RESUMO

Pain after burns is a very common problem in children. Severe pain will not only hinder treatment, but also damage children's mental health if not handled in time. Therefore, pain management is very important in treating children with burns. As a safe, effective, and convenient non-drug therapy, music therapy has great advantages in relieving pain and is widely used in a variety of clinical fields. This paper focused on music therapy and its mechanism of pain relief, the current status of research on pain management of pediatric burns, the application and prospect of music therapy in pain management of pediatric burns, etc., to provide reference for clinical application.

OBJECTIVE: The aim of this study was to evaluate the impact of particulate matter 2.5 (PM2.5) on liver function at the animal level and to study its impact targets. MATERIALS AND METHODS: 60 male and female BALB/c mice of SPF grade, aged 6-8 weeks, were randomly divided into four groups, with 15 mice in each, including the normal saline control group, the PM2.5 low dose group [2 µg/(100 g/d)], the PM2.5 medium dose group [8 µg/(100 g/d)] and the PM2.5 high dose group [16 µg/(100 g/d)]. Each day, 0.9% saline or PM2.5 particles were administered through the nasal route, and samples were taken after 3 weeks of continuous exposure. Hematoxylin-eosin staining (HE) was used to observe the liver damage caused by PM2.5. Serum alanine aminotransferase (ALT) and aspartate aminotransferase (AST) levels were detected by using an automatic biochemical analyzer to detect the content of liver glycogen and blood glucose. Multiple indicators were observed, including plasma tumor necrosis factor (TNF-α) and interleukin-6 (IL-6) levels, oxidative stress response indicators reactive oxygen species (ROS), malondialdehyde (MDA), superoxide dismutase (SOD) detection, RT-PCR and Western blot detection of glycogen synthase (GS), glucokinase (GK), nuclear factor erythroid 2-related factor 2 (Nrf2) expression and phosphorylation level of phospho-c-Jun N-terminal kinases (p-JNK). RESULTS: PM2.5 can cause damage to the liver by increasing PM2.5 concentrations, raising the metabolic rate of liver cells, resulting in a substantial amount of inflammatory infiltration and vacuolar degeneration of cells, and increasing the liver/body weight. TNF-α and IL-6 inflammatory factor expression increased (p<0.05). An increase in the serum ALT and AST levels were also observed. The blood glucose of mice increased, whereas the content of liver glycogen declined (p<0.05). ROS, MDA, and SOD levels all increased considerably. PM2.5 can drastically lower the expression of GS and GK, increase the expression of Nrf2, and raise the phosphorylation level of p-JNK (p<0.05). CONCLUSIONS: PM2.5 can induce oxidative stress in mouse liver through the Nrf2/JNK pathway, induce liver inflammation in mice, and inhibit glycogen synthesis.

In addition to offering methionine, 2-hydroxy-4-methylthiobutyric acid (HMTBa) is also an organic acid and shows excellent bacteriostasis. Therefore, 3 experiments were conducted to determine the influence of drinking water supplemented HMTBa in combination with acidifier on performance, intestinal development, and microflora in broilers. The addition of different concentration (0.02-0.20%) of the blend of HMTBa and other acids significantly reduced the pH of water and exerted antimicrobial activity in dose-dependent manner in vitro. The outcomes from animal trial consisting of the drinking water with blended acidifier at 0.00, 0.05, 0.10, 0.15, and 0.20% indicated that the water with 0.15 or 0.20% acidifier resulted in linear and quadratic higher body weight at 42 d, gain and water consumption during 1 to 42 d (P < 0.05). In experiment 3, responding to graded blended acidifier in drinking water, birds receiving 0.10, 0.15, and 0.20% acidifier decreased the internal pH of gastrointestinal tract and muscle, and exhibited increased duodenal weight, length, villus high, and the ratio of villus high to crypt depth. Drinking water with 0.2% blended acidifier increased the abundance of probiotics (Bacteroidaceae, Ruminococcaceae, and Lachnospiraceae) and decreased the account of pathogenic bacteria such as Desulfovibrionaceae. Alternations in gut microflora were closely related to the metabolism of carbohydrate, amino acid, and vitamins. These findings, therefore, suggest that drinking water with 0.10 to 0.13% the combination HMTBa with acidifier might benefit to intestinal development and gut microbiota, and the subsequent produce a positive effect on the performance of broilers.

