The objectives of this study were (1) to analyse the relationship between the colour of the protectors and the outcome of 895 matches in the six Olympic Games in which taekwondo has been included, and (2) to analyse the effect of confounding factors through the different degrees of asymmetry between contestants. Data were recorded on the colour of each athlete's protectors, the scoring system, the sex of the athlete, the weight category, the round of competition, the winner of the match, the method of victory and the points scored by each athlete. Among the main results, a relationship emerged between male athletes wearing red and winning the match (p = 0.034) under the manual scoring system. There were relationships between female athletes wearing blue and winning the match in the quarterfinal (p = 0.014) and last 16 rounds (p = 0.021) using the manual and electronic scoring systems respectively. In female athletes, relationships emerged between wearing blue and winning the match with small (p = 0.008) and medium (p = 0.047) asymmetry under the manual system and with large (p = 0.036) asymmetry under the electronic system. The electronic system had a positive impact on the fairness of Olympic competition. Red tends to gain in importance as the asymmetry between the two athletes decreases, but not enough to give a competitive advantage. The results for Olympic competition held in the now concluded era of the manual system confirmed the presence of the colour effect as a result of psychological factors attributable to referees and judges.

The intestinal morphology and function can be compromised in pigs exposed to heat stress (HS), partly due to increased production of reactive-oxygen species. Because methionine (Met) functions as intracellular antioxidant, the requirement of Met may be increased in HS-pigs. The effect of dietary supplementation with dl-Met above requirement on performance, small intestine morphology, antioxidant enzymes activity, amino acid transporters expression, and serum concentration (SC) of free AA in HS-pigs was evaluated. A basal wheat-soybean meal diet was formulated to meet 100% Met requirement with the other indispensable AA exceeding at least 20% their requirement. Sixty individually housed pigs (23.0 ± 2.4 kg BW, 12 pigs per treatment) were randomly assigned to five treatments: TN100, thermal-neutral (22.7 °C) housed pigs fed the basal diet; HS100, HS120, HS140, HS160; HS-pigs (29.6 °C to 39.4 °C) fed the basal diet supplemented with dl-Met to contain 0%, 20%, 40%, and 60% dl-Met above the requirement, respectively. Pigs had free access to feed and water during the 21-d trial. Blood samples were collected on day 18 to analyze the absorptive AA-SC. The effect of ambient temperature (HS100 vs. TN100), as well as the linear and quadratic effects of increasing Met levels in the diets for HS-pigs were analyzed. The HS100 pigs gained less weight than TN100 and HS120 pigs (P < 0.01); gain:feed was also higher in HS120 pigs than in HS100 pigs (P ≤ 0.05). Feed intake of TN100 pigs was higher than that of HS-pigs fed the dl-Met supplemented diets (P < 0.05). Villi height reduced in pigs HS, but Met supplementation quadratically increased it (P < 0.05). Superoxide dismutase and catalase activities, reduced glutathione concentration, and relative expression of B0AT2 in ileum decreased (P < 0.05), but glutathione peroxidase activity increased in HS-pigs. dl-Met supplementation linearly affected catalase and glutathione peroxidase activities, as well as the relative expression of b0,+AT in jejunum (P < 0.05) of HS-pigs. The SC of Ile, Leu, Lys, Phe, and Val were higher in HS100 pigs than in TN100 pigs (P < 0.05). Graded levels of supplemental dl-Met in diets for HS-pigs linearly decreased SC of Ile, Leu, and Val (P < 0.05), tended to decrease His, Lys, and Thr (P < 0.10), and increased Met (P < 0.01). In conclusion, HS had negative effect on weight gain and intestinal morpho-physiology; however, it was ameliorated by adding 20% Met above the requirement in diets for growing pigs.


