RESUMO

Plants synthesize abundant terpenes through glandular trichomes (GTs), thereby protecting themselves from environmental stresses and increasing the economic value in some medicinal plants. However, the potential mechanisms for simultaneously regulating terpenes synthesis and GTs development remain unclear. Here, we showed that terpenes in Conyza blinii could be synthesized through capitate GTs. By treating with appropriate intensity of UV-B, the density of capitate GTs and diterpene content can be increased. Through analyzing corresponding transcriptome, we identified a MYB transcription factor CbMYB108 as a positive regulator of both diterpene synthesis and capitate GT density. Transiently overexpressing/silencing CbMYB108 on C. blinii leaves could increase diterpene synthesis and capitate GT density. Further verification showed that CbMYB108 upregulated CbDXS and CbGGPPS expression in diterpene synthesis pathway. Moreover, CbMYB108 could also upregulated the expression of CbTTG1, key WD40 protein confirmed in this study to promote GT development, rather than through interaction between CbMYB108 and CbTTG1 proteins. Thus, results showed that the UV-B-induced CbMYB108 owned dual-function of simultaneously improving diterpene synthesis and GT development. Our research lays a theoretical foundation for cultivating C. blinii with high terpene content, and broadens the understanding of the integrated mechanism on terpene synthesis and GT development in plants.

Red radish sprout become a popular dietary vegetable because of its unique flavor, abundant nutrients and short production cycle. As a cruciferous plant, it has strong ability to absorb and assimilate Se which can promote the content of anthocyanin in plants. However, the mechanisms of Se on anthocyanin accumulation are still unclear. In this study, we explored that appropriate Se promoted growth, antioxidant system and nutrients in radish sprouts. The enhancement of photosynthesis by Se treatment resulted in more sucrose synthesis in radish sprouts. And the transport of sucrose from cotyledon to hypocotyl promoted by Se through up-regulating the gene expression of sucrose transporters, and more sucrose increased the expression of anthocyanin biosynthesis genes to promote anthocyanin accumulation in hypocotyl. These results reveal the beneficial effect of Se on radish sprouts quality, and provide a new insight into the function of Se on sucrose-induced anthocyanin accumulation in radish sprouts.

Soybean sprouts are a flavorful microgreen that can be eaten all year round and are widely favored in Southeast Asia. In this study, the regulatory mechanism of calcium on anthocyanin biosynthesis in soybean sprouts under blue light was investigated. The results showed that blue light, with a short wavelength, effectively induced anthocyanin accumulation in the hypocotyl of soybean sprout cultivar "Dongnong 690." Calcium supplementation further enhanced anthocyanin content, which was obviously inhibited by LaCl3 and neomycin treatment. Moreover, exogenous calcium changed the metabolism of anthocyanins, and seven anthocyanin compounds were detected. The trend of calcium fluorescence intensity in hypocotyl cells, as well as that of the inositol 1,4,5-trisphosphate and calmodulin content, was consistent with that of anthocyanins content. Specific spatial distribution patterns of calcium antimonate precipitation were observed in the ultrastructure of hypocotyl cells under different conditions. Furthermore, calcium application upregulated the expression of genes related to anthocyanin biosynthesis, and calcium inhibitors suppressed these genes. Finally, transcriptomics was performed to gain global insights into the molecular regulation mechanism of calcium-associated anthocyanin production. Genes from the flavonoid biosynthesis pathway were distinctly enriched among the differentially expressed genes, and weighted gene co-expression network analysis showed that two MYBs were related to the accumulation of anthocyanins. These results indicated that calcium released from apoplast and intracellular stores in specific spatial-temporal features promote blue light-induced anthocyanin accumulation by upregulation of the expression of genes related to anthocyanin synthesis of "Dongnong 690" hypocotyl. The findings deepen the understanding of the calcium regulation mechanism of blue light-induced anthocyanin accumulation in soybean sprouts, which will help growers produce high-quality foods beneficial for human health.

This study aimed to investigate the effect of Se (Selenium) treatment on nutritional quality in radish sprouts. The results showed that 15 µM sodium selenite significantly increased phenolics compounds, flavonoids compounds, anthocyanins, and some essential amino acid content, while improving the total antioxidant capacity of radish sprouts. Besides, the Se-enriched radish sprouts significantly alleviated the liver damage caused by carbon tetrachloride (CCl4 ) in mice and improved the antioxidant capacity of the liver in mice, whereas the Se-enriched radish sprouts alleviated the inflammatory reaction and apoptosis caused by CCl4 . These results imply that Se-enriched radish sprouts have a positive impact on mice with CCl4 -induced liver injury, and that in future Se-enriched radish sprouts could be developed into an effective food and health care product for the liver injury prevention. PRACTICAL APPLICATION: Because selenium is an essential trace element in the human body, selenium-enriched sprouts can help eliminate free radicals in the body, relieve aging, and selenium-deficient diseases. They are easy to grow and have low costs. Hence, selenium-enriched sprouts have a great potential of being widely consumed.

