The aim of this study was to determine the Se concentration in the main tissues of beef cattle and to evaluate the differences in tissue distribution between animals with different selenium status. Selenium concentration was determined in the serum, longissimus dorsi muscle, semitendinosus muscle, kidney, heart, liver, spleen and lungs of cows, heifers and beef bulls, using spectrofluorimetric method. Despite receiving supplementation, 55.6% animals demonstrated an optimal Se level, while 44.4% were deficient. The mean serum Se concentration was significantly higher (p < 0.05) in animals with a normal Se status than in Se-deficient animals. Differences in Se tissue distribution were observed between Se-deficient animals and those with normal Se status. The organs most susceptible to Se deficiency are the semitendinosus muscle, lungs, heart and liver. In both normal and Se-deficient animals, significantly higher Se concentrations were observed in the kidney than other organs (p < 0.05), and the lowest in the muscles. As Se deficiencies can be found among supplemented animals, the level of Se should be monitored in beef cattle in order to detect possible Se deficiencies, which may have negative health effects for animals and reduce the value of animal products as a source of Se in the human diet.

The aim of this study was to evaluate the effect of serum selenium on PPAR-γ and the selected proinflammatory cytokines (IL-1ß, IL-6, TNF-α) in relation to depressive symptoms and obesity in middle-aged women. The research procedure was as follows: a survey was performed using the authors' questionnaire and the BDI, anthropometric measurements, and the analysis of blood for the levels of selenium, cytokines, and genetic analysis of the PPAR-γ polymorphism (n = 443). It was found that the BMI increased along with the concentration of IL-6. No moderating effect of selenium was observed, although the cut-off values for "p" were established for IL-ß*Se (p = 0.068) and IL-6*Se (p = 0.068), so there was a potential association with these two markers. At high selenium levels, the effect of higher IL-ß levels on a decrease in BMI was stronger, as was the effect of an increase in IL-6 levels on an increase in BMI. No effect of selenium on PPAR-γ was found in relation to depressive symptoms and obesity. Higher selenium levels may have a beneficial effect on BMI even at high IL-ß concentrations, however, at high IL-6 concentrations, this effect was not observed. Selenium levels had no impact on depressive symptoms.

BACKGROUND: Selenium plays an important role in metabolic homeostasis. It has been suggested that it may also affect the expression and activity of PPAR-γ. The aim of study was to analyze the relationships between these variables in the context of the health of women, for whom the risk of MetS increases with age. MATERIAL AND METHODS: The study involved 390 women in middle age. The stages of study: a survey-based part; anthropometric measurements; analysis of biological material (blood) in terms of glycemia, triglyceride, HDL, and selenium levels, as well as genetic analysis of the PPAR-γ polymorphisms. RESULTS: It was found that selenium may moderate the effect of the G allele of the PPAR-γ gene on the occurrence of elevated waist circumference (OR=1.030, 95%CI 1.005-1.057, p=0.020); and the effect of the C (OR=1.077, 95%CI 1.009-1.149, p=0.026) and the G alleles (OR=1.052, 95%CI 1.025-1.080, p<0.000) on the odds of elevated blood pressure. Women in whom HDL levels were not significantly reduced, had higher selenium levels (p=0.007). CONCLUSIONS: 1. The effect of selenium on MetS and its components has not been demonstrated. 2. The effect of individual alleles of the PPAR-γ gene on MetS and its components was not demonstrated. 3. The concentration of selenium may affect waist circumference in carriers of the G allele, and arterial hypertension in carriers of the C and G alleles by affecting the expression of PPAR-γ. 4. Higher selenium concentrations increased the odds of higher HDL levels in the group of subjects meeting the MetS criteria.

