To maximize the efficiency of dietary P utilization in swine production, understanding the mechanisms of P utilization in lactating sows is relevant due to their high P requirement and the resulting high inorganic P intake. Gaining a better knowledge of the Ca and P quantities that can be mobilized from bones during lactation, and subsequently replenished during the following gestation, would enable the development of more accurate P requirements incorporating this process of bone dynamics. The objective was to measure the amount of body mineral reserves mobilized during lactation, depending on dietary digestible P and phytase addition and to measure the amount recovered during the following gestation. Body composition of 24 primiparous sows was measured by dual-energy x-ray absorptiometry 2, 14, 26, 70 and 110 days after farrowing. Four lactation diets were formulated to cover nutritional requirements, with the exception of Ca and digestible P: 100% (Lact100; 9.9 g Ca and 3.0 g digestible P/kg), 75% (Lact75), 50% without added phytase (Lact50) and 50% with added phytase (Lact50 + FTU). The gestation diet was formulated to cover the nutritional requirements of Ca and digestible P (8.2 g Ca and 2.6 g digestible P/kg). During the 26 days of lactation, each sow mobilized body mineral reserves. The mean amount of mobilized bone mineral content (BMC) was 664 g, representing 240 g Ca and 113 g P. At weaning, the BMC (g/kg of BW) of Lact50 sows tended to be lower than Lact100 sows (-12.8%, linear Ca and P effect × quadratic time effect) while the BMC of Lact50 + FTU sows remained similar to that of Lact100 sows. During the following gestation, BMC returned to similar values among treatments. Therefore, the sows fed Lact50 could recover from the higher bone mineral mobilization that occurred during lactation. The P excretion was reduced by 40 and 43% in sows fed Lact50 and Lact50 + FTU, respectively, relative to sows fed Lact100. In conclusion, the quantified changes in body composition during the lactation and following gestation of primiparous sows show that bone mineral reserves were mobilized and recovered and that its degree was dependent on the dietary P content and from phytase supplementation during lactation. In the future, considering this potential of the sows' bone mineralization dynamics within the factorial assessment of P requirement and considering the digestible P equivalency of microbial phytase could greatly limit the dietary use of inorganic phosphates and, thus, reduce P excretion.

This study investigated the ability of replacement gilts to adapt their calcium and phosphorus utilization and their kinetics in bone mineralization to compensate for modified intake of these nutrients by applying a novel Ca and P depletion and repletion strategy. A total of 24 gilts were fed according to a two-phase feeding program. In the first phase, gilts (60-95 kg BW) were fed ad libitum a depletion diet providing either 60% (D60; 1.2 g digestible P/kg) or 100% (D100; 2.1 g digestible P/kg) of the estimated P requirement. In the second phase, gilts (95-140 kg BW) were fed restrictively (aim: 700-750 g/d BW gain) a repletion diet. Half of the gilts from each depletion diet were randomly assigned to either a control diet or a high-P diet (R100 and R160; with 2.1 and 3.5 g digestible P/kg, respectively) according to a 2 × 2 factorial design, resulting in four treatments: D60-R100, D60-R160, D100-R100 and D100-R160. Dual-energy X-ray absorptiometry was used to measure whole-body bone mineral content (BMC), bone mineral density (BMD) and lean and fat tissue mass on each gilt at 2-week intervals. The depletion and repletion diets, fed for 5 and 8 weeks, respectively, did not influence growth performance. The D60 gilts had a reduced BMC and BMD from the second week onwards and ended (95 kg BW) with 9% lower values than D100 gilts (P < 0.001). During repletion, D60 gilts completely recovered the deficit in bone mineralization from the second and fourth week onwards, when fed R160 (D60-R160 vs D100-R160) or R100 (D60-R100 vs D100-R100) diets, respectively (treatment × time interaction, P < 0.001); thus, the depletion diets did not affect these values at 140 kg BW. These results illustrate the rapid homeostatic counter-regulation capacity of dietary Ca and P, and they show the high potential to limit dietary digestible P concentration by completely excluding the use of mineral phosphates during the depletion phase, representative of the fattening period, without causing any detrimental effects to gilts at mating. The gilts were able to recover their BMC deficit between their selection at 95 kg BW and first mating at 140 kg BW by increasing their dietary Ca and P efficiency. Finally, excess dietary digestible P, requiring increased amounts of mineral phosphates, further increased the gilts' BMC.

