RESUMO

Background: Ulcerative colitis (UC) is a chronic inflammatory disorder of the colon with a continuously remitting and relapsing course. Its etiology is closely related to abnormal interactions between host and gut microbiota. The mucus barrier lining the gastrointestinal tract is necessary to coordinate host and gut microbiota interaction by nourishing and modulating the microbiota. Differential effects of the anti-inflammatory fatty acids docosahexaenoic acid (DHA) and eicosapentaenoic acid (EPA) on UC progression in mice were firstly addressed by our previous work; here, the mechanism for their respective effects were further uncovered from host-microbiome crosstalk based on mucus barrier modulation to pave the way for UC therapy. Methods: Assessment of the disease activity index and histopathology score was conducted in mice with dextran sodium sulfate (DSS)-induced colitis pre-treated with different doses of EPA and DHA. Mucin generation, glycosylation and secretion were evaluated by a combination of electron microscopy, specific mucous staining, and qPCR. Western blotting was used to analyze the underlying molecular events. Fecal short chain fatty acids were detected using gas chromatography, and the gut microbial composition was analyzed using 16S rRNA sequencing. Results: Compared with DHA, the more potent inhibitory effect of high dose EPA on DSS-induced colitis was reconfirmed, which was underlain by a reinforced mucus layer as indicated by increased mucin granule release, mucus layer stratification and markedly upregulated expression of the key modulators involved in goblet cell differentiation. In turn a remarkably enhanced mucus barrier in the EPA group functioned to modulate the gut microbiome, as demonstrated by the enriched abundance of the phylum Bacteroidetes and mucin-degrading bacterium Akkermansia muciniphila producing acetic and propionic acids. Conclusions: EPA and DHA differentially coordinate the interaction between the host and the gut microbiota and relieve mucus barrier disruption in DSS-induced colitis. EPA may develop into a promising adjunctive therapy for UC.

A comparative study was carried out on the chemical composition and nutritional value of six cultivated Lentinula edodes strains that are widely appreciated in China. The results demonstrated that all investigated L. edodes were good sources of protein (14.87%-27.13%), carbohydrates (62.03%-75.56%), and dietary fiber (35.88%-42.49%) and had low ash (5.24%-6.38%) and low fat (0.80%-1.70%) content. There were significant differences among different cultivars. Shenxiang 215 had high crude protein and dietary fiber contents. Potassium was the most abundant mineral element, followed by phosphorus. Different cultivars exhibited distinct fatty acid compositions and free amino acid profiles. Shenxiang 215 had high essential amino acid content. Polyunsaturated fatty acids were predominant in cultivars 0912, Huxiang F4, and Huxiang F2; however, monounsaturated fatty acids were predominant in other strains. Cultivar 0912 had a better mineral and polyunsaturated fatty acid profile. The amino acid profile and protein quality were systematically investigated referring to the latest version of international amino acid reference patterns, including the amino acid score, ratio coefficient of amino acid, ratio coefficient score of amino acid, essential amino acid index, and protein digestibility - corrected amino acid score. The results demonstrated that Shenxiang 18 had better protein quality. These findings provide a reference for breeders to select parents for directional quality breeding.

OBJECTIVE: To examine perceptions of mindful eating and mindful food parenting among parents of elementary school children. METHODS: Four focus groups were conducted with 19 Mexican-American parents and 13 non-Hispanic white parents of children from a northern California elementary school. RESULTS: Themes emerging from this research included food traditions during mealtime, perceptions of mindful eating, mindful food parenting and portion control, mindful food parenting practices through gardening, and mindful food parenting with traditional foods. Mexican-American participants identified cooking traditional foods as an important part of their mindful eating and food parenting practices. Mexican-American participants also highlighted the importance of using their senses to appreciate food. Both Mexican-American and non-Hispanic white participants emphasized the importance of involving children in gardening and meal preparation. Mexican-American parents had different perceptions of food portion control than the non-Hispanic white participants. CONCLUSIONS: The current study indicates the importance of mindfulness in broad food-related practices such as gardening, cooking and preserving food traditions in mindful eating practices. More research is needed to further understand how culture shapes and impacts these food practices among different cultural groups.

Saccharopine dehydrogenase (EC 1.5.1.7) regulates the last step of fungal lysine biosynthesis. The gene (Fvsdh) encoding saccharopine dehydrogenase was identified and cloned from the whole genome of Flammulina velutipes. The genomic DNA of Fvsdh is 1257 bp, comprising three introns and four exons. The full-length complementary DNA of Fvsdh comprises 1107 bp with a deduced amino acid sequence of 368 residues. A 1,000-bp promoter sequence containing the TATA box, CAAT box, and several putative cis-acting elements was also identified. The results of tissue expression analysis showed that the expression level of the Fvsdh gene was higher in the pileus than in the stipe whether in the elongation or maturation stage. Further research showed that the lysine contents were 3.03 and 2.95 mg/g in maturation-pileus and elongation-pileus, respectively. In contrast, the lysine contents were 2.49 and 2.07 mg/g in elongation-stipe and maturation-stipe, respectively. To study the function of Fvsdh, we overexpressed Fvsdh in F. velutipes and found that Fvsdh gene expression was increased from 1.1- to 3-fold in randomly selected transgenic strains. The lysine contents were also increased from 1.12- to 1.3-fold in these five transformants, except for strain T3, in which the lysine contents were the same as the control. These results indicate that the expression of the Fvsdh gene can affect the lysine content of F. velutipes.

China is home to rich wild and cultivated strains of Lentinus edodes, an important edible and medicinal mushroom. Artificial selection of L. edodes has a long history, and the widely cultivated strains belong to populations different from those of most wild strains. Internal transcribed spacer (ITS) regions have been used as good markers to identify L. edodes populations. But because ITS regions exhibit incomplete concerted evolution, the use of an ITS to identify L. edodes populations has been questioned. The objective of this study was to determine whether the ITS region is suitable for identifying L. edodes populations and which populations the widely cultivated strains and the most wild strains belong to by investigating intraindividual and differential ITS polymorphisms between 44 cultivars and 44 wild strains of L. edodes in China. Intraindividual ITS polymorphism is common in L. edodes strains, and most strains possessed 2 different ITS sequences, which came from their heterokaryons. The genetic polymorphisms of ITS1, 5.8S, and ITS2 in L. edodes strains are distinct. All strains were divided into one 5.8S type (5.8S-A), 2 ITS1 types (ITS1-A and ITS1-B), and 2 ITS2 types (ITS2-A and ITS2-B), which were subdivided into 2 branches (ITS2-A1 and ITS2-A2; ITS2-B1 and ITS2-B2). ITS1/5.8S/ITS2 could be used as a good marker in preliminary classification of L. edodes strains in China. It not only exhibited classified information of ITS1, 5.8S, and ITS2 for each strain at the same time, it also indicated whether the strain was heterozygous. The 44 cultivated strains were mainly the A/A/A1 type, and the 44 wild strains were mainly the A/A/A2 and other mixed types.