The aim of this study was to a) evaluate the concentration of Se and Hg and their relationship in the tissues of 4 species of wild mammals, including Se:Hg molar ratios, and b) evaluate the effect of the analysed elements and their mutual proportions expressed as Se:Hg molar ratio, on the antioxidant system in the tissues of the tested animals. The study was performed on 31 animals belonging to four species: wild boar, red fox, roe deer, brown hare. Determination of Hg in liver, kidney and muscle of animals was performed using an AMA 254 mercury analyser. Total Se concentrations were determined using the spectrofluorometric method. In omnivores demonstrated higher Se concentrations in all analysed organs compared to the herbivores. The highest concentration of Hg was found in the kidneys of the tested animals, and the lowest in the muscles. High and moderate correlation between Se and Hg was observed in the liver of omnivorous, while in herbivores this correlation was weak. In all analysed samples, the Se:Hg molar ratios were above 1 (min: liver 5.9, max: kidney 110). Generally, the highest Se:Hg ratio values were found in kidney and the lowest in liver of tested animals. No significant correlation was found between GPx, GST and SOD activity and Se or Hg concentration in analysed organs. But it was observed that Se:Hg molar was negatively correlated with CAT activity in the most samples. The obtained results may suggest that omnivorous animals demonstrate greater Hg sequestration in the liver than herbivores, which has been proposed as one of the mechanisms of Se antagonistic action towards Hg. The ratio between Se and Hg, rather than the concentration of these elements in organs, affected the antioxidant status in the animal organism, specifically the CAT activity.

The aim of this study was to determine the effect of probiotics on gut microbiota, on carcass and meat quality and on mineral contents in the longissimus lumborum (LL) muscle in pigs. The research was carried out with 120 hybrid pigs deriving from Naïma sows and P-76 boars. Pigs from the experimental group received the EM®Bokashi probiotic (Greendland Technologia EM®, Janowiec n/Wisla, Poland) in their feed (containing Saccharomyces cerevisiae, Lactobacillus casei and Lactobacillus plantarum). The study showed that EM®Bokashi probiotic supplementation resulted in a significantly higher count of lactic acid bacteria (LAB) and yeasts in the feed, a lower number of Clostridium in the mucosa and colorectal digesta as well as a lower Enterobacteriaceae count in the colorectal digesta. The research showed that carcasses of the pigs who received the EM®Bokashi probiotic had a higher lean percentage and lower fat content than the carcasses of the control fatteners. Diet supplementation with the EM®Bokashi probiotic resulted in a lower pH and technological yield (TY) and a higher drip loss and shear force at a lower protein content in LL muscle. Moreover, the administration of the probiotic to fatteners resulted in higher yellowness (b*) and saturation (C*) and higher concentrations of Na, Mg and Se in meat.

This study examined the concentration of total mercury (THg) and selenium (Se), as well as the molar ratio of Se:THg in hair samples of terrestrial animals. THg and Se concentrations were measured from the hair of raccoons (Procyon lotor) and European wildcats (Felis s. silvestris) from Germany and Luxembourg. Median THg concentrations in hair from raccoons and wildcats were 0.369 and 0.273 mg kg-1 dry weight (dw), respectively. Se concentrations were higher in the hair of raccoons than of wildcats (0.851 and 0.641 mg kg-1 dw, respectively). Total mercury concentration in hair of raccoons from Luxembourg was almost 5× higher that found in hair of raccoons from Germany; however, Se concentration was similar. Thus, molar ratio of Se:THg was ~4× higher in the hair of raccoons from Germany than those from Luxembourg. Significant negative correlation was found between THg concentration and Se:THg molar ratio in both wildcats and raccoons.

The aim of the study was to evaluate Hg and Se concentrations and Se:Hg molar ratios in the placenta, umbilical cord and fetal membranes, and to examine the relationship between the concentrations of the elements and selected factors. The study material consisted of the placenta, umbilical cord and fetal membranes obtained from 91 healthy women from northwestern and central Poland. In our study mean Hg and Se concentrations in afterbirth were ~ 0.01 mg/kg dry weight (dw) and ≤ 0.5 mg/kg dw, respectively. Correlation analysis showed negative relationships between placenta weight and Se concentration in the placenta and umbilical cord, as well as between placenta length and Se levels in the umbilical cord. We found negative correlations between THg concentration in the placenta and birth weight and between Se concentration in the placenta and umbilical cord and the morphological parameters of the placenta. Furthermore, we noted new types of interactions in specific parts of the afterbirth. In our study, Se:THg molar ratios ranged from 5 to 626; these values indicate protection against Hg toxicity.

The aim of this study was to compare selenium concentration in the liver and kidneys of wild boar inhabiting industrialized and non-industrialized regions of Poland. Selenium concentrations in organs were determined using spectrofluorometric method. In all the animals studied, Se concentrations were a few times lower than in kidneys which may indicate too low content of this element either in the boar's diet or the presence of a poorly absorbable form of Se. No statistically significant differences were noted in the mean Se concentrations in the liver and kidney of wild boar from industrialized and non-industrialized areas. In the case of wild boar, it seems that the level of selenium in their organs is more dependent on geochemical conditions in the specific feeding ground than on the scale of regional industrialization, and that this situation is most likely related to the specificity of wild boar feeding.