Solanum glaucophyllum leaves contain high levels of glycosidically bound 1,25 dihydroxyvitamin D3, the most important vitamin D metabolite. The tolerance to this source was evaluated during six weeks with fifty weaned pigs fed increasing levels (0, 2.5, 5, 10 and 20µg 1,25(OH)2D3/kg diet). The diet contained, per kg, 9.7g Ca, 3.5g digestible P and 2000IU cholecalciferol. Ten additional pigs were fed a diet containing 1000IU cholecalciferol/kg, without 1,25(OH)2D3. Weekly plasma and final kidney, bone and urinary mineral contents, bone density and breaking strength served as indicators for possible adverse effects of the supplement. All animals grew well and remained clinically healthy. The measured parameters remained unchanged when 1000 replaced 2000IU cholecalciferol/kg and when 1,25(OH)2D3 was fed up to 10µg/kg. Twenty µg 1,25(OH)2D3 increased plasma Ca and decreased plasma P from the 2nd and the 4th experimental week onwards, respectively. Twenty µg 1,25(OH)2D3 increased final plasma Ca and 1,25(OH)2D3 and reduced final plasma P by respectively 19, 56 and 13%. Twenty µg 1,25(OH)2D3 also increased kidney Ca and urinary Ca by 43 and 69%, respectively, reduced bone breaking strength by 12% and tended to decrease bone ash by 3%. To conclude, 2000IU D3 was not beneficial compared to 1000IU cholecalciferol; up to 10µg 1,25(OH)2D3 per kg diet did not lead to observed adverse effects; 20µg 1,25(OH)2D3 altered the homeostatic regulation of Ca and P thus, may lead to first signs of possible adverse effects, such as soft tissue calcification.

In a 2×2 factorial experiment the hypotheses tested were that the metabolic acid load caused by benzoic acid (BA) added to the feed affects bone mineralization of weanling pigs, and that a wide dietary calcium (Ca) to phosphorus (P) ratio in phytase-supplemented feeds with a marginal P concentration has a positive effect on bone mineralization. The four experimental diets, which contained 0.4% P and were supplemented with 1,000 FTU phytase/kg, contained either 5 g BA/kg or no BA and either 0.77% Ca or 0.57% Ca. The 68 four-week-old Large White pigs were fed the experimental diets ad libitum for six weeks and were then slaughtered. Benzoic acid increased feed intake (p = 0.009) and growth rate (p = 0.051), but did not influence the feed conversion ratio (p>0.10). Benzoic acid decreased the pH of the urine (p = 0.031), but did not affect breaking strength and mineralization of the tibia (p>0.10). The wide Ca:P ratio decreased feed intake (p = 0.034) and growth rate (p = 0.007) and impaired feed the conversion ratio (p = 0.027), but increased the mineral concentration in the fat-free DM of the tibia (p = 0.013) without influencing its breaking strength (p>0.10). The observed positive effect of the wide Ca:P ratio on bone mineralization may be attributed, at least in part, to the impaired feed conversion ratio, i.e. to the higher feed intake and consequently to the higher mineral intake per kg BW gain. The negative impact on animal performance of the wide dietary Ca:P ratio outweighs its potentially positive effect on bone mineralization, precluding its implementation under practical feeding conditions.

