Objective @#In the theories of pulse disgnosis in traditional Chinese medicine (TCM), it is emphasized that pulse manifestations at the radial artery within the wrist (called Cunkou) signify the physiological and pathological conditions of different internal organs in the human body. However, different opinions exist among researchers about the objectiveness of the pulse diagnosis technique. Some researchers mentioned that no significant differences were observed in pulse manifestations at various Cunkou areas, hence there might be some difficulty in evaluating the status of different organs through checking pulse manifestations at Cunkou. This research aims to analyze the pulse response at Cunkou from the aspect of the characteristics of tactile sensing, thus to give a preliminary explanation to the above question.@*Methods@#This research utilized the Weber-Fechner law to model the tactile sensing as a dynamic low-pass signal filter with varying bandwidths under different compression levels during pulse diagnosis. The model was applied to analyzing the clinical data collected previously by our group. The arterial pressures measured invasively with equipment from 14 patients with aorta coarctation were processed to simulate different pulse manifestations at Cun, Guan, and Chi positions of Cunkou when different compression levels were applied.@*Result@#Due to the characteristics of tactile sensing, significant variations were observed in pulse manifestations at different pulse-depths under the application of changing compression levels; while no such changes in pulse manifestations were observed from the employment of transducer only, without tactile sensing involved. The results explained why different understandings on pulse manifestations were formed between direct pulse-taking technique in TCM and modern sphygmography using transducers. The features of pulse manifestations at Cunkou, using direct pulse-taking technique but at different depths, superficial, middle, and deep positions, respectively, predicted by the developed tactile sensing model were in line with those described in TCM pulse theories.@*Conclusion@#Based on the developed tactile sensing model, this study preliminarily explains the phenomenon that pulse manifestation at Cunkou changes in response to the compression force applied during TCM pulse-taking. Integrating the tactile sensing model with the study of TCM pulse diagnosis opens a new chapter for visualizing and quantitatively interpreting pulse manifestations. This not only expands the scope of pulse diagnosis study effectively, but also provide a scientific basis for further technical upgrading and optimization of existing pulse diagnosis equipment.

This study aimed to explore the effect of Ganmai Dazao Decoction on the ethology of rats with posttraumatic stress disorder(PTSD) and study the related mechanism through the changes in magnetic resonance imaging and protein expression. Sixty rats were randomly divided into 6 groups, namely the normal group, the model group, the low(1 g·kg~(-1)), medium(2 g·kg~(-1)), and high-dose Ganmai Dazao Decoction groups(4 g·kg~(-1)), and the positive control group(intragastric administration with 10.8 mg·kg~(-1) of fluoxetine), with 10 rats in each group. Two weeks after inducing PTSD by single-prolonged stress(SPS) in rats, the positive control group was given fluoxetine hydrochloride capsule by gavage, the low, medium, and high-dose groups were given Ganmai Dazao Decoction by gavage, and both the normal group and the model group were given the same volume of normal saline by gavage, each for 7 days. The open field experiment, elevated cross elevated maze, forced swimming experiment, and new object recognition test were carried out for the behavioral test. Three rats in each group were selected to detect the expression of neuropeptide receptor Y1(NPY1R) protein in the hippocampus by Western blot. Then, the other three rats in each group were selected to use the 9.4T magnetic resonance imaging experiment to observe the overall structural changes in the brain region and the anisotropy fraction of the hippocampus. The results of the open field experiment showed that the total distance and central distance of rats in the model group were significantly lower than those in the normal group, and the total distance and central distance of rats in the middle and high-dose Ganmai Dazao Decoction groups were higher than those in the model group. The results of the elevated cross maze test showed that medium and high-dose Ganmai Dazao Decoction remarkably increased the number of open arm entries and the residence time of open arm of rats with PTSD. The results of the forced swimming experiment showed that the immobility time in the water of the model group rats was significantly higher than that of the normal group, and Ganmai Dazao Decoction hugely reduced the immobility time in the water of rats with PTSD. The results of the new object recognition test showed that Ganmai Dazao Decoction significantly increased the exploration time of new objects and familiar objects in rats with PTSD. The results of Western blot showed that Ganmai Dazao Decoction significantly reduced the expression of NYP1R protein in the hippocampus of rats with PTSD. The 9.4T magnetic resonance examination found that there was no significant difference in the structural image among the groups. In the functional image, the fractional anisotropy(FA value) of the hippocampus in the model group was significantly lower than that in the normal group. The FA value of the hippocampus in the middle and high-dose Ganmai Dazao Decoction groups was higher than that in the model group. Ganmai Dazao Decoction reduces the injury of hippocampal neurons by inhibiting the expression of NYP1R in the hippocampus of rats with PTSD, thereby improving the nerve function injury of rats with PTSD and playing a neuroprotective role.