The exposure of pigs to ambient temperature above their comfort zone affects several functions of the small intestine, especially those related with digestion of feed and absorption of nutrients, which in turn reduces the availability of nutrients for growth. Amino acids such as methionine are involved in multiple functions of intestinal cells. Thus, methionine supplementation may help pigs to overcome the negative impact of their exposure to high ambient temperature. Indeed, methionine supplementation to the diet increased growth rate and feed efficiency of pigs housed under heat stress, which was presumably associated with an improvement in the utilization of the absorbed amino acids.

Electromyographic biofeedback (EMG-BF) is a therapeutic technique that has been used successfully in the rehabilitation of injuries. Although it has been applied to athletes, its use in this field is not very widespread. The objective of this study is to analyze its effectiveness in the recovery of electromyographic activity of the quadriceps after meniscectomy, evaluated through isometric contraction of the vastus lateralis. The sample comprised ten professional footballers in the Spanish League (2nd Division A) who had previously suffered a meniscus injury in their knee and had undergone a meniscectomy. The intervention consisted of EMG-BF treatment lasting between 6 and 10 sessions. The electromyographic signal was recorded using a Thought Technology ProComp Infiniti 8-channel biofeedback unit with a sampling rate of 2048 samples/second. For each session, a within-subject ABA design of 6 or 10 trials per session was used, with three pre- and three post-measures, which determined the gain for each session. The results indicated (1) improvements in all cases, (2) EMG-BF was effective, (3) the working model was statistically significant with an explained variance of between 67% and 75%, and (4) the generalizability analysis showed that the results are reliable and generalizable. The results indicate that EMG-BF is effective in neuromuscular rehabilitation after this type of intervention.

Although research on the effect of color in taekwondo has involved several international competitions, no previous study has investigated the presence of this phenomenon in national-level competitions. The main objective of this study was to analyze the relationship between the color protectors and success in 1155 taekwondo matches of the Italian and Uzbekistan Senior Championships (ITA-SC and UZB-SC) (2019 and 2021). The results showed no relationship between the color protectors and the match outcome, in both ITA-SC and UZB-SC (p = 0.71, V = 0.01; p = 0.61, V = 0.02). Moreover, no relationship emerged between the color protectors and the match outcome in the four editions of the SC. Stratifying analyses by weight category and sex, males showed positive relationships between the color blue/red and winning the match in 3 and 1 of 16 weight categories, respectively. Contrary, females showed positive relationships between the color blue/red and winning the match in 1 and 3 of 16 weight categories, respectively. Analyzing the two national contexts found that, in both the Italian and Uzbek contexts, matches in 2 and 2 of 16 weight categories were won by athletes wearing blue and red protectors, respectively. Significant relationships emerged between the color blue and winning the match with small asymmetry in the men's UZB-SC and between the color red and winning the match with large asymmetry in the female ITA-SC. The implementation of the electronic point recording system for the body and head has had a positive impact on fairness in national taekwondo competitions, did not detect any effect of color related to cultural context, and did not allow for the color red to tip the scales between losing and winning in matches between athletes of similar ability and strength.

Resumen Se realiza una revisión narrativa que plantea una reflexión acerca del rol de la oxigenación hiperbárica en la recuperación de los intoxicados con monóxido de carbono (ICO). La relación presión de tratamiento de oxigenación hiperbárica (TOHB), o sea dosis de oxígeno, y demora en su implementación son descriptas en esta revisión. Se presentan 9 casos de pacientes con ICO tratados con TOHB a 1,45 ATA (Atmósferas absolutas) por falta de acceso a TOHB de alta presión. Si bien es necesario investigación adicional, sugerimos que esta modalidad terapéutica a 1,45 ATA para ICO debe ser elegida frente al oxígeno normobárico, y considerada cuando las instalaciones de alta presión no están disponibles a distancias razonables.


Abstract A narrative review that raises a reflection about the role of hyperbaric oxygenation in the recovery of monoxide carbon (CO) poisoning is carried out. A description of the relationship of the pressure of hyperbaric oxygen therapy (HBOT), oxygen dosage, and the delay in its implementation was done. Nine cases of intoxications treated with HBOT at 1.45 ATA due to lack of access to high-pressure HBOT were presented. While additional research is necessary, we suggest that this therapeutic modality at 1.45 ATA (Absolute Atmospheres) should be chosen instead of normobaric oxygen therapy for CO poisoning, and considered when high pressure facilities are not available at reasonable distances.

