RESUMO

The present study was designed to assess Messastrum gracile SVMIICT7 potential in treating dairy wastewater (autoclaved (ADWW) and raw (DWW)) with relation to nutrient removal, in-vivo Chl-a-based biomass, and bio-oil synthesis. Chlorophyll a fluorescence kinetics revealed improved photochemical efficiency (0.639, Fv/Fm) in M. gracile when grown with DWW. This may be owing to enhanced electron transport being mediated by an effective water-splitting complex at photosystem (PSII) of thylakoids. The increase in ABS/RC observed in DWW can be attributed to the elevated chlorophyll content and reduced light dissipation, as evident by higher values of ETo/RC and a decrease in non-photochemical quenching (NPQ). M. gracile inoculated in DWW had the highest Chl-a-biomass yield (1.8 g L-1) and biomolecules while maximum nutrient removal efficiency was observed in ADWW (83.7% TN and 60.07% TP). M. gracile exhibited substantial bio-oil yield of 29.6% and high calorific value of 37.19 MJ kg-1, predominantly composed of hydrocarbons along with nitrogen and oxygen cyclic compounds. This research offers a thorough investigation into wastewater treatment, illustrating the conversion of algal biomass into valuable energy sources and chemical intermediates within the framework of a biorefinery.

The mosquito repellent Nepetalactone rich Nepeta cataria L. (catmint) plant has a variety of therapeutic and industrial potential. Reports on the genetic diversity of N. cataria germplasm are minimal globally and need attention for adding a new variety into commercial cultivation. The present study, therefore, assessed the genetic diversity among thirteen half-sib genotypes of N. cataria using agro economic and phytochemical traits. The experimental set has shown substantial variation for agro economic traits studied. Among all the studied populations, fresh herb-based essential oil content ranged from 0.1 % to 0.3 %, with a grand mean of 1.67 %. However, the estimated oil yield ranged from 44.4 kg/h to 120.73 kg/h with an average of 71.34 kg/h. Among the eleven phytochemical constituents detected in different concentrations in the essential oil of experimental sets, 4aα,7α,7aα-Nepetalactone (67.9-87.5 %) constituted the significant proportion of essential oil. Altogether, based on mean comparison, the population NC8 was found to be promising for estimated oil yield and 4aα,7α,7aα-Nepetalactone content. The greater heritability estimates (h2 bs) and genetic advance as percent of mean (GAM) were observed for important economic parameters, i. e., oil content, herb yield, and oil yield. The cluster analysis revealed the least interactions between various agro economic and phytochemical variables. The microscopic study of trichome showed a positive correlation of abaxial leaf surface with essential oil content. The promising antimicrobial potential of catmint oil was also observed against human health-related pathogens. The results infer from our study provide valuable insight for genetic improvement and product development in the catmint germplasm.

Food commodities are often contaminated by microbial pathogens in transit or during storage. Hence, mitigation of these pathogens is necessary to ensure the safety of food commodities. Globally, researchers used botanicals as natural additives to preserve food commodities from bio-deterioration, and advances were made to meet users' acceptance in this domain, as synthetic preservatives are associated with harmful effects to both consumers and environments. Over the last century, the genus Curcuma has been used in traditional medicine, and its crude and nanoencapsulated essential oils (EOs) and curcuminoids were used to combat harmful pathogens that deteriorate stored foods. Today, more research is needed for solving the problem of pathogen resistance in food commodities and to meet consumer demands. Therefore, Curcuma-based botanicals may provide a source of natural preservatives for food commodities that satisfy the needs both of the food industry and the consumers. Hence, this article discusses the antimicrobial and antioxidant properties of EOs and curcuminoids derived from the genus Curcuma. Further, the action modes of Curcuma-based botanicals are explained, and the latest advances in nanoencapsulation of these compounds in food systems are discussed alongside knowledge gaps and safety assessment where the focus of future research should be placed.

