RESUMO

OBJECTIVE: To explore the underlying mechanism of acupuncture on nerve repair by investigating its effect on the differentiation of glial cells and the repair of glial scars. METHODS: Sprague-Dawley rats were randomly allocated to three groups: normal group, model group, and acupuncture group. Acupuncture was applied at Renzhong (GV26), Baihui (GV20), Fengfu (GV16), Yamen (GV15) and Hegu (LI4) within 12 h after TBI modeling with a frequency of one session per day for 4 weeks. Neurobehavioral assessment, hematoxylin and eosin staining, immunofluorescence detection, and magnetic resonance imaging scanning were performed on days 3, 7, 14, and 28 after modeling of traumatic brain injury (TBI). RESULTS: Acupuncture promoted the proliferation of glial cells and glial scars at an early stage but inhibited the proliferation of glial cells and glial scars at a late stage. Morphological observations and immunofluorescence histochemistry showed that the morphology of the perilesional cortex in the acupuncture group was improved and the number of neurons was increased when compared with the model group. The lesion size of ipsilateral brain parenchyma in the acupuncture group was smaller than in the model group on days 7, 14, and 28 ( < 0.05) after TBI modeling. CONCLUSIONS: Acupuncture might have a bidirectional regulatory effect on glial scar repair after TBI by promoting the proliferation of glial cells and glial scars to limit the injured area and relieve nerve injury during the early stages, and by inhibiting glial scar hyperplasia to benefit the regeneration and repair of neurons and axons and promote neurological function recovery during the later stages.

Objective@#To observe the pharmacodynamic effect of Xiaobanxia Decoction (XBXD) and categorized formula on cisplatin-induced delayed chemotherapeutic vomiting in rats, and to explore its prevention and treatment mechanism.@*Methods@#Sixty Wistar rats were randomly divided into blank group, model group, ondansetron group, XBXD group, Ginger Pinellia Decoction group and Pinellia and Dried Ginger Powder group, with 10 rats in each group. The ondansetron group was given 2.6 mg/(kg•d) ondansetron, the XBXD group was given XBXD 22 g/(kg•d), the Ginger Pinellia Decoction group was given 31.5 ml/(kg•d) Ginger Pinellia Decoction, and Pinellia and Dried Ginger Powder group was given 0.63 g/(kg•d) Dried Ginger Powder. The blank group and the model group were intragastrically administered with normal saline. All the treattmens were last for three days and twice per day. After the first administration, except the blank group, the rats in all groups were intraperitoneally injected with 6 mg/kg cisplatin to establish chemotherapy-induced vomiting model. The kaolin intake of the chemotherapical rats were weighed every 24 h, and the ileum and medulla 5-HT, 5-HIAA, TPH and MAOA levels were detected by ELISA.@*Results@#Compared with the model group, the kaolin intake of the chemotherapy rats in each treatment group significantly decreased (P<0.05). The levels of ileum 5-HT (308.04 ± 29.90 ng/L, 355.97 ± 19.16 ng/L, 389.97 ± 24.86 ng/L vs. 498.95 ± 13.92 ng/L) and medulla 5-HT (375.32 ± 19.33 ng/L, 395.18 ± 16.12 ng/L, 406.68 ± 12.09 ng/L vs. 478.52 ± 13.88 ng/L) in the XBXD group, Ginger Pinellia Decoction group and Pinellia and Dried Ginger Powder significantly decreased (P<0.05). The levels of ileum TPH (35.14 ± 2.68 ng/L vs. 47.31 ± 0.83 ng/L) in the XBXD group significantly decreased (P<0.05). The levels of medulla TPH (33.68 ± 2.79 ng/L, 38.19 ± 1.74 ng/L vs. 43.68 ± 1.53 ng/L) in the Ginger Pinellia Decoction group and Pinellia and Dried Ginger Powder significantly decreased (P<0.05). The levels of MAOA, 5-HIAA in the ileum and medulla of the XBXD group, Ginger Pinellia Decoction group and Pinellia and Dried Ginger Powder significantly decreased (P<0.05).@*Conclusions@#XBXD and categorized formula can effectively prevent and treat delayed chemotherapy-induced vomiting in rats, and its mechanism may be related to decrease the TPH level and inhibiting 5-HT synthesis.

