RESUMO

Diffuse pollution, including that in the lower and middle reaches of the Yangtze River, is the primary source of pollution in several agricultural watersheds globally. As the largest river basin in China, the Yangtze River Basin has suffered from total phosphorus (TP) pollution in the past decade owing to diffuse pollution and aquatic ecology destruction, especially in the midstream tributaries and mid-lower reaches of the lakes. However, the transport dynamics of diffuse pollutants, such as phosphorus (P) from land to water bodies have not been well evaluated, which is of great significance for quantifying nutrient loss and its impact on water bodies. In this study, diffuse pollution estimation with remote sensing (DPeRS) model coupled with Soil and Water Assessment Tools (SWAT) was utilized to simulate the transport dynamics of P, investigate the spatial heterogeneity and P sources in the Poyang Lake Basin. Additionally, the P transport mechanism from land to water and the migration process in water bodies were considered to investigate the impact of each loss unit on the water body and evaluate the load generated by diverse pollution types. The estimated diffuse TP loss was 6016 t P·yr-1, and the load to inflow rivers and to Poyang Lake were 11,619 and 9812 t P·yr-1, respectively. Gan River Basin (51.09%) contributed most TP to Poyang Lake among five inflow rivers, while waterfront area demonstrated the highest TP load per unit area with 0.057 t km-2·yr-1. Our study also identified P sources in the sub-basins and emphasized agricultural diffuse sources, especially planting, as the most significant factor contributing to TP pollution. Additionally, to improve the aquatic environment and water ecological conditions, further nutrient management should be applied using a comprehensive approach that encompasses the entire process, from source transportation to the water body.

Rainfall plays a crucial role in influencing the loss of agricultural diffuse pollution. The middle Yangtze River region is well-know for its humid climate and numerous agricultural activities. Thus, this study quantitatively analyzed the concentration and distribution of nitrogen (N) and phosphorus (P) load and loss in a major tributary of the middle Yangtze River under different rainfall patterns by using sampling analysis and SWAT model simulation. The total nitrogen (TN) and nitrate-nitrogen (NO3-) concentrations were 1.604-3.574 and 0.830-2.556 mg/L, respectively. The total phosphorous (TP) and Soluble Reactive Phosphorus (SRP) were 2-148 and 2-104 µg/L, respectively. The modeling results demonstrated that higher rainfall intensity led to greater load and loss flux of diffuse pollutant at the outlet. Organic nitrogen (ORGN) is the main nitrogen form transported from the subbasin to the reach, while organic phosphorus (ORGP) and inorganic phosphorus (INORGP) were transported at similar amounts. Under the condition of conventional rainfall, the outlet reaches mainly transported NO3-, and ORGN gradually increased when rainstorm events occurred. The ratio of INORGP to ORGP was relatively stable. During extreme rainfall event, rainfall is the dominant element of agricultural diffuse pollution. A strong positive correlation exists between rainfall intensity and pollution loss during rainstorms. Storm rain events were the main source of TN and TP losses. Few storm rain days generated pollutants that accounted for a large proportion of the total loss, and their impact on TP loss was significantly greater than that of TN. The influence of storm rain on TN is mainly the increase in runoff, while TP is sensitive to the runoff and sediment transport promoted by rainfall. By setting different precipitation scenarios, it was confirmed that under the same rainfall amount, short-term storm rain has the most significant impact on the TN load, whereas TP load may be influenced more by the combined effects of rainfall duration and intensity. Therefore, to reduce the impact of agricultural diffuse pollution, it is important to take targeted measures for the rainstorm days.