Silicon (Si) is the most abundant element on earth after oxygen and is very important for plant growth under stress conditions. In the present study, we inspected the role of Si in the mitigation of the negative effect of salt stress at three concentrations (40 mM, 80 mM, and 120 mM NaCl) in two wheat varieties (KRL-210 and WH-1105) with or without Si (0 mM and 2 mM) treatment. Our results showed that photosynthetic pigments, chlorophyll stability index, relative water content, protein content, and carbohydrate content were reduced at all three salt stress concentrations in both wheat varieties. Moreover, lipid peroxidation, proline content, phenol content, and electrolyte leakage significantly increased under salinity stress. The antioxidant enzyme activities, like catalase and peroxidase, were significantly enhanced under salinity in both leaves and roots; however, SOD activity was drastically decreased under salt stress in both leaves and roots. These negative effects of salinity were more pronounced in WH-1105, as KRL-210 is a salt-tolerant wheat variety. On the other hand, supplementation of Si improved the photosynthetic pigments, relative water, protein, and carbohydrate contents in both varieties. In addition, proline content, MDA content, and electrolyte leakage were shown to decline following Si application under salt stress. It was found that applying Si enhanced the antioxidant enzyme activities under stress conditions. Si showed better results in WH-1105 than in KRL-210. Furthermore, Si was found to be more effective at a salt concentration of 120 mM compared to low salt concentrations (40 mM, 80 mM), indicating that it significantly improved plant growth under stressed conditions. Our experimental findings will open a new area of research in Si application for the identification and implication of novel genes involved in enhancing salinity tolerance.

The genus Cajanus (Family: Fabaceae) consists of approximately 37 species, and Cajanus cajan (C. cajan) is a significant member of the genus. It is a commercial legume crop widely grown in sub-tropical and semi-arid tropical areas of the world. C. cajan is well known for its folk medicinal uses to treat various disorders, such as toothache, dizziness, diabetes, stomachache, female ailments and chronic infections. These properties have been linked to the presence of several value-added nutritional and bioactive components. Different solvent extracts from C.cajan (leaves, root, stem and seeds) have been evaluated for their phytochemical and biological activities, namely antioxidant, antimicrobial, antidiabetic, neuroprotective, and anti-inflammatory effects. Taken together, and considering the prominent nutraceutical and therapeutic properties of C. cajan, this review article focuses on the important details including ethnomedicinal uses, chemical composition, biological applications and some other medicinal aspects related to C.cajan nutraceutical and pharmacological applications.

Among conventional cancer therapies, radio-frequency magnetic hyperthermia (MHT) has widely been investigated for use with magnetic nanoparticles (MNPs). However, the majority of in vivo biodistribution studies have tested very low MNP dosages (equivalent to magnetic resonance imaging (MRI) applications) to check for clearance rate; which is far below the clinical dose of MHT. Due to this poor validation in preclinical scenarios, quite a few MNPs already in clinical use were later discontinued, on grounds of unexpected clinical outcomes in terms of inflammation, and prolonged clearance in vivo. By exploiting an economical method of synthesis, we have developed chitosan-coated Fe3O4 nanoparticles with high heating efficiency performance. Their anti-tumor response was evaluated in an ectopic tumor model of C6 glioblastoma by MHT. The intratumoral injection of MNPs on days 1 and 7 resulted in rapid tumor inhibition rate of 69.4% within 8 days, with complete inhibition within 32 days, and no recurrence recorded over a 5-month follow-up. Notably, the MNP-mediated MHT therapy achieved the highest degree of therapeutic efficacy required for complete tumor ablation by combining controlled temperature range (<44 °C), reduced MNP dosage; much lower than in most reported studies, and AMF parameters (time of exposure and frequency) within the clinical safety limit. Periodic body weight measurements confirmed negligible adverse side effects in rats. The anti-tumor activity was validated by severe apoptosis (TUNEL, cleaved Caspase-3), reduced proliferation (Ki 67) and disrupted vasculature (CD 31) in the Fe3O4-MHT-treated group. Real-time gene expression of pro-inflammatory cytokines (IL-6, TNF-α, IL-1α, IL-1ß) confirmed the intratumoral activation of IL-6, suggesting the role of immunomodulation in triggering the adaptive immune response for faster tumor regression in the treated group. In addition, the biodistribution and clearance rate of MNPs monitored using ICP-OES confirmed their time-dependent biodegradation via excretion (urine, feces), phagocytosis (liver) and circulatory system (blood), with negligible deposition in other major organs (kidney, heart, lungs). Although we could not show complete clearance of our MNPs within the time frame tested, future studies should focus on combining MHT with immunotherapy, and target tumors at a much-reduced iron dose, consequently improving in vivo clearance rate, and hence overcoming the limitations of MHT in clinical therapy.

