RESUMO

This study investigated the effectiveness and mechanism for the control of internal phosphorus (P) liberation from sediment by hydrous zirconium oxide (HZrO2) combined with calcite, bentonite and zeolite. The results suggested that coexisting calcite, calcium-modified bentonite (CaBT) and calcium-modified zeolite (CaZ) all had the ability to promote the adsorption of phosphate (PO43-) onto HZrO2. The mechanisms of PO43- elimination by HZrO2/calcite mixture involved the adsorption of PO43- on calcite, the precipitation of PO43- with Ca2+, and the inner-sphere complexation of PO43- with HZrO2. The amendment of sediment with HZrO2/calcite, HZrO2/CaBT or HZrO2/CaZ mixture can effectively prevent the sedimentary P release, and the immobilization of mobile P in the sediment and the uptake of dissolved reactive P (DRP) from the interstitial water by the amendment material played a key role in the control of P release from sediment by the combined amendment. Capping sediment with HZrO2/calcite, HZrO2/CaBT or HZrO2/CaZ mixture also can effectively intercept sediment P release, and the formation of P static layer attributed to the uptake of interstitial water DRP and DGT (diffusive gradient in thin-films)-unstable P in the upper sediment by the capping material was a key to the inhibition of sedimentary P migration into the overlying water by the combined capping. The great majority of P immobilized by the HZrO2/calcite, HZrO2/CaBT or HZrO2/CaZ combined covering layer is stable P and it has a low re-releasing risk under dissolved oxygen-deficit and pH 5-9 condition. The stability of P bound by the combined covering layer was larger than that by the single HZrO2 covering layer. The results of this research show that the combined use of HZrO2 and calcite, HZrO2 and CaBT, or HZrO2 and CaZ as a capping material has great potential in the reduction of sediment P loading.

This study developed novel active capping systems with recycling convenience using ferrihydrite (Fh) combined with magnetite (Mag), and investigated the effectiveness and mechanism for the restriction of endogenous phosphorus movement from sediment into overlying water (OW) by the combined use of Fh and Mag. The Fh/Mag combined amendment effectively hindered endogenous phosphorus release from sediment to OW in dissolved oxygen (DO)-deficit environment, and the immobilization of diffusion gradient in thin film-labile phosphorus (LPDGT) and mobile phosphorus in the sediment played a key role in the control of endogenous phosphorus liberation by the Fh/Mag combined amendment. Combined capping sediment with Fh and Mag effectively hindered endogenous phosphorus release from sediment to OW in anoxic environment, and the inactivation of LPDGT in the upper sediment played a key part in the control of sediment phosphorus release by the Fh/Mag mixture capping. The stability of phosphorus immobilized by the Fh/Mag combined covering layer was related to its construction way, and the majority (around 90%) of P immobilized to the Fh/Mag mixture covering layer had low risk of release in common pH (5-9) and DO-deficit environments. The Fh/Mag mixture amendment or capping did not increase the risk of sediment iron release, and it also did not produce a large impact on the diversity and richness of bacterial community in the sediment. The combined utilization of Fh and Mag as a composite amendment or capping material to prevent the internal phosphorus from being moved to OW can make full use of their respective advantages. The Fh/Mag mixture capping wrapped by permeable fabric has high potential to reduce the risk of endogenous phosphorus from sediment into OW due to its advantages of high internal phosphorus release suppression efficiency, environmental friendliness, application convenience and sustainability.

A 45-day fattening trial was conducted using iso-nitrogenous and iso-fatty fattening diets, among which 0%, 25%, 50%, 75%, and 100% of fish oil were, respectively, replaced by blending vegetable oils (canola oil: soybean oil = 1:1), recorded as Diet 1 ~ Diet 5, respectively. The results showed that the meat of crabs (150 ± 25 g) cultured by five fattening feed were distinguished effectively with the electronic tongue. The total free amino acids and taste nucleotides in the meat of crab cultured by Diet 1 were higher than others. The contents of betaine (taste activity value <1) and umami amino acids in the meat of Diet 3 were highest. The contents of sweet amino acids in the meat of crab cultured by Diet 5 was the highest among the different diet. The equivalent umami concentration of Diet 1 was the highest, suggesting that the umami taste is better. In conclusion, 50% of fish oil could be replaced by vegetable oil in fattening feeds. PRACTICAL APPLICATIONS: This paper provided technical support for blending vegetable oils instead of fish oil in fattening feed for swimming crab by using electronic tongue, automatic amino acid analysis, and high performance liquid chromatography to research the changes in the contents of free amino acids, nucleotides, and betaine in female meat of swimming crab, and the taste impacts were evaluated by taste activity values and equivalent umami concentration methods.

