RESUMO

Nanoparticles are applied as versatile platforms for drug/gene delivery in many applications owing to their long-retention and specific targeting properties in living bodies. However, the delivery mechanism and the beneficial effect of nanoparticle-retention in many organisms remain largely uncertain. Here, the transport and metabolism of mineral nanoparticles in mammary gland during lactation are explored. It is shown that maternal intravenous administration of iron oxide nanoparticles (IONPs; diameter: ≈11.0 nm, surface charge: -29.1 mV, surface area: 1.05 m2 g-1 ) provides elevated iron delivery to mammary gland and increased iron secretion into breast milk, which is inaccessible by classical iron-ion transport approaches such as the transferrin receptor-mediated endocytic pathway. Mammary macrophages and neutrophils are found to play dominant roles in uptake and delivery of IONPs through an unconventional leukocyte-assisted iron secretion pathway. This pathway bypasses the tight iron concentration regulation of liver hepcidin-ferroportin axis and mammary epithelial cells to increase milk iron-ion content derived from IONPs. This work provides keen insight into the metabolic pathway of nanoparticles in mammary gland while offering a new scheme of nutrient delivery for neonate metabolism regulation by using nanosized nutrients.

BACKGROUND: Depression is the most common mental disorder in patients with advanced cancer, which may lead to poor prognosis and low survival rate. This study aims to explore the serial multiple mediating roles of social support and spiritual coping between hope and depression among patients with advanced cancer. METHODS: A cross-sectional study was conducted in China between May and August 2020. A total of 442 advanced cancer patients were investigated by the following self-reported questionnaires: Herth Hope Index (HHI), Spiritual Coping Questionnaire (SCQ, Chinese version), Social Support Rating Scale (SSRS), Hospital Anxiety and Depression Scale (HADS). RESULTS: Depression was negatively correlated with hope, social support, and positive spiritual coping (P < 0.01), and positively correlated with negative spiritual coping (P < 0.01). Hope explained 16.0% of the variance in depression. Bootstrap analyses of the hope--social support--positive spiritual coping--depression showed that there were direct [B = -0.220, 95%CI(- 0.354, - 0.072)] and indirect effects of hope on depression mediated solely by social support [B = -0.122, 95%CI(- 0.200, - 0.066)] and positive spiritual coping [B = -0.112, 95%CI(- 0.217,-0.025)], or by both together [B = -0.014, 95%CI(- 0.038,-0.003)]. Similarly, the hope--social support--negative spiritual coping--depression showed that there were direct [B = -0.302, 95%CI(- 0.404, - 0.190)] and indirect effects of hope on depression mediated solely by social support [B = -0.126, 95%CI(- 0.205, - 0.071)] and negative spiritual coping [B = -0.033, 95%CI(- 0.080,-0.002)], or by both together [B = -0.010, 95%CI(- 0.030,-0.001)]. CONCLUSIONS: This study proves the hypothesis that social support and spiritual coping play intermediary roles between hope and depression. Interventions established through hope, social support and spiritual coping can effectively prevent depression from occurring.

Erigeron breviscapus is an important medicinal plant in Compositae and the first species to realize the whole process from the decoding of the draft genome sequence to scutellarin biosynthesis in yeast. However, the previous low-quality genome assembly has hindered the optimization of candidate genes involved in scutellarin synthesis and the development of molecular-assisted breeding based on the genome. Here, the E. breviscapus genome was updated using PacBio RSII sequencing data and Hi-C data, and increased in size from 1.2 Gb to 1.43 Gb, with a scaffold N50 of 156.82 Mb and contig N50 of 140.95 kb, and a total of 43,514 protein-coding genes were obtained and oriented onto nine pseudo-chromosomes, thus becoming the third plant species assembled to chromosome level after sunflower and lettuce in Compositae. Fourteen genes with evidence for positive selection were identified and found to be related to leaf morphology, flowering and secondary metabolism. The number of genes in some gene families involved in flavonoid biosynthesis in E. breviscapus have been significantly expanded. In particular, additional candidate genes involved in scutellarin biosynthesis, such as flavonoid-7-O-glucuronosyltransferase genes (F7GATs) were identified using updated genome. In addition, three candidate genes encoding indole-3-pyruvate monooxygenase YUCCA2 (YUC2), serine carboxypeptidase-like 18 (SCPL18), and F-box protein (FBP), respectively, were identified to be probably related to leaf development and flowering by resequencing 99 individuals. These results provided a substantial genetic basis for improving agronomic and quality traits of E. breviscapus, and provided a platform for improving other draft genome assemblies to chromosome-level.

