RESUMO

ETHNOPHARMACOLOGICAL RELEVANCE: Shentong Zhuyu Decoction (STZYD) is a traditional prescription for promoting the flow of Qi and Blood which is often used in the treatment of low back and leg pain clinicall with unclear mechanism. Neuropathic pain (NP) is caused by disease or injury affecting the somatosensory system. LncRNAs may play a key role in NP by regulating the expression of pain-related genes through binding mRNAs or miRNAs sponge mechanisms. AIM OF THE STUDY: To investigate the effect and potential mechanism of STZYD on neuropathic pain. METHODS: Chronic constriction injury (CCI) rats, a commonly used animal model, were used in this study. The target of STZYD in NP was analyzed by network pharmacology, and the analgesic effect of STZYD in different doses (H-STZYD, M-STZYD, L-STZYD) on CCI rats was evaluated by Mechanical withdrawal thresholds (MWT) and thermal withdrawal latency (TWL). Meanwhile, RNA-seq assay was used to detect the changed mRNAs and lncRNAs in CCI rats after STZYD intervention. GO analysis, KEGG pathway analysis, and IPA analysis were used to find key target genes and pathways, verified by qPCR and Western Blot. The regulatory effect of lncRNAs on target genes was predicted by co-expression analysis and ceRNA network construction. RESULTS: We found that STZYD can improve hyperalgesia in CCI rats, and H-STZYD has the best analgesic effect. The results of network pharmacological analysis showed that STZYD could play an analgesic role in CCI rats through the MAPK/ERK/c-FOS pathway. By mRNA-seq and lncRNA-seq, we found that STZYD could regulate the expression of Cnr1, Cacng5, Gucy1a3, Kitlg, Npy2r, and Grm8, and inhibited the phosphorylation level of ERK in the spinal cord of CCI rats. A total of 27 lncRNAs were associated with the target genes and 30 lncRNAs, 83 miRNAs and 5 mRNAs participated in the ceRNA network. CONCLUSION: STZYD has the effect of improving hyperalgesia in CCI rats through the MAPK/ERK/c-FOS pathway, which is related to the regulation of lncRNAs to Cnr1 and other key targets.

The vertical sequestration of dissolved organic matter (DOM) by iron minerals along the soil profile is assumed to be central to the long-term storage of the soil organic matter (SOM) pool. However, there is limited information available about how the interaction between DOM and natural iron-bearing minerals shape mineral SOM associations quantitatively and qualitatively in forest subsoils. Here, we systematically investigated the influences of forest organic layer-pyrolyzed biochar-derived DOM (BDOM) and leached DOM (LDOM) on quantity, molecular composition, and diversity of deposition layer-derived iron minerals-associated OM by using Fourier transform ion cyclotron resonance mass spectrometry and other complementary spectroscopy. Results indicated natural iron minerals (FeOx1 and FeOx2) had a greater capacity for sorbing LDOM with higher aromaticity and molecular weight than those of BDOM, and the higher proportion of goethite and short-order-range phase in natural iron minerals was closely related to the increased OM adsorption capacity. We also observed the preferential sorption of oxygen/nitrogen-rich polycyclic aromatic compounds and carboxylic-containing compounds in LDOM and concurrent the potential release of lignin-like/aromatics compounds and carboxyl/nitrogen-less aliphatic compounds from native OM coprecipitates into the solution. However, unsaturated and oxidized phenolic compounds in BDOM had a stronger affinity for FeOx through hydrophobic partitioning and specific polar interactions, and concomitantly the partial release of nitrogen-free aliphatic and other carboxyl-rich compounds. More nitrogen structures in aromatic-containing compounds can improve the saturation level and polarity of BDOM. Compared with BDOM, LDOM exerted a stronger control over the exchange of native OM from subsoil natural iron-bearing minerals and substantially enhanced the molecular diversity of the reconstituted mineral-associated OM during the adsorptive fractionation. Overall, these findings suggest the compositional evolution of DOM profoundly shapes SOM formation and persistence in forest subsoils, which is the key to understanding DOM cycling and contaminant fate during its passage through the soil.