OBJECTIVE: Inflammatory diseases are risk factors for osteoporosis. We aimed to explore whether fibrinogen, which is linked to chronic inflammation, is associated with bone mineral density (BMD) in menopausal women. METHODS: In this cross-sectional study, we analyzed 339 menopausal women from Zhejiang Province between January 2016 and October 2019. Linear regression analysis was performed to assess the relationship between fibrinogen and BMD. RESULTS: Significant inverse association was observed between the serum fibrinogen level and BMD in menopausal women. The mean BMD in each quartile of fibrinogen level was 0.901, 0.897, 0.892, and 0.855 g/cm2, respectively (p = 0.027). After adjusting for age, body mass index, metabolic profiles, blood inflammatory factors, and serum levels of estradiol, calcium, phosphorus, and alkaline phosphatase, fibrinogen levels remained significantly associated with BMD (regression coefficients for quartiles 1-3 vs. quartile 4 were 0.046, 0.027, and 0.036, respectively; p for trend <0.05). CONCLUSIONS: Higher fibrinogen levels were associated with lower BMD in menopausal women, which was independent of age, body mass index, estradiol, and other factors. Therefore, serum fibrinogen can be used as a new predictor of reduced BMD in menopausal women.

Genetic selection and intensive nutrition for increased growth rate in meat-type ducks has resulted in an imbalance between pectorales increment and sternal mass, which is detrimental to productivity and welfare. Reducing body weight and increasing sternal mass probably reverses these adverse effects. Therefore, 2 experiments (Expt.) were conducted to investigate the effects of 25-hydroxycholecalciferol (25-OH-D3), a vitamin D3 metabolites, on sternal mass. In Expt. 1, 512 1-day-old male ducks were randomly assigned to 4 low-nutrient density diets and received following treatments in a 2 × 2 factorial arrangement: (i) NRC or China Agricultural industry standards (NY/T) vitamin premixes and (ii) 0.069 mg/kg 25-HyD in feed or not. At 49 D of age, regardless of 25-OH-D3, NY/T vitamin regimen inhibited bone turnover and consequently increased sternal trabecular bone volume and mineral deposition compared with NRC vitamin premix. Supplementing 25-OH-D3 to NRC but not NY/T vitamin regimen significantly improved sternal microarchitecture and mineral content, which companied by decreased serum bone resorption markers concentration, as well as downregulation of the gene expressions of osteoclast differentiation and activity. In Expt. 2, 256 1-day-old male ducks were fed a standard nutrient density diet contained NRC vitamin premix with 0 or 0.069 mg/kg of 25-OH-D3. Results also showed that 25-OH-D3 treatment significantly improved sternal mineral accumulation and microarchitecture, along with decreasing osteoblast and osteoclast numbers in bone surface, declining serum bone turnover markers levels, and increasing serum Ca concentration. Collectively, these findings indicated that the dietary administration of 25-OH-D3 increased sternal mass in NRC vitamin diet by suppressing bone resorption in 49-day-old meat duck.