Plants are a rich source of secondary metabolites that have been found to have medicinal properties. The present study was conducted to evaluate the phytochemical screening, antibacterial activities and heavy metal analysis of seven medicinal plants i.e., Nigella sativa (seeds), Trigonella foenum-graecum (seeds), Brassica campestris (seeds), Pistacia integerrima (galls), Linum usitatissimum (seeds), Hyssopus officinalis (flowers), Ephedra vulgaris (dry branches) and its two recipes which are used by hakims (Practitioners of local herbal medicines), against different diseases particularly respiratory tract infections. The obtained results revealed that alkaloids (30%) and flavonoids (41%) were in maximum quantity in P. integerrima (galls) while saponins (10.9%) were in maximum quantity in Recipe 1. The antibacterial activity was determined by the agar well disc diffusion method using methanol, ethanol, chloroform and deionized water extracts. Each plant extract was tested against one Gram-positive (Streptococcus pneumonia) and two Gram-negative (Pseudomonas aeruginosa and Klebsiella pneumonia) bacteria. Maximum zones of inhibition in methanol, ethanol, chloroform and aqueous extract were seen in T. foenum-graecum against S. pneumonia (20.06 ± 0.16 mm), B. campestris against S. pneumonia (22.40 ± 0.24 mm), Recipe 2 against K. pneumonia (20.06 ± 0.16 mm) and N. sativa against S. pneumonia (20.23 ± 0.16 mm), respectively. The concentrations of heavy metals were determined by atomic absorption spectrophotometer and showed the existence of high concentration of Iron (Fe), Lead (Pb) and Chromium (Cr). Thus, it has been found that medicinal plants individually as well as their recipes are potentially active against various diseases particularly respiratory tract infections.

The aim of this study was to explore the phytochemical composition, heavy metals analysis and the antibacterial activity of six medicinal plants i.e., Terminalia chebula Retz (fruits), Aegle marmelos L., (fruits), Curcuma longa L., (rhizomes), Syzygium aromaticum L., (flower buds), Piper nigrum L., (seeds), Cinnamomum cassia L., (barks) and its two remedial recipes (recipe 1 and 2) used against diarrhea obtained from the local herbal practitioners (Hakeems). A preliminary phytochemical screening of the above-mentioned plants extract in methanol, chloroform, n-hexane and distilled water revealed the presence of various constituents such as alkaloids, flavonoids, tannins and saponins by using standard procedures. The quantitative phytochemical studies shows that alkaloids, flavonoid and saponins were in maximum amount in Terminalia chebula. The concentration of Cd, Ni, Pb, Fe, Cr, Cu and Zn were investigated by using an atomic absorption spectrometer. The obtained analysis shows that Cr, Fe and Pb were present in the highest concentration in medicinal plants and their recipes. The antibacterial activities of the crude extract found in the recipes of methanol, chloroform, n-hexane and distilled water were analyzed by using agar well disc diffusion assay and minimum inhibitory concentration (MIC) by broth dilution method against four bacterial strains, namely, E. coli, Salmonella, Shigella and Methicillin-resistant Staphylococcus aureus (MRSA), respectively. The maximum zones of inhibition in methanol, water, chloroform and n-hexane extracts were seen in recipe 2 against Shigella (22.16 ± 0.47 mm), recipe 2 against Shigella (20.33 ± 0.24 mm), recipe 1 against Shigella (20.30 ± 0.29 mm) and recipe 2 against E. coli (30.23 ± 0.12 mm), respectively. Furthermore, the recipe extracts are more active against the tested bacterial strains than the extracts from individual plants. Therefore, it is concluded that the use of herbal plants and their recipes are the major source of drugs in a traditional medicinal system to cure different diseases.

