RESUMO

Studies have underscored a growingdemand for innovative practices in the cultivation of seedlings from forest species, with a notable emphasis on the utilization of organic waste, inoculation with arbuscular mycorrhizal fungi (AMF), and phosphate fertilization. This study aimed to evaluate the impact of organic residues, inoculation with AMF, and phosphorus on the growth and quality of Peltophorum dubium (Spreng.) Taub. seedlings. Two independent experiments were conducted. In Experiment I, treatments included inoculation with various AMF species: control (without inoculation), Clareoideoglomus etunicatum, Rhizophagus heterosporum, Rhizophagus clarum, and MIX (a combination of the three AMF species), all in conjunction with varying doses of phosphorus (0, 60, 120, 180, and 240 mg kg-1 soil). In Experiment II, treatments comprised a control group (without AMF) and inoculation with Glomus clarum, Gigaspora margarita, Gigaspora albida, Clareoideoglomus etunicatum, and MIX (a combination of the four AMF species), each associated with four substrates: S1) soil + coarse sand - SCS, S2) SCS + poultry manure, S3) SCS + cattle manure, and S4) SCS + sheep manure. Peltophorum dubium seedlings exhibited heightened growth with the inoculation of R. heterosporum, R. clarum, and MIX. Positive responses were observed in seedlings when exposed to organic residues, particularly sheep manure, resulting in increased biomass production and enhanced Dickson quality index. The AMF inoculation, specifically with R. heterosporum, R. clarum, and MIX, provided optimal growth conditions for P. dubium seedlings. Remarkably, the utilization of organic residues, notably substrates with chicken manure and cattle manure, exerted substantial positive effects on both growth and quality of P. dubium seedlings.

Scaffolds for bone tissue regeneration should provide the right cues for stem cell adhesion and proliferation, but also lead to their osteogenic differentiation. Hydrogels of modified platelet lysates (PLMA) show the proper mechanical stability for cell encapsulation and contain essential bioactive molecules required for cell maintenance. We prepared a novel PLMA-based nanocomposite for bone repair and regeneration capable of releasing biofactors to induce osteogenic differentiation. Human bone marrow-derived mesenchymal stem cells (hBM-MSCs) were encapsulated in PLMA hydrogels containing bioactive mesoporous silica nanoparticles previously loaded with dexamethasone and functionalized with calcium and phosphate ions. After 21 d of culture, hBM-MSCs remained viable, presented a stretched morphology, and showed signs of osteogenic differentiation, namely the presence of significant amounts of alkaline phosphatase, bone morphogenic protein-2 and osteopontin, hydroxyapatite, and calcium nodules. Developed for the first time, PLMA/MSNCaPDex nanocomposites were able to guide the differentiation of hBM-MSCs without any other osteogenic supplementation.

This report describes the inhibition of the proteolytic activity of Haemonchus contortus lysate by a mixture of n-alkyl ferulates isolated from Maprounea guianensis. These compounds were evaluated in vitro for the ability to inhibit the H. contortus proteases using SDS-PAGE copolymerized with gelatine (zymography) and by assaying the hemoglobinolytic activity of these enzymes in solution. The ferulates abrogated the proteolysis compared to E-64, suggesting the activity is the result of a cysteine protease.

Hydrolysable tannins, total phenols and flavonoids in Eugenia uniflora leaves were monthly analysed for one year. The results were correlated with climate conditions (rainfall, humidity, cloudiness and mean temperature) through chemometric methods. Principal component analysis revealed high levels of hydrolysable tannins in the rainy season, whereas flavonoids were mainly produced in the dry season. These facts suggest that climatic changes may be one of the factors affecting phenol levels in Eugenia uniflora.


Taninos hidrolisáveis, fenóis totais e flavonóides presentes em folhas de Eugenia uniflora foram quantificados mensalmente durante um ano. Os resultados foram correlacionados com as condições climáticas (pluviosidade, umidade, nebulosidade e temperatura média) através de métodos quimiométricos. Análise de componentes principais revelou a ocorrência de altos teores de taninos hidrolisáveis durante a estação de chuvas, enquanto os flavonóides foram produzidos principalmente na estação seca. Estes fatos sugerem que mudanças climáticas podem ser um dos fatores que afetam os níveis de fenóis em Eugenia uniflora.

