementation.

Several processes have been suggested to protect lipids from bioactivity of the rumen microorganisms. The majority of experiments with conjugated linoleic acid (CLA) were conducted using calcium salts of CLA. The objectives of this study were to determine the effects of encapsulated CLA (E-CLA) that was supplemented during days 21 to 100 post partum (PP), on milk fat depression, recovery rate and performance parameters. Forty-two multiparous Israeli-Holstein cows were divided at day 21 PP into two treatment groups: (i) control - supplemented with 43 g/day per cow of calcium salts of fatty acids (FAs). (ii) E-CLA - supplemented with 50 g/day per cow of encapsulated lipid supplement providing 4.7 g/day per cow of trans-10, cis-12 CLA. Post-treatment cows were followed for recovery rate until 140 days PP. Dry matter intake (DMI) during the treatment period was reduced by 2.5%, and milk yield was enhanced by 4.5% in the E-CLA cows. Milk fat percentage and yield were reduced by 13% and 9%, respectively, in the E-CLA treatment as compared with the control. The energy-corrected milk output was 3.6% higher in the control group than in the E-CLA group. Yields of trans-10, cis-12 CLA isomer in milk was 2.13-fold higher in the E-CLA cows than in the controls. Full recovery to milk fat percentage of the control group occurred 4 to 5 weeks after cessation of the E-CLA supplementation. No differences between groups were observed in any fertility parameter that was tested. In conclusion, the E-CLA supplement decreased DMI, enhanced milk yield, and decreased energy output in milk, and was effective in depressing milk fat. Full recovery to the milk fat content, but not yield, of the control group in the E-CLA group was relatively slow and occurred 4 to 5 weeks after termination of the supplementation.

The current study examined the effects of live yeast (LY) supplementation to dairy cows during the summer season on milk production, feed efficiency and ration digestibility. Forty-two dairy cows (14 primiparous and 28 multiparous) were fed either a control lactating diet or supplemented with 1 g of LY (Saccharomyces cerevisiae, Biosaf, Lesaffre) per 4 kg of dry matter consumed. The LY amounts were adjusted twice a week. Four rumen samples were taken from 30 cows in 2-h periods and ammonia concentrations were determined. Fecal grab samples from 30 cows were collected during 3 consecutive days, to determine the apparent digestibility of diets. The daily dry matter intake in the LY group was 2.5% greater compared with the control group (24.7 and 24.1 kg, respectively). The daily average milk yield of the LY group was greater by 1.5 kg (4.1%) compared with the control group (37.8 vs. 36.3 kg, respectively). There were no significant differences in the milk fat and protein percentages, but fat yield was greater in the LY group than in the control. The fat-corrected milk 4% was 2.0 kg (6.1%) greater in the LY group than in the control group (34.8 vs. 32.8 kg, respectively). The efficiency of using dry matter to produce 4% fat-corrected milk was 3.7% greater in the LY group compared with the control group. The ruminal ammonia concentrations after feeding were greater in the control group than in the LY group (151.9 vs. 126.1 mg/l, respectively). No differences were observed among groups in the total tract apparent digestibility of dry matter and other diet components. The pH values in the rumen that were determined in a companion trial using 4 fistulated cows tended to be higher in cows that were supplemented with LY than in the control (6.67 vs. 6.54, respectively). It may be concluded that LY supplementation to dairy cows during the hot season improved the rumen environment in a way that increased the dry matter intake and in consequence enhanced the productivity and efficiency.

Dietary unsaturated fatty acids (UFAs) have been implicated in several reproductive processes in dairy cows through a variety of mechanisms. This study examined the effects of periparturient supplementation of rumen bypass fats low or high in proportion of UFAs (oleic and linoleic) on preovulatory follicle characteristics. Forty-two 256-day pregnant dairy cows were divided into three groups and were fed a control diet (n=14) or supplemented with fats either low (LUFA; n=14) or high (HUFA; n=14) in UFAs. At 14-15 days following behavior estrus, the cows received a prostaglandin F(2)(alpha) injection and 48 h later >7 mm follicles were aspirated. Progesterone (P(4)), androstenedione (A(4)), and estradiol (E(2)) were determined in the follicular fluid. Out of 75 follicles, 37 follicles that were aspirated between 55 and 70 days post partum were regarded as E(2)-active follicles (E(2)/P(4) ratio >1) and subjected for further analysis. The diameter of preovulatory follicles was greater in cows fed HUFA than in those fed control or LUFA. The concentrations and content of A(4) and E(2) in follicles and E(2)/P(4) ratio were higher in the HUFA group than in the other two groups. The P450 aromatase mRNA expression in granulosa cells that were collected from the aspirated preovulatory follicles was also higher in the HUFA cows than in the other groups. A significant correlation was observed between E(2) concentrations in preovulatory follicles and E(2) concentrations in plasma at aspiration. In conclusion, dietary UFA increased the size of and elevated steroid hormones in preovulatory follicles, which may be beneficial to consequent ovarian function.