Genetic progress and increasing nutrient density for greater body mass and meat yield in poultry has inadvertently led to an imbalance between pectorales mass and sternal development which may or may not be detrimental to productivity and welfare. Slowing weight gain while promoting bone mineralization could positively influence sternal health. Thus, the present study aimed to evaluate the effect of graded calcium (Ca) supplementation in low nutrient density (LND) diets on sternal mass and bone turnover in meat ducks. Male meat ducks (720, 15-day-old) were randomly assigned and fed a standard nutrient density positive control (PC) diet, and 4 LND diets with 0.5, 0.7, 0.9, and 1.1% Ca, respectively. Metabolic energy (ME) was reduced in the LND by 9.5 and 16.3% at 15-35 D and 36-56 D compared to PC diet, respectively, while maintaining proportionate essential nutrient proportions to energy similar as in the PC diet. Although the 0.9% Ca LND diet decreased body weight and sternal dimension, it increased the relative sternum weight, the trabecular bone volume/tissue volume (BV/TV) and Ca content of the sternum compared with PC diet. Feeding 0.7% or more Ca with the LND diet significantly increased the mineral content, bone density, BV/TV, and trabecular number of the sternum for 49-days-old ducks. Furthermore, the LND diet with 0.7% or more Ca-increased osteocyte-specific gene mRNA and osteoprotegerin (OPG) expression, and it blocked the expression of cathepsin K and decreased osteoclasts number per bone surface. Tartrate-resistant acid phosphatase (TRAP) staining also revealed that the addition 0.7% or more Ca to the LND diet significantly decreased the number of osteoclasts compared with the 0.5% Ca LND diet. Meanwhile TRAP activity in serum was significantly decreased in 0.7% or more Ca-treated groups. We concluded that LND diet with 0.7% or more Ca may maintain optimal sternal mass through suppressing bone resorption for meat duck.

The native places of the famous Chinese medicine doctors listed in the Volume on History of Medicine and Medical Literature of the Great Dictionary of Traditional Chinese Medicine were analyzed and counted, and the political, cultural, social andeconomic factors influencing the regional distributions of the famous Chinese medicine doctors were briefly analysed. Such distributions appear to be obviously regional, Zhejiang, Jiangsu and Anhui are the 3 provinces with the largest number of famous Chinese medicine doctors. Meanwhile, the distributions of the famous Chinese medicine doctors are centered on certain cities and counties even within those 3 provinces. From the historical distributions of the famous Chinese medicine doctors, it reveals that the doctors' experiences of rising to fame in the Chinese medicine have relatively close connection to the regional distributions and running within the families of the famous Chinese medicine doctors, the traditional method of transmission from master to disciple in the family should still be paid much attention to in training the students of Chinese medicine in modern times.

This study was conducted to determine the DE and ME content of 25 samples of corn distillers dried grains with solubles (DDGS) fed to growing pigs and to generate prediction equations for DE and ME based on chemical analysis. The 25 samples included 15 full-oil (no oil extracted; ether extract [EE] > 8%) DDGS and 10 reduced-oil (oil extracted; EE < 8%) DDGS collected from 17 ethanol plants in China. A corn­soybean meal diet constituted the basal diet and the other 25 diets replaced a portion of the corn, soybean meal, and lysine of the basal diet with 28.8% of 1 of the 25 corn DDGS sources. Seventy-eight barrows (initial BW = 42.6 ± 6.2 kg) were used in the experiment conducted over 2 consecutive periods (n = 6 per treatment) using a completely randomized design. For each period, pigs were placed in metabolism cages for a 5-d total collection of feces and urine following a 7-d adaptation to the diets. Among the 25 corn DDGS samples, EE, NDF, DE, and ME content (DM basis) ranged from 2.8 to 14.2%, 31.0 to 46.6%, 3,255 to 4,103 kcal/kg, and 2,955 to 3,899 kcal/kg, respectively. Using a stepwise regression analysis, a series of DE and ME prediction equations were developed not only among all 25 DDGS but also only within 15 full-oil DDGS and 10 reduced-oil DDGS samples. The best fit equations of DE (kcal/kg DM) for the complete set of 25 DDGS, 15 full-oil DDGS, and 10 reduced-oil DDGS were 2,064 ­ (38.51 × % NDF) + (0.64 × % GE) ­ (39.70 × % ash), ­(87.53 × % ADF) + (1.02 × % GE) ­ (22.99 × % hemicellulose), and 3,491 ­ (40.25 × % NDF) + (46.95 × % CP), respectively. The best fit equations for ME (kcal/kg DM) for the complete set of 25 DDGS, 15 full-oil DDGS, and 10 reduced-oil DDGS were 1,554 ­ (44.11 × % NDF) + (0.77 × % GE) ­ (68.51 × % ash), 7,898 ­ (42.08 × % NDF) ­ (136.17 × % ash) + (101.19 × % EE) (103.83 × % CP), and 4,066 ­ (46.30 × % NDF) + (45.80 × % CP) ­ (106.19 × % ash), respectively. Using the sum of squared residuals to compare the accuracy of the 3 groups of prediction equations revealed that separate equations for full-oil DDGS and reduced-oil DDGS each provided a better fit than a single equation for the entire set of DDGS sources. These results indicated that the DE and ME values in corn DDGS are related to the chemical composition, primarily the EE and fiber concentrations. Specific prediction equations derived from full-oil and reduced-oil DDGS are better than equations derived from the entire set of DDGS.

