RESUMO

Scyphozoan jellyfish, known for their evolutionary position and ecological significance, are thought to exhibit relatively notable resilience to ocean acidification. However, knowledge regarding the molecular mechanisms underlying the scyphozoan jellyfish response to acidified seawater conditions is currently lacking. In this study, two independent experiments were conducted to determine the physiological and molecular responses of moon jellyfish (Aurelia coerulea) polyps to within- and trans-generational exposure to two reduced pH treatments (pH 7.8 and pH 7.6). The results revealed that the asexual reproduction of A. coerulea polyps significantly declined under acute exposure to pH 7.6 compared with that of polyps at ambient pH conditions. Transcriptomics revealed a notable upregulation of genes involved in immunity and cytoskeleton components. In contrast, genes associated with metabolism were downregulated in response to reduced pH treatments after 6 weeks of within-generational acidified conditions. However, reduced pH treatments had no significant influence on the asexual reproduction of A. coerulea polyps after exposure to acidified conditions over a total of five generations, suggesting that A. coerulea polyps may acclimate to low pH levels. Transcriptomics revealed distinct gene expression profiles between within- and trans-generational exposure groups to two reduced pH treatments. The offspring polyps of A. coerulea subjected to trans-generational acidified conditions exhibited both upregulated and downregulated expression of genes associated with metabolism. These physiological and transcriptomic characteristics of A. coerulea polyps in response to elevated CO2 levels suggest that polyps produced asexually under acidified conditions may be resilient to such conditions in the future.

BACKGROUND: The mass occurrence of scyphozoan jellyfish severely affects marine ecosystems and coastal economies, and the study of blooming jellyfish population dynamics has emerged in response. However, traditional ecological survey methods required for such research have difficulties in detecting cryptic life stages and surveying population dynamics owing to high spatiotemporal variations in their occurrence. The environmental DNA (eDNA) technique is an effective tool for overcoming these limitations. RESULTS: In this study, we investigated the biodiversity and spatial distribution characteristics of blooming jellyfish in the Bohai Sea of China using an eDNA metabarcoding approach, which covered the surface, middle, and bottom seawater layers, and sediments. Six jellyfish taxa were identified, of which Aurelia coerulea, Nemopilema nomurai, and Cyanea nozakii were the most dominant. These three blooming jellyfish presented a marked vertical distribution pattern in the offshore regions. A. coerulea was mainly distributed in the surface layer, whereas C. nozakii and N. nomurai showed a upper-middle and middle-bottom aggregation, respectively. Horizontally, A. coerulea and C. nozakii were more abundant in the inshore regions, whereas N. nomurai was mainly distributed offshore. Spearman's correlation analysis revealed a strong correlation between the eDNA of the three dominant blooming jellyfish species and temperature, salinity, and nutrients. CONCLUSIONS: Our study confirms the applicability of the eDNA approach to both biodiverstiy evaluation of blooming jellyfish and investigating their spatial distribution, and it can be used as a supplementary tool to traditional survey methods.

Understanding the mechanisms underlying diapause formation is crucial for gaining insight into adaptive survival strategies across various species. In this study, we aimed to uncover the pivotal role of temperature and food availability in regulating diapausing podocyst formation in the jellyfish Aurelia coerulea. Furthermore, we explored the cellular and molecular basis of diapause formation using single-cell RNA sequencing. Our results showed cell-type-specific transcriptional landscapes during podocyst formation, which were underscored by the activation of specific transcription factors and signalling pathways. In addition, we found that the heat shock protein-coding genes HSC70 and HSP90a potentially act as hub genes that regulate podocyst formation. Finally, we mapped the single-cell atlas of diapausing podocysts and identified cell types involved in metabolism, environmental sensing, defence and development that may collectively contribute to the long-term survival and regulated excystment of diapausing podocysts. Taken together, the findings of this study provide novel insights into the molecular mechanisms that regulate diapause formation and contributes to a better understanding of adaptive survival strategies in a variety of ecological contexts.

Purpose: This study aimed to investigate the lower limb inter-joint coordination and variability during Tai Chi movements compared with normal walking in older adults. Methods: A total of 30 female Tai Chi practitioners (70.9 ± 5.2 years) were recruited in this study. Herein, each participant performed three trials of the normal walking and Tai Chi movements. The lower limb kinematics data were collected with Vicon 3D motion capture system. The continuous relative phase (CRP) includes both spatial and temporal information of two adjacent joints, which was calculated to assess the inter-joint coordination of lower limbs. Coordination amplitude and coordination variability were assessed with mean absolute relative phase (MARP) and deviation phase (DP). MANOVOA was used to analyze inter-joint coordination parameters between different movements. Results: The CRP values of hip-knee and knee-ankle segments in the sagittal plane of the Tai Chi movements changed frequently. The MARP values of the hip-knee (p < 0.001) and knee-ankle segments (p = 0.032) as well as the DP values of the hip-knee segment (p < 0.001) were significantly lower in Tai Chi than in normal walking. Conclusion: More consistent and stable inter-joint coordination patterns of Tai Chi movements found in this study may be one of the critical factors that Tai Chi could be a suitable coordinated exercise for older adults.