Herpetospermum pedunculosum (Ser.) C. B. Clarke is an important traditional Tibetan medicinal plants in the genus of Herpetospermum, Cucurbitaceae. To better determine its phylogenetic location with respect to the other Cucurbitaceae species, the complete plastome of H. pedunculosum will be reported, which is the first species with plastid genome sequence in the genus of Herpetospermum. Its whole genome is 156,531 bp in length, consisting of a pair of inverted repeats (IRs) of 26,147 bp, one large single-copy (LSC) region of 85,878 bp, and one small single-copy (SSC) region of 18,359 bp. There are 128 genes, including 83 protein-coding genes, 36 transfer RNA (tRNA) genes, and 8 ribosomal RNA (rRNA) genes in the plastome. Phylogenetic analysis based on 13 complete plastomes of Cucurbitaceae species showed sisterhood of H. pedunculosum and a clade containing Trichosanthes kirilowii and Hodgsonia macrocarpa, suggesting the close relationship between tribe Schizopeponeae and tribe Sicyoeae in the family Cucurbitaceae.

Many studies have examined changes in soil microbial community structure and composition by carbon nanomaterials (CNMs). Few, however, have investigated their impact on microbial community functions. This study explored how fullerene (C60) and multi-walled carbon nanotubes (M50) altered functionality of an agricultural soil microbial community (Archaea, Bacteria and Eukarya), using microcosm experiments combined with GeoChip microarray. M50 had a stronger effect than C60 on alpha diversity of microbial functional genes; both CNMs increased beta diversity, resulting in functional profiles distinct from the control. M50 exerted a broader, severer impact on microbially mediated nutrient cycles. Together, these two CNMs affected CO2 fixation pathways, microbial degradation of diverse carbohydrates, secondary plant metabolites, lipids and phospholipids, proteins, as well as methanogenesis and methane oxidation. They also suppressed nitrogen fixation, nitrification, dissimilatory nitrogen reduction, eukaryotic assimilatory nitrogen reduction, and anaerobic ammonium oxidation (anammox). Phosphorus and sulfur cycles were less vulnerable; only phytic acid hydrolysis and sulfite reduction were inhibited by M50 but not C60. Network analysis suggested decoupling of nutrient cycles by CNMs, manifesting closer and more hierarchical gene networks. This work reinforces profound impact of CNMs on soil microbial community functions and ecosystem services, laying a path for future investigation in this direction.

TiO2 nanoparticles (nTiO2) have been widely used in many disciplines. However, whether they can be used to improve crops growth and nutritional quality is unknown. In this study, coriander (Coriandrum sativum L.) was treated with 0, 50, 100, 200, and 400 mg/L nTiO2 to evaluate their possible benefit to plant growth and nutritional quality under hydroponic conditions. Our observations showed that 50 mg/L nTiO2 only slightly but insignificantly increased the root and shoot fresh biomass by 13.2 % and 4.1 %, respectively, relative to the control. nTiO2 at this level promoted shoot K, Ca, Mg, Fe, Mn, Zn, and B accumulation, while spatial distribution of K, Ca, Fe, Mn, Cu and Zn in coriander leaves was not affected. No nTiO2 internalization or translocation to shoots occurred. 400 mg/L nTiO2 significantly reduced root fresh biomass by 15.8 % and water content by 6.7 %. Moreover, this high dose induced root cell membrane wrinkling, attributable to their aggregation and adsorption on root surfaces. At 100-400 mg/L, antioxidant defense systems (SOD, CAT and APX) in plant were triggered to alleviate oxidative stress. At an appropriate dose (50 mg/L), nTiO2 can improve nutrient quality of edible tissues without exerting toxicity to plant or posing health risk to consumers.