We investigated dexamethasone therapy for preventing delayed encephalopathy after carbon monoxide (CO) poisoning. Eighty healthy male rats were exposed to CO and randomly divided into four groups: hyperbaric oxygen treatment (H), treatment (D), combined hyperbaric and dexamethasone treatment (C), and a control (M) group in which the rats inhaled CO to coma in the hyperbaric oxygen chamber, then were removed without further treatment. Twelve rats were put into the hyperbaric oxygen chamber and treated with air for 60 min (N) group. An eight arm maze was used to evaluate cognitive and memory abilities of these mice. Serum myelin basic protein (MBP) levels were evaluated using ELISA, and magnetic resonance imaging was used to observe brain demyelination and morbidity associated with delayed encephalopathy. A sample of the hippocampus from each group was examined by light microscopy. Cognitive and memory functions decreased in the control group M. Three days after CO poisoning, the serum MBP level of each group increased significantly. On Day 10 after CO poisoning, the MBP levels in groups C and D decreased significantly, but returned to normal on Day 18. MBP levels in the M and H groups were elevated at all time points. Brain MRIs showed significant differences among C, D, H and control M groups. Hematoxylin & eosin staining of the hippocampus showed greater damage in the control M and H groups. Early dexamethasone treatment may be useful for preventing delayed encephalopathy after CO poisoning and may reduce serum MBP levels.

Two experiments were conducted to evaluate the efficacy of feeding an Escherichia coli (E. coli) derived phytase to pigs fed P deficient, corn-soybean meal diets. In Exp. 1, one hundred and twenty crossbred piglets (9.53±0.84 kg) were allocated to one of five treatments which consisted of four low P diets (0.61% Ca, 0.46% total P and 0.24% non-phytate P) supplemented with 0, 500, 1,000, or 20,000 FTU/kg E. coli phytase as well as a positive control formulated to be adequate in all nutrients (0.77% Ca, 0.62% total P and 0.42% non-phytate P). The treatments were applied to six pens with four pigs per pen for 28 days. In Exp. 2, ten crossbred pigs (19.66±1.16 kg) fitted with ileal T-cannula were used in a nutrient balance study. The pigs were assigned to treatments similar to those used in Exp. 1 in a doubly replicated 5×4 incomplete Latin square design (5 diets with 4 periods). Each period consisted of a 5-d adjustment period followed by a 3-d total collection of feces and urine and then a 2-d collection of ileal digesta. Supplementation with phytase linearly increased (p<0.05) weight gain, feed intake, feed efficiency, bone breaking strength and fat-free dry and ash bone weight. There were linear increases (p<0.01) in the apparent ileal digestibility (AID) of DM, GE, CP, Ca, total P, inositol hexaphosphate (IP6) and some AA with increasing dose of E. coli phytase. Pigs fed 20,000 FTU/kg had a greater (p<0.05) AID of IP6 (80% vs 59% or 64%, respectively) than pigs fed diets with 500 or 1,000 FTU/kg phytase. There were linear increases (p<0.05) in the total tract digestibility of Ca, total P, Na, K, Mg, and Zn as well as in the retention of Mg and Zn with increased phytase dose. The retention and utilization of Cu, and the total tract digestibility of CP and Cu quadratic increased (p<0.05) with increased phytase dose. In conclusion, supplementation of 500 FTU of phytase/kg and above effectively hydrolyzed phytate in low-P corn-soybean diets for pigs. In addition, a super dose of phytase (20,000 FTU/kg) hydrolyzed most of the IP6 and consequently further improved mineral use, protein utilization and performance.