Phosphorus is an integral component of marine biogeochemistry. This research investigated the environmental behavior of P in Laizhou Bay using high-resolution sampling, P fractionation, and isotherm adsorption. The total dissolved P (TDP) concentration ranged from 8.4 to 61.0 µg/L in the bay water, while total P (TP) concentration ranged from 311.6 to 654.5 mg/kg in the sediment. The TDP concentration in the water was high in the estuarine area of the Yellow River and the southwestern bay under the combined effects of riverine inputs, direct wastewater discharge, and limited water exchange ability. High TP concentrations in the sediment were observed near the mouth of the Yellow River and central bay, mainly due to the movement and settlement of fine suspended particles under the influence of ocean currents. The P in the bay sediment was predominantly in the calcium-bound fraction and was associated with small particles such as silt and clay. The equilibrium P concentration (EPC0) ranged from 1.6 to 131.4 µg/L, and P partition coefficient or buffer intensity (Kd) ranged from 104 L/kg to 880 L/kg. The EPC0 decreased from the northeastern to southwestern area, while Kd showed an inverse distribution; therefore, the southwestern bay sediment had high buffer intensity for external P loads. Additionally, ECP0 increased linearly, and Kd decreased with exchangeable P (Exc-P) and Fe-bound P (Fe-P) concentrations in the sediment, demonstrating that P sediment-water exchange in LZB was dominated by contributions from Exc-P and Fe-P. These results can aid the understanding of the P sources and geochemistry of coastal ecosystems, particularly sediment P release potential.

The leaf litter decomposition is the important chain for the nutrient cycle in forest ecosystem, but its degradation dynamics and pulse discharge patterns in freeze-thawing watershed needed complete understanding. By integrating field observations and MODIS data, the temporal-spatial distributions of snow coverage and forest leaf litter biomass were analyzed. The critical period for snowmelt runoff under warming temperature and the relatively slow degradation patterns were identified. The on-site observations snowmelt runoff showed discharge concentration and fraction dynamics of typical forest leaf litter nutrients (carbon, nitrogen, and phosphorus) in thawing period. The snowmelt runoff flow and nutrient flux observed the linear regressions with the increased temperature from -8 °C to 6 °C (r2 = 0.443-0.987). The concentration of TOC, TN, and TP reached summit value around 50.0, 6.0, and 0.5 mg L-1 in the snowmelt runoff, respectively. The fraction analysis proved that the much high composition of dissolved organic fraction and the biggest organic phosphorus percentage was 94%. The comparison experiments of forest soil with or without leaf litter cover demonstrated that the leaf litter caused a lower discharge load in the snowmelt flow, and the leaf litter cover can decease the potential transport capability of the snowmelt runoff. Coupled with remote sensing data, the watershed leaf litter nutrient discharge model was developed with snowmelt hydrological process mode. The watershed averaged discharge of TOC, TN, and TP from deciduous broad-leaved forest leaf litter was around 851.99, 75.05, and 9.78 mg·m-2, respectively. The yearly simulation showed the spatial distribution variance of the nutrient discharge loads were held by different forest types, elevations, and slopes. The critical loss area identification provided new mitigations solution. The findings suggested that seasonal discharge of forest leaf litter nutrient in thawing period acted as a key contributor to watershed water pollution.

In this study, we investigated whether transcutaneous electrical acupoint stimulation (TEAS) could improve cognitive function in VD rats by regulating PINK1/Parkin-mediated mitophagy. VD rat model was prepared by modified 2-vessel occlusion (2-VO) and randomly divided into four groups: Sham group (Sham), Model group (Model), TEAS group (TEAS), and TEAS + 3-MA group (T +3 -MA). In the T +3 -MA group, autophagy inhibitor (3-MA) was injected into the lateral ventricle. After modeling, Y maze (YM), new object recognition test (NORT), Morris water maze (MWM), immunofluorescence, and Western blot were used to observe the effects of TEAS on VD rats. Behavioral experiments revealed that TEAS effectively improved the learning and memory ability of VD rats. Immunofluorescence results showed that TEAS could upregulate LC3 expression. Western blot results showed that TEAS upregulated the expression of PINK1, Parkin, and LC3-II, and downregulated the expression of LC3-I and p62 in VD rats. T +3 -MA group shows the opposite trend to TEAS group. This study demonstrates that TEAS ameliorates cognitive function through PINK1/Parkin-mediated mitophagy in VD rats.