Carotenoids are known to be involved in the regulation of the antioxidative capability, immune response and stress resistance in crustacean species; however, very limited information is available on their underlying molecular mechanisms. This study performed transcriptome sequencing of hemolymph and hepatopancreas of juvenile Chinese mitten crabs (Eriocheir sinensis) that fed with three diets, i.e. diet A containing 90 mg kg-1 dry weight of astaxanthin, diet B containing 200 mg kg-1 dry weight of ß-carotene and control diet without supplementation of dietary carotenoids. The results showed that there were 2955 and 497 differentially expressed genes (DEGs) in the hemolymph between the astaxanthin treatment and control groups, and between the ß-carotene treatment and control groups, respectively. Moreover, compared with the control group, 833 and 1886 DEGs were obtained in the hepatopancreas of the astaxanthin treatment and the ß-carotene treatment groups, respectively. The DEGs in the three groups were enriched in 255 specific KEGG metabolic pathways according to KEGG enrichment analysis. Through this study, a series of key genes involved in Nrf2 signalling, ROS production, intracellular antioxidant enzymes and chaperones were significantly affected by dietary carotenoids. Dietary carotenoids also significantly altered the expression levels of immune-related molecules associated with signal transduction, prophenoloxidase cascade, apoptosis, pattern recognition proteins/receptors and antimicrobial peptides. In conclusion, this transcriptomic study provides valuable information for understanding the molecular mechanism and potential pathway of dietary carotenoids improved the antioxidative capability and immunity of juvenile E. sinensis.

Green alga Haematococcus pluvialis is an important source of natural astaxanthin (Ast), which have been shown to be beneficial for the color formulation, survival, antioxidation, immunity and stress resistance of many crustacean. This study was conducted to investigate the effects of dietary supplementation of H. pluvialis meal on growth, antioxidant status, ammonia resistance, color parameters, and carotenoids composition of juvenile Chinese mitten crab Eriocheir sinensis. Five diets were formulated to contain 0, 30, 60, 90 and 120 mg/kg dry diets of natural Ast (defined as Diet 1-5) using H. pluvialis meal as astaxanthin source. The results showed that: (1) Although all treatments with Ast supplementation had the relatively higher growth performance and survival than the control (Diet 1 treatment), no significant differences were found on growth performance, feed conversion ratio and hepatosomatic index among all treatments. (2) The highest total antioxidant capacity (T-AOC) in hepatopancreas and hemolymph were observed in Diet 4 and 3 treatments respectively, while the lowest malondialdehyde (MDA) contents in hepatopancreas and hemolymph were also found in these two treatments. Furthermore, the significantly positive relationships were detected on acid phosphatase (ACP) activities and dietary Ast contents for hepatopancreas and hemolymph. (3) Diet 3 treatment had the highest mRNA levels of EsLecA, EsTrx, and EsPrx6 in hepatopancreas, while both Diet 3 and 4 treatments reached the peaks for mRNA expression levels of EsMyd88 and EsHc, respectively. (4) The stress test with ammonia-N indicated Diet 1 treatment had the highest mortality among all treatments, and the lowest mortality was found on Diet 3 treatment during the stress test. (5) Dietary Ast significantly improved the redness (a*) of carapace and hepatopancreas, which were consistent with the Ast contents in these tissues from the different treatments. Ast concentrations in carapace reached the plateau for Diet 3 treatment while hepatopancreatic Ast concentration kept increasing with elevating dietary Ast contents. In conclusion, natural astaxanthin could enhance the antioxidative capability, non-specific immunity, tissue Ast contents and stress resistance to ammonia-N, and these results suggested the optimal diet micro-algal astaxanthin was around 60 mg/kg for juvenile E. sinensis.

Five different diets with different ratio of fish oil to vegetable oil were prepared. The biological index and proximate composition of Eriocheir sinensis fed with different diets were compared, and then sensory analysis, electronic nose (E-nose) and headspace-solid phase micro-extraction combined with gas chromatography-mass spectrometer (HS-SPME-GC-MS) analysis were applied to determine the odor profile of E. sinensis. The results showed that partial replacement (50%-75%) of fish oil by vegetable oil (FO/VO) was beneficial to the weight increment, nutrition accumulation, and odor-active compounds (OACs) formation of E. sinensis. A total of 7 and 11 OACs were detected in the hepatopancreas and gonad, respectively, these OACs contributed greatly to the overall odor profiles of E. sinensis when the dietary replacement levels were at 50% and 75%, respectively. The results could provide the guide for dietary fish oil replacement as well as improving the odor quality of E. sinensis. Practical application The objective of this research is to compare the effects of dietary replacement of fish oil by vegetable oil on proximate composition and odor profiles of E.sinensis. The results obtained from this study would not only chose an optimal dietary replacement level and serve as a useful database for the odor of female and crabs, but also provide some guide for the improvement of Chinese mitten crab aquaculture.