Observational methodology uses validated observational tools to collect information in sports with multiple variables that interact in the sporting context. Given the importance of data quality for observational tools, the purpose of this study was to design, validate, and test the reliability of a mixed observational instrument combining field formats and category systems for analyzing technical and tactical actions in an Olympic taekwondo (TKD) tournament. The instrument collects information of six criteria and 25 categories of the tactical and technical actions, kicking zone, laterality, kicking leg, guard, and score. A total of 2 374 actions were analyzed from 10 bouts involving 12 competitors in the 2016 Rio de Janeiro Olympic Games. The coefficients derived from the generalizability analysis (G coefficients) were used to check the accuracy, validity, and reliability of the tool created, while lag-sequential and polar-coordinate analysis were used to determine technical and tactical patterns of TKD athletes. The results showed optimal G coefficients and provide us with objective, valid, and reliable information on the bout situation analyzed, allowing us to extrapolate the data from our results to the sample population. The lag-sequential and polar-coordinate analyses showed that the competitors scored one point through direct attacks with linear techniques to the chest and performed circular actions with the back leg before and after scoring one point. Three points are scored by indirect attacks and subsequent counterattacks with spinning kicks to the trunk and head (using linear and circular techniques). These results reveal technical and tactical implications derived from the latest modifications of the regulations of this sport, as well as the need to adapt athletes' training for the next Olympic Games.

Identification of druggable vulnerabilities is a main objective in triple-negative breast cancer (TNBC), where no curative therapies exist. Gene set enrichment analyses (GSEA) and a pharmacological evaluation using a library of compounds were used to select potential druggable combinations. MTT and studies with semi-solid media were performed to explore the activity of the combinations. TNBC cell lines (MDAMB-231, BT549, HS-578T and HCC3153) and an additional panel of 16 cell lines were used to assess the activity of the two compounds. Flow cytometry experiments and biochemical studies were also performed to explore the mechanism of action. GSEA were performed using several data sets (GSE21422, GSE26910, GSE3744, GSE65194 and GSE42568), and more than 35 compounds against the identified functions were evaluated to discover druggable opportunities. Analyses done with the Chou and Talalay algorithm confirmed the synergy of dasatinib and olaparib. The combination of both agents significantly induced apoptosis in a caspase-dependent manner and revealed a pleotropic effect on cell cycle: Dasatinib arrested cells in G0/G1 and olaparib in G2/M. Dasatinib inhibited pChk1 and induced DNA damage measured by pH2AX, and olaparib increased pH3. Finally, the effect of the combination was also evaluated in a panel of 18 cell lines representative of the most frequent solid tumours, observing a particularly synergism in ovarian cancer. Chenopodium ambrosioides L. has been used for centuries as traditional medicine in many clinical situations. The objectives of this study were first to assess the nutraceutical potential of C. ambrosioides L. extract through analyses of its chemical composition and antioxidant properties, followed by assessing toxicity and antioxidative activities on fish splenocytes. The second one was to perform an in vivo study using dietary C. ambrosioides L. extract (0.0, 0.5, 1.0 and 2.0%; w/w) for 15 and 30 days (2-week and 4-week treatments) to assess associated-intestine health status by short-chain fatty production, antioxidant enzyme activities and anti-inflammatory effects on Pacific red snapper (Lutjanus peru). Non-polar and polar fractions were detected by gas chromatography/mass spectrometry (GC-MS) in C. ambrosioides, of which the most abundant compounds were carvacrol, phytol, squalene, vitamin E and sucrose. The extract of C. ambrosioides L. enhanced a considerable antiradical and reducing power; fish splenocytes responded positively with higher (88%) cell viability than control. The production of nitric oxide and superoxide anion, as well as superoxide dismutase and catalase activities, were also enhanced in splenocytes treated with C. ambrosioides L. The in vivo study results showed that acetate was the major short-chain fatty acid found in fish receiving C. ambrosioides L. after week four. Pro-inflammatory cytokine gene expression in intestine was modulated in fish fed with C. ambrosioides L. at any time of the experimental trial. In addition, the histological findings suggested that its extract did not cause inflammatory damage in intestine. Overall, the results suggest that C. ambrosioides L. is safe for immune cells and promoting intestinal health status of fish through antioxidant and anti-inflammatory effects, making it an interesting additive in functional diets.

