RESUMO

Iron deficiency anemia (IDA) is a common nutritional disease suffered by 1 billion people. To develop a new drug which avoids the side effects of traditional oral iron supplementation for IDA treatment, we have designed Fe3O4@ Astragalus polysaccharide core-shell nanoparticles (Fe3O4@APS NPs) and demonstrated them to be an efficient therapeutic drug for IDA treatment in vivo. The Fe3O4@APS NPs have been successfully synthesized with good water solubility and stability, especially in imitated digestion. Cytotoxicity assessment in cells and pathological tests in mice justify their good biocompatibility and low toxicity. The IDA treatment in rats shows that they have efficient therapeutic effect, which is contributed to both the iron element supplement from Fe3O4 and the APS-stimulated hematopoietic cell generation. Moreover, the superparamagnetic Fe3O4@APS NPs are capable for use as a magnetic resonance imaging contrast agent. This study presents the possibility of nanocomposites involving purified natural products from Chinese herb medicine for biomedical applications.

RESUMO

An incubation experiment was carried out to study the effect of montmorillonite on pH, the fractions and availability of phosphorus in a sandy yellow soil (SY) and a mineral flavicant soil (MF) applied with biogas residues of swine manure (BSR). The rates of montmorillonite applied were 0%, 5%, 7.5% and 10%, respectively. The results indicated that soil pH was not affected by BSR, but was significantly increased by montmorillonite. In both soils, application of BSR caused an increase in the content and proportion of Al-P during the incubation, and an increase in the content but a slight decrease in the proportion of Fe-P in later period of incubation. The concentration of O-P was not markedly changed, but its proportion was decreased by applying BSR in both soils. The changes of Ca-P varied with soils. BSR had no significant influence on Ca-P content, but decreased the proportion of Ca-P in SY, meanwhile, it increased both Ca-P content and proportion in MF. Addition of montmorillonite in soils applied with BSR decreased the contents and proportions of Al-P and O-P, and the proportion of Fe-P did not change the contents of Fe-P, but increased the content and proportion of Ca-P in SY. In MF, montmorillonite could decrease the contents and proportions of Fe-P and O-P, and the proportion of Al-P did not affect the content of Al-P, but increased the contents and proportions of Ca-P greatly. The contents of available phosphorus (A-P) were enhanced by 17.9%-38.0% and 17.1%-33.7% in SY and MF respectively, and the phosphorus activation coefficient (PVC) was significantly improved in both soils by applying BSR. The contents of A-P were reduced by 8.8%-35.5% and 1.1%-11.6% in SY and MF respectively. Correlation analysis showed that montmorillonite reduced the availability of phosphorus through increasing pH and contents of Ca-P, decreasing the content of Al-P in SY, and through increasing pH and decreasing content of Fe-P in MF. The availability of phosphorus in SY was influenced more obviously than that in MF by montmorillonite.

RESUMO

Pot experiments were carried out to investigate the influence of different selenium (Se) levels (0, 0.5 and 1.0 mg x L(-1)) on the plant growth, concentration of malonaldehyde (MDA), activities of antioxidant enzymes, accumulation and chemical forms of cadmium (Cd) in cucumber when exposed to Cd (20 mg x kg(-1)). The results showed that dry weights of leaf, stem, root, fruit and plant, and concentrations and accumulation of Cd significantly differed between two varieties of cucumber Yanbai and Jinyou 1. With increasing levels of Se, the contents of MDA in the leaves of Yanbai increased, but the contents of MDA in the leaves of Jinyou 1 decreased. The contents of MDA in the roots of Yanbai first increased and then decreased, while the contents of MDA in Jinyou 1 first decreased and then increased. The variation trends of CAT, SOD and POD in the leaves and roots of both varieties were different with increasing levels of Se. The concentrations of different chemical forms of Cd in the fruit decreased after spraying of Se, compared with the control. Cadmium concentrations in the leaves, stem, roots and fruit of both varieties decreased by 3.2%-17.9%, 14.6%-28.2%, 5.1%-18.5% and 60.6%-75.8% in the presence of Se when exposed to Cd. Accumulation of Cd in the plant of both varieties was in order of Jinyou 1 > Yanbai in the presence or absence of Na2SeO3.

RESUMO

There were two phases for the recognition and treatment of hypochondriac pain before the Ming Dynasty, and the Song Dynasty being their demarcation line. Before the Song Dynasty, hypochondriac pain was regarded generally as a concomitant symptom, generally attributed to pathogen of coldness, and the materia medica with the warm nature were always used for the treatment. This recognition was related with the traditional idea paying attention predominantly to the yang of the yin-yang theory. Since the Song Dynasty, especially the Jin-Yuan Period, the hypochondriac pain was regarded gradually as an independent symptom. Physicians got more and more recognition about its causes and pathogenesis. The treatment was changed from dispelling cold to activating qi, and the idea of treatment based on syndrome differentiation on hypochondriac pain was finally established, reflecting the influence of spirit of medical innovation during the Jin-Yuan Period on the recognition of disease.