The levels of ileum 5-HT (308.04 ± 29.90 ng/L, 355.97 ± 19.16 ng/L, 389.97 ± 24.86 ng/L vs. 498.95 ± 13.92 ng/L) and medulla 5-HT (375.32 ± 19.33 ng/L, 395.18 ± 16.12 ng/L, 406.68 ± 12.09 ng/L vs. 478.52 ± 13.88 ng/L) in the XBXD group, Ginger Pinellia Decoction group and Pinellia and Dried Ginger Powder significantly decreased ( P<0.05). The levels of ileum TPH (35.14 ± 2.68 ng/L vs. 47.31 ± 0.83 ng/L) in the XBXD group significantly decreased ( P<0.05). The levels of medulla TPH (33.68 ± 2.79 ng/L, 38.19 ± 1.74 ng/L vs. 43.68 ± 1.53 ng/L) in the Ginger Pinellia Decoction group and Pinellia and Dried Ginger Powder significantly decreased ( P<0.05). The levels of MAOA, 5-HIAA in the ileum and medulla of the XBXD group, Ginger Pinellia Decoction group and Pinellia and Dried Ginger Powder significantly decreased ( P<0.05). Conclusions:XBXD and categorized formula can effectively prevent and treat delayed chemotherapy-induced vomiting in rats, and its mechanism may be related to decrease the TPH level and inhibiting 5-HT synthesis.

<p><b>OBJECTIVE</b>To observe the difference in the therapeutic effects on infantile herpetic angina among the treatments with western medication, acupuncture and the combined therapy of acupuncture and western medication.</p><p><b>METHODS</b>One hundred and twenty children of herpetic angina were randomized into a western medication group, an acupuncture group and a combined therapy group, 40 cases in each one. Besides the basic treatment with symptomatic support, in the western medication group, ribavirin granules were prescribed for oral administration, 10 mg/kga day, three times a day, for 3 days. In the acupuncture group, acupuncture was applied at Fengchi (GB 20), Hegu (LI 4), Quchi (LI 11), Xiaguan (ST 7), Jiache (ST 6), Dicang (ST 4), Shaoshang (LU 11) and Shangyang (LI 1), once a day, for 3 days. In the combined therapy group, the treatments as the western medication group and the acupuncture group were used, in which, the oral administration with ribavirin granules and acupuncture were adopted for 3 days. The clinical therapeutic effects were compared after treatment and the follow-up visit was conducted since the 4th day at the end of treatment to compare the antifebrile time, herpes relieving time and the use frequency of ibuprofen oral liquid in the children among the three groups.</p><p><b>RESULTS</b>After treatment, the total effective rate was 95.0% (39/40) in the combined therapy group, was 62.5% (25/40) in the western medication group and was 80.0% (32/40) in the acupuncture group. The result in the combined therapy group was better than those in the acupuncture group and the western medication group, indicating the significant differences (both<0.05). The antifebrile time and the herpetic relief time in the combined therapy group were earlier than those in the acupuncture group and the western medication group, indicating the significant differences (both<0.05). The use frequency of ibuprofen oral liquid in the combined therapy group was lower than those in the acupuncture group and the western medication group, indicating the significant difference (both<0.05).</p><p><b>CONCLUSIONS</b>The combined therapy with acupuncture and western medication relieve fever and herpes much more quickly and reduces the use frequency of ibuprofen oral liquid as compared with the simple use of acupuncture or western medication in the treatment of infantile herpetic angina, which achieve the much better therapeutic effects.</p>