Erigeron breviscapus (Vant.) Hand.-Mazz. is a famous traditional Chinese medicine that has positive effects on the treatment of cardiovascular and cerebrovascular diseases. With the increase of market demand (RMB 500 million per year) and the sharp decrease of wild resources, it is an urgent task to cultivate high-quality and high-yield varieties of E. breviscapus. However, it is difficult to obtain homozygous lines in breeding due to the self-incompatibility (SI) of E. breviscapus. Here, we first proved that E. breviscapus has sporophyte SI (SSI) characteristics. Characterization of the ARC1 gene in E. breviscapus showed that EbARC1 is a constitutive expression gene located in the nucleus. Overexpression of EbARC1 in Arabidopsis thaliana L. (Col-0) could cause transformation of transgenic lines from self-compatibility (SC) into SI. Yeast two-hybrid (Y2H) and bimolecular fluorescence complementation (BiFC) assays indicated that EbARC1 and EbExo70A1 interact with each other in the nucleus, and the EbARC1-ubox domain and EbExo70A1-N are the key interaction regions, suggesting that EbARC1 may ubiquitinate EbExo70A to regulate SI response. This study of the SSI mechanism in E. breviscapus has laid the foundation for further understanding SSI in Asteraceae and breeding E. breviscapus varieties.

This study was aimed at investigating the effect of four types of catalysts on the distribution characteristics of 16 polycyclic-aromatic hydrocarbon (PAHs) in bio-oils from the pyrolysis of dry sewage sludge. The pyrolysis experiments of sewage sludge were conducted in a tubular reactor at the high and low temperatures of 850 °C and 450 °C, respectively. CaO, KCl, Na2CO3, and Fe2O3were selected as catalysts for the catalyzed pyrolysis. In the non-catalyzed sludge pyrolysis, PAHs concentrations in bio-oil increased with temperature, reaching a maximum value of 15.25 µg/g at 850 °C. This value was 4.5 times higher than PAHs concentration from the non-catalyzed pyrolysis at 450 °C. With the presence of catalysts at 850 °C, an evident reduction of PAHs concentration was observed for all the bio-oil samples. The added catalysts proved to effectively inhibit PAHs formation in bio-oil and reduce the toxic equivalent quantity (TEQ) of PAHs in the experiment at high temperature. The lowest observed PAHs concentration in bio-oil was 4.23 µg/g, obtained with the use of KCl catalyst. However, an opposite trend was observed for catalyzed pyrolysis at 450 °C. The added catalysts promoted PAHs formation, up to the concentration of 8.89 µg/g with the use of CaO catalyst. Catalyst loading also influenced PAHs concentration and TEQ, with the lowest output found at 20% loading for all catalysts. Bio-oil yields, mass ratio of 16 PAHs concentrated into bio-oil, the TEQ of 16 PAHs and their subtotal group classified by ring number were separately examined to analyze their effects on fractional distribution of the total PAH amount. Overall, the presence of the selected catalysts was found to have the ability to inhibit PAHs production at higher pyrolysis temperature and to promote PAHs production at lower pyrolysis temperature.

In the light of optical theory, we advanc an ultra-wideband impulse radiating antenna (IRA) which is composed of an ellipsoidal reflector and a cone radiator. The high-intensity ultra-short electric pulses radiated by IRA can be transferred into the deep target in tissue non-invasively and be focused effectively. With the focused picosecond electric pulses, the organelles (mitochondria) transmembrane potential shall change to collapse under which the tumor cells will be targetly induced to apoptosis, so the method of non-invasive treatment of tumors would be achieved. Based on the time-domain electromagnetic field theory, the propagation characteristics of picosecond electric pulses were analyzed with and without the context of biological tissue, respectively. The results show that the impulse characteristics of input pulse were maintained and the picosecond electric pulses can keep high resolution in target areas. Meanwhile, because of the dispersive nature of medium, the pulse amplitude of the pulses will attenuate and the pulse width will be broadened.

The effects of copper accumulation and resistance in two biotypes of Cynodon dactylon were studied. Results showed that at a low concentration of copper (<100 mg/kg), the growth of Cynodon dactylon was generally unaffected. As copper concentration increased, negative effects on the growth of Cynodon dactylon became apparent. The critical concentration at which the plant exhibited poisoning symptoms was different for the two biotypes of Cynodon dactylon. At 500 mg/kg copper concentration in soil, the biotype from the polluted area showed significantly higher tolerance of copper than the biotype from the unpolluted area.