The molecular mechanisms of the differences among the Cd tolerance and accumulation of different pak choi cultivars are essential to further breed Cd-safe genotypes pak choi. In our research, via morphological comparison, qRT-PCR and yeast function complementary approaches, we explored the differences of Cd tolerance and capacity for Cd uptake in nine various pak choi varieties. Results showed that higher expressions of BcZIPs involved in Cd uptake in 'Kang Re605' may lead to its higher capacity for Cd accumulation. The lowest expressions of transporter gene in 'Wu Yueman' were consistent with its fewest ability to uptake Cd. Beyond that, the difference of resistance was very great among varieties. Meanwhile, the expressions of the BcGSTUs were differentially induced by Cd exposure in different pak choi varieties, and 'Kang Re605' performed the highest BcGSTUs expression overall. To verify the role of GSTUs played in Cd resistance of pak choi, four BcGSTUs, BcGSTU4, BcGSTU11, BcGSTU12 and BcGSTU22 in a high-Cd accumulation and tolerance variety 'Kang Re605' were cloned, quantitated and transferred to Cd-sensitive yeast mutant strain. And finally found that BcGSTU11 increased the Cd tolerance of yeast, which may associate with a high Cd resistance of 'Kang Re605'. Simultaneously, less BcGSTUs abundance in 'Shang Haiqing' may result in its weak tolerance to Cd. These findings will help us to comprehend the roles of BcZIPs and BcGSTUs in Cd absorption and detoxification as well as promote our understanding of the Cd-resistant and Cd-accumulated mechanisms in pak choi.

Polysaccharides from Trichosanthes peel (TPP) were obtained by ultrasound-assisted extraction. TPP-1 was separated from the TPP by Sephadex G-100 column chromatography. Phosphorylation of TPP-1 was carried out and phosphorylated TPP-1 was named as PTTP-1. The results of infrared spectra, 13C nuclear magnetic resonance spectra and 31P nuclear magnetic resonance spectra showed that the main structure of PTPP-1 was similar to that of TPP-1 and -H2PO3 groups which were conjugated to C-6 of →4)-α-D-Manp-(1→, C-4 of →6)-α-D-Galp-(1→, C-2 and C-3 of →1)-α-L-Araf, C-2 of →1)-α-L-Araf-(3→, and C-6 and C-3 of →1)-α-D-Glcp. In vivo antiaging activity results proved that TTP-1 and PTTP-1 could both significantly improve the body weight, spleen index, and thymus index of the D-galactose-induced aging mice, increase the levels of superoxide dismutase, catalase, glutathione peroxidase, and reduce malondialdehyde contents in the liver, brain, and serum of aging mice. These results indicated that both TPP-1 and PTTP-1 presented significant antiaging activity. Moreover, PTTP-1 showed stronger antiaging effects in aging mice, indicating that phosphorylation improved antiaging effect.

Soil salinity remains a major threat to global food security, and the progress in crop breeding for salinity stress tolerance may be achieved only by pyramiding key traits mediating plant adaptive responses to high amounts of dissolved salts in the rhizosphere. This task may be facilitated by studying natural variation in salinity tolerance among plant species and, specifically, exploring mechanisms of salinity tolerance in halophytes. The aim of this work was to establish the causal link between mesophyll ion transport activity and plant salt tolerance in a range of evolutionary contrasting halophyte and glycophyte species. Plants were grown under saline conditions in a glasshouse, followed by assessing their growth and photosynthetic performance. In a parallel set of experiments, net K+ and H+ transport across leaf mesophyll and their modulation by light were studied in control and salt-treated mesophyll segments using vibrating non-invasive ion selective microelectrode (the MIFE) technique. The reported results show that mesophyll cells in glycophyte species loses 2-6 fold more K+ compared with their halophyte counterparts. This decline was reflected in a reduced maximum photochemical efficiency of photosystem II, chlorophyll content and growth observed in the glasshouse experiments. In addition to reduced K+ efflux, the more tolerant species also exhibited reduced H+ efflux, which is interpreted as an energy-saving strategy allowing more resources to be redirected towards plant growth. It is concluded that the ability of mesophyll to retain K+ without a need to activate plasma membrane H+-ATPase is an essential component of salinity tolerance in halophytes and halophytic crop plants.
