RESUMO

Cervical cancer ranks fourth in incidence among women worldwide. Cisplatin is currently the first-line drug of treatment for cervical cancer; however, it causes serious adverse effects. Therefore, it is crucial to explore natural products for cervical cancer treatment. Prosopis laevigata is a medicinal plant frequently used for ophthalmological and gastrointestinal infections. In this study, we used the MTT cell viability assay to evaluate the cytotoxic effect of a wood extract from Prosopis laevigata (Extract T7) in SiHa, HeLa, Ca Ski, and C-33 A cancer cell lines. Phosphatidylserine translocation and cell cycle evaluations were performed to determine the mechanism of cellular death. The extract's safety was evaluated using the Ames test with Salmonella typhimurium strains, in vivo acute toxicity assay, and repeated dose toxicity assay in mice. We also identified phenolic compounds of Extract T7 through liquid chromatography/mass spectrometry. Naringin, catechin, and eriodictyol demonstrated a higher concentration in Extract T7. Additionally, Extract T7 exhibited a cytotoxic effect against cervical cancer cells, where C-33 A was the most sensitive (IC50= 22.58 ± 1.10 µg/mL and 14.26 ± 1.11 µg/mL at 24 h and 48 h respectively). Extract T7 induced death by apoptosis and cell cycle arrest in the G2 phase in C-33 A. Extract T7 was not mutagenic. No toxicological effects were observed during acute toxicity and repeated dose toxicity for 28 days. Therefore, further evaluations of Extract T7 should be conducted to identify the complete mechanism of action for potential anti-tumoral activity and safety before conducting studies in animal models.

RESUMO

The study of medicinal plants for cancer treatment has gained attention due to an increasing incidence of cancer worldwide and antineoplastics-related undesirable secondary effects. Most of the natural products of medicinal plants that have been evaluated for cytotoxic activity, are derived from leaves, bark, roots and flowers. However, natural products derived from wood have demonstrated a cytotoxic effect with promising results. Moreover, some fractions and compounds have been isolated of wood in order to increase the effect. This review presents in vitro experimental evidence of cytotoxic effect of natural products from wood against cancer cell lines. It also provides considerations and recommendations to obtain herbal medicines over time.

Quercus sideroxyla is a wood species whose bark has phenolic compound and should be considered to be bioactive; the hypoglycemic and genotoxic properties of Q. sideroxyla bark were evaluated in this study. Total phenolic compound was determined in crude extract (CE) and organic extract (OE). The OE has the highest amount of phenols (724.1 ± 12.0 GAE/g). Besides, both CE and OE demonstrated effect over the inhibition of α-amylase in vitro. Hypoglycemic activity was assessed by glucose tolerance curve and the area under curve (UAC); OE showed the highest hypoglycemic activity. In addition, diabetes was induced by streptozotocin (65 mg/kg) and the extracts (50 mg/kg) were administered for 10 days; OE showed hypoglycemic effect compared with diabetic control and decreased hepatic lipid peroxidation. Acute toxicity and genotoxicity were evaluated in CE; results of acute toxicity did not show any mortality. Besides, the comet assay showed that CE at a dose of 100 mg/kg did not show any genotoxic effect when evaluated at 24 h, whereas it induced slight damage at 200 mg/kg, with the formation of type 1 comets.

Evaluation of the phenolic compounds and antioxidant activity of Cedrela odorata L. and Juglans regia L. bark extracts was performed in vitro. Juglans regia showed greater extract concentration and higher antioxidant activity. Hypoglycemic activity in rats was assessed by generating a glucose tolerance curve and determining the area under the curve (AUC). Diabetes was later induced by an injection with streptozotocin (65 mg/kg of b.w.) and confirmed after 24 hours. The extract was administered (200 mg/kg b.w.) over 10 days, and blood glucose was monitored and compared with a control group. The glucose AUC showed a hypoglycemic effect of J. regia and C. odorata in normal rats. Both extracts reduced hepatic lipid peroxidation in diabetic rats. Polyphenolic extracts reduced cholesterol levels in a hypercholesterolemic mouse model and decreased hepatic lipid peroxidation. Polyphenolic extract doses of 100 and 200 mg/kg b.w. were administered alone or with cyclophosphamide (CPA) 50 mg/kg ip, which was used as a positive control. Analyses were performed using leukocytes in a comet assay after 4 and 24 h of treatment. Genotoxic effects were evaluated by the comet assay, which showed that while J. regia extract had no effect, C. odorata extract induced slight damage at 200 mg/kg, with the formation of type 0 and 1 comets.

The oily (hexane) and aqueous extracts from apple seeds (Malus domestica Borkh –Rosaceae-) cultivars Winter Banana (WB), Winter Permain (WP) and Blanca de Asturias (BA) have shown significant differences in phenolic content and antioxidant activity. BA seed oily extracts and WB seed aqueous extracts have presented the best scavenging capacity of DPPH• radical and inhibition of hydroxyl radicals. In the biological assay apple seed extracts showed protection against low density lipoproteins (LDL) oxidation, which might fight the risk of atherosclerosis. The BA oily seed extract may have potential use as nutraceutical ingredient.


La fracción oleosa (obtenida con hexano) y extractos acuosos de harina de semilla de manzana (Malus domestica Borkh–Rosaceae-) variedades Winter Banana (WB), Winter Permain (WP) y Blanca de Asturias (BA) mostraron diferencias significativas en el contenido de polifenoles independientemente de la variedad. Las diferentes fracciones mostraron capacidad de atrapamiento del radical DPPH• y de inhibición del radical hidroxilo, siendo la fracción oleosa de las semillas de BA la mejor en los dos análisis; en el caso de los extractos acuosos, las semillas de WB presentaron la mejor respuesta. En la evaluación biológica los diferentes extractos mostraron un efecto protector contra la oxidación de las lipoproteínas de baja densidad (LDL) lo cual puede disminuir el riesgo de ateroesclerosis. La fracción oleosa de semillas de BA mostró mayor potencial como ingrediente nutracéutico.