Individuals with Down syndrome (DS), a genetic condition due to triplication of Chromosome 21, are characterized by intellectual disability that worsens with age. Since impairment of neurogenesis and dendritic maturation are very likely key determinants of intellectual disability in DS, interventions targeted to these defects may translate into a behavioral benefit. While most of the neurogenesis enhancers tested so far in DS mouse models may pose some caveats due to possible side effects, substances naturally present in the human diet may be regarded as therapeutic tools with a high translational impact. Linoleic acid and oleic acid are major constituents of corn oil that positively affect neurogenesis and neuron maturation. Based on these premises, the goal of the current study was to establish whether treatment with corn oil improves hippocampal neurogenesis and hippocampus-dependent memory in the Ts65Dn model of DS. Four-month-old Ts65Dn and euploid mice were treated with saline or corn oil for 30 days. Evaluation of behavior at the end of treatment showed that Ts65Dn mice treated with corn oil underwent a large improvement in hippocampus-dependent learning and memory. Evaluation of neurogenesis and dendritogenesis showed that in treated Ts65Dn mice the number of new granule cells of the hippocampal dentate gyrus and their dendritic pattern became similar to those of euploid mice. In addition, treated Ts65Dn mice underwent an increase in body and brain weight. This study shows for the first time that fatty acids have a positive impact on the brain of the Ts65Dn mouse model of DS. These results suggest that a diet that is rich in fatty acids may exert beneficial effects on cognitive performance in individuals with DS without causing adverse effects.

ß-Glucans are naturally occurring polysaccharides that are produced by bacteria, fungi and yeast. They are considered immunostimulants in fish acting on non-specific defense mechanism. Yeast-derived glucans from cell wall (Sterigmatomyces halophilus, ß-Gluc/Sh) have been used for this purpose in this study. Therefore, an in vitro assay using peripheral blood leucocytes (PBLs) from Pacific red snapper was performed to evaluate the stimulant effects of ß-Gluc/Sh and zymosan A (positive control) for 12 and 24 h and after bacterial challenge with Aeromonas hydrophila at 24 h. In addition, structural characterization of this marine yeast glucan was performed by proton nuclear magnetic resonance (NMR) revealing structures containing (1-6)-branched (1-3)-ß-D-glucan. PBLs responded positively to ß-Gluc/Sh where cell viability was higher than 80%. After challenge, ß-Gluc/Sh was able to inhibit cytotoxicity caused by A. hydrophila, highlighting that the PBLs incubated with ß-Gluc/Sh significantly increased the non-specific immune response, such as phagocytic activity, respiratory burst, nitric oxide and peroxidase activities followed by an increase in superoxide dismutase and catalase activities after 12 and 24 h post-stimulation and after challenge with the pathogen. Regarding induction of antioxidant gene expression, it was more pronounced in stimulated ß-Gluc/Sh leucocytes compared to other groups at all experimental times of the trial and after bacterial challenge. Indeed, our results clearly showed the ability of leucocytes to strongly react to ß-Gluc/Sh with an increase in cytokine gene expression, particularly the IL-1ß, IL-10 and IL-17 genes. These results confirm that S. halophilus yeast-derived ß-glucan, isolated from an extreme marine environment, is beneficial for increasing innate immune response and enhancing resistance against A. hydrophila in vitro.