Due the long-term nature of joint tissue remodeling processes, knee structures including meniscus and anterior cruciate ligament (ACL) can be a model for studying the bioaccumulation of mercury (Hg) and selenium (Se). The accumulation and retention of Hg in human tissues can have a negative effect on the proper functioning of homeostasis-affecting organisms. A factor of chronic poisoning with Hg forms is probably the Se:Hg ratio in tissues. Se:Hg molar ratios below one may increase Hg toxicity potentials, while molar ratios that approach or exceed one effectively may protect against Hg toxicity. Therefore, the aim of the study was to determine total mercury (THg), Se, and Se:THg molar ratios in the cartilage, meniscus and ACL of patients with osteoarthritis (OA) from northwestern Poland. In all studied samples (n=95), we observed higher Se than THg concentration. Taking into consideration the biological factors, we found significantly higher THg levels in the cartilage of women, patients under 65 years of age, patients without hypertension and in the ACL of patients with spinal degenerative disease. We found higher Se levels in the meniscus in women than in men. In all studied parts of the knee joint, we found the Se:THg molar ratio higher than one, which suggests that the joint forming structures are not much exposed to THg. Moreover the results reported here may provide a basis for establishing reference values for the meniscus and ACL in patients with OA who had undergone knee replacement surgery.

The aim of this study was to investigate and compare total mercury (Hg), selenium (Se), and Se:Hg molar ratios in fish muscles (phytophages n=3; benthophages n=32; predators n=5) and semiaquatic carnivores, including piscivores (the European otter n=8, the feral American mink n=7) and the omnivorous raccoon (n=37) from a riverine European ecosystem in a Se-deficient area. The Hg concentration in fish reached 0.337µg/g dry weight, dw (0.084µg/g wet weight, ww). We found significant differences among Hg levels in tested vertebrate groups (predators vs benthophages: 0.893 vs 0.281µg/g; piscivores vs omnivores: 6.085 vs 0.566µg/g dw). Fish groups did not differ in Se concentrations, with a mean value of 0.653µg/g dw. Significant differences were revealed between Se levels in piscivorous and omnivorous carnivores (0.360 vs 0.786µg/g dw, respectively). Fish Se:Hg molar ratio values were >2.2. Benthophages had higher the ratio than predators but similar to phytophages. Among carnivores, piscivores had much lower the ratio than raccoon (0.14 vs 3.75) but raccoon and fish medians did not significantly differ. We found almost two times higher Se levels in fish and raccoons compared to piscivores, possibly resulting from lower fish Se digestibility by piscivores in contrast to higher absorption of plant Se by many fish and omnivorous raccoons. Considering that a tissue Se:Hg molar ratio <1 may be connected with a Hg toxicity potential increase, we assume that piscivores in Se-deficient area are in worse situation and more exposed to Hg than fish and omnivores.

The aim of the study was to evaluate the effect of low concentrations of selenium in the environment on the activity of selected antioxidant enzymes: Se-GSHPx, total GSHPx, SOD, CAT, and GST as well as fatty acid profile in the livers of brown hares during winter and spring. Liver tissues obtained from 20 brown hares collected in the north-eastern Poland in the winter and spring season were analyzed. In the tissue analyzed, a significantly lower level of selenium was noticeable in the spring compared to winter; however, values measured in both seasons indicated a deficiency of this element in the analyzed population of brown hares. There were no differences found that could indicate the influence of Se deficiency on the activity of antioxidant enzymes. The determined activity of antioxidant enzymes and fatty acid composition suggest a negligible impact of the low concentration of Se on the analyzed biochemical parameters of brown hare livers.