OBJECTIVE: The purpose of the current work was to comparatively assess 1470 nm diode laser enucleation of the prostate (DiLEP) and plasmakinetic resection of the prostate (PKRP) for treating benign prostatic hyperplasia (BPH). PATIENTS AND METHODS: From January 2016 to March 2017, 157 individuals with bladder outflow obstruction caused by BPH were randomized to DiLEP and PKRP groups, for prospective analysis. Of these, 152 cases were evaluated before operation and at 3, 6, and 12 months postsurgery. Patient baseline properties, presurgery data, and postsurgical outcomes were comparatively assessed, as well as complications. RESULTS: There were no significant preoperative differences between surgical groups. DiLEP-treated cases showed remarkable reduced operative time, postsurgical bladder irrigation time, catheterization duration, and hospital stay compared with the PKRP group (P < 0.001). Hemoglobin amount decrease was markedly less pronounced after DiLEP (P = 0.004). However, no patients needed blood transfusion in either group. The decrease in sodium level showed no marked differences between the DiLEP and PKRP groups (P = 0.380). In addition, complications were comparable and no significant differences in both groups. At 3, 6, and 12 months, International Prostate Symptom Score (IPSS), quality of life (QoL), maximum flow rate (Qmax), and postvoid residual (PVR) were similar in both groups (P > 0.05). CONCLUSIONS: DiLEP and PKRP are similar in efficacy and safety for relieving obstruction and low urinary tract symptoms. Compared with PKRP, DiLEP has decreased risk of hemorrhage, operative time, bladder irrigation time, catheterization duration, and hospital stay. However, IPSS, QoL, Qmax, and PVR were similar for both procedures within 12 postoperative months.

To examine the exact role of flavored Guilu Erxian decoction, a Traditional Chinese Medicine (TCM) in the treatment of cisplatin-induced side-effects in bone marrow mesenchymal stem cells (BM-MSCs). BM-MSCs were isolated from bone marrow collected from SD rats and identified by flow cytometry. Cells were cultivated in MEM alpha medium containing 5% (TCM-L), 10% (TCM-M) and 20% (TCM-H) dosages of flavored Guilu Erxian decoction with or without cisplatin. Cell viability was determined through CCK-8 and thymidine analog 5-ethynyl-2'-deoxyuridine (EdU) staining assay. Flow cytometry was used to determine cell cycle and apoptosis. The expression of p21 and cleaved-caspase-3 were examined using Western blot assay. The PI3K-AKT-mTOR pathway associated proteins, including p-PI3K, p-AKT and p-mTOR, were also examined by Western blot assay. CCK-8 and EdU staining assay demonstrated that cisplatin could inhibit cell proliferation in BM-MSCs in a dose and time dependent manner. Further, cisplatin could induce apoptosis through increasing G0/G1 cell cycle arrest, p21 and cleaved-caspase-3 expression. However, these phenomena would be significantly alleviated when adding the serum containing flavored Guilu Erxian decoction. Furthermore, the PI3K-AKT-mTOR pathway activation could be inhibited by cisplatin in BM-MSCs, while flavored Guilu Erxian decoction treatment successfully abrogated this effect. Combination of flavored Guilu Erxian decoction and cisplatin could reduce the damage to BM-MSCs. This indicates that the flavored Guilu Erxian decoction can enhance the possibility of BM-MSCs repairing and rehabilitating the normal function of injured tissues induced by cisplatin, which could provide a new direction for therapeutic applications.

Alkylated PAHs (APAHs) have been shown to be more toxic and persistent than their non-alkylated parent compounds. However, little is known about the extent of soil contamination by these pollutants. To help understand agricultural soil pollution by these compounds at a regional scale, a total of 18 methylated PAHs (MPAHs, a major class of APAHs) in 243 soil samples were analyzed. These soil samples were collected from 11 sites in the Yangtze River Delta (YRD) region, a representative fast developing area in China. The total concentration of MPAHs (∑18MPAHs) ranged from 5.5 to 696.2 ng/g dry soil, with methylnaphthalenes (M-NAPs) and methylphenanthrenes (M-PHEs) accounting for more than 70% of the compositional profile. Relatively high concentrations of ∑18MPAHs were found in Jiaxing and Huzhou areas of Zhejiang province, as well as on the border between the cities of Wuxi and Suzhou. Different MPAH groups showed dissimilar spatial distribution patterns. The spatial distribution of lower molecular weight MPAHs was related to agricultural straw burning and emissions/depositions from industrial activities, whereas that of higher molecular weight MPAHs was much more a function of the total organic carbon (TOC) content of soil. Although coal, biomass (crop straw and wood), and petroleum combustion were identified to be the major emission sources for most of the sampling sites, the areas with relatively severe pollution with ∑18MPAHs resulted from the localized hotspots of petroleum leakage. Isomeric MPAHs with methyl group substituted at 2- (ß) position exhibited significantly higher concentrations than those substituted at 1- (α) position. Results of this work help to understand soil pollution by MPAHs, and are useful for designing effective strategies for pollution control so as to ensure food safety in areas with fast economic growth.