Phosphorus contamination in urbanized bays has been a major concern because the bay restoration is often hindered by complex P sources and behaviors. This study examined the spatiotemporal changes of P species and exchange potential in/between the water and sediment of the Jiaozhou Bay. The results indicated that dissolved P (TDP) and inorganic P (DIP) in the water ranged from 7.8-128.7 and 1.8-14.1 µg/L, respectively; while total P (TP) in the sediment ranged from 213.4-638.7 mg/kg. The TDP and DIP concentrations in the water were high in winter and low in summer, and generally decreased from northeastern or northern areas to southwestern or southern areas mainly due to phytoplankton bloom cycles and riverine and wastewater inputs. TP in the sediment was lower in the northwestern area due to solid dilution effect by the settlement of settled coarser suspended particles. Changes in aquatic geochemical conditions from rivers to bay caused P accumulation in estuarine sediment, with higher P partition in organic fraction (40%). Compared to the estuarine sediment, higher fractions of P were associated with carbonate (34%) and iron oxide (17%) minerals in the bay sediment. Equilibrium P concentrations at zero sorption (EPC0) were 4.1-149.8 µg/L, which was substantially higher than the DIP concentration, demonstrating P release potential from the sediment. In addition, the P release potential was high in the northeastern area while P partition coefficient or buffer intensity (Kd) was high in the northwestern area. EPC0 was significantly positively correlated with soluble and exchangeable P in the sediment while Kd was significantly negatively correlated. These results can provide improved insights into P behaviors in an urbanized bay, particularly the P release potential and spatiotemporal change.

River delta-front estuaries (DEs) are vital interfaces for fluxes between terrestrial and marine environments. However, deep uncertainty exists in estimating the sedimentary pollutant flux from terrestrial environments in DEs due, in part, to a lack of direct measurements in these dynamic and complicated regions and uncertainty in the calculation method. Due to its high sediment content, the Yellow River (YR) has a strong ability to adsorb phosphorus; therefore, it reliably reflects estuarine sedimentary processes. Here, through the comprehensive analysis of field samples, monitoring data and remote sensing images, we conclude that riverine fine particles control the deltaic estuary pollution status and that particle size is the key factor. Based on the stable relationships between phosphorus and heavy metals, with r2 values of 0.990, 0.992, and 0.639 for As, Cd, and Cr, respectively, we estimated that the P flux reached 22.68 g/m2 yr in 2017. Analysis of the YR high-silt sediment load, which has a strong phosphorus adsorption ability and constitutes a substantial fraction of global fluvial sediment transport, revealed a negative correlation between the riverine sediment load and the estuarine phosphorus flux.

The synergetic loss mechanism of heavy metals (HMs) and phosphorus (P), as well as the relationships between their different geochemical fractions remain unclear. This study employs field research, source identification and sedimentary geochemistry in Yellow River basin to investigate the internal mechanisms of the different geochemical fractions, terrestrial source signatures, and synergetic loss fluxes. The average contents of As, Cd, Cr, Cu, Pb, Zn and P in the basin were 8.29 mg/kg, 0.15 mg/kg, 47.52 mg/kg, 11.78 mg/kg, 10.65 mg/kg, 46.56 mg/kg and 578.78 mg/kg, respectively. Based on Pearson's correlation and redundancy analyses, the impact factors on the transport of HMs and P, and the internal relationships between different geochemical fractions were analyzed. According to the constant rate of supply (CRS) model, the terrestrial losses of As, Cd, Cr and Cu showed significant positive relationships with the TP flux, with r2 value of 0.981, 0.991, 0.996 and for 0.984, respectively. It has been proven that the extensive fine particles in the Yellow River basin carry a large amount of diffuse pollutants, thus ultimately increasing the estuarine pollutant load. This research provides new insights from the level of microscopic fractions to macroscopic fluxes to investigate the impacts of anthropogenic activity on regional environmental changes.