Two experiments were conducted to explore the effects of a sodium saccharin-based dietary sweetener (Sucram) on growth performance, health, and physiological responses of feedlot steers. In Exp. 1, 173 newly-received male calves purchased from auction barns were fed 0, 100, 200, or 300 g of Sucram/t of DM over 56 d. Overall, ADG and G:F (P > 0.10) were not different among treatments, but steers receiving 200 g Sucram/t displayed numerically greater ADG (23%). In addition, DMI was 17% greater for steers receiving 200 g of Sucram/t compared to steers fed the control diet (cubic effect, P = 0.09). The morbidity rate for respiratory disease did not differ (P > 0.50) among treatments. In Exp. 2, 15 steers (initial BW = 261 ± 28 kg) were used to evaluate the effects of Sucram on apparent total tract digestibility, plasma metabolite concentrations, and urine monoamine metabolite concentrations. Treatments consisted of ad libitum access to a 60% concentrate diet (Control), ad libitum access to Control + 200 g of Sucram/t of DM (Adlib), and Control + 200 g of Sucram/t of DM with feed intake paired to the Control (Paired). By design, steer DMI during the metabolism period did not differ (P = 0.34) between Paired and Control, but DMI tended (P = 0.14) to be 8.2% greater for Adlib than for Control. Treatments did not alter (P > 0.17) apparent total tract nutrient digestibility. Postprandial plasma citrulline concentration was lower (P = 0.03) for Adlib than for Control and tended to be lower (P = 0.13) for Paired than for Control. Plasma homocysteine concentration was reduced (P < 0.03) by feeding Sucram. Urinary concentrations of ethylmalonic acid, vanillymandelic acid, and 5-hydroxyindolacetic acid were greater (P < 0.06) for Adlib than for Control; Paired steers had a greater (P = 0.02) urine vanillymandelic acid concentration than Control steers and tended (P < 0.12) to have a greater urinary concentration of ethylmalonic and 5-hydroxyindolacetic acid than Control steers. Serum insulin was greater for Adlib than for Control steers (P = 0.04) and tended to be greater for Paired than for Control steers (P = 0.14), but serum prolactin area did not differ (P > 0.22) among treatments. Supplementation with Sucram may increase feed intake by newly-received, stressed feedlot calves. Saccharin supplementation reduced plasma homocysteine and increased urinary excretion of vanillymandelic acid, suggesting an improved activity of the dopamine reward system.

Dry cow diets based on grassland forage from intensive production contain high amounts of K and could be responsible for a reduced ability to maintain Ca homoeostasis. The aim of this study was to determine whether a moderate anionic salt supplementation to a forage-based pre-calving diet with varying native K content affects the mineral and acid-base status in transition cows. Twenty-four dry and pregnant Holstein cows, without antecedent episodes of clinical hypocalcemia, were assigned to two diets during the last 4 weeks before estimated calving date. Twelve cows were fed a hay-based diet low in K (18 g K/kg DM), and 12, a hay-based diet high in K (35 g K/kg DM). Within each diet, six cows received anionic salts during the last 2 weeks before the estimated calving day. After calving, all cows received the high K diet ad libitum. Blood samples were taken daily from day 11 pre-partum to day 5 post-partum. Urine samples were taken on days 7 and 2 pre-partum and on day 2 post-partum. The anionic salt did not alter feed intake during the pre-partum period. Serum Ca was not influenced by the dietary treatments. Feeding pre-partum diets with low K concentrations induced a reduced metabolic alkalotic charge, as indicated by reduced pre-partum urinary base-acid quotient. Transition cows fed the low K diet including anionic salts induced a mild metabolic acidosis before calving, as indicated by higher urinary Ca, lower urinary pH and net acid-base excretion. Although serum Ca during the post-partum period was not affected by dietary treatment, feeding a low K diet moderately supplemented with anionic salts to reach a dietary cation-anion difference close to zero permitted to obtain a metabolic response in periparturient cows without altering the dry matter intake.

Zinc (Zn) is essential for swine and poultry and native Zn concentrations in feedstuffs are too low to meet their Zn requirement. Dietary Zn bioavailability is affected by phytate, phytase and Zn supplemented in organic form is considered as more bioavailable than inorganic sources. A meta-analysis using GLM procedures was processed using broiler and piglet databases to investigate, within the physiological response of Zn, (1) the bioavailability of inorganic and organic Zn sources (Analysis I); (2) the bioavailability of native and inorganic Zn dependent from dietary phytates, vegetal and supplemental phytase activity (Analysis II). Analysis I: the bioavailability of organic Zn relative to inorganic Zn sources ranged, depending on the variable, from 85 to 117 never different from 100 (P > 0.05). The coefficients of determination of the regressions were 0.91 in broilers and above 0.89 in piglets. Analysis II: in broilers, bone Zn was explained by supplemental Zn (linear and quadratic, P < 0.001) and by supplemental phytase (linear, P < 0.001). In piglets, the interaction between dietary Zn and phytates/phytases was investigated by means of a new variable combining dietary phytic phosphorus (PP) and phytase activity. This new variable represents the remaining dietary PP after its hydrolysis in the digestive tract, mainly due to phytase and is called non-hydrolyzed phytic phosphorus (PP(NH)). Bone Zn was increased with native Zn (P < 0.001), but to a lower extent in high PP or low phytase diets (ZN(N) × PP(NH), P < 0.001). In contrast, the increase in bone zinc in response to supplemental Zn (P < 0.001) was not modulated by PP(NH) (P > 0.05). The coefficients of determination of the regressions were 0.92 in broilers and above 0.92 in piglets. The results from the two meta-analyses suggest that (1) broilers and piglets use supplemented Zn, independent from Zn source; (2) broiler use native Zn and the use is slightly enhanced with supplemental phytase; (3) however, piglets are limited in the use of native Zn because of the antagonism of non-hydrolyzed dietary phytate. This explains the higher efficacy of phytase in improving Zn availability in this specie.