Ensaios para avaliar o potencial antibacteriano de Rhizophora mangle (mangue-vermelho), coletada no município de Conde, Bahia, foram realizados com cepas bacterianas Gram-positivas (Staphylococcus aureus ATCC 6538 e Micrococcus luteus ATCC 9341) e Gram-negativas (Echerichia coli ATCC 10536, Salmonella Cholerea-suis 10708, Klebsiela pneumoniae ATCC 700603 e Pseudomonas aeruginosa ATCC 15442). A atividade antibacteriana foi avaliada pelos métodos de difusão em disco e concentração inibitória mínima (CIM), e os resultados analisados estatisticamente. Os resultados do ensaio de difusão em disco mostraram diferença significativa quanto à sensibilidade dos micro-organismos frente aos extratos testados (p<0,05). A CIM do extrato da folha (313 µg mL-1) apresentou o melhor desempenho para inibir o crescimento das cepas Gram-positivas, enquanto o extrato da casca foi mais eficaz para as cepas Gram-negativas. De acordo com os dados levantados por este estudo, R. mangle apresentou propriedade antibacteriana para cepas Gram-positivas e Gram-negativas, podendo tornar-se alternativa terapêutica tanto para o uso popular quanto para a indústria farmacêutica.


Assays to evaluate the antibacterial potential of Rhizophora mangle (red mangrove), sampled at Conde Municipality, Bahia State, Brazil, were performed against Gram-positive (Staphylococcus aureus ATCC 6538 and Micrococcus luteus ATCC 9341) and Gram-negative (Echerichia coli ATCC 10536, Salmonella Cholerea-suis 10708, Klebsiela pneumoniae ATCC 700603 and Pseudomonas aeruginosa ATCC 15442) bacteria. Antibacterial activity was evaluated by disc diffusion and minimal inhibitory concentration (MIC) and results were statistically analyzed. The results of disc diffusion assay showed a significant difference as to the sensitivity of microorganisms against the tested extracts (p<0.05). The MIC of leaf extract (313 µg mL-1) indicated the best performance to inhibit the growth of Gram-positive strains, while bark extract had a better efficacy against Gram-negative strains. Based on the presented data, R. mangle showed antibacterial properties against both Gram-positive and Gram-negative strains and can be used as an alternative therapy for popular use or for the pharmaceutical industry.

In recent years, the search for natural plant products to fight viral diseases has been increasing. In this work, two Spondias species, namely S. mombin and S. tuberosa, found in Ceará state (Brazil), and their main phenolic components were evaluated against dengue virus. In vitro antiviral tests were performed against type-2 dengue virus by the MTT method and standard cytopathic effect reduction assay in C6/36 cells. Cytotoxicity was also evaluated by MTT. The presence of phenolic compounds quercetin, rutin, and ellagic acid in plant extracts was characterized by HPLC analysis. Both Spondias species extracts and components were nontoxic to the cells whereas rutin and quercetin displayed relevant antiviral activity with IC50 of 362.68 µg/mL and 500 µg/mL, respectively.

Solanum paniculatum L. is a plant species widespread throughout tropical America, especially in the Brazilian Savanna region. It is used in Brazil for culinary purposes and in folk medicine to treat liver and gastric dysfunctions, as well as hangovers. Because of the wide use of this plant as a therapeutic resource and food, the present study aimed at evaluating the mutagenic and cytotoxic effects of S. paniculatum ethanolic leaf and fruit extracts using the mouse bone marrow micronucleus test. Our results indicate that neither S. paniculatum ethanolic leaf extract nor its ethanolic fruit extract exhibited mutagenic effect in mice bone marrow; however, at higher doses, both extracts presented cytotoxic activity.

Solanum paniculatum L. is a plant species widespread throughout tropical America, especially in the Brazilian Savanna region. Solanum paniculatum L., popularmente conhecida como jurubeba, ocorre em toda a América tropical, especialmente no Cerrado. No Brasil, é utilizada para fins culinários e na medicina popular para o tratamento de distúrbios gástricos e hepáticos, além de ressacas. Devido à grande utilização desta planta pela população como recurso terapêutico e alimentício, o presente estudo teve como objetivo avaliar as atividades mutagênica e citotóxica dos extratos etanólico das folhas e frutos de S. paniculatum utilizando o teste do micronúcleo em medula óssea de camundongos. Os resultados indicam que os extratos etanólicos tanto das folhas quanto dos frutos de S. paniculatum não apresentaram ação mutagênica em medula óssea de camundongos, porém, em doses mais elevadas, ambos os extratos exibiram atividade citotóxica.