Introduction: Trigunas are three basic mental attributes of a personality according to Indian Vedic literature that explains the relationship between mental attributes and human behavior. The three attributes are Tamas (tendency toward lethargy and rigidity), Rajas (tendency toward ambition and activity) and Sattva (tendency toward selfless service), respectively. Satisfaction with life and perceived stress are the important determinants of one's quality of life. Aim: A cross-sectional study assessed the relationship between Trigunas, life satisfaction and perceived stress. Materials and methods: The study recruited 121 willing healthy university students (75 females) with general health questionnaire scores ≤3. All participants were assessed using standardized psychometric tools. Results: Spearman two-tailed correlation test revealed Sattva to have positive correlation with life satisfaction (r = 0.503) and negative correlation with perceived stress (r = -0.302) and other two Gunas (Tamas: R = -0.77; Rajas: R = -0.75), respectively. On the other hand, both Rajas and Tamas correlated positively with perceived stress (Rajas: R = 0.183; Tamas: R = 0.321) and negatively with life satisfaction (Rajas: R = -0.40; Tamas: R = -0.36). Conclusion: This cross-sectional study on university students in India suggests an association of Vedic personality traits (Sattva, Rajas and Tamas Gunas) with life satisfaction, and perceived stress.

Aim: To explore the optimum temperature for cancer cell death using magnetic hyperthermia (MH), which in turn will affect the mode of cell death. Method: The focus of this study is to improve upon the existing methodology for the synthesis of chitosan encapsulated Fe3O4. MH was done at different temperatures. The cell death pathway was explored using flow cytometry and western blot. Results: Coated Fe3O4 exhibited low cytotoxicity, high stability and heating efficiency. MH at 43°C was the optimum temperature for robust cell death. Cell death pathway suggested that during the initial stages of recovery, apoptosis was the main mode of cell death. While at later stages, major apoptosis and minor necrosis were observed. Conclusion: It is important to find out the long-term effect of hyperthermia treatment on cancer cells and their consequences on surrounding healthy cells.

The human response to Mycobacterium tuberculosis (Mtb) infection is affected by the availability of iron (Fe), which is necessary for proper immune cell function and is essential for the growth and virulence of bacteria. Increase in host Fe levels promotes Mtb growth and tuberculosis (TB) pathogenesis, while Fe-supplementation to latently infected, asymptomatic individuals is a significant risk factor for disease reactivation. However, the effect of Fe-supplementation on the host immunity during latent Mtb infection remains unclear, due partly to the paucity in availability of animal models that recapitulate key pathophysiological features seen in humans. We have demonstrated that rabbits can develop non-progressive latency similar to infected humans. In this study, using this model we have evaluated the effect of Fe-supplementation on the bacterial growth, disease pathology, and immune response. Systemic and lung Fe parameters, gene expression profile, lung bacterial burden, and disease pathology were determined in the Mtb-infected/Fe- or placebo-supplemented rabbits. Results show that Fe-supplementation to Mtb-infected rabbits did not significantly change the hematocrit and Hb levels, although it elevated total Fe in the lungs. Expression of selected host iron- and immune-response genes in the blood and lungs was perturbed in Mtb-infected/Fe-supplemented rabbits. Iron-supplementation during acute or chronic stages of Mtb infection did not significantly affect the bacterial burden or disease pathology in the lungs. Data presented in this study is of significant relevance for current public health policies on Fe-supplementation therapy given to anemic patients with latent Mtb infection.