The study was conducted to evaluate the effects of dietary zinc addition (0 or 15 mg/kg of Zn as inorganic or organic zinc) to three maize-soybean meal basal diets varying in their native Zn, phytic P contents and phytase activity (expressed in kg of feed: P- with 25 mg Zn and 1.3 g phytic P, P+ with 38 mg Zn and 2.3 g phytic P or P+/ENZ being P+ including 500 units (FTU) of microbial phytase per kg) in two monogastric species (piglets, broilers). Measured parameters were growth performance, zinc status (plasma, and bone zinc) and soluble zinc in digesta (stomach, gizzard and intestine). The nine experimental diets were fed for 20 days either to weaned piglets (six replicates per treatment) or to 1-day-old broilers (10 replicates per treatment). Animal performance was not affected by dietary treatments (P > 0.05) except that all P- diets improved body weight gain and feed conversion ratio in piglets (P < 0.05). Piglets fed P- diets had a better Zn status than those fed P+ diets (P < 0.05). In both species, Zn status was improved with supplemental Zn (P < 0.05), irrespective of Zn source. Phytase supplementation improved piglet Zn status to a higher extent than adding dietary Zn, whereas in broilers, phytase was less efficient than supplemental Zn. Digestive Zn concentrations reflected the quantity of ingested Zn. Soluble Zn (mg/kg dry matter) and Zn solubility (% of total Zn content) were highest in gizzard contents, which also presented lower pH values than stomach or intestines. The intestinal Zn solubility was higher in piglet fed organic Zn than those fed inorganic Zn (P < 0.01). Phytase increased soluble Zn in piglet stomach (P < 0.001) and intestine (P = 0.1), but not in broiler gizzard and intestinal contents. These results demonstrate (i) that dietary zinc was used more efficiently by broilers than by piglets, most probably due to the lower gizzard pH and its related higher zinc solubility; (ii) that zinc supplementation, irrespective of zinc source, was successful in improving animal's zinc status; and (iii) suggest that supplemented Zn availability was independent from the diet formulation. Finally, the present data confirm that phytase was efficient in increasing digestive soluble Zn and improving zinc status in piglets. However, the magnitude of these effects was lower in broilers probably due to the naturally higher Zn availability in poultry than in swine.

Two experiments were carried out with broiler breeders (experiment 1) and laying hens (experiment 2) to study the effects of Se sources, in interaction with dietary level of Se or dietary fats on performance, Se incorporation into tissues (blood, liver, breast muscle, and egg) and eggs, hatchability, and glutathione peroxidase (GPX) activities in tissues and blood. Both experiments involved a 3 x 2 factorial arrangement of 3 Se sources (selenite, Se yeast, or B-Traxim Se) and either 2 levels of each source (0.1 or 0.3 mg/kg) or 2 fats (fresh or oxidized). Egg production was not affected by Se source or dietary fat in both experiments. Egg production was greater (P < 0.01) in breeder hens fed 0.3 mg/kg of Se in experiment 1. Hatchability of eggs from hens fed 0.1 mg/kg of Se was lower (P < 0.05) in hens fed Se yeast, whereas from hens fed 0.3 mg/kg of Se, it was comparable across treatments. Selenium in egg, liver, and breast muscle was greater (P < 0.01 or <0.05) in hens fed the greater concentration of Se. Eggs from breeder hens fed organic Se sources had greater (P < 0.01) Se content than those of inorganic source. Egg albumen from breeder fed Se yeast had the greatest Se (P < 0.01), whereas egg yolk from hens fed B-Traxim Se had the greatest Se (P < 0.05). These parameters were affected by interaction between dietary Se level and source (P < 0.01 or < 0.05). Selenium contents in liver and breast muscle were greater (P < 0.01) in hens fed Se yeast compared with hens fed other sources of Se. In experiment 1, liver GPX was greater (P < 0.01) in hens fed selenite or Se yeast, whereas plasma GPX was greater (P < 0.01) in hens fed selenite compared with B-Traxim Se or Se yeast. Supplementation with oxidized fat increased (P < 0.05) GPX in blood and liver. B-Traxim Se decreased (P < 0.05) malondialdehyde content in breast muscle of layers. It is concluded that broiler breeders require supplementation of 0.3 mg/kg of Se, and that there are numerous measurable advantages in using organic rather than inorganic sources for both breeders and layers.