The ethyl acetate extract (SsAcOEt) from Serjania schiedeana, select fractions (F-6, F-12, F-13, F-14), and one isolated compound, were evaluated in 12-O-tetradecanoylphorbol 13-acetate (TPA) ear edema and kaolin/carrageenan (KC)-induced monoarthritis assays. SsEtOAc induced edema inhibition of 90% (2.0 mg/ear), fractions showed activity within a range of 67-89%. Due to the fact F-14 showed the highest effect, it was separated, yielding a proanthocyanidin-type called epicatechin-(4ß â†’ 8)-epicatechin-(4ß â†’ 8, 2ß â†’ O → 7) epicatechin (ETP). This compound (2.0 mg/ear) provoked 72% of edema inhibition (ED50 = 0.25 mg/ear, Emax = 52.9%). After 9 days of treatment, joint inflammation was decreasing, and on the last day, SsEtOAc (400 mg/kg), F-14 and ETP (10 mg/kg), SsEtOAc (200 mg/kg), methotrexate (MTX) 1.0 mg/kg and meloxicam (MEL) 1.5 mg/kg, produced an inhibition articulate edema of 94, 62, 36, 21, 80, and 54%, respectively. In the joint, pro-inflammatory molecules were elevated in animals without treatment (vehicle group, VEH). Treatments from S. schiedeana induced a decrease in the concentration of interleukin (IL)-1ß, IL-17, and IL-6, and SsEtOAc at a higher dose diminished tumor necrosis factor (TNF-α). IL-10 and IL-4 were fewer in the VEH group in comparison with healthy mice; the animals with treatments from S. schiedeana induced an increment in the levels of these cytokines in joint and spleen.

El objetivo es identificar las acciones técnicas de las karatekas que consiguen puntuar en competición y explorar si el comportamiento técnico puede discriminar entre diferentes niveles competitivos. Se ha registrado el comportamiento técnico de 35 karatekas, modalidad de kumite femenino, en un total de 40 combates (18 de nivel regional/nacional y 22 de nivel internacional). Se ha utilizado un diseño observacional situado en el cuarto de cuadrante de carácter nomotético y de seguimiento multidimensional. El registro se ha realizado con el programa Lince y una herramienta de observación 'ad hoc' formada por un sistema mixto de formatos de campo y un sistema de categorías denominado FCSCSHIAIKUMITE. Se ha utilizado la prueba de Chi-cuadrado (p≤ 0,05) y se ha realizado un análisis de coordenadas polares, escogiendo como conductas focales aquellas relativas a la puntuación y como conductas condicionadas todas aquellas relativas a los demás criterios. Los resultados muestran que solo el criterio puntuación discrimina entre ambos niveles de rendimiento, sumando 152 puntos en nivel internacional frente a 78 en nivel nacional (p= 0,002). El Yuko es la forma de obtener punto más frecuentada en ambos niveles (76,4% vs 86,8%), no obstante, el Ippon es eficiente y frecuentado a nivel internacional (21,7%), y solo representa un 4,4% en combates de nivel nacional. En conclusión, hay dos aspectos que podrían ser de especial atención, el perfeccionamiento de las acciones técnicas de punto más habituales y de las acciones defensivas a esas técnicas, así como la eficacia para obtener más puntuación al utilizarlas (AU)


O objectivo é identificar as acções técnicas das karatekas que conseguem puntuar em competição e explorar se o comportamento técnico pode discriminar entre diferentes níveis competitivos. Registou-se o comportamento técnico de 35 karatekas, modalidade de kumite feminino, num total de 40 combates (18 de nível regional/nacional e 22 de nível internacional). Utilizou-se um desenho observacional situado no quarto de quadrante de carácter nomotético e de rastreamento multidimensional. O registro realizou-se com o programa Lince e uma ferramenta de observação 'ad hoc' formada por um sistema misto de formatos de campo e um sistema de categorias denominado FCSCSHIAIKUMITE. Utilizou-se a prova de Chi-quadrado (p=0,05) e realizou-se uma análise de coordenadas polares, escolhendo como condutas focais aquelas relativas à pontuação e como condutas condicionadas todas aquelas relativas aos demais critérios. Os resultados mostram que só o critério pontuação discrimina entre ambos níveis de rendimento, somando 152 pontos em nível internacional em frente a 78 em nível nacional (p= 0,002). O Yuko é a forma de obter ponto mais frequentada em ambos níveis (76,4% vs 86,8%), não obstante, o Ippon é eficiente e frequentado a nível internacional (21,7%), e só representa um 4,4% em combates de nível nacional. Em conclusão, há dois aspectos que poderiam ser de especial atenção, o aperfeiçoamento das acções técnicas de ponto mais habituais e das acções defensivas a essas técnicas, bem como a eficácia para obter mais pontuação ao as utilizar (AU)