During a survey of potato scab pathogens in China from 2003 to 2012, a new pathogen was found in Shanxi and Neimenggu provinces. The incidence was approximately 20% of all recovered strains. The lesions caused by the pathogen were slightly raised and similar to those caused by Streptomyces scabies (3). Lesions were excised (approximately 10 mm3) from 40 infected tubers, surface-disinfested with 0.3% NaOCl for 30 s, rinsed in sterile water three times, cut into 5 mm3, then sliced into 1-mm pieces, and plated on water agar amended with ampicillin (50 µg/ml). Plates were incubated at 28°C in the dark for 4 days. The spores of Streptomyces sp. strains growing from the tuber pieces were collected from single bacterial colonies and cultured on oatmeal agar. To fulfill Koch's postulates, one strain, CPS-2, was grown at 28°C for 10 days and the spores were washed from the plates as inoculum. One hundred milliliters of inoculum (1 × 105 CFU/ml) was mixed with autoclaved soil and vermiculite (1:1) in each pot (15 cm in diameter). Cut tubers were planted in the pots (potato cv. Favorita, one plant per pot, five replicates) and grown under greenhouse conditions (22 ± 5°C). Typical common scab symptoms consisting of small, brown, raised lesions developed on potato tubers 12 weeks after planting. The same strain was re-isolated from the lesions of the new scabby tubers. Non-inoculated plants, treated as described above, but without strain CPS-2, remained healthy. The CPS-2 strain was identified based on morphological and physiological characterization and 16S rDNA sequence. On yeast-malt extract agar, the test strain produced grayish-white aerial hypha, reddish brown substrate mycelium and pigments, and loose spiral spore chains. Spores were smooth and were 0.8 to 0.9 × 1.1 to 1.2 µm in size (diameter and length). The ability of the strain to use single sources of carbon and nitrogen was verified according to the International Streptomyces project (4). The strain grew in media supplemented with L-arabinose, D-fructose, D-glucose, rhamnose, raffinose, meso-inositol, sucrose, and D-xylose, but not D-mannitol. It used L-hydroxyproline, L-methionine, and L-histidine, and produced melanin on tyrosine and peptone yeast extract agar. The strain did not grow at a pH less than 5.0 and was sensitive to streptomycin (20 µg/ml), phenol (0.1%), and crystal violet (0.5 µg/mL), but not to penicillin (10 IU/ml). The strain also produced hydrogen sulfide. The biological characteristics of strain CPS-2 were in accord with Streptomyces galilaeus. CPS-2 produced thaxtomin A in oatmeal liquid medium and the txt AB gene fragment was successfully amplified using specific primers (2). The 16S rDNA sequence of CPS-2 was amplified by PCR with primers 16S1-F: 5'-CATTCACGGAGAGTTTGATCC-3' and 16S1-R: 5'-AGAAAGGAGGTGATCCAGCC-3' (1) and sequenced. A BLAST search of the 16S rDNA sequence for CPS-2 was conducted using the NCBI GenBank database, resulting in 99.8% similarity to S. galilaeus (NR_040857). The 16S rDNA sequence for CPS-2 (1,388 bp) was deposited in GenBank (AY621378). To our knowledge, this is the first report of S. galilaeus causing common scab of potato in China. References: (1) R. A 42-d experiment was conducted with 276 Arbor Acres broilers (1 d of age, weighing 41.1 ± 1.5 g) to determine the effects of Forsythia suspensa extract (FSE) and berberine (BE) on growth performance, immunity, antioxidant activities, and intestinal microbiota stocked at high stocking density (28 kg of BW/m(2) as the normal density, 46 kg of BW/m(2) as the high density). The feeding program consisted of a starter diet from d 1 to 21 of age and a finisher diet from d 22 to 42. Dietary treatments included (1) negative control group (NC), stocked at high density; (2) positive control group (PC), stocked at normal density; (3) FSE, NC + 100 mg/kg of FSE; (4) BE, NC + 100 mg/kg of BE; and (5) FSE + BE, NC + 100 mg/kg of FSE + 100 mg/kg of BE. Birds had free access to diets and water. Body weight and feed intake were measured at d 21 and 42. Blood, spleen and bursa, and intestinal contents were collected at d 42 for analysis. The results showed that growth performance did not differ among treatments in the starter period; however, in the finisher and overall periods, birds in FSE + BE, FSE, BE, and PC had greater final BW, ADG, and ADFI than the birds in NC (P < 0.05). The birds in FSE + BE, FSE, BE, and PC had greater bursa weight, bursa weight/BW ratio, serum total antioxidant capacity, and superoxide dismutase activity than birds in NC (P < 0.05). The FSE + BE, FSE, and BE birds had lower serum malondialdehyde level than NC birds (P < 0.05). The FSE + BE, FSE, BE, and PC birds had lower counts of Escherichia coli but greater Lactobacillus in the cecum than that of NC birds (P < 0.05). Dietary supplementation with FSE, BE, or both can improve the growth performance possibly by enhancing immunity, reducing oxidative stress, and promoting intestinal colonization by healthy microbiota of broilers under high stocking density.