Two experiments were conducted to investigate the iron bioavailability of different sources and supply levels in piglets. In experiment 1, the influence of feeding an iron deficient basal diet or the basal diet supplemented with 28 ppm Fe either in form of Fe-chelate, crystalline Fe-glycinate, or Fe-sulphate on apparent iron digestibility and on blood parameters was determined. In experiment 2, a dose-response approach was used to determine the effects of supplementing an iron-deficient basal diet with 30, 60, or 90 ppm of Fe either in form of crystalline Fe-glycinate or Fe-sulphate on digestibility of Fe, growth and blood parameters. In experiment 1, comparison of iron sources revealed a tendency (0.05 < p < 0.1) for a higher Fe digestibility from Fe-glycinate (40.9%) compared with Fe-chelate (30.8%) or Fe-sulphate (30.7%). Fe-balance was higher (p < 0.05) for Fe-glycinate compared with Fe-sulphate but iron balance was similar for iron sulphate and iron chelate. Those results, however, were not reflected in data of blood parameters. In experiment 2, iron digestibility, blood parameters and performance were significantly (p < 0.05) influenced by iron supply. Effects of iron source on digestibility of iron were lower than observed in the first experiment.

Sixty Angus (n = 29) and Angus-Sim-mental cross (n = 31) steers, averaging 9 mo of age and 277 kg of initial BW, were used in a 148-d study to determine the bioavailability of copper glycinate (CuGly) relative to feed-grade copper sulfate (CuSO(4)) when supplemented to diets high in S and Mo. Steers were blocked by weight within breed and randomly assigned to 1 of 5 treatments: 1) control (no supplemental Cu), 2) 5 mg of Cu/kg of DM from CuSO(4), 3) 10 mg of Cu/kg of DM from CuSO(4), 4) 5 mg of Cu/kg of DM from CuGly, and 5) 10 mg of Cu/kg of DM from CuGly. Steers were individually fed a corn silage-based diet (analyzed 8.2 mg of Cu/kg of DM), and supplemented with 2 mg of Mo/kg of diet DM and 0.15% S for 120 d (phase 1). Steers were then supplemented with 6 mg of Mo/kg of diet DM and 0.15% S for an additional 28 d (phase 2). Average daily gain and G:F were improved by Cu supplementation regardless of source (P = 0.01). Final ceruloplasmin, plasma Cu, and liver Cu values were greater (P < 0.05) in steers fed supplemental Cu compared with controls. Plasma Cu, liver Cu, and ceruloplasmin values were greater (P < 0.05) in steers supplemented with 10 mg of Cu/kg of DM vs. those supplemented with 5 mg of Cu/kg of DM. Based on multiple linear regression of final plasma Cu, liver Cu, and ceruloplasmin values on dietary Cu intake in phase 1 (2 mg of Mo/kg of DM), bioavailability of Cu from CuGly relative to CuSO(4) (100%) was 140 (P = 0.10), 131 (P = 0.12), and 140% (P = 0.01), respectively. Relative bio-availability of Cu from CuGly was greater than from CuSO(4) (P = 0.01; 144, 150, and 157%, based on plasma Cu, liver Cu, and ceruloplasmin, respectively) after supplementation of 6 mg of Mo/kg of DM for 28 d. Results of this study suggest that Cu from CuGly may be more available than CuSO(4) when supplemented to diets high in S and Mo.