Although the relationship between mercury (Hg) and selenium (Se) has been studied in wild birds in areas with sufficient or excessive Se levels, little is known about this relationship in areas where the supply of Se is limited. As Hg detoxification is based on the production of biologically inactive Hg-Se complexes, the aim of this study was to investigate the relationships between the concentrations of total mercury (THg), methylmercury (MeHg), inorganic mercury (InHg=THg-MeHg), percent MeHg of THg, Se and molar ratios (THg:Se, MeHg:Se, InHg:Se) in the breast muscle (n=16) of the piscivorous common mergansers (Mergus merganser) from a Se-deficient and moderately Hg-polluted area in Poland. Mergansers were divided into two groups differing in condition (A-very good condition; C-moderate condition). Concentrations of THg, MeHg and Se were determined by atomic absorption spectrometry, modified gas chromatography atomic fluorescence spectroscopy, and spectrofluorometric methods, respectively. In all studied mergansers, mean concentrations of THg, MeHg, InHg, and Se in muscle were 2.63, 1.92, 0.46, and 0.54µgg(-1)dw, respectively. THg and MeHg concentrations in the muscle of group A mergansers were greater than in group C. The ratio of THg:Se was higher in group A than in group C (2.32 vs 1.36; p<0.01), as well as the molar ratio of MeHg:Se (A vs C: 1.98 vs 1.03; p<0.05). Comparisons between mergansers from Poland and Canada showed similar THg and percent MeHg in the muscle, but mergansers from Poland had several times lower Se levels and higher THg:Se ratios (>2.0) than the Canadian mergansers and other European and North American waterbirds. We found statistically significant positive correlations (MeHg-THg, percent MeHg/THg-MeHg, percent InHg/THg-InHg, THg:Se-THg, MeHg:Se-THg, THg:Se-MeHg, MeHg:Se-MeHg, InHg:Se-InHg, MeHg:Se-THg:Se) and some negative correlations (percent InHg/THg-MeHg, percent MeHg/THg-InHg, THg:Se-Se, MeHg:Se-Se). As THg and percent MeHg in the studied mergansers were similar to populations living in non-Se-deficient areas, it is likely that different mechanisms of muscle Hg detoxification have evolved in mergansers populations living in Se-deficient areas.

The subject of the present study was the piscivorous common mergansers (Mergus merganser). The total mercury (THg), methylmercury (MeHg), selenium (Se) inorganic mercury (InHg; THg - MeHg), percentage of THg that is MeHg (%MeHg), molar ratios (THg:Se, MeHg:Se, InHg:Se), and their mutual relations in livers and kidneys were determined in ducks from an Se-deficient area in Poland. The authors verified a hypothesis that, as a result of living in an Se-deficient region, mergansers from Poland should have higher THg:Se ratios than other waterbirds with similar THg tissue levels. Although a comparison of healthy mergansers from Poland and Canada showed similar THg tissue contents, the group in the present study had a few times lower Se levels and higher THg:Se ratios (overall means >1.7 in both livers and kidneys in all studied individuals) than the Canadian group and other European and North American waterbirds. The authors found significant correlations between various relations, including MeHg-THg, InHg-THg, Se-THg, %MeHg-THg, InHg/%THg-THg, %MeHg/THg-InHg, %InHg/THg-InHg, MeHg:Se-THg:Se, InHg:Se-THg:Se, InHg:Se-MeHg:Se in liver and InHg-THg, Se-THg, Se-InHg, %MeHg-THg, %MeHg-InHg, %InHg/THg-InHg, THg:Se-THg, InHg:Se-THg, MeHg:Se-MeHg, THg:Se-InHg, InHg:Se-InHg, and InHg:Se-THg:Se in kidney. It is likely that the main factor responsible for the high value of THG:Se ratio (>1) in mergansers from Poland is Se deficiency in central and northern Europe. Therefore, this element is unlikely to participate in the detoxification of Hg in these birds.

The aim of the study was to evaluate the activity of glutathione peroxidase (GSH-Px) and the concentration of selenium in Holstein-Friesian cows at different stages of lactation. Selenium was determined spectrofluorimetrically and GSH-Px activity using a Sigma CGP1 Glutathione Peroxidase Cellular Activity Assay kit. Mean serum selenium concentration was highest in early-lactation multiparous cows (0.18 µg/ml) and the lowest in dry cows (0.111 µg/ml). In early lactation, serum selenium concentration was significantly (P ≤ 0.01) higher in multiparous cows than in cows from the other groups. Mean GSH-Px activity in the serum of dry cows was over twice lower than in late-lactation cows (P ≤ 0.01) and over four times lower than in first-calving heifers and multiparous cows in early lactation (P ≤ 0.01). The coefficients of Spearman's rank correlation between GSH-Px activity and selenium concentration in the cows at different stages of lactation were not significant. A significant (P ≤ 0.01) mean positive correlation (0.46) was found between GSH-Px activity and serum selenium concentration for all the cows analysed together. The highest Se concentration and GSH-Px activity found in the serum of cows during the first stage of lactation may suggest that the generation of reactive oxygen species and their derivatives was higher during this period compared to the other stages, thus placing the cows at a greater risk of oxidative stress. It is therefore essential to give particular attention during this period to meeting the cows' requirement for selenium and other feed components that increase, directly or indirectly, the capacity of the body's antioxidant system.