This work addresses the functionalization of chitosan thin films and its nanoliposomes blend films by a microwave-excited Ar/N2/H2 surface-wave plasma treatment which was found an effective tool to modify surface properties. Changes in the film properties (wettability, chemical composition, morphology) induced by the plasma treatment are studied using water contact angle measurements, X-ray photoelectron spectroscopy and scanning probe microscopy. The results suggest that hydrophilicity of the films is improved by plasma treatment in a plasma condition dependency manner. Water contact angle of chitosan films before and after plasma treatment are, respectively, 101° and 27°. Besides chemical changes on the surface, the nanoliposomes incorporation and plasma treatment also induce morphological modifications. Moreover, a correlation is found between the nanoliposomes composition and size, and the effects of plasma treatment. It is shown that the plasma treatment significantly improves the chitosan film functionalization. The effect of N2 content (88% and 100%) in the plasma gas mixture on the film etching is also pointed out.

Silymarin is used by many patients with chronic hepatitis B, but its efficacy remains unknown. The aim of this investigation was to conduct a meta-analysis to determine the efficacy and safety of silymarin and its combination therapy for the treatment of chronic hepatitis B. We searched Chinese and English reports from January 1966 to December 2011, using 12 databases. Two reviewers independently screened all reports to identify randomized placebo-controlled trials that evaluated silymarin and its combination therapy for the treatment of chronic hepatitis B. Twelve trials satisfied the eligibility criteria for this meta-analysis. Silymarin was equivalent to antiviral drug or protection liver drugs in serum transaminases, viral load and hepatic fibrosis markers. But silymarin combined with antiviral drug or antiviral drug and protection liver drugs significantly reduced the level of serum transaminases, hepatic fibrosis markers and serum TGF-ß1, TNF-α, IL-6 versus antiviral drug or protection liver drugs. Silymarin combined with protection liver drugs significantly reduced the level of serum transaminases, TBIL, hepatic fibrosis markers and was equivalent to protection liver drugs in the normalisation rates of serum transaminases, TBIL, but protection liver drugs significantly increased the improvement rates of hepatic fibrosis markers. Silymarin combined with antiviral drug or antiviral drug and protection liver drugs may have potential therapeutic value. Treatment with silymarin appears to be safe and well tolerated. The data are too limited to exclude a substantial benefit or harm of silymarin and its combination therapy on serum transaminases, and also to support recommending this herbal compound for the treatment of chronic hepatitis B.