The aim is to identify the technical actions of the karatekas that achieve to mark in competition and explore if the technical behaviour can discriminate between different competitive levels. It has registered the technical behaviour of 35 karatekas, modality of kumite feminine, in a total of 40 fights (18 of national regional/level and 22 of international level). It has used an observational design situated in the chamber of quadrant of nomothetical character and of multidimensional follow-up. The register has made with the program Lince and a tool of observation 'Ad hoc' formed by a mixed system of formats of field and a system of categories designated FCSCSHIAIKUMITE. It has used the proof of Chi-square (p≤ 0,05) and it has made an analysis of polar coordinates, choosing like focal behaviours those relative to the punctuation and like behaviours conditioned all those relative to the other criteria. The results show that only the criterion punctuation discriminates between both levels of performance, adding 152 points in international level in front of 78 in national level (p= 0,002). The Yuko is the form to obtain point more frequented in both levels (76,4% vs 86,8%), nevertheless, the Ippon is efficient and frequented to international level (21,7%), and only represents 4,4% in fights of national level. In conclusion, there are two appearances that could be of special attention, the improvement of the technical actions of point more usual and of the defensive actions to these technicians, as well as the efficiency to obtain more punctuation when using them (AU)

The use of electromyographic biofeedback (EMG-BF) in injury rehabilitation is amply referenced in psychological literature. Despite the pioneering work in the early eighties, in sports, it has hardly transcended its usage in sports injury rehabilitation. A study of twelve cases on professional players from the Spanish league grouped into two different types of pathologies (fractures in the upper limbs and knee injuries) that have been treated to EMG-BF parallel with an established rehabilitation plan is presented by each club’s medical team. An ABA design is used. Each session is recorded online pre and post to determine the EMG gain reached at each point of the session. The procedure is considered to be completed when the player stably reaches criteria established by the hemilateral member. In all cases more than one session tracking is outtaken. All cases include notoriously stable improvements. The overall results support the conclusion that the EMG - BF is effective in these pathologies (AU)


La utilización del biofeedback electromiográfico (BF-EMG) en la rehabilitación de lesiones está sobradamente referenciada en la literatura psicológica. A pesar de los trabajos pioneros en la década de los años ochenta, en el ámbito deportivo, apenas ha transcendido su utilización en la rehabilitación de lesiones deportivas. Se presenta un estudio de doce casos de futbolistas profesionales de la liga española agrupados en dos tipos de patologías distintas (fracturas en los miembros superiores y lesiones de rodilla) que han sido sometidos a un tratamiento de BF-EMG, paralelo al plan de rehabilitación establecido por los servicios médicos de cada club. Se utiliza un diseño ABA. En cada sesión se registra una línea pre y post que permite determinar la ganancia EMG adquirida en cada punto de la sesión. La intervención se da por finalizada cuando el futbolista alcanza de forma estable el criterio establecido por el miembro hemilateral. En todos los casos se realiza una sesión más de seguimiento. Todos los casos presentan mejorías notorias y estables. Los resultados globales permiten considerar que el BF-EMG es eficaz en estas patogías (AU)