Mechanochemical methods with co-milling reagents have been widely used to degrade organic pollutants. In this study, calcium oxide and persulfate were employed as co-milling reagents in a mechanochemical process that showed highly effective degradation of hexachlorobenzene in contaminated soil. The influences of soil particle size and organic matter content were also investigated. The interaction between different factors was analyzed by response surface methodology, and a multi-variate regression equation was obtained relating the soil-to-oxidant mass ratio, rotation speed and organic matter content. The existence of SO4- and OH during the mechanochemical reaction was proved by the indirect detection of benzoquinone and p-hydroxybenzoic acid for the first time, providing a new method for testing free radicals in solid-phase reactions. Finally, a possible activation mechanism and hexachlorobenzene degradation pathway were proposed. This study successfully presents a mild degradation method in the field of hexachlorobenzene contaminated site remediation.

OBJECTIVES: This study aims to provide a way to estimate the volume of the thyroid remnant and determine its relationship with the outcome of radioiodine (RAI) therapy in depth. MATERIALS AND METHODS: A retrospective analysis was performed on patients who underwent initial RAI therapy between January 2010 and January 2016. The patients were divided into five groups based on the thyroid remnant estimated by post-therapy whole-body scan(post-Rx WBS), thyroid scintigraphy and ultrasonography. The relationship between the volume of thyroid remnant and the outcome of RAI therapy were evaluated by univariate analysis and multivariate analysis. RESULTS: Of 703 patients, the majority could be found different size of thyroid remnants using the three imaging methods, and only few patients(2.1%) could reach no thyroid remnant. There was no association between the volume of thyroid remnant and the outcome of RAI therapy in univariate analysis (χ2 = 1.633, P = 0.652) and multivariate analysis (P > 0.05). In the subgroup of patients with high-risk factors, there was still no significant difference (intermediate risk subgroup: P = 0.338 vs high risk subgroup: P = 0.263). CONCLUSION: Different sizes of thyroid remnants were left after surgery. However, in high radioiodine activity, the volume of thyroid remnants may not affect the outcome of RAI therapy even in patients with some high-risk factors, so the high radioiodine activities may resolve the the problem caused by thyroid remnants in some cases.

The formation and transportation processes of non-point source (NPS) pollution varied among the studied watersheds in the Northeastern China, so we hypothesized that the driving force behind NPS pollution followed the spatial scale effect. With a watershed outlet sedimentary flux analysis and a distributed NPS pollution loading model, we investigated the temporal dynamics of NPS and the differences in driving forces. Sediment core samples were collected from two adjacent watersheds, the smaller Abujiao watershed and the larger Naoli watershed. The natural climatic conditions, long-term variations in the distribution of land use, soil properties and tillage practices were the same in the two watersheds. The vertical distributions of total nitrogen, total phosphorus, Zn and As at 1-cm intervals in the section showed clear differences between the watersheds. There were higher concentrations of total nitrogen and total phosphorus in the larger watershed, but the heavy metals were more concentrated in the smaller watershed. Lead-210 (210Pb) analyses and the constant rate of supply model provided a dated sedimentary flux, which was correlated with the corresponding yearly loading of NPS total nitrogen and total phosphorus in the two watersheds. The total phosphorus showed a stable relationship in both watersheds with an R2 value that ranged from 0.503 to 0.682. A rose figure comparison also demonstrated that the pollutant flux in the sediment was very different in the two watersheds, which had similar territorial conditions and different hydrological patterns. Redundancy analysis further indicated that expanding paddy areas had a large impact on the sedimentary flux of nitrogen and phosphorus in the smaller watershed, but precipitation had a direct impact on NPS loading in the larger watershed. We concluded that the spatial scale effect affected the NPS pollution via the transport processes in the waterway, which was mainly influenced by branch length and drainage density.