BACKGROUND: Patients with chronic prostatitis/chronic pelvic pain syndrome (CP/CPPS) frequently show depressive symptoms clinically and increasing evidence indicates a correlation between CP/CPPS and depression. However, the underlying mechanisms of CP/CPPS-related depression remain poorly understood. Here, we sought to determine the role of hippocampal microglial activation and neurobiological changes in a mouse model of experimental autoimmune prostatitis (EAP)-induced depression and the treatment efficacy of Chinese herb extract baicalein. METHODS: EAP was induced through intradermal injection of prostate antigen and adjuvant twice. Then, mice were assessed for affective behaviors in the open field test, elevated plus maze, forced swim test, and tail suspension test. The morphology and function of microglia and astrocytes were detected by immunofluorescence, Western blotting, and transmission electron microscopy. Proinflammatory mediators along with serotonin transporter (SLC6A4/SERT) and indoleamine 2,3-dioxygenase (IDO) were quantified with reverse transcription-polymerase chain reaction (RTPCR), and serum serotonin concentrations were measured by enzyme-linked immunosorbent assay (ELISA). Proton magnetic resonance spectroscopy (1H-MRS) was performed to measure hippocampal glutamate levels. In addition, baicalein was used in a subset of EAP mice to test its anti-depressant action. RESULTS: EAP was successfully established and induced depressive- and anxiety-like behavior in mice. Increasing levels of co-expressed ionized calcium-binding adapter molecule 1 (Iba1) and glial fibrillary acidic protein (GFAP) and ultrastructural observations suggested microglial activation and reactive astrocytosis in the hippocampus. These activated microglia resulted in increased expressions of multiple proinflammatory cytokines. Simultaneously, EAP mice showed higher gene expressions of SLC6A4 and IDO and lower concentrations of serotonin. 1H-MRS indicated a decrease in the glutamate + glutamine (Glx)/total creatine (tCr) ratio in EAP mice. Furthermore, baicalein treatment alleviated the depressive-like behavior and neuroinflammation by suppressing the nuclear factor-kappa B (NF-κB) pathway. CONCLUSION: Our data indicate that EAP-induced depressive-like behavior is linked to microglia activation and subsequent neurotransmitter metabolism. Moreover, baicalein attenuates behavioral changes by inhibiting neuroinflammation via downregulation of the NF-κB pathway.

Chemodynamic therapy (CDT) was widely exploited for cancer therapy and expected to replace traditional anticancer drug therapies. Generally, CDT needs to combine with extra therapeutic methods for obtaining the optimal therapeutic efficacy of cancer. Herein, a multifunctional theranostic platform combing CDT with limotherapy was developed via nanoselenium (nano-Se)-coated manganese carbonate-deposited iron oxide nanoparticle (MCDION-Se). MCDION-Se could release abundant of Mn2+ ions that catalyzed H2O2 into hydroxyl radicals (·OH) via a Fenton-like reaction, effectively inducing the apoptosis of cancer cells. Besides, nano-Se coated onto MCDION-Se also dramatically activated superoxide dismutase (SOD) and promoted the generation of superoxide anion radicals (SOARs) in tumor tissue. Subsequently, a high content of H2O2 was produced via SOD catalysis of SOARs, further enhancing CDT efficiency. Meanwhile, the nano-Se and Mn2+ ions inhibited the generation of adenosine triphosphate (ATP), thus starving cancer cells. In addition, in vitro and in vivo experiments showed that MCDION-Se could effectively enhance the contrast of tumor tissue and improve the quality of magnetic resonance imaging (MRI). Overall, this work provided a nanoplatform that combined CDT with limotherapy for cancer therapy and simultaneously utilized MRI for monitoring the treatment of tumors.

In this study, the induction of hairy roots of Bupleurum chinense DC. was explored and established after experiments at different conditions: A. rhizogenes A4 was used to infect the leaves bases of B. chinense tube seedlings. The explants were co-cultured on Phytagel-solidified media for 3 days and then, were turned into solid media, similar with the co-culture media except that bacteriostat was added. After 10 days, rootlets began to appear and after 4 to 5 weeks, rootlets can be converted into liquid shaking culture stage. Plants regeneration from hairy root was useful for the research of new germplasm production and the variety improvement breeding. In the present study, the regenerated plants were obtained. One approach was to continuously culture under light conditions the seedlings which parting off spontaneously from the hairy roots during liquid shaking culture. The other approach was to culture the callus-like tissues produced by hairy roots with the optimized regeneration media for the induction of regenerated plants. The results of present study provide a technique to induce hairy roots and plantlet regeneration of B. chinense and this technique is helpful for the researches on metabolism, especially on the Agrobacterium-mediated genetic transformation of B. chinense.