<p><b>OBJECTIVE</b>To explore the effects of acupuncture and medication on PI3K/Akt/mTOR signaling pathway in rats with premature ovarian failure.</p><p><b>METHODS</b>Ten of fifty SPF-grade female SD rats were randomly selected into a normal group, and the remaining 40 rats were treated with intraperitoneal injection of cyclophospha mide (30 mg/kg) for consecutive 5 days to establish rat model of premature ovarian failure. Thirty five successful rat models were randomly divided into a model group (9 cases), a medication group (9 cases), an acupuncture group A (9 cases) and an acupuncture group B (8 cases). The rats in the model group and normal group did not receive any treatment. The rats in the medication group were treated with intragastric administration of diethylstil bestrol, once a day. The rats in the acupuncture group A and acupuncture group B were respectively treated with acupuncture at different acupoints, twice a day. All the treatment was given for 4 weeks. After the treatment, enzyme-linked immunosorbent assay (ELISA) was applied to test the levels of estradiol (E2), progesterone (P), follicle stimulating hormone (FSH) and luteotropic hormone (LH). The ovarian tissue sample was processed with hematoxylin eosin (HE) staining as well as RNA and protein extraction to test the mRNA expression of estrogen receptor alpha (ERalpha), estrogen receptor beta (ERP), phosphatidylinositol 3-kinase/serine/threonine kinase (PI3K), protein kinase B (Akt) and mammalian target of rapamycin (mTOR).</p><p><b>RESULTS</b>High-dose short-term in- tervention of cyclophosphamide could establish rat model of premature ovarian failure with a successful rate of 87.5%. Compared with the normal group, the vaginal smear in the model group was featured with signs of estro gen deficiency, early-follicle reduction, structural damage to the follicle, and reducing number of mature follicles; the level of E2 was significantly reduced (P<0.05), levels of P, FSH and ILH were increased (all P<0.05), and mRNA expression of estrogen-related ERP3, PI3K, Akt and mTOR were all reduced (all P<0.05). Compared with the model group, the number of mature follicle was increased in the medication group and acupuncture groups, the levels of E2 was obviously increased (all P<0.05). level of FSH was reduced (all P<0.05), and mRNA expression of PI3K, Akt and mTOR all showed an increasing trend (all P<0.05). The differences of each index result between acupuncture groups and medication group were not significant (all P>0.05).</p><p><b>CONCLUSION</b>Acupuncture has certain advantage for the treatment of premature ovarian failure, which achieves similar therapeu tic effect as estrogen; the possible mechanism may be related to up-regulation of gene and protein expression in PI3K/Akt/mTOR signaling pathway.</p>

Objective: To observe the influence of scalp acupuncture plus point injection on intelligence quotient in microcephaly children. Methods: Twenty-five children with microcephaly were treated by scalp acupuncture as main therapy plus point injection. The children's intelligence was tested by China-Wechsler's intelligence scale before and after treatment. Results and Conclusion: Acupuncture can improve intelligence and especially has significant effect on verbal intelligence quotient in microcephaly children.

Objective To observe the effect of acupuncture on the infracted area and apoptosis of rats with brain contusion.Methods Feeney's method was used to establish brain contusion model.The 40 rats were randomized into sham-operation,model,acupuncture and naloxone (NX) group with 10 in each.Acupuncture was performed for the rats of acupuncture group and NX was injected intraperitoneally for the rats of NX group.The treatment was done once everyday,7 days altogether.Then the brain histotomy was prepared after all the treatment finished.Computer image analysis system was used to detect the infracted area.TUNEL method was adopted to detect the apoptosis.Results The brain contusion in the model group was severe,there were lots of apoptotic cells and the infracted areas were bigger than the injured areas (P0.05).Conclusion Acupuncture can decrease the infracted areas and apoptosis of the rats with brain contusion,being effective in anti-secondary brain injuries.