This study investigated the molecular mechanism of brain impairment induced by drinking fluoridated water and selenium intervention. Results showed that the learning and memory of rats in NaF group significantly decreased. Moreover, the number of apoptotic cells, the expression levels of Cytc mRNA and protein, and the expression levels of Caspase-9 and Caspase-3 mRNA significantly increased; by contrast, Caspase-9 and Caspase-3 protein levels significantly decreased. Compared with the NaF group, the mRNA levels of Cytc and Caspase-9, as well as the protein levels of Cytc in NaF+Se group, significantly decreased. Conversely, the protein levels of Caspase-3 and Caspase-9, as well as the mRNA levels of Caspase-3, significantly increased. Thus, the mitochondrial CytC-Caspase-9-Caspase-3 apoptosis pathway in the hippocampus was one of the mechanisms leading to fluorosis-induced brain damage. Furthermore, the Cytc signaling molecules were possibly the key target molecules in fluorosis-induced apoptosis, and selenium could alleviate fluorosis-induced brain injury.

The farmland tillage practices changed the soil chemical properties, which also impacted the soil respiration (R s ) process and the soil carbon conservation. Originally, the farmland in northeast China had high soil carbon content, which was decreased in the recent decades due to the tillage practices. To better understand the R s dynamics in different land use types and its relationship with soil carbon loss, soil samples at two layers (0-15 and 15-30 cm) were analyzed for organic carbon (OC), total nitrogen (TN), total phosphorus (TP), total carbon (TC), available nitrogen (AN), available phosphorus (AP), soil particle size distribution, as well as the R s rate. The R s rate of the paddy land was 0.22 (at 0-15 cm) and 3.01 (at 15-30 cm) times of the upland. The average concentrations of OC and clay content in cultivated areas were much lower than in non-cultivated areas. The partial least squares analysis suggested that the TC and TN were significantly related to the R s process in cultivated soils. The upland soil was further used to test soil CO2 emission response at different biochar addition levels during 70-days incubation. The measurement in the limited incubation period demonstrated that the addition of biochar improved the soil C content because it had high concentration of pyrogenic C, which was resistant to mineralization. The analysis showed that biochar addition can promote soil OC by mitigating carbon dioxide (CO2) emission. The biochar addition achieved the best performance for the soil carbon conservation in high-latitude agricultural area due to the originally high carbon content.

OBJECTIVE: To assess the impact of tea consumption on the risk of osteoporotic hip fractures. METHODS: Between January 2008 and June 2012, 581 (148 males, 433 females) incident cases of hip fractures were enrolled from four hospitals in Guangdong province, with 581 sex- and age-matched (± 3 years) controls from either hospitals or communities. Face-to-face interviews were conducted to collect data pertaining to tea drinking and various covariates. RESULTS RESULTS: from univariate conditional logistic analyses showed that an inverse association was observed in tea drinking and hip fracture risk. Longer time, greater frequency and dosage of tea consumption were dose-dependently associated with lower risk of hip fractures (P-trend < 0.05). Compared to non-drinkers, the odd ratios related to regular tea drinkers, subgroups with different length, frequency, dosage, type of tea consumption were ranged between 0.54 and 0.74 (all P < 0.05). After adjustment for factors as age, daily energy intake, BMI, education levels, passive smoking, calcium supplement and physical activity, the dose-dependent associations among above said factors still remained significant. However, the strength of the association lowered slightly. The beneficial effect of tea was significant only in men but not in women. Similar effects were found in subjects with different education levels. CONCLUSION: Regular tea drinking habit might decrease the risk of osteoporotic hip fractures in the elderly males.

