RESUMO

Trophic status in surface waters has been mostly monitored by measuring soluble reactive phosphorus (SRP) and total phosphorus (TP). Additional to these common parameters, a two-dimensional ion chromatography mass spectrometry (2D-IC-MS) method was used to simultaneously measure soluble phosphate (Pi), pyrophosphate (PPi), and eleven phosphate-containing metabolites (P-metabolites) in Lake Ontario and its tributaries. From the additional P species, PPi, adenosine 5'-monophosphate (AMP), glucose 6-phosphate (G-P), D-fructose 6-phosphate (F-P), D-fructose 1,6-biphosphate (F-2P), D-ribulose 5-phosphate (R-P), D-ribulose 1,5-bisphosphate (R-2P), and D-(-)-3-phosphoglyceric acid (PGA) were detected and quantified in the lake and river samples. The additional multivariate statistical analysis identified similarities between samples collected at different locations. The presence of R-P, R-2P, and F-2P in Lake Ontario tributaries seems to be mainly related to the Calvin cycle, while the lack of all these three P-metabolites and higher PGA levels than G-P in Toronto Harbour samples seems to be the result of depleted Calvin cycle, pentose phosphate, and glycolysis metabolic pathways.

Trace-level phosphate analysis and phosphorus speciation in surface water remained challenging due to adsorption and phosphate uptake by microorganisms. In this study a two-dimensional ion chromatography separation coupled to electrospray ionization high-resolution mass spectrometry (2D-IC-ESI-MS) allowed isotope dilution quantitation of phosphate with simultaneous analysis of 11 phosphate-containing metabolites and two inorganic condensed phosphates. Samples were collected from Lake St. Clair, Lake Ontario, and Georgian Bay (ON, Canada). Comparative experiments showed lower phosphate results for samples not immediately spiked and for external calibration quantitation. Field spiking with 18O-labeled phosphate combined with isotope dilution quantitation allows measurement of the phosphate levels existent at the collection time instead of the phosphate concentrations remaining in the samples at the analysis time. This is a significant advantage against the traditional colorimetric and ion chromatographic (IC) analysis methods, which are unable to compensate for the adsorption loss occurring in standards and samples, especially when phosphate is present at levels below 20 µg L-1 as P (61 µg L-1 as PO43-). Two phosphate-containing metabolites, adenosine 5'-monophosphate (AMP) and d-glucose 6-phosphate (Glucose-P), were detected in a subset of samples collected from Lake St. Clair, with no statistically significant correlation between them and the simultaneously measured phosphate. Directly bioavailable P (phosphate), indirectly bioavailable P (phosphatase-hydrolyzed P) and nonbioavailable P (nonhydrolizable P) fractions were quantified by measuring phosphate, phosphate after phosphatase addition and total phosphorus. The proposed 2D-IC-ESI-MS method developed for a QExactive MS instrument with field spiking of the internal standard provides accurate phosphate results and eliminates quantitation errors caused by phosphate adsorption. This setup allows simultaneous collection of targeted and nontargeted analysis data and thus the detection of trace polar organic phosphorus metabolites as well.

The reliable analysis of highly toxic hexavalent chromium, Cr(VI), at ultra-trace levels remains challenging, given its easy conversion to non-toxic trivalent chromium. This work demonstrates a novel analytical method to quantify Cr(VI) at low ngL(-1) concentration levels in environmental water samples by using speciated isotope dilution (SID) analysis and double-spiking with Cr(III) and Cr(VI) enriched for different isotopes. Ion chromatography tandem mass spectrometry (IC-MS/MS) was used for the analysis of Cr(VI) as HCrO4(-) → CrO3(-). Whereas the classical linear multipoint calibration (MPC) curve approach obtained a method detection limit (MDL) of 7ngL(-1) Cr(VI), the modified SID-MS method adapted from U. S. EPA 6800 allowed for the quantification of Cr(VI) with an MDL of 2ngL(-1) and provided results corrected for Cr(VI) loss occurred after sample collection. The adapted SID-MS approach proved to yield more accurate and precise results than the MPC method, allowed for compensation of Cr(VI) reduction during sample transportation and storage while eliminating the need for frequent external calibration. The developed method is a complementary tool to routinely used inductively-coupled plasma (ICP) MS and circumvents typically experienced interferences.

BACKGROUND: Coagulation disorders and reperfusion of ischemic myocardium are major causes of morbidity and mortality. Lectin pathway initiation complexes are composed of multimolecular carbohydrate recognition subcomponents and 3 lectin pathway-specific serine proteases. We have recently shown that the lectin pathway-specific carbohydrate recognition subcomponent mannose-binding lectin plays an essential role in the pathophysiology of thrombosis and ischemia/reperfusion injury. Thus, we hypothesized that the endogenous mannose-binding lectin (MBL)/ficolin-associated protein-1 (MAP-1) that inhibits complement activation in vitro also could be an in vivo regulator by attenuating myocardial schema/reperfusion injury and thrombogenesis when used at pharmacological doses in wild-type mice. METHODS AND RESULTS: In 2 mouse models, MAP-1 preserves cardiac function, decreases infarct size, decreases C3 deposition, inhibits MBL deposition, and prevents thrombogenesis. Furthermore, we demonstrate that MAP-1 displaces MBL/ficolin-associated serine protease (MASP)-1, MASP-2, and MASP-3 from the MBL complex. CONCLUSIONS: Our results suggest that the natural, endogenous inhibitor MAP-1 effectively inhibits lectin pathway activation in vivo. MAP-1 at pharmacological doses represents a novel therapeutic approach for human diseases involving the lectin pathway and its associated MASPs.

This paper aim is to present the case of a male patient, age 63 admitted in hospital for non-specific gastroenterologic symptoms easily attributed to colecyst calculosis; the anamnesis and careful clinical examination have avoided a useless cholecystectomy and permitted to establish indication for laparotomy; during that was found an ileal tumor with massive metastasis in the base of mesentery. The surgical attitude was dictated by histological diagnostic difficulties (prostate adenocarcinoma metastasis), so it has given up the idea of tumor excision (unjustified intraoperative risk for a metastasis), performing a bypass of the ileal tumor by an enteroenterostomy, then the patient was sent to the Oncological Department. Postoperative evaluation of the patient finally permitted to establish the neuroendocrine origin of the tumor; after oncological treatment (chemotherapy), about 10 months after operation the patient didn't present any sign of tumor (tumor markers and CT scan within normal). The paper emphasizes the problem of practicing an aggressive surgical treatment for carcinoid intestinal tumors with mesenteric metastasis but also brings attention to nonsymptomatic forms of cholecyst calculosis that may hide associated malignant tumors undetected by laparoscopic cholecystectomy.