Mentha is a genus from the family Lamiaceae, whose essential oils has long been used in various forms such as in management of plant pathogens and insect pests, in traditional medicine as well as in culinary and cosmetics. Its major chemical components such as menthol, carvone have now been successfully commercialized in the industry as antimicrobials/insecticidal agents. Current review focuses on chemical composition of essential oils of some Mentha species from different geographical regions with their insecticidal (repellent, antifeedant, and ovicidal) and antimicrobial efficacies against bacterial, fungal plant pathogens and insects of stored products. Reports of the researchers on chemical analysis of essential oils of Mentha species revealed that most of the oils being rich in pulegone, menthon, menthol, carvone, 1, 8-cineole, limonene and ß-caryophyllene. Reviewed literature revealed that, essential oils from different Mentha species possess potential antimicrobial activity against plant pathogens and have insecticidal activity against stored product insects. Thus, antimicrobial and insecticidal properties of essential oils of Mentha species offer the prospect of using them as natural pesticides with a commercial value, having social acceptance due to its sustainability and being environment friendly.

INTRODUCTION: In recent years, there has been increasing interest in diagnosing various components of chronic obstructive pulmonary disease (COPD) using high-resolution computed tomography (HRCT). The present study was undertaken to evaluate HRCT features in patients with COPD. MATERIALS AND METHODS: Fifty patients of COPD (confirmed on Spirometry as per the GOLD guidelines 2014 guidelines) were enrolled, out of which 35 patients got a HRCT done. The Philips computer program for lung densitometry was used with these limits (-800/-1, 024 Hounsfield unit [HU]) to calculate densities, after validating densitometry values with phantoms. We established the area with a free hand drawing of the region of interest, then we established limits (in HUs) and the computer program calculated the attenuation as mean lung density (MLD) of the lower and upper lobes. RESULTS: There was a significant correlation between smoking index and anteroposterior tracheal diameter (P = 0.036). Tracheal index was found to be decreasing with increasing disease severity which was statistically significant (P = 0.037). A mild linear correlation of pre-forced expiratory volume in the first second (FEV1) was observed with lower lobe and total average MLD while a mild linear correlation of post-FEV1 was observed with both coronal (P = 0.042) and sagittal (P = 0.001) lower lobes MLD. In addition, there was a linear correlation between both pre (P = 0.050) and post (P = 0.024) FEV1/forced vital capacity with sagittal lower lobe MLD. CONCLUSION: HRCT may be an important additional tool in the holistic evaluation of COPD.

Alzheimer's disease (AD) is a neurodegenerative disorder that has a higher prevalence and incidence in people older than 60 years. The need for improved AD therapies is unmet as the current therapies are symptomatic with modest efficacy. Partial agonists of the 5-HT4 receptor (5-HT4R) offer both symptomatic and disease-modifying treatments as they shift amyloid-precursor-protein (APP) processing from the amyloidogenic pathway to the nonamyloidogenic pathway by activating the α-secretase enzyme. In addition, they also offer symptomatic treatment by increasing levels of the neurotransmitter acetylcholine in the brain. Because of this fascinating dual mechanism of action, several chemical scaffolds having 5-HT4R pharmacophores were designed and evaluated. Most of the synthesized compounds showed potent in vitro affinities and in vivo efficacies. Upon analysis of focused structure-activity relationships, compound 4o was identified as a potent 5-HT4R partial agonist with favorable ADME properties and good in vivo efficacy. GR-125487, a selective 5-HT4R antagonist, attenuated the activity of compound 4o in the novel-object-recognition-test cognition model.

Worldwide, tuberculosis (TB) is still a serious and significant health concern, more so with the emergence of multidrug-resistant-TB. The inability of mankind to control this infection stems from the fact that the vaccines and drugs that were once effective against TB are no longer efficacious. This has led to a search for new antituberculous agents and adjuvant therapy. Vitamins are being revisited for their role in pathogenicity as well as for their antimycobacterial properties. Vitamins such as biotin and thiamin are essential for Mycobacterium tuberculosis and are required for establishment of infection. On the other hand, vitamins such as Vitamin C and Vitamin D have been shown to possess antimycobacterial properties. To combat M. tuberculosis, innovative strategies need to be devised, keeping in mind the efficacy of the agent to be used. Vitamins can prove to be useful agents capable of modifying the life cycle and biology of M. tuberculosis. We present here a brief overview of the available knowledge on thiamin, biotin, Vitamin C, and Vitamin D, keeping TB treatment and control in perspective.