Two experiments examined the effects of a saccharin-based artificial sweetener (Sucram) on health, performance, and dietary preference of feedlot cattle. In Exp. 1, 200 steer calves (initial BW = 190.4 +/- 1.47 kg) were fed a 65% concentrate diet supplemented with or without 200 mg of Sucram/kg (DM basis) during a 56-d receiving-growing period. Feeding Sucram did not affect overall (P = 0.19) DMI; however, from d 29 to 56, there was a trend (P = 0.10) for increased DMI with Sucram (5.71 vs. 6.02 kg/d, respectively). From d 0 to 28 and d 0 to 56, there were trends (P = 0.11 and 0.12, respectively) for increased ADG and for increased d-56 BW (P = 0.07) for calves fed Sucram. No differences were detected (P = 0.82) for receiving (REC) period morbidity. During the finishing (FIN) period, 180 steers from the REC period were assigned (9 pens/treatment in a 2 x 2 factorial design) to the following treatments: 1) control REC/control FIN; 2) control REC/Sucram FIN; 3) Sucram REC/control FIN; and 4) Sucram REC/ Sucram FIN. Over the FIN period, ADG tended (P = 0.12) to be greater for Sucram; however, carcass-adjusted ADG did not differ among treatments. Daily DMI was affected by a REC x FIN interaction (P = 0.08), which was the result of greater DMI by cattle in the Sucram REC/Sucram FIN treatment and decreased DMI by cattle in the Sucram REC/control FIN treatment. In general, changes in carcass characteristics were minor. In Exp. 2, 12 steers (initial BW = 395.6 +/- 6.17 kg) were used in a simultaneously replicated 3 x 3 Latin square preference test. Each square consisted of 3 pens, with 2 steers/pen, and 3 time periods. Bunks had dividers at their midpoint, and equal quantities of diet (as-fed basis) were delivered randomly on either side of the divider daily. Treatments were: 1) control; 2) Sucram = basal diet supplemented with 200 mg of Sucram/kg of DM; and 3) choice = control and Sucram on separate sides of the divider. Dietary preference differed on d 1 (P = 0.01) and d 3 (P = 0.02) for control vs. choice and Sucram vs. choice, with the choice group consuming 0.49 and 1.72 kg of DM more of the Sucram diet than the control diet, respectively. This effect, however, was not consistent across days, and average DMI did not differ (P = 0.81) among treatments. Addition of Sucram to the diet of newly received cattle tended to increase receiving period ADG; however, its effects on morbidity, finishing performance, and dietary preference were limited.

The objective of this study was to quantify the bioavailability of zinc (Zn) from sulphate and glycinate as representatives of inorganic and organic zinc sources. The semi-synthetic basal diet contained 2 microg/g of native Zn and was fortified with pure sodium-phytate (8 g/kg) in order to simulate conditions of common cereal-based meals. The basal diet was supplemented with either 53 microg/g of Zn from sulphate (control) or 10 microg/g of Zn from either sulphate (ZnSulphate) or glycinate (ZnGly). Twenty-four (65)Zn-labelled, growing rats weighing 133 g were allotted to the three diets (eight animals per treatment) and were kept pair-fed to ZnSulphate for 15 days. Zn contents in blood plasma, femur and whole body, as well as, plasma alkaline phosphatase activities were reduced compared with control indicating a zinc deficiency in ZnSulphate and ZnGly treatment. This allowed their differentiation in zinc bioavailability. True absorption of dietary Zn was significantly higher in ZnGly than in ZnSulphate (51% vs. 44%) while losses of endogenous faecal Zn and urinary Zn were not affected to a quantitatively relevant extent (mean: 17% and 2% of intake). This resulted in a +30% significantly improved Zn retention for ZnGly (33% vs. 25%) and a lower severity on Zn deficiency symptoms compared with ZnSulphate. Metabolic utilization accounted for 95% of absorbed dietary Zn for both Zn sources. Overall, the bioavailability of zinc glycinate was significantly superior by 16% to zinc sulphate (49% vs. 42%), mainly because of a higher absorptive potential at presence of a strong anti-nutritive component (phytate) in the diet.

The effects of calf age and dam breeds of different milk yield potential on turnover of energy and nutrients were followed in 16 Simmental and 16 Angus beef cows with Angus-sired calves. Calf ages investigated were 1, 4, 7, and 10 mo. The forage offered for ad libitum access consisted of hay for the calves and of a constant mixture of grass silage, meadow hay, and straw (1:0.7:0.3 on a DM basis) for the cows. Calves of 10 mo of age received an additional 2.6 kg DM/d of crushed barley. The animals were kept in groups of four cows and four calves except in the respiration chambers, where only one cow (tethered) and her calf (loose) were grouped together. Indicator techniques were applied to obtain individual data on feces and urine volumes during group housing. In the Simmental cows, heavier on average by 22 kg, voluntary DMI was higher than in the Angus cows (14.0 vs 12.3 kg/d). In calves, DMI from supplementary feeds was 1.6, 3.9, and 6.3 kg/d, on average, at 4, 7, and 10 mo of age, respectively. Dam breed had no significant effect on DMI and ADG of calves and on BW changes of cows. System retention of energy, N, and P showed a curvilinear development with calf age. System energy expenditure, which linearly increased with calf age, was higher with Simmental than with Angus dams (11%), even when adjusted for metabolic BW (8%). Energy loss through methane linearly increased with NDF intake and, consequently, with calf age from 18 to 30 MJ/d (446 to 751 L/d) for cows and calves together. Similarly, fecal and urinary N excretion and fecal P excretion steadily increased with calf age. In calves, the easily volatile N percentage of manure N rapidly decreased from very high levels in young calves. The resulting changes in inclination to gaseous N loss during manure storage for 8 wk were more than compensated by alterations in N intake of the calves, resulting in an increased total system N loss with progressing lactation. Overall, the present results indicate a difference between Angus and Simmental in efficiency of nutrient and energy utilization that might be explained by the difference in amount of delivery of nutrients to the calves via milk vs forage. Suggestions are made as to how the emissions of N, P, and methane, which were excessive in relation to productivity, could be reduced by nutritional means.

