RESUMO

Species of the Diospyros L. genus (Ebenaceae family) have been largely used in traditional medicine for the treatment of several diseases, especially infectious ones. To date, active major compounds such as naphthoquinones, triterpenoids, and tannins have been isolated and pharmacologically validated from Diospyros species. The present study summarizes the information available in the literature on the species described in the Flora of Mozambique. To do so, scientific databases (e.g., PubMed, Scopus, Web of Science, and Google Scholar) were searched using various keywords and Boolean connectors to gather and summarize the information. Of the 31 native and naturalized species in the Flora of Mozambique, 17 are used in different regions of Africa and were described for their traditional uses. They were reported to treat more than 20 diseases, mostly infectious, in the gastrointestinal and oral cavity compartments. This work provides an overview of the therapeutical potential of Diospyros species and explores novel insights on the antimicrobial potential of extracts and/or isolated compounds of these Mozambican species.

The use of amphiphilic block copolymers to generate colloidal delivery systems for hydrophobic drugs has been the subject of extensive research, with several formulations reaching the clinical development stages. However, to generate particles of uniform size and morphology, with high encapsulation efficiency, yield and batch-to-batch reproducibility remains a challenge, and various microfluidic technologies have been explored to tackle these issues. Herein, we report the development and optimization of poly(ethylene glycol)-block-(ε-caprolactone) (PEG-b-PCL) nanoparticles for intravenous delivery of a model drug, sorafenib. We developed and optimized a glass capillary microfluidic nanoprecipitation process and studied systematically the effects of formulation and process parameters, including different purification techniques, on product quality and batch-to-batch variation. The optimized formulation delivered particles with a spherical morphology, small particle size (dH < 80 nm), uniform size distribution (PDI < 0.2), and high drug loading degree (16 %) at 54 % encapsulation efficiency. Furthermore, the stability and in vitro drug release were evaluated, showing that sorafenib was released from the NPs in a sustained manner over several days. Overall, the study demonstrates a microfluidic approach to produce sorafenib-loaded PEG-b-PCL NPs and provides important insight into the effects of nanoprecipitation parameters and downstream processing on product quality.

INTRODUCTION: Performance tests of the upper limb have been frequently used in the evaluation of individuals to identify risk of injury and to determine improvement in physical performance. Understanding the variables related to the performance of the tests is important for better applicability and interpretation of test results. The aim of this cross-sectional study was to investige the correlation between isometric strength of scapular, shoulder and spine muscles with physical performance test of upper limb in men and women. EXPERIMENTAL: Forty-nine healthy and physically conditioned individuals, men (n = 24) (22,7 ± 3,2 years; 74,6 ± 8,1 Kg; 176,7 ± 7,1 cm) women (n = 25) (23 ± 2,7 years; 61,5 ± 9,1 Kg; 163,2 ± 5,6 cm), were evaluated for shoulder abductor isometric strength (S-ABD); Middle (MT) and lower (LT) trapezium; lateral shoulder rotators (S-LR); flexors (T-FLEX), lateral incliners (T-INCL), and trunk extenders (T-EXT). Evaluation of upper limb physical performance was carried out by Upper Quarter Y Balance test (YBT-UQ) and Chain upper extremity stability test (CKC-UEST). Pearson's test (r values) p ≤ 0.05 was used to analyze the correlation. RESULTS: The results reveal strong correlations between S-ABD and CKC-UEST in male group; and strong correlations in female group between S-ABD strength with CKC-UEST; and S-LR with all YBT-UQ directions and score and between T-EXT strength with CKC-UEST. CONCLUSION: Therefore, strength of shoulder and spine stabilizers influences performance in upper limb tests, but with different correlations as to sex. To improve test performance, it may be important to increase the strength of the shoulder and spine muscles.

BACKGROUND: Understanding the biogenesis pathways for the functional expression of recombinant proteins, in particular membrane proteins and complex multidomain assemblies, is a fundamental issue in cell biology and of high importance for future progress in structural genomics. In this study, we employed a proteomic approach to understand the difference in expression levels for various multidomain membrane proteins in L. lactis cells grown in complex and synthetic media. METHODOLOGY/PRINCIPAL FINDINGS: The proteomic profiles of cells growing in media in which the proteins were expressed to high or low levels suggested a limitation in the availability of branched-chain amino acids, more specifically a too limited capacity to accumulate these nutrients. By supplying the cells with an alternative path for accumulation of Ile, Leu and/or Val, i.e., a medium supplement of the appropriate dipeptides, or by engineering the transport capacity for branched-chain amino acids, the expression levels could be increased several fold. CONCLUSIONS: We show that the availability of branched chain amino acids is a critical factor for the (over)expression of proteins in L. lactis. The forward engineering of cells for functional protein production required fine-tuning of co-expression of the branched chain amino acid transporter.

The pollen specificity of the Arabidopsis arabinogalactan protein (AGP) genes AGP6 and AGP11 suggests that they are integral to pollen biogenesis, and their high percent of sequence similarity may indicate a potential for overlapping function. Arabidopsis agp6 agp11 double null mutants have been studied in our laboratory, and in the present work, we characterize the germination and growth of its pollen. When compared to wild type, mutant agp6 agp11 pollen displayed reduced germination and elongation, both in vivo and in vitro, and precocious germination inside the anthers, provided that sufficient moisture was available. This characteristic was not observed in wild type plants, even in water content conditions which for the mutant were sufficient for pollen germination. Therefore, an additional distinctive phenotypic trait of arabinogalactan proteins AGP6 and AGP11 may be to avert untimely germination of pollen. Such AGPs may control germination through water uptake, suggesting an important biological function of this gene family in pollen.