There are two kinds of land policies, the smallholding land policy (SLP) and the farmland policy (FLP) in China. The farmland nutrient dynamics under the two land policies were analysed with the soil system budget method. The averaged nitrogen (N) input of the SLP and the FLP over sixteen years increased about 23.9% and 33.3%, respectively and the phosphorus (P) input climbed about 39.1% and 42.3%, respectively. The statistical analysis showed that the land policies had significant impacts on N and P input from fertilizer and manure, but did not obviously affect the N input from seeds and biological N fixation. The efficiency percentage of N of the SLP and the FLP climbed about 54.5% and 59.4%, respectively, and the P efficiency improved by 52.7% and 82.6%, respectively. About the nutrient output, the F-test analysis indicated that the land polices had remarkable impacts on N output by crop uptake, ammonia volatilisation, denitrification, leaching and runoff, and P output by uptake, runoff, and leach. The balance showed that the absolute loss of N from land deceased about 43.6% and 46.0%, respectively, in the SLP and the FLP, and P discharge reduced about 34.2% and 75.2%, respectively. The F-test analysis of N and P efficiency and balance of between two polices both indicated that the FLP had significant impact on nutrient dynamic. With the Mitscherlich model, the correlations between nutrient input and crop uptake, usage efficiency and loss were analysed and showed that was a threshold value for the optimal nutrient input with the highest efficiency rate. For the optimal nutrient efficiency, the space for extra P addition was bigger than the N input. The FLP have more advantage than the SLP on the crop yield, nutrient efficiency and environmental discharge.

Dynamics of livestock and poultry manure nutrient was analyzed at a provincial scale from 2002 to 2008. The nutrient capacity of 18 kinds of croplands and grasslands to assimilate nutrients was assessed in the same temporal-spatial scale. Manure nitrogen (N) had increased from 5.111 to 6.228 million tons (MT), while manure phosphorus (P) increased from 1.382 to 1.607 MT. Manure N and P share similar spatial patterns of yields, but proportion of specialized livestock husbandry and contribution of leading livestock categories (swine, cattle, cow, sheep, layer chicken, broiler chicken) were different. The nutrients generated from dominant seven provinces took more than about half of total manure N in China. After subtracting the chemical fertilizers, there were some manure nutrient capacities in western part of China. Risk analysis of manure nutrient pollution overload in eastern and southern parts of China was serious, which should restrict livestock's developments. Amount of chemical fertilizers applied should be reduced to make room for manure nutrients. For the sake of greenhouse effects, the emission of methane (CH4) and nitrous oxide (NO x ) emissions in China is serious for the global change, thus merits further statistics and studies. The spatial and temporal pattern of Chinese manure nutrient pollution from livestock and the assimilation capacity of cropland and grassland can provide useful information for policy development on Chinese soil environment and livestock.

Managing storm rainfall runoff is paramount in semi-arid regions with urban development. In Beijing, pollution prevention in urban storm runoff and storm water utilization has been identified as the primary strategy for urban water management. In this paper, we sampled runoff during storm rainfall events and analyzed the concentration of chemical oxygen demand (COD), total suspended solids (TSS) and total phosphorus (TP) in the runoff. Furthermore, the first flush effect of storm rainfall from diverse underlying surfaces was also analyzed. With the Storm Water Management Model (SWMM), the different impervious rates of underlying surfaces during the storm runoff process were expressed. The removal rates of three typical pollutants and their interactions with precipitation and underlying surfaces were identified. From these rates, the scenarios regarding the urban storm runoff pollution loading from different designs of underlying previous rates were assessed with the SWMM. First flush effect analysis showed that the first 20% of the storm runoff should be discarded, which can help in utilizing the storm water resource. The results of this study suggest that the SWMM can express in detail the storm water pollution patterns from diverse underlying surfaces in Beijing, which significantly affected water quality. The scenario analysis demonstrated that impervious rate adjustment has the potential to reduce runoff peak and decrease pollution loading.