As a micro-wound and target-aimed technology without special limitation, Electric Pulses have been widely researched in tumor treatment and the effects have been demonstrated by a series of experiments, yet the mechanism has not been explained clearly. In this experiment, energy controllable steep pulse (ECSP) was used to treat nude mice bearing human ovarian tumor, and the result was compared with that of the control group. The expression of an important coagulant factor-tissue factor (TF) was analyzed, as TF was also a tumor indicator of invasion and metastasis, the result may indicate the relationship among ECSP, thrombosis and tumor invasion. In this study, to shed light on the mechanism of tumor treatment in electrical fields, nude mice bearing ovarian tumors were randomly divided into the treated group and the untreated group. We treated the former group and took out the tumor instantly. The thrombosis and necrosis of ovarian tumor were observed under microscope. The expression of TF was analyzed by SP immunohistochemistry and RT-PCR. Lower level of TF expression was noticed in the tumor tissue treated by ECSP, and more apparent thrombosis was also seen in this group. The results make it clear that ECSP can accelerate thrombosis and consume coagulant factors such as TF, and that low expression of TF in tumor tissue can cut out the signal paths of tumor invasion. So it is suggested that ECSP may restrain tumor invasion and metastasis by modulating thrombosis.

The study on the Cu forms in Cynodon dactylon rhizosphere soil of copper tailings yard in Tongling City, Anhui Province showed that among the test Cu forms, the amount of residual form occupied the majority, while that of exchangeable form was relatively low. Compared with non-rhizosphere soil, rhizosphere soil had a higher organic matter content but a lower pH. With the growth of C. dactylon, the contents of organically combined and exchangeable Cu in rhizosphere soil increased by 7.89% and 5%, respectively, while those of carbonate-combined and Fe-Mn oxides-combined Cu decreased. The growth of C. dactylon accelerated the transformation of Cu forms in rhizosphere soil, and decreased the rhizosphere soil Cu content through its absorption.

This experimental study was designed to investigate the effects and the expressions of microvessel density (MVD), vascular endothelial growth factor (VEGF) on the transplanted tumor in the rat model with Walker-256 after energy controllable steep pulse(ECSP). The experiment revealed that the steep pulse electrical field has better effect on tumor, compared with the control. The positive cell staining intensity of VEGF in the control group was significantly higher than that in ECSP group (P < 0.05). The number of MVD in the tumor tissues of ECSP group was significantly lower than that of tumor control group (P < 0.05). These results showed that ECSP could inhibit the growth and angiogenesis of tumor and its pathway is to down-regulate the expression of VEGF possibly.

By the method of solution culture, this paper studied the coercion and damage of Cu pollution on the growth of Artemisia lavandulaefolia. The Cu concentration was set as 2.5, 5, 10, 20 and 40 mg x L(-1), experimental duration was 14 days, and the growth and physiological indices of plants were tested. The results showed that the growth of A. lavandulaefolia was stimulated at low Cu concentration (2.5 mg x L(-1)), while inhibited at higher Cu concentrations (5 to approximately 40 mg x L(-1)). There was a significant negative correlation between each growth index and Cu concentration, and pigments contents had the similar trend. The sensibility of various photosynthetic pigments to Cu was in the order of chlorophyll a > chlorophyll a + b > chlorophyll b > carotenoid. Cell membrane permeability, O2 generation rate, and MDA content decreased slightly at 2.5 mg Cu x L(- 1) and then increased with increasing Cu concentration. The activities of POD, SOD and CAT increased first but decreased then with the increasing Cu concentration, and the endurance index of root showed the same trend, being > 0.5 at the Cu concentration less than 20 mg x L(-1) while decreased to 0.36 at 40 mg Cu x L(-1).

The study on the soil and Paeonia ostii Cu, Cd, Pb and Zn at the copper-tailings reservoir of Tongling City, Anhui Province showed that the forms and contents of test heavy metals were differed in Paeonia ostii-planted and control soils. The Cu and Cd contents in control soil were 4.36-14.43 and 3.67-3.86 times of the 2nd national standard for soil environmental quality, respectively, and the composite pollution index of the heavy metals in Paeonia ostii-planted soil was up to 5.607. The heavy metals content in Paeonia ostii leaf and root-stem was in order of Cu>Zn>Pb>Cd, and that in Paeonia ostii stem and root- peel was Zn>Cu>Pb>Cd. The comparison of the heavy metals contents in Paeonia ostii with the criterion about herbal medicine showed that the Cu content in Paeonia ostii was 1.43-2.53 times of the criterion, and the Cd content was 1.17-3.17 times of the criterion.