RESUMO

The dopant local structure and optical properties of Cu-doped ZnSe (ZnSe:Cu) and Cu and Al co-doped ZnSe (ZnSe:Cu,Al) nanocrystals (NCs) were studied with an emphasis on understanding the impact of introducing Al as a co-dopant. Quantum-confined NCs with zinc blende crystal structure and particle size of 6 ± 0.6 Å were synthesized using a wet chemical route. The local structure of the Cu dopant, studied by extended X-ray absorption fine structure, indicated that Cu in ZnSe:Cu NCs occupies a site that is neither substitutional nor interstitial and is adjacent to a Se vacancy. Additionally, we estimated that approximately 25 ± 8% of Cu was located on the surface of the NC. Al(3+) co-doping aids in Cu doping by accounting for the charge imbalance originated by Cu(+) doping and consequently reduces surface Cu doping. The Cu ions remain distorted from the center of the tetrahedron to one of the triangular faces. The lifetime of the dopant-related photoluminescence was found to increase from 550 ± 60 to 700 ± 60 ns after Al co-doping. DFT calculations were used to obtain the density of states of a model system to help explain the optical properties and dynamics processes observed. This study demonstrates that co-doping using different cations with complementary oxidation states is an effective method to enhance optical properties of doped semiconductor NCs of interest for various photonics applications.

RESUMO

To further know the seasonal variation characteristics of phosphorus species in lake sediment, the contents of different phosphorus species in the surface sediment sampling from Changshou Lake were determined by using the SMT sequential extraction method in July and November, 2009, and March, 2010. The contents of total phosphorus (TP) basically show an order of the low-water period, high-water period, and normal-water period from high to low, and the maximum appeared in Shoudao (2960.29 mg/kg) while the minimum in Guanjiakou (586.05 mg/kg). The mass fractions of phosphorus associated with hydrous ferric/aluminum oxides (Fe/Al-P) and organic phosphorus (OP) in TP at all sampling sits are in an order of the normal-water period, high-water period, and low-water period from high to low, and the maximum of Fe/Al-P appeared in Lewen (40.01%) while that of OP in the dam mouth (72.44%). The mass fraction of phosphorus bound to calcium salt (Ca-P) in TP is in an order of the low-water period, normal-water period, and high-water period, and the maximum appeared in Baibudang (15.52%). These results suggested that the phosphorus speciation in the sediment of Changshou Lake shows obviously temporal and spatial distribution characteristics, and Fe/Al-P is mainly influenced by pH of the overlying water, while OP is not apparently influenced by dissolved oxygen (DO).

RESUMO

PURPOSE: To determine the anti-inflammatory and anti-oxidant effects of epigallocatechin gallate (EGCG), the major polyphenol component of green tea, in human corneal epithelial cells (HCEpiC). METHODS: HCEpiC were challenged with interleukin-1ß (IL-1ß) for 18 h or hyperosmolarity (440 mOsm) for 24 h. Luminex technology was used to determine the effects of EGCG (0.3-30 µM) on IL-1ß- or hyperosmolar-induced cytokine release into the medium. Cell metabolic activity was measured using the alamarBlue assay. Effects of EGCG on mitogen-activated protein kinase (MAPK) phosphorylation were determined by cell-based enzyme-linked immunosorbent assay (ELISA) and western blotting. Effects of EGCG on nuclear factor kappa B (NFκB) and activator protein-1 (AP-1) transcriptional activity were assessed by reporter gene assay. The effects of EGCG on glucose oxidase (GO)-induced reactive oxygen species (ROS) production was determined using the ROS probe CM-H2DCFDA. RESULTS: Treatment of HCEpiC with 1 ng/ml IL-1ß for 18 h significantly increased release of the cytokines/chemokines granulocyte colony-stimulating factor (G-CSF), granulocyte-macrophage colony-stimulating factor (GM-CSF), interleukin-6 (IL-6), interleukin-8 (IL-8), and monocyte chemotactic protein-1 (MCP-1), while hyperosmolarity-induced release of IL-6 and MCP-1. When cells were treated with IL-1ß and EGCG or hyperosmolarity and EGCG there was a dose-dependent reduction in release of these cytokines/chemokines, with significant inhibition observed at 3-30 µM. There was no effect of EGCG on cell metabolic activity at any of the doses tested (0.3-30 µM). EGCG significantly inhibited phosphorylation of the MAPKs p38 and c-Jun N-terminal kinase (JNK), and NFκB and AP-1 transcriptional activities. There was a significant dose-dependent decrease in GO-induced ROS levels after treatment of HCEpiC with EGCG. CONCLUSIONS: EGCG acts as an anti-inflammatory and anti-oxidant agent in HCEpiC and therefore may have therapeutic potential for ocular inflammatory conditions such as dry eye.

RESUMO

Besifloxacin is a new fluoroquinolone anti-infective developed for ophthalmic use. Besifloxacin ophthalmic suspension 0.6% (Besivance) was recently approved for the treatment of bacterial conjunctivitis. The objective of this article is to provide a comprehensive overview of microbiological, pharmacokinetic/pharmacodynamic and clinical studies with besifloxacin. Microbiological studies have demonstrated that besifloxacin has wide-spectrum and potent activity against common ocular pathogens, including Gram-negative and Gram-positive pathogens associated with bacterial conjunctivitis, and retained activity against fluoroquinolone-resistant staphylococci and multidrug-resistant strains. In preclinical and human studies, topically applied besifloxacin had a prolonged ocular concentration and minimal systemic exposure. In clinical studies, patients randomized to besifloxacin ophthalmic suspension 0.6% experienced significantly higher rates of clinical resolution and microbial eradication than patients randomized to vehicle. Besifloxacin ophthalmic suspension 0.6% was also found to be as effective and well tolerated as moxifloxacin ophthalmic solution 0.5%. The low minimum inhibitory concentrations and high attainment of pharmacodynamic targets with besifloxacin may contribute to a lower risk for the emergence of bacterial resistance, although further studies are needed. These data indicate that besifloxacin ophthalmic suspension 0.6% is an important new option for the treatment of bacterial conjunctivitis.