Introduction. Converging evidence suggests that PDE-4 (phosphodiesterase subtype 4) plays a crucial role in regulating cognition via the PDE-4-cAMP cascade signaling involving phosphorylated cAMP response element binding protein (CREB). Objective. The primary endpoint was to examine the neurocognitive effects of extract Sceletium tortuosum (Zembrin) and to assess the safety and tolerability of Zembrin in cognitively healthy control subjects. Method. We chose the randomized double-blind placebo-controlled cross-over design in our study. We randomized normal healthy subjects (total n = 21) to receive either 25 mg capsule Zembrin or placebo capsule once daily for 3 weeks, in a randomized placebo-controlled 3-week cross-over design. We administered battery of neuropsychological tests: CNS Vital Signs and Hamilton depression rating scale (HAM-D) at baseline and regular intervals and monitored side effects with treatment emergent adverse events scale. Results. 21 subjects (mean age: 54.6 years ± 6.0 yrs; male/female ratio: 9/12) entered the study. Zembrin at 25 mg daily dosage significantly improved cognitive set flexibility (P < 0.032) and executive function (P < 0.022), compared with the placebo group. Positive changes in mood and sleep were found. Zembrin was well tolerated. Conclusion. The promising cognitive enhancing effects of Zembrin likely implicate the PDE-4-cAMP-CREB cascade, a novel drug target in the potential treatment of early Alzheimer's dementia. This trial is registered with ClinicalTrials.gov NCT01805518.

Docosahexaenoic acid (DHA, 22:6n-3) is a major constituent of nerve cell membrane phospholipids. Besides a role in membrane architecture, DHA is a pleiotropic molecule involved in multiple facets of neuronal biology and also in neuroprotection. We show here that supplementation with DHA (but not arachidonic acid) to mouse hippocampal HT22 cells modulates the expression of genes encoding for antioxidant proteins associated with thioredoxin/peroxiredoxin and glutathione/glutaredoxin systems. Thus, within the thioredoxin system, DHA increased Txn1-2, Trxrd1-2, Prdx3, and Srxn1 gene expression. Paralleling these changes, DHA increased thioredoxin reductase activity, the main enzyme involved in thioredoxin regeneration. For the glutathione system, the most important change triggered by DHA was the upregulation of Gpx4 gene, encoding for the nuclear, cytosolic and mitochondrial isoforms of phospholipid-hydroperoxide glutathione peroxidase (PH-GPx/GPx4, the main enzyme protecting cell membranes against lipid peroxidation), which was followed by a significant increase in total glutathione peroxidase and GPx4 activities. Noticeably, DHA also upregulated a new Gpx4 splicing variant that retained part of the first intronic region. Finally, we demonstrate that DHA treatment, under the same time course, protects HT22 cells from the oxitoxic exposure to amyloid beta (Aß25-35 ) peptide. Altogether, our data pinpoint to a role of DHA as Indirect Antioxidant that modulates neuronal defences in neuroprotection. DHA improves the antioxidant capacity of cultured hippocampal HT22 cells. We propose that DHA supplementation induces the remodelling of membrane phospholipids, and also triggers a transcriptional program to increase the expression of members of the glutathione and thioredoxin systems. We postulate that this transcriptional effect is mediated by a signal arising from non-enzymatic oxidation of DHA.

INTRODUCTION:"Obesity" is considered a feature of youth with DS but whether "low physical fitness" is also a feature is unknown. OBJECTIVE: The aim of this case-control study was to compare the levels of fatness and fitness in adolescents with and without DS. METHODS: Participants included 17 (5 girls) adolescents with DS aged 12-18 years and a control group of 94 (45 girls) adolescents without DS aged 12-16 years. The ALPHA health-related fitness test battery for children and adolescents was selected to assess fatness and fitness in both groups. RESULTS: There were no differences in levels of fatness between groups (all P > 0.27). Adolescents with DS had lower levels of fitness in all the tests than adolescents without DS (all P < 0.001).CONCLUSION: Adolescents with DS have similar levels of fatness and lower levels of fitness than their peers without DS(AU)


