BACKGROUND: Ganoderma lucidum ( G . lucidum ) is a traditional Chinese herbal medicine that has shown potential as an alternative adjuvant therapy for cancer patients. However, the mechanisms and adjuvant therapeutic effects of G . lucidum in cancer treatment remain unclear. METHODS: In this work, G . lucidum spore oil (GanoOil), a newly developed oily G . lucidum spore extract was used to investigate the mechanisms and adjuvant therapeutic effects of GanoOil in conjunction with the chemotherapeutic drug cyclophosphamide (CTX) for preventing breast cancer metastasis. RESULTS: In the model of lung metastasis, orally administered GanoOil increased the population of CD8 + T cells and interleukin (IL)-6 cytokine levels in mouse blood, whereas also enhancing the activity of natural killer cells in the spleen. Furthermore, the combination of GanoOil and CTX effectively suppressed the lung metastasis of circulating breast cancer cells, alleviated CTX-induced weight loss, and reduced the ratio of lung and spleen weight to body weight in mice. Moreover, high concentrations of GanoOil exhibited no significant toxicity or side effects in both in vitro and in vivo experiments. CONCLUSION: In conclusion, GanoOil is a safe drug that can enhance immune activity in mice to achieve therapeutic effects on cancer, and can also synergistically inhibit tumor metastasis with CTX.

Methicillin-resistant Staphylococcus (MRS) is a multi-drug resistant bacteria that pose a serious threat to human health. Antibacterial nanomaterials are becoming a promising antibiotic substitute or antibiotic adjuvants. In this work, selenium nanowires were modified with nano­silver (Ag NPs) with antibacterial activity and [Ru(bpy)2dppz]2+ with fluorescent labeling of DNA (SRA), and the antibacterial activity, antibacterial mechanism and biological toxicity of SRA synergistic antibiotics were studied. In vitro, antibacterial results show that SRA (12 µg/mL) improves the antibacterial activity of various antibiotics against resistant bacteria and significantly slows the development of bacterial resistance to antibiotics. Studies on antibacterial mechanisms have shown that SRA synergistic antibiotics destroy drug-resistant bacteria through a combination of physical (physical damage) and chemical pathways (destruction of biofilm, membrane depolarization, cell membrane destruction, adenosine triphosphate consumption and reactive oxygen species production). Transcriptomics analysis found that SRA affects bacterial activity by affecting bacterial biosynthesis, ATP synthesis and biofilm formation. Furthermore, SRA synergistic antibiotics can accelerate wound healing of bacterial infection by reducing the inflammatory response. The toxicity evaluation results show that SRA has extremely low cellular and in vivo toxicity. SRA has the potential of clinical application as multiple antibiotic adjuvants to deal with resistant bacterial infections.

Leukopenia or thrombocytopenia is sometimes observed in end-stage renal disease (ESRD) patients, but the association between chronic leukopenia or thrombocytopenia and hemodialysis (HD) is still unclear. We aimed to investigate the incidence of chronic leukopenia or thrombocytopenia in patients with ESRD who received HD and to determine the risk factors of this complication. We retrospectively analyzed ESRD patients treated with HD at Ditmanson Medical Foundation Chia-Yi Christian Hospital in 2018. The risk factors for the occurrence of chronic leukopenia and thrombocytopenia were analyzed by Cox regression models. Of the 473 patients in our study cohort, 46 (9.7%) patients had a hematologic abnormality, including 18 patients with chronic leukopenia, 18 with chronic thrombocytopenia, and 10 with pancytopenia. Multivariate analysis revealed that patient age ≥60 years at the initiation of dialysis was a significant predictor for both chronic leukopenia [adjusted hazard ratio (aHR), 2.71; 95% confidence interval (CI), 1.06-6.89] and chronic thrombocytopenia (aHR, 2.83; 95% CI, 1.08-7.35). Chronic liver disease (aHR, 3.31; 95% CI, 1.27-8.61) and serum ferritin levels >800 mg/dl (aHR, 3.29; 95% CI, 1.29-8.39) were risk factors for chronic thrombocytopenia. A trend showed that vitamin D from intravenous supplementation (aHR, 0.13; 95% CI, 0.01-1.16, P = 0.066) and serum phosphorous level (aHR, 0.73; 95% CI, 0.53-1.02, P = 0.068) may be associated with chronic thrombocytopenia. Our study demonstrated that hematological abnormality was a long-term complication of HD. These results reveal that older patients with HD and high serum ferritin levels are at an elevated risk for chronic cytopenia. Healthcare professionals should be aware of this risk when treating HD patients in order to improve their prognosis.