Transplantation of haematopoietic stem cells from human leucocyte antigen (HLA)-disparate parental donors presents a promising new approach for the treatment of patients lacking a HLA-matched donor. Success against major obstacles such as graft-versus-host disease (GvHD) and graft rejection has recently been demonstrated, so that immune reconstitution is one of the prime factors that determines the long-term prognosis following transplantation. Twenty children transplanted with megadoses of highly purified CD34(+) haematopoietic stem cells after rigorous T-cell depletion were prospectively monitored for their immune reconstitution during the first post-transplant year. Natural killer (NK) cells showed a marked increase on d +30. T and B cells began to reconstitute on d +72 and +68 respectively. During extended follow-up, their numbers and proliferative capacity upon mitogen stimulation continually increased. Early reconstituting T cells were predominantly of a primed, activated phenotype with severely skewed T-cell receptor (TCR)-repertoire complexity. Naive T cells emerged 6 months post transplantation, paralleled by an increase in TCR-repertoire diversity. All patients self-maintained sufficient immunoglobulin levels after d +200. This study demonstrates that paediatric recipients of highly purified, haploidentical stem cells are able to reconstitute functioning T-, B- and NK-cell compartments within the first post-transplant year. This, together with the absence of significant GvHD, provides a strong indication for this approach to be considered in children who lack a HLA-matched donor.

Octavolateral sensory organs (auditory and lateral line organs) of cave salamander Proteus anguinus are highly differentiated. In the saccular macula of the inner ear the complex pattern of hair cell orientation and the large otoconial mass enable particle displacement direction detection. Additionally, the same organ, through air cavities within the body, enables detection of underwater sound pressure changes thus acting as a hearing organ. The cavities in the lungs and mouth of Proteus are a resonators that transmit underwater sound pressure to the inner ear. Behaviourally determined audiograms indicate hearing sensitivity of 60 dB (rel. 1 microPa) at frequencies between 1 and 10 kHz. The hearing frequency range was between 10 Hz and 10 kHz. The hearing sensitivities of depigmented Proteus and black Proteus were compared. The highest sensitivities of the depigmented animals (N=4) were at frequencies 1.3-1.7 kHz and it was 2 kHz in black animals (N=1). Excellent underwater hearing abilities of Proteus are sensory adaptations to cave habitat.

PG27, an active fraction purified from an extract of a Chinese herb, Tripterygium wilfordii hook f, was used to prevent graft-versus-host disease (GVHD) in a murine model. Lethally irradiated BALB/c (H-2(d)) recipients of B10.D2 (H-2(d)) donor grafts were given daily intraperitoneal injections of PG27 (40 mg/kg per day) for the first 35 days after transplantation. Control mice were given daily injections of solvent vehicle (Ethanol and Cremophor EL). All the control recipients (15/15) died of GVHD within 90 days, but all the recipients given prophylactic treatment with PG27 (15/15) survived beyond 100 days without any signs of GVHD. Furthermore, the GVHD-free recipients were used as donors, and their bone marrow and spleen cells were transplanted into lethally irradiated normal BALB/c (same party) or lethally irradiated normal C3H (H-2(k), third party) mice. Although 10 of 10 same-party recipients survived more than 100 days without any signs of GVHD, 10 of 10 third-party C3H recipients died of GVHD within 40 days. Further studies of PG27 in the murine BCL1 leukemia/lymphoma model demonstrated that animals treated with PG27 partially retained the graft-versus-leukemia (GVL) effect of the graft without GVHD. These results suggest that treatment with PG27 induces host-specific tolerance and retains the GVL effect of allogeneic marrow grafts. (Blood. 2000;95:705-710)