BACKGROUND: Organophosphate pesticide residues are harmful to human health because of their potential mutagenic and carcinogenic properties. Therefore, it is of great importance to development an accurate and reliable analytical method to prevent their uncontrolled effects on environmental pollution and human health. RESULTS: This study reports a new method of molecularly imprinted solid-phase extraction coupled with high-performance liquid chromatography (MISPE-HPLC) for simultaneous determination of two organophosphate pesticides residues. Two types of molecularly imprinted polymers (MIPs) were prepared using the trichlorfon and monocrotophos as the template molecule, respectively, methacrylic acid as the functional monomer, and ethylene glycol dimethacrylate as the cross-linker. The recognition ability and adsorption-desorption dynamic of each imprinted polymer toward the trichlorfon or monocrotophos were characterised. Using the mixture of trichlorfon-MIP and monocrotophos-MIP (20:80, wt/wt) as solid-phase extraction sorbent, the factors affecting the pre-concentration on the analytes and the sensitivity of the MISPE-HPLC method were optimised. Under optimal condition, the linear range was 0.005-1.0 mg L⁻¹. The limit of detection was 4.2 µg g⁻¹ for trichlorfon, and 1.2 ng g⁻¹ for monocrotophos. The peak area precision [Relative standard deviation (RSD)] for three replicates was 2.9-4.5%. The blank rape and cauliflower samples spiked with trichlorfon and monocrotophos at 0.05 and 0.005 µg g⁻¹ levels were extracted and determined by this method with recoveries ranging from 88.5% to 94.2%. Moreover, this method was successfully applied to the quantitative detection of the trichlorfon and monocrotophos residues in leek samples. CONCLUSION: With good properties of high sensitivity, simple pre-treatment and low cost, this MISPE-HPLC method could provide a new tool for the rapid determination of multi-pesticide residues in the complicated food samples.

BACKGROUND: Acrylamide has attracted worldwide concerns because of its demonstrated neurotoxicity, genetoxicity and reproductive-development toxicity. It is necessary to control acrylamide production during food processing and protect human health. RESULTS: In this study, a functionalised material was synthesised by immobilising an ionic liquid onto an activated silica gel surface. The adsorption ability of the material towards acrylamide was evaluated, and the results showed that it had high adsorption capacity. Scatchard analysis indicated that the binding sites in the prepared material had two distinct groups (high and low affinity binding sites). The saturated adsorption capacity (Q(max,1)) was 7.9 mg g(-1) due to the high affinity binding sites, and another saturated adsorption capacity (Q(max,2)) was 2.3 mg g(-1) due to the low affinity binding sites. This prepared material also offered fast kinetics for adsorption of the acrylamide. Using this material as sorbent, a method of solid-phase extraction coupled with high-performance liquid chromatography (SPE-HPLC) for analysis of acrylamide in foods was developed. Under optimal conditions, the limit of detection (S/N = 3) of this method for acrylamide was 2.1 µg kg(-1), and the RSD for five replicate extractions of 50 µL(-1) acrylamide was 4.5%. The blank potato and bread crumb samples spiked with acrylamide at different levels of 10.0 and 15.0 µg kg(-1) were extracted and determined respectively by this developed method, and recoveries ranging from 83.0% to 89.1% were obtained. Finally, this method was applied to quantitative detection of acrylamide in bread crust and cracker samples. CONCLUSION: With high sensitivity and pre-treatment simplicity, this SPE-HPLC method could provide a new tool for the rapid determination of acrylamide in the food samples.