This study evaluated the transference of maternal-specific antibodies against Haemonchus contortus and the humoral immune response of 32 goat kids to natural infection by the parasite, from birth to 5 months of age. All serum samples collected immediately after birth and before colostrum ingestion were negative for H. contortus antibodies, as demonstrated by ELISA. However, 30 and 60 days after colostrum and milk ingestion, antibody detection was positive and faecal egg count (FEC) was negative, suggesting the transference of humoral immunity by colostrum. All animals were FEC-negative for Strongylodea until four months of age, consistent with the stage of animal development where nourishment comes only from breastfeeding. Animals also presented a high serum IgA concentration, associated with lower levels of total IgG. This work demonstrated that passive humoral immunity against H. contortus in goat kids occurs via the colostrum and not by transplacental transference of antibodies.

Várias plantas nativas do bioma Cerrado são utilizadas como plantas medicinais. Dentre elas, o pequi possuí ação moluscicida utilizada no combate a esquistossomose. Objetivamos neste trabalho a verificação da toxicidade de frações de pequi sobre outros organismos aquáticos, antes da utilização destas em mananciais. Para isso, analisamos alterações no índice mitótico das células epiteliais das brânquias de Guaru (Poecilia vivipara) expostas às frações da folha e da casca do caule de pequi extraídas com acetato de etila. Constatamos que nenhuma das frações se mostrou letal aos peixes. Os animais expostos à fração acetato de etila da folha não apresentaram modificações significativas no índice mitótico em relação ao grupo controle, mas os animais expostos à fração acetato de etila da casca do caule apresentaram aumento do índice mitótico das células epiteliais em duas regiões dos filamentos branquiais. Desta forma, a fração acetato de etila da folha poderia ser utilizada como moluscicida em mananciais, enquanto que a fração acetato de etila da casca do caule necessitaria passar por outros testes mais específicos.

Alcoholic extracts of six Brazilian Cerrado medicinal plants were evaluated for their molluscicidal activity against Biomphalaria glabrata, intermediate host of schistosomiasis. Stryphnodendron polyphyllum bark extract, rich in condensed tannins, was the most promising as molluscicide.

The essential oils of Hyptis suaveolens plants collected from 11 localities of the Brazilian Cerrado region were investigated by GC-MS. Sabinene, limonene, biclyclogermacrene, beta-phellandrene and 1,8-cineole were the principal constituents. The results were submitted to principal component and chemometric cluster analysis which allowed three groups of essential oils to be distinguished with respect to the content of p-mentha-2,4(8)-diene, limonene/beta-phellandrene/gamma-terpinene and germacrene D/bicyclogermacrene. In patterns of geographic variation in essential oil composition indicated that the sesquiterpenes are mainly produced in the samples grown at lower latitudes.

From the leaves of Eucalyptus ovata 13 known compounds, four flavonol glycosides and nine tannins, including the trimer gallocatechin-(4alpha-8)-gallocatechin-(4alpha-8)-catechin (1), were isolated. The spectral data of the peracetate derivative of the trimer are reported.

RESUMO

From the leaves of Eucalyptus consideniana and E. viminalis, known hydrolysable tannins, galloyl esters and ellagitannins, were isolated. Flavonol glycosides were also isolated from the leaves of E. consideniana. The spectral data of the dimeric ellagitannin, oenothein B (1), are reported.

The essential oil from the hydrodistillation of Eugenia dysenterica leaves consisted mainly of beta-caryophyllene and alpha-humulene as the major sesquiterpene, while limonene and alpha-thujene were the major monoterpene hydrocarbons. The main oxygenated mono and sesquiterpene constituents were alpha-terpineol and beta-caryophyllene oxide, respectively. The oil was investigated against eight strains of Candida albicans, 35 strains of Cryptococcus neoformans var. neoformans, and two C. neoformans var. gattii isolated from HIV-infected individuals with candidosis or cryptococcal meningitis using the agar dilution method. Based on the minimal inhibitory concentration (MIC) values, the most significant results were obtained against Cryptococcus strains. It was observed that 22 strains were inhibited at a concentration of 250 microg/ml, whereas four exhibited potent inhibition with MIC values below 125 microg/ml against 10(6) UFC/ml organisms. We found MICs > or = 3.12 microg/ml for 91.6, 50 and 30% of all Cryptococcus strains in relation of amphotericin B, fluconazole and itraconazole, respectively.

From the aerial parts of the Myrica gale catechin, epicatechin, gallocatechin, epigallocatechin, epigallocatechin-3-O-gallate, gallocatechin-(4 alpha-8)-epicatechin, gallocatechin-(4 alpha-8)-epigallocatechin, and gallocatechin-(4 alpha-8)-gallocatechin-(4 alpha-8)-gallocatechin (1), were isolated. The 1H- and 13C-NMR data of the peracetate derivative of the trimer are reported.