Introducción: La obesidad es considerada una característica de los jóvenes con SD, sin embargo se desconoce si la "baja condición física" también lo es. Objetivo: Comparar los niveles de obesidad y condición física en adolescentes con y sin SD. Métodos: Participaron 17 adolescentes (5 niñas) con SD de 12 a 18 años y un grupo control de 94 (45 niñas) adolescentes sin SD de 12-16 años de edad. La batería de condición física ALPHA relacionada con la salud para niños y adolescentes fue seleccionada para evaluar la obesidad y la condición física en ambos grupos. Resultados: No se encontraron diferencias en los niveles de obesidad entre grupos (P > 0,27). Los adolescentes con SD tuvieron niveles más bajos de condición física en todos los test en comparación con los adolescentes sin SD (P < 0,001). Conclusión: Los adolescentes con SD tienen niveles similares de obesidad y menores de condición física que sus compañeros sin SD (AU)

In view of the clear evidence that urokinase type plasminogen activator (uPA) plays an important role in the processes of tumor cell metastasis, aortic aneurysm, and multiple sclerosis, it has become a target of choice for pharmacological intervention. The goal of this study was thus to determine the presence of inhibitors of uPA in plants known traditionally for their anti-tumor properties. Crude methanol extracts were prepared from the leaves of plants (14) collected from the subtropical dry forest (Guanica, Puerto Rico), and tested for the presence of inhibitors of uPA using the fibrin plate assay. The extracts that tested positive (6) were then partitioned with petroleum ether, chloroform, ethyl acetate and n-butanol, in a sequential manner. The resulting fractions were then tested again using the fibrin plate assay. Extracts from leaves of Croton lucidus (C. lucidus) showed the presence of a strong uPA inhibitory activity. Serial dilutions of these C. lucidus partitions were performed to determine the uPA inhibition IC50 values. The chloroform extract showed the lowest IC50 value (3.52 µg/mL) and hence contained the most potent uPA inhibitor. Further investigations revealed that the crude methanol extract and its chloroform and n-butanol partitions did not significantly inhibit closely related proteases such as the tissue type plasminogen activator (tPA) and plasmin, indicating their selectivity for uPA, and hence superior potential for medicinal use with fewer side effects. In a further evaluation of their therapeutic potential for prevention of cancer metastasis, the C. lucidus extracts displayed cytostatic activity against human pancreatic carcinoma (PaCa-2) cells, as determined through an MTS assay. The cytostatic activities recorded for each of the partitions correlated with their relative uPA inhibitory activities. There are no existing reports of uPA inhibitors being present in any of the plants reported in this study.

3,4-Methylenedioxymethamphetamine (MDMA) administration to rats produces acute hyperthermia and 5-HT release. Interleukin-1beta (IL-1beta) is a pro-inflammatory pyrogen produced by activated microglia in the brain. We examined the effect of a neurotoxic dose of MDMA on IL-1beta concentration and glial activation and their relationship with acute hyperthermia and 5-HT depletion. MDMA, given to rats housed at 22 degrees C, increased IL-1beta levels in hypothalamus and cortex from 1 to 6 h and [(3)H]-(1-(2-chlorophenyl)-N-methyl-N-(1-methylpropyl)3-isoquinolinecarboxamide) binding between 3 and 48 h. Increased immunoreactivity to OX-42 was also detected. Rats became hyperthermic immediately after MDMA and up to at least 12 h later. The IL-1 receptor antagonist did not modify MDMA-induced hyperthermia indicating that IL-1beta release is a consequence, not the cause, of the rise in body temperature. When MDMA was given to rats housed at 4 degrees C, hyperthermia was abolished and the IL-1beta increase significantly reduced. The MDMA-induced acute 5-HT depletion was prevented by fluoxetine coadministration but the IL-1beta increase and hyperthermia were unaffected. Therefore, the rise in IL-1beta is not related to the acute 5-HT release but is linked to the hyperthermia. Contrary to IL-1beta levels, microglial activation is not significantly modified when hyperthermia is prevented, suggesting that it might be a process not dependent on the hyperthermic response induced by MDMA.