Two experiments were conducted to investigate the potential for Forsythia suspensa extract (FSE) to substitute for antibiotic in broiler chicken. First, a well-diffusion assay procedure and a 2-fold dilution method were used to determine the bacteriostatic activity of FSE on Escherichia coli K88, staphylococcus aureus, and salmonella was assayed. An inhibitory effect of FSE was observed on the growth of these bacteria. This effect seems to be dose depended, which disappeared after 25.00, 12.50, 1.56 mg/ml. Second, a 42-d trial with 252 broiler chickens (d 1, 38.7±1.1 g BW) was conducted to evaluate the effect of dietary supplementation of FSE in broiler chicken. The feeding program consisted of a starter diet from d 1 to 21 and a finisher diet from d 22 to 42. Dietary treatments included were: i) NC: negative control fed a corn-soybean meal based diet; ii) PC: positive control group fed based diet with chlortetracycline; and iii) FC: a test group fed with 100 mg FSE/kg diet. In this study growth performance did not differ among treatments during the starter period. However, dietary supplemental chlortetracycline and FSE increased (p<0.05) average daily gain (ADG), average daily feed intake (ADFI) compared with NC during the finisher and overall phase. Apparent digestibility of calcium on d 21, digestibility of energy and calcium on d 42 of FC was greater (p<0.05) than NC. Moreover, cecal Escherichia coli counts for birds from FC were lower (p<0.05) than NC. Dietary FSE supplementation also improved (p<0.05) villus height and villus height to crypt depth ratios in both duodenum and ileum and decreased (p<0.05) crypt depth in the duodenum. Duodenum villus height and villus height to crypt depth ratio in both duodenum and ileum from the FC group were also greater (p<0.05). Serum growth hormone and IGF-1 were not influenced by different treatments. Apparently, FSE has the potential to substitute for antibiotic in broiler chicken.

This study was conducted to evaluate the effects of 4 Chinese herbal polysaccharides on the production of serum antibodies and the proliferation of peripheral T lymphocytes, including subpopulations in vaccinated chickens. A total of 450 chickens were randomly assigned to 9 groups at 14 d of age and vaccinated first with live Newcastle disease (ND)-infectious bronchitis virus vaccine, and second with ND-infectious bronchitis oil adjuvant vaccine at 28 d of age. At the same time as the first vaccination, the chickens in groups 1 to 8 were intramuscularly injected with 4 polysaccharides at high and low dosages, respectively, once a day for 3 successive days starting on the day of the first vaccination. Group 9 (control group) was injected in the same manner with saline instead of a polysaccharide. On d 7, 14, 21, 28, 35, 42, and 49 after the first vaccination, the temporal changes in serum ND hemagglutination inhibition antibody titer were determined by the micromethod. On d 10, 20, 30, 40, and 50 after the first vaccination, the proliferation of peripheral blood mononuclear cells in response to concanavalin A stimulation as well as the proportions of CD3(+), CD4(+), and CD8(+) peripheral blood mononuclear cells were determined by the 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide method and flow cytometry, respectively. The results showed that astragalus polysaccharide and isatis root polysaccharide at low dosages, and achyranthes root polysaccharide and Chinese yam polysaccharide at high dosages significantly enhanced the ND antibody titers, concanavalin A-induced proliferation of peripheral blood lymphocytes, and ratio of CD4(+) to CD8(+) (P <0.05). Collectively, these findings indicate that the 4 polysaccharides possess significant immune-enhancing properties in chickens. This finding may have direct application in vaccine design and other strategies designed to potentiate immune system development and function in chickens.

As the cost of drug development is always many times more than that of drug discovery, predictive methods aiding in the screening of bioavailable drug candidates are of profound significance. In this paper, a novel method for screening bioactive compounds from traditional Chinese medicines (TCMs) was developed by using living cell extract and gas chromatography (GC)-mass spectrometer (MS). The method was validated by using elemene emulsion injection (EEI), a typical TCM with known active compound, to interact with murine ascites hepatocarcinoma cell strain with high metastatic potential (HCa-F). Finally, the method was applied to screen the bioactive compounds from multi-component zedoary turmeric oil and glucose injection (ZTOGI). After HCa-F cells was incubated in ZTOGI, ethyl acetate (EtOAc) was used to extract the compounds in the cells for GC-MS analysis. Fourteen compounds were detected in the desorption eluate of HCa-F cell extract of ZTOGI, and further identified by MS. Curzerene and beta-elemene were found to be two major bioactive compounds in ZTOGI. These results show that the method developed may be applied to quickly screen the potential bioactive components in TCMs interacting with the target cells.