To better understand the spatial dynamics of non-point source (NPS) phosphorus loading with soil property at watershed scale, integrated modeling and soil chemistry is crucial to ensure that the indicator is functioning properly and expressing the spatial interaction at two depths. Developments in distributed modeling have greatly enriched the availability of geospatial data analysis and assess the NPS pollution loading response to soil property over larger area. The 1.5 km-grid soil sampling at two depths was analyzed with eight parameters, which provided detailed spatial and vertical soil data under four main types of landuses. The impacts of landuse conversion and agricultural practice on soil property were firstly identified. Except for the slightly bigger total of potassium (TK) and cadmium (Cr), the other six parameters had larger content in 20-40 cm surface than the top 20 cm surface. The Soil and Water Assessment Tool was employed to simulate the loading of NPS phosphorus. Overlaying with the landuse distribution, it was found that the NPS phosphorus mainly comes from the subbasins dominated with upland and paddy rice. The linear correlations of eight soil parameters at two depths with NPS phosphorus loading in the subbasins of upland and paddy rice were compared, respectively. The correlations of available phosphorus (AP), total phosphorus (TP), total nitrogen (TN) and TK varied in two depths, and also can assess the loading. The soil with lower soil organic carbon (SOC) presented a significant higher risk for NPS phosphorus loading, especially in agricultural area. The Principal Component Analysis showed that the TP and zinc (Zn) in top soil and copper (Cu) and Cr in subsurface can work as indicators. The analysis suggested that the application of soil property indicators is useful for assessing NPS phosphorus loss, which is promising for water safety in agricultural area.

BACKGROUND AND PURPOSE: Cardiac toxicity is a major concern in drug development and it is imperative that clinical candidates are thoroughly tested for adverse effects earlier in the drug discovery process. In this report, we investigate the utility of an impedance-based microelectronic detection system in conjunction with mouse embryonic stem cell-derived cardiomyocytes for assessment of compound risk in the drug discovery process. EXPERIMENTAL APPROACH: Beating of cardiomyocytes was measured by a recently developed microelectronic-based system using impedance readouts. We used mouse stem cell-derived cardiomyocytes to obtain dose-response profiles for over 60 compounds, including ion channel modulators, chronotropic/ionotropic agents, hERG trafficking inhibitors and drugs known to induce Torsades de Pointes arrhythmias. KEY RESULTS: This system sensitively and quantitatively detected effects of modulators of cardiac function, including some compounds missed by electrophysiology. Pro-arrhythmic compounds produced characteristic profiles reflecting arrhythmia, which can be used for identification of other pro-arrhythmic compounds. The time series data can be used to identify compounds that induce arrhythmia by complex mechanisms such as inhibition of hERG channels trafficking. Furthermore, the time resolution allows for assessment of compounds that simultaneously affect both beating and viability of cardiomyocytes. CONCLUSIONS AND IMPLICATIONS: Microelectronic monitoring of stem cell-derived cardiomyocyte beating provides a high throughput, quantitative and predictive assay system that can be used for assessment of cardiac liability earlier in the drug discovery process. The convergence of stem cell technology with microelectronic monitoring should facilitate cardiac safety assessment.

Cardiac safety testing of lead drug candidates is an important part of the drug discovery and development process. All new chemical entities need to be subjected to extensive preclinical assessment for cardiac liability, especially for a potentially fatal form of ventricular arrhythmia referred to as Torsades de Pointes. We have developed an innovative label-free, real-time system, the xCELLigence RTCA Cardio System, which is designed to monitor contractility of cardiomyocytes based on impedance measurement. The assay is performed using specially designed microtiter plates that are integrated with gold microelectrodes. The system was validated using mouse embryonic stem cell-derived cardiomyocytes, human-induced pluripotent stem cell-derived cardiomyocytes, and rat neonatal primary cardiomyocytes by applying a variety of tool compounds and drugs with known mechanisms of action. Our data show that the time resolution in the assay can provide important information about compound action. Furthermore, the impedance-based beating profile in response to compound treatment can provide mechanistic toxicity information regarding the target being modulated and may be able to flag pro-arrhythmic compounds. We believe the real-time and kinetic aspect of this technology combined with beat-to-beat measurement of cardiomyocyte contraction would make this instrument an important part of preclinical cardiac safety assessment.