PIP: Physicians in China have developed a new technique to perform vasectomies which improves men's acceptance of vasectomy. It is called no-scalpel vasectomy. Before performing the vasectomy, the surgeon should determine whether the patient is indeed interested in permanent sterilization. The surgeon should also gather information on patient's age, marital status, and medical history and perform a physical examination. Counseling should consist of clear, simple language to diminish any fears. The surgeon must inform the patient of alternative nonpermanent means of contraception and stress that vasectomy is essentially irreversible. 2 physicians recommend external spermatic fascia injection using 2-3 ml of 2% plain lidocaine to induce vasal nerve blockage. After properly fixing the vas deferens with a ring clamp, the surgeon pierces the scrotal skin, vas sheath, and vas deferens in the midline with a curved dissecting clamp held at a 45 degree angle from horizontal. The surgeon then rotates the clamp 180 degrees to prepare the vas for cutting. The surgeon cuts out a 1 cm segment and then occludes the ends of the vas. The vas is then returned to the scrotal sac via the same puncture hole. No sutures are needed for the puncture hole. The same procedure is followed for the other vas. Hands-on training requires 10-15 procedures to develop proficiency. The no-scalpel technique takes about 40% less time than conventional techniques. The complication rate for the no-scalpel technique is 0.4 events/100 procedures compared to 3.1/100 procedures for conventional techniques. Both the ring clamp and dissecting clamp were developed in China. These instruments are provided through a company in Georgia and through the Association for Voluntary Surgical Contraception (AVSC). AVSC helps medical institutions coordinate physician training of the no-scalpel technique.^ieng

The sensitivity of 94 inferior collicular (IC) neurons of Eptesicus fuscus and Myotis lucifugus to spatial location of the acoustic stimulus were studied under free-field stimulus conditions. The best frequency (BF) and minimum threshold (MT) of each neuron were determined with sound delivered in front of the bat. Then the variation in discharge rate of the neuron was measured with a BF sound broadcast from a moving loudspeaker at different angular positions along the horizontal, vertical or diagonal plane of the frontal auditory space. A wide range of stimulus intensities above the MT of the neuron was used. Neurons were classified into 3 classes on the basis of their spatial sensitivity: (1) omnisensitive neurons (15%) were broadly tuned to sound delivered in the frontal auditory space and their responses did not show any correlation with sound location; (2) stimulus intensity-dependent neurons (28%) varied their discharge rates with sound location and intensity so that the peak of their spatial response profiles also varied with stimulus intensity; and (3) stimulus intensity-independent neurons (57%) varied their discharge rates only with sound location over a wide range of stimulus intensities so that their peak discharge always appeared at the same or a small range of angle. In most cases, the medial limbs of the spatial sensitivity curve for these neurons were extremely sharp and congruent. By moving the loudspeaker along the horizontal, vertical and diagonal planes, it was possible to approximate the boundary of the spatial response area of a neuron. Most IC neurons responded to sound delivered within 20 degrees ipsilateral, 60 degrees contralateral, 45 degrees up and 40 degrees down of the frontal auditory space, confirming previous similar studies. In general, an increasing stimulus repetition rate appeared to sharpen the spatial sensitivity curve of a neuron. Conversely, an increasing moving velocity of the stimulus decreased its response. The possible role of these 3 classes of neurons in echolocation and neural mechanisms underlying the spatial sensitivity of these neurons is discussed.

The treatment of osteomyelitis essentially depends on a bactericidal concentration of antibiotic in bone tissue with an appropriate spectrum. Ciprofloxacin is effective against the main organisms causing osteomyelitis (staphylococcus aureus, pseudomonas aeruginosa) at low MIC. We investigated the concentration of ciprofloxacin in human bone. After a single dose (200 mg ciprofloxacin i.v., preoperatively) the concentration in cortical bone was 6.9 +/- 0.8 mcg/g and 9.7 +/- 0.8 mcg/g in cancellous bone. In any case the concentration of ciprofloxacin exceeded the MIC for staph. aureus and pseudomonas aeruginosa. For the treatment of osteomyelitis we transferred the antibiotic bound to autologous cancellous bone grafts to the site of inflammation after surgical debridement. First clinical results are documented.