Essential oils of aromatic and medicinal plants generally have a diverse range of activities because they possess several active constituents that work through several modes of action. The genus Artemisia includes the largest genus of family Asteraceae has several medicinal uses in human and plant diseases aliments. Extensive investigations on essential oil composition, antimicrobial, insecticidal and antioxidant studies have been conducted for various species of this genus. In this review, we have compiled data of recent literature (2012-2017) on essential oil composition, antimicrobial, insecticidal and antioxidant activities of different species of the genus Artemisia. Regarding the antimicrobial and insecticidal properties we have only described here efficacy of essential oils against plant pathogens and insect pests. The literature revealed that 1, 8-cineole, beta-pinene, thujone, artemisia ketone, camphor, caryophyllene, camphene and germacrene D are the major components in most of the essential oils of this plant species. Oils from different species of genus Artemisia exhibited strong antimicrobial activity against plant pathogens and insecticidal activity against insect pests. However, only few species have been explored for antioxidant activity.

Isoliquiritigenin (ISL) and liquiritigenin (LTG) are structurally related flavonoids found in a variety of plants. Discovery of novel antimicrobial combinations for combating methicillin-resistant Staphylococcus aureus (MRSA) infections is of vital importance in the post-antibiotic era. The present study was taken to explore the in vitro and in vivo combination effect of LTG and ISL with ß-lactam antibiotics (penicillin, ampicillin and oxacillin) against mec A-containing strains of MRSA. Minimum inhibitory concentration (MIC) of both LTG and ISL exhibited significant anti-MRSA activity (50-100 µg/mL) against clinical isolates of MRSA. The result of in vitro combination study showed that ISL significantly reduced MIC of ß-lactam antibiotics up to 16-folds [∑ fractional inhibitory concentration (FIC) 0.312-0.5], while LTG reduced up to 8-folds (∑FIC 0.372-0.5). Time kill kinetics at graded MIC combinations (ISL/LTG + ß-lactam) indicated 3.27-9.79-fold and 2.59-3.48-fold reduction in the growth of clinical isolates of S. aureus respectively. In S. aureus-infected Swiss albino mice model, combination of ISL with oxacillin significantly (p < 0.05, p < 0.01, p < 0.001) lowered the systemic microbial burden in blood, liver, kidney, lung and spleen tissues in comparison with ISL, oxacillin alone as well as untreated control. Considering its synergistic antibacterial effect, we suggest both ISL and LTG as promising compounds for the development of novel antistaphylococcal combinations. During mycofloral analysis of green gram (Vigna radiata (L.) R. Wilczek) seed samples taken from different grocery stores by agar and standard blotter paper methods, 5 fungal species were identified, of which Aspergillus flavus exhibited higher relative frequency (75.20% to 80.60%) and was found to produce aflatoxin B1 . On screening of 11 plant essential oils against this mycotoxigenic fungi, Lippia alba essential oil was found to be most effective and showed absolute inhibition of mycelia growth at 0.28 µL/mL. The oil of L. alba was fungistatic and fungicidal at 0.14 and 0.28 µL/mL, respectively. Oil had broad range of fungitoxicity at its MIC value and was absolutely inhibited the AFB1 production level at 2.0 µL/mL. Chemical analysis of this oil revealed geranial (36.9%) and neral (29.3%) as major components followed by myrcene (18.6%). Application of a dose of 80 µL/0.25 L air of Lippia oil in the storage system significantly inhibited the fungal proliferation and aflatoxin production without affecting the seed germination rate. By the virtue of fungicidal, antiaflatoxigenic nature and potent efficacy in storage food system, L. alba oil can be commercialized as botanical fungicide for the protection of green gram seeds during storage.