The influence of diet ingredients and its supplementation with chosen B group vitamins on concentration of selenium in blood serum and tissues and activity of glutathione peroxidase in blood and liver of male rats was examined in the conducted experiment. The animals, aged 5 months, were divided into three groups and fed ad libitum with granulated mixes. Group I with basic mix containing among other things full grains, Group II with modified mix in which full grains were exchanged for wheat flour and in part with saccharose and Group III with modified mix supplemented in excess with vitamins B1, B2, B6 and PP. The experiment was conducted for six weeks during which the amount of consumed feeding stuff was calculated currently and once a week body mass of the animals was checked. When the experiment was finished the activity of GSH-Px was determined by spectrophotometric method in blood and liver whereas concentration of selenium in blood serum, muscles and in liver by fluorometric method. It was ascertained that the change of diet ingredients and its supplementation with chosen group B vitamins was in favour of lowering the amount of selenium in the examined tissues, and the decrease was not only the result of lower amount of the consumed element, but also of its increased usage, forced by the changes taking place under the influence of diet components and its supplementation.

The present study was undertaken to evaluate the concentration of selenium and selected heavy metals and their possible relationship in serum of 25 healthy lactating cows (Montbéliarde) reared in a non-polluted area, in the western part of Poland. Cadmium, lead, copper, and zinc concentrations were determined by inductively coupled plasma-atomic emission spectrometry and Se concentration was determined fluorimetrically. The content of Se, Zn, and Cu was 0.083 ± 0.026, 0.629 ± 0.413, and 0.152 ± 0.042 µg/mL, respectively. The presence of the Cd and Pb was found in all serum samples. The mean concentration of these metals was 0.0009 ± 0.0008 and 0.018 ± 0.016 µg/mL, respectively. Analysis of correlations between Se and toxic metals showed a negative and significant (P < 0.05) relationship between selenium concentration and lead and cadmium concentration in the serum of the animals studied, with correlation coefficients of r = -0.595 and r = -0.618, respectively. For copper and zinc, this relationship was also negative but not significant (r = -0.255 and r = -0.203). Our study demonstrated that the level of toxic metals decreased as serum selenium concentration increased. It seems necessary to conduct further research on the interactions between these elements in blood, serum, and target organs concomitantly with the determination of their intake.

The purpose of this study was to determine glutathione peroxidase (GSHPx) activity in the liver of red deer in relation to selenium concentrations in the liver, as well as to evaluate changes in GSHPx activity according to sex, body weight and season of the year. Total selenium concentration in the liver of red deer averaged 0.095 ± 0.018 µg/g of wet weight. GSHPx activity in the liver of red deer ranged widely from 4.4 to 45.8 U/g of protein. Females were characterized by higher GSHPx activity compared to males (21.2 vs. 17.0 U/g protein). The highest GSHPx activity was recorded in autumn and the lowest in summer. The lowest GSHPx activity in the liver was found in the heaviest animals (>100 kg body weight), averaging 14.0 U/g protein. Animals weighing <66 kg and 66-100 kg were characterized by similar activity of 25.1 and 24.5 U/g, respectively. Despite the differences in GSHPx activity according to sex, body weight and season of the year, these factors had no significant effect on the activity of this enzyme. The main factor regulating GSHPx activity in the liver of examined red deer was selenium concentration.

The aim of the study was to compare selenium concentrations in different organs of roe deer from northwestern Poland. Samples of liver, kidneys, heart and lungs, collected from 74 roe deer shot during the hunting seasons of 2008-2009 in northwestern Poland, were studied. Selenium concentration in the organs was determined spectrofluorimetrically. Mean selenium concentration was 0.06 µg/g w.w. in the liver, 0.41 µg/g w.w. in the kidneys and 0.05 µg/g w.w. in the heart and lungs. Season had a significant effect on selenium concentration in the liver, kidneys, lungs and heart. In all the organs, the highest selenium concentration was found in spring and the lowest in autumn and winter. All animals studied were deficient in selenium. The low selenium concentration in the liver or heart can disturb their function, and in the future, it may be a factor contributing to the population decline of roe deer in the northwestern part of Poland.