OBJECTIVE: To investigate the feasibility and effectiveness of ATAS acupuncture (Acupoints-Time-Space Acupuncture) as a non-pharmacological intervention to prevent or relieve chemotherapy-induced fatigue in breast cancer patients undergoing taxane chemotherapy. METHODS: A pilot study in Kunming center with the aim of evaluating 40 patients randomized to 3 groups: ATAS, sham and non-acupuncture with an unequal randomization of 2:1:1. Participants with stage I-III breast cancer were scheduled to receive adjuvant EC4P4 chemotherapy. Participants in the ATAS and sham acupuncture arms received 20 sessions of acupuncture over 20 weeks, non-acupuncture arm received usual care. Evaluation scales, including VAS-F, MFI-20, HDAS, ISI, and blood samples were collected at four timepoints (T1-T4). mRNA sequencing was performed to detect the mechanism of acupuncture. RESULTS: A total of 581 sessions of acupuncture were performed on patients in the acupuncture group. There was no difference between the three groups in terms of clinical characteristics. Patients randomized to ATAS acupuncture had improved symptoms including fatigue, anxiety and insomnia during the whole process of chemotherapy compared with the other two groups. The VAS-F score of ATAS acupuncture group was decreased compared with non-acupuncture group (P=0.004). The score of MFI-20 in ATAS acupuncture group was kept at low level, while the other two groups' scores kept climbing during chemotherapy (P=0.016; P=0.028, respectively). The mechanism of ATAS acupuncture which reduced fatigue and depression may be related to ADROA1, by regulating cGMP/PKG pathway. CONCLUSION: This pilot study has demonstrated that ATAS acupuncture can significantly reduce fatigue induced by chemotherapy. Mulching can effectively maintain and improve soil health and functionality. The mechanisms, however, have not been fully elucidated. Therefore, the effects of temperature on the biogeochemical properties of soil were investigated in the present study in relation to nitrogen management and soil functionality. The results of the 3-year field experiments showed that integrated straw mulch (S) and nitrogen fertilizer (N) treatments enhanced the activities of soil urease, invertase, alkaline phosphatase, and catalase by >1.8, 2.1, 2.0 and 1.4 fold, respectively, compared with the control treatment. Furthermore, these treatments increased soil available N by 28%, phosphorus by 45%, and potassium by 55%. In general, the soil organic carbon, dissolved carbon and labile organic carbon content in the treated plot were approximately 1.2-2.9 folds greater than in the control plot. These improvements in soil fertility and carbon indices increased the biomass and grain yield of soybean (67 and 75%, respectively) during the three-year study period. During 2015-2017, the straw mulch and nitrogen fertilizer treatment increased the soil moisture (23%) and decreased the soil temperature (8%) in comparison to the control in the 0-0.2 m soil depth and, therefore, improved soil enzyme activities, nutrient availability, and carbon stocks, and ultimately, soil functionality and sustainability, in the semiarid region.

This study was designed to evaluate the effect of oral administration of ginseng stem-and-leaf saponin (GSLS) on immune responses in chickens vaccinated with live Newcastle disease (ND) vaccines. In experiment 1, chickens were orally administered GSLS (2.5, 5, 10, and 20mg/kg) in drinking water for 3 days, and then intranasally vaccinated with ND vaccine. Hemagglutination inhibition (HI) test showed that the optimal dose of GSLS for enhancing HI titers was 5mg/kg. In experiment 2, chickens were fed with GSLS (5mg/kg) for 7, 5 or 3 days before vaccination. Result indicated that GSLS enhanced HI titers in a time-dependent manner. The order of the duration for drinking GSLS on increasing HI titers was 7 days>5 days>3 days. In experiment 3, chickens were fed with GSLS before or after immunization. Serum was sampled at a week interval for 5 weeks for HI test, splenic lymphocytes were prepared at weeks 2 and 4 after the booster immunization for lymphocyte proliferation test (LPT) and intestinal tissues were sampled at week 4 after the booster immunization for immunohistochemistry and histological examination. Results showed that HI titer, LPT, IgA+ cells and intestinal intraepithelial lymphocytes (iIELs) were significantly higher in chickens drinking GSLS before vaccination than those after vaccination. Therefore, oral administration of GSLS in drinking water at a dose of 5mg/kg for 7 days significantly increased systemic as well as gut mucosal immunity in chickens immunized with ND vaccine. Considering the immune potentiating effect of GSLS on ND vaccine, GSLS should be evaluated for its oral adjuvant effect on the immune response against other diseases such avian influenza and infectious bursal disease.

A simple and efficient method based on micellar electrokinetic capillary electrophoresis (MECC) with electrochemical detection for the determination of catechin, rutin, hyperin, and kaempferol in Plumula Nelumbini is described. Under optimum conditions, all analytes were well separated within 20 min at a separation voltage of 16 kV in a 60-mmol/L borate running buffer (pH 8.0) containing 20 mmol/L sodium dodecyl sulfate (SDS). The current response was linear over two orders of magnitude with detection limits (S/N = 3) ranging from 6.90 x 10(-8) g/mL to 1.30 x 10(-7) g/mL for all analytes. This work provides a useful method for the analysis of Plumula Nelumbini.

Capillary zone electrophoresis with electrochemical detection has been used for the separation and determination of scopoletin, hyperin, chlorogenic acid, and quercetin in Mutouhui. The effects of several important factors, including running buffer acidity, separation voltage, and working potential, were evaluated to achieve the optimum conditions. The working electrode was a 300-microm carbon disk electrode at a working potential of + 0.95 V (versus saturated calomel electrode). Under the optimum conditions, the analytes can be well separated within 20 min in a 75-cm-long fused-silica capillary. The current response was linear over two orders of magnitude with detection limits (S/N = 3) ranging from 2.70 x 10(-8) g/mL to 1.30 x 10(-7) g/mL for all analytes. This method was used successfully in the analysis of Mutouhui, and the assay results were satisfactory.