Dry multiparous cows were used to investigate the effects on intake, production, and metabolism of either a supplement containing 55% dry propylene glycol (PGLY), a prilled fat supplement (PrFA) containing a low proportion of unsaturated fatty acids (FA), or calcium soaps of FA supplement (CaLFA) containing a high proportion of unsaturated FA. Fifty-three dry cows (256 d pregnant) were stratified into 4 groups and began one of the following dietary treatments: 1) control cows were fed a dry cow diet and at postpartum were fed a lactating cow diet; 2) diets of cows in the PGLY group were supplemented with 500 g/d per cow of dry PGLY until 21 d in milk (DIM); 3) diets of cows in the PrFA group were supplemented with 230 g/d per cow of PrFA until 100 DIM; 4) diets of cows in the CaLFA group were supplemented with 215 g/d per cow of CaLFA until 100 DIM. Prepartum DMI was lower in the PrFA and CaLFA groups than in the control and PGLY groups, whereas postpartum DMI in the PrFA group was higher than that in the control group. Milk production until 100 DIM in both fat-supplemented groups was 4.5% higher than that in the control group. Plasma glucose concentrations pre- and postpartum were higher in the PGLY group than in the PrFA and CaLFA groups, but were similar to those in the control group. Prepartum nonesterified FA (NEFA) concentrations in plasma were increased by 43 and 70% in the PrFA and CaLFA groups, respectively, as compared with the control and PGLY groups. Both fat supplements increased plasma beta-hydroxybutyrate concentrations over those of the PGLY and control groups pre- and postpartum. Peripartum plasma insulin concentrations in the control group were 1.7-fold higher than in the PrFA group and 2.1-fold higher than in the CaFA group. Differences between the PrFA and CaLFA groups were observed: DMI was higher pre- and postpartum in the PrFA group than in the CaLFA group, and prepartum plasma NEFA concentrations were 19% higher and insulin concentrations were 21% lower in the CaLFA group than in the PrFA group. No significant differences were observed in DMI, plasma glucose, NEFA, and beta-hydroxybutyrate concentrations between the control and PGLY groups. Feeding fat to cows during late pregnancy decreased the DMI and negatively affected the metabolic status of the cows, as reflected by plasma metabolites. Furthermore, protected fat with a high proportion of unsaturated FA (CaLFA) was more pronounced in increasing plasma NEFA concentrations and depressing plasma insulin concentrations than fat with a low proportion of unsaturated FA (PrFA).

Eighty multiparous cows were used to test the effects of feeding a supplement containing 55% dry propylene glycol (PGLY), prilled fat (PrFA) containing a low proportion of unsaturated fatty acids (FA), or calcium soaps of long-chain FA (CaLFA) containing a high proportion of unsaturated FA on energy balance (EB), blood metabolites, and early postpartum (PP) ovarian follicles. Dry cows (256 d pregnant) were divided into 6 groups and began the following dietary treatments: 1) control group, fed a dry cow diet and fed a lactating cow diet PP; 2) PGLY group, diet supplemented with 500 g/d per cow of dry PGLY prepartum through 21 d in milk; 3) PrFA:control group, diet supplemented with 230 g/d per cow of PrFA prepartum and fed the control diet PP; 4) PrFA:PrFA group, diet supplemented with 230 g/d per cow of PrFA prepartum through 21 d in milk; 5) CaLFA:control, supplemented with 215 g/d per cow of CaLFA prepartum and fed the control diet PP; 6) CaLFA:CaLFA, supplemented with 215 g/d per cow of CaLFA prepartum through 21 d in milk. Follicular fluid was aspirated from follicles > or = 6 mm on d 12 PP. The daily average calculated EB during the first 21 d in milk was lower in the PrFA:PrFA (-4.16 Mcal/d) and CaLFA:CaLFA (-3.64 Mcal/d) groups than in the control (-1.71 Mcal/d) and PGLY (-2.19 Mcal/d) groups. Postpartum plasma beta-hydroxybutyrate was higher, and insulin concentrations were lower in the PrFA:PrFA (6.2 mg/dL and 126.1 pg/mL, respectively) and CaLFA:CaLFA (7.0 mg/dL and 130.7 pg/mL) groups than in the control (4.5 mg/dL and 274.5 pg/mL) and PGLY (4.3 mg/dL and 272.6 pg/mL) groups, whereas nonesterified FA concentrations were higher only than the control group. Postpartum nonesterified FA were 21% lower and insulin plasma concentrations were 86% higher in the CaLFA:control group as compared with the PrFA:control group. The progesterone concentrations in the follicular fluid of estradiol-active follicles were higher in the CaLFA:CaLFA (200.7 ng/mL) group than in all other groups (57.3 to 92.4 ng/mL), and androstenedione and estradiol concentrations were higher (54.2 and 1,049.1 ng/mL, respectively) than in the PGLY (15.5 and 440.1 ng/mL), PrFA:control (22.6 and 314.1 ng/mL), and CaLFA:control (17.5 and 451.9 ng/mL) groups. In conclusion, supplementation of protected fat during the peripartum period negatively affected the EB status of the cows. Neither fat supplementation nor PGLY influenced the development of ovarian follicles during the early PP period, but feeding fat containing a high ratio of unsaturated FA (CaLFA) increased progesterone concentrations in estradiol-active follicles that were aspirated at 12 d in milk.