Twenty-nine Kanamycin resistant clinical isolates of Mycobacterium tuberculosis from Northern India were screened to evaluate genetic mutations in rrs gene, eis gene with its promoter, and whiB7 gene along with its 5'UTR. 14 strains (~48.0%) collectively exhibited mutations in rrs, eis or whiB7 target regions. While the highest frequency of mutations was found in rrs gene, eis and whiB7 loci displayed novel mutations. The novel mutations displayed by eis and whiB7 loci were found to be associated specifically with the Kanamycin resistance as none of the twenty nine Kanamycin sensitive strains harbor them. The inclusion of novel mutations of eis and whiB7 loci will be useful in improving the specificity of future diagnostics.

Elderly patients (>65 years old) represent the fastest growing population among the ESRD patients and those awaiting kidney transplantation. There is ample evidence to suggest that kidney transplant in the elderly population offers the best chance of survival and improves health-related quality of life compared to remaining on dialysis. Although all these emerging facts are encouraging, this population brings with them complex medical problems including frailty, cognitive impairment, and multiple comorbidities. These issues can be barriers to transplantation and threaten the well-being of the patients after transplantation. Furthermore, aging results in changes to the immune system and affects the pharmacokinetics of immunosuppressants. All these changes can increase risk of complications such as infections and malignancy. Because death with a functioning graft is a common cause of graft loss, the new kidney allocation system has been implemented in an attempt to maximize allograft utilization and minimize unrealized graft years. This may result in longer wait-times for the elderly. In this review, we will highlight the barriers to kidney transplant, characterize transplant-related issues in the elderly, and propose alternative strategies under the new allocation system.

Aromatic and medicinal plants produce essential oils in the form of secondary metabolites. These essential oils can be used in diverse applications in food, perfume, and cosmetic industries. The use of essential oils as antimicrobials and food preservative agents is of concern because of several reported side effects of synthetic oils. Essential oils have the potential to be used as a food preservative for cereals, grains, pulses, fruits, and vegetables. In this review, we briefly describe the results in relevant literature and summarize the uses of essential oils with special emphasis on their antibacterial, bactericidal, antifungal, fungicidal, and food preservative properties. Essential oils have pronounced antimicrobial and food preservative properties because they consist of a variety of active constituents (e.g., terpenes, terpenoids, carotenoids, coumarins, curcumins) that have great significance in the food industry. Thus, the various properties of essential oils offer the possibility of using natural, safe, eco-friendly, cost-effective, renewable, and easily biodegradable antimicrobials for food commodity preservation in the near future.

In vitro antibacterial activity of 16 essential oils was investigated by disc diffusion method against two Gram positive bacteria Bacillus subtilis and Staphylococcus aureus and two Gram negative bacteria, Shigella flexneri and Escherichia coli. Oils of Cymbopogon citratus and Ocimum basilicum showed highest antibacterial activity. Gram positive bacteria were found to be more sensitive than Gram negative. Antioxidant activities were tested by 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging assay and ABTS radical cation decolourization assay while Folin-Ciocalteu method was used to determine the total phenolic content. In DPPH assay, highest antioxidant activity was observed in 0. basilicum oil followed by Azeratum conyzoides, A. marmelos and C. citratus, with percent inhibition and IC50 value ranging from 66.11-71.93% and 14.10-17.92 µl ml(-1) respectively. In ABTS assay, similar results were obtained but with higher percent inhibition which ranged from 67.48-76.23% and lower IC50 value (12.12-17.21 µ ml(-1)). Moreover, radical scavenging activity of essential oils was lower than that observed for the synthetic antioxidant BHA and BHT. The total phenolic content of the essential oils as GAE in mg 100 µl(-1) of EO was found to be highest in O. basilicum (0.406) oil followed byA. conyzoides (0.322), A. marmelos (0.238) and C. citratus (0.231). The results provide evidence that the oils of C. citratus and O. basilicum can be further commended for treatment of infections caused by these bacterial pathogens and are potential source of natural antioxidants having appreciable amount of total phenolic content.