Huang Qi (root of Astragalus membranaceus) and Dang Gui ( Angelica sinensis), two of the most widely used herbs in traditional Chinese medicine, have been proven to be effective in the treatment of diabetes mellitus (DM) although the underlying molecular mechanisms are not fully elucidated. This study was designed to investigate the protective effect of Dang Gui and Huang Qi mixture (GQM) on the development of diabetic nephropathy in rats with streptozotocin (STZ)-induced DM and the possible underlying molecular mechanism. The diabetic animal model was made by a single intraperitoneal injection of STZ and then treated with GQM or benazepril. Blood glucose, triglyceride (TG), cholesterol (CHO), high density lipoprotein (HDL), serum creatinine (Scr), creatinine clearance rate (Ccr), blood urea nitrogen (BUN), urine beta (2)-microglobin (beta (2)-MG), kidney/body weight (K/B) ratio, glomerular area (GA), renal transforming growth factor-beta (1) (TGF-beta (1)) mRNA expression and blood and renal angiotensin II (AngII) expression were determined 8 weeks after the treatment. The blood glucose, CHO and TG levels, BUN, SCr, Ccr. K/B ratio, GA, the excretion of beta (2)-MG, renal TGF-beta (1) mRNA expression and blood and renal AngII expression were significantly increased while the HDL level was decreased 8 week after STZ injection. The changes in blood glucose, TG, CHO and HDL were reversed by GQM, not by benazepril, whereas the changes in other variables were reversed by both GQM and benazepril. Our results suggest that GQM alleviates the disorder in blood glucose and lipids, protects against the progression of renal nephropathy in diabetic rats, probably by inhibiting the expression of AngII and TGF-beta (1) mRNA.

BACKGROUND: Several clinical trials have suggested that a metabolic cocktail of glucose-insulin-potassium (GIK) decreases mortality rates in patients with acute myocardial infarction (AMI). It has also been reported that Fas-mediated apoptosis plays an important role in ischaemic/reperfusion injury in the rat model. This study was designed to evaluate the interaction of ischaemic/reperfusion and reperfusion therapy coadministered with high-dose GIK treatment on soluble Fas/APO-1 (sFas) and Fas ligand (sFasL) plasma concentration in patients with AMI. MATERIALS AND METHODS: Seventy-four patients presenting with AMI who underwent reperfusion therapy were randomized into a GIK group (n = 35) receiving high-dose GIK for 24 h or a vehicle group (n = 39). Thirty-four control subjects were also enrolled in the present study. Strepavidin-biotin ELISA was used to determine the soluble sFas and sFasL plasma concentration at baseline, 24 h (h), 3 day (d), 7 d and 14 d. RESULTS: Soluble Fas and sFas-L serum concentrations ([sFas] and [sFas-L]) of patients with AMI were significantly elevated at baseline as compared with normal controls (NCs; P < 0.01 vs. NC). The sFas in the GIK and vehicle groups markedly decreased 24 h after the GIK infusion (10.7-->5.9 ng mL(-1) and 9.7-->6.5 ng mL(-1); P < 0.01 vs. baseline) and then increased during the 3-7-d period (5.9-->12.1 ng mL(-1) and 6.5-->11.1 ng mL(-1); P < 0.01 vs. 24 h). The GIK group demonstrated reduced sFas (12.1-->5.9 ng mL(-1)) at 14 d (P < 0.01 vs. 7 d), with no concomitant changes in the vehicle group. The sFas-L in the GIK and vehicle groups was not significant different during the 14-d period. CONCLUSIONS: These results indicate that the sFas and sFasL in patients with AMI increased significantly compared with NC. Owing to the cardioprotective effects reported here and by others, a high-dose GIK infusion co-administered with the timely re-establishment of nutritive perfusion should be strongly considered as a treatment of choice for AMI. Additionally, sFas may be a valuable marker of the physiological response to ischaemic/reperfusion injury and reperfusion associated with high-dose GIK treatment.