PURPOSE: Postmenopausal osteoporosis (PMO) is usually managed by conventional drug treatment. However, prolonged use of these drugs cause side effects. Gut microbiota may be a potential target for treatment of PMO. This work was a three-month intervention trial aiming to evaluate the added effect of probiotics as adjunctive treatment for PMO. METHODS: Forty patients with PMO were randomized into probiotic (n = 20; received Bifidobacterium animalis subsp. lactis Probio-M8 [Probio-M8], calcium, calcitriol) and placebo (n = 20; received placebo material, calcium, calcitriol) groups. The bone mineral density of patients was measured at month 0 (0 M; baseline) and month 3 (3 M; after three-month intervention). Blood and fecal samples were collected 0 M and 3 M. Only 15 and 12 patients from Probio-M8 and placebo groups, respectively, provided complete fecal samples for gut microbiota analysis. RESULTS: No significant change was observed in the bone mineral density of patients at 3 M. Co-administering Probio-M8 improved the bone metabolism, reflected by an increased vitamin D3 level and decreased PTH and procalcitonin levels in serum at 3 M. Fecal metagenomic analysis revealed modest changes in the gut microbiome in both groups at 3 M. Interestingly, Probio-M8 co-administration affected the gut microbial interactive correlation network, particularly the short-chain fatty acid-producing bacteria. Probio-M8 co-administration significantly increased genes encoding some carbohydrate metabolism pathways (including ABC transporters, the phosphotransferase system, and fructose and mannose metabolism) and a choline-phosphate cytidylyltransferase. CONCLUSIONS: Co-administering Probio-M8 with conventional drugs/supplements was more efficacious than conventional drugs/supplements alone in managing PMO. Our study shed insights into the beneficial mechanism of probiotic adjunctive treatment. To determine the ecological effects of atmospheric wet deposition of dissolved nutrients on the coastal waters around the Yangma Island, rain and snow samples were collected and analyzed at a shore-based site for one year. The wet deposition fluxes of dissolved inorganic nitrogen and phosphorus (DIN and DIP) and dissolved organic nitrogen and phosphorus were 69.2, 0.136, 13.3 and 0.143 mmol m-2 a-1, respectively. In summer, the new production fueled by wet deposition accounted for 19.3 % of that in seawater and 16.4 % of the amount of particulate organic carbon ingested by the scallops cultivated in the study area, indicating the potential contribution of wet deposition to fishery resources. Meanwhile, precipitation increased the seasonal average DIN/DIP ratios in surface seawater by 17.7 %, 16.3 %, 23.4 % and 6.5 % in spring, summer, autumn and winter, respectively, which could change the composition of ecological community and cause obvious negative impact on the ecosystem and mariculture.

OBJECTIVE: The objective of this study was to compare the effectiveness and safety of local anesthesia (LA) and epidural anesthesia (EA) for percutaneous transforaminal endoscopic discectomy (PTED) and provide reference data for clinical decision-making. METHODS: We searched PubMed, EMBASE, the Cochrane library, Web of Science, Medline, Science Direct, and China National Knowledge Infrastructure from inception to March 2022 to identify randomized and nonrandomized controlled trials comparing LA and EA for PTED. Studies that assessed at least 2 of the following indicators were considered eligible: surgical duration, X-ray exposure time, satisfaction rate, visual analog scale scores for pain, Oswestry Disability Index, and complications. Meta-analysis was conducted using Review Manager 5.3.3 software. RESULTS: Five randomized controlled trials and 5 retrospective cohort studies involving a total of 1660 patients were included. The LA and EA groups included 803 and 857 patients, respectively. Meta-analysis revealed significant intergroup differences in the intraoperative lumbar visual analog scale scores (P < 0.00001) and anesthesia satisfaction rate (P < 0.00001). There were no significant intergroup differences in the surgical duration, X-ray exposure time, postoperative Oswestry Disability Index, and complication rate. CONCLUSIONS: EA is as safe as LA and produces better anesthetic effects than LA in patients undergoing PTED. Therefore, EA should be promoted as a reliable anesthetic technique for PTED.

Analysis of the common and most influential natural and anthropogenic activities on the spatiotemporal variation in nutrients at a multiannual scale is important. Eleven cruises from 2015 to 2017 were carried out to better elucidate the seasonal and spatial variations in nutrients, as well as the impact factors on dissolved inorganic nitrogen (DIN), phosphorus (DIP) and silicate (DSi). Both nutrient concentrations and forms showed similar and significant seasonal variations over the 3 years, and were closely related to the biomass and species of phytoplankton. Terrestrial inputs had significant effects on the spatial distribution of nutrients throughout the year, especially in the surface water, which showed DIN > DIP>DSi. In summer, shellfish aquaculture and hypoxia jointly affected the spatial distribution of nutrients. The bottom water nutrient concentrations in the aquaculture area were 1.1-2.3 times higher than those outside of the aquaculture area. Seasonal hypoxia can increase the release of DSi and NH4+ from the sediment to the water. In summary, anthropogenic activities and physical conditions jointly influenced the nutrient distributions.

The monthly magnitudes of dissolvable nutrients through atmospheric dry deposition (ADD) and their ecological effects to the coastal waters around the Yangma Island, North Yellow Sea, were investigated for one year. The results indicated that anthropogenic activities were the major sources of dissolvable inorganic and organic nitrogen (DIN and DON); dust events were the major sources of inorganic phosphorus (DIP) and silicate (DSi); however, organic phosphorus (DOP) could be originated from marine biological activities. The annual ADD fluxes of DIN, DON, DIP, DOP and DSi were 21.8, 2.7, 0.10, 0.30 and 0.73 mmol m-2 yr-1, respectively. Overall, the new production supported by the bioavailable nitrogen through ADD in winter was up to 9.14 mg C m-2 d-1. Notably, the annual molar ratio of DIN/DIP through ADD was 216 ± 123, which was much higher than that of the dissolved inorganic nitrogen to phosphorus in seawater and might exacerbate their unbalance in some coastal waters.

In situ field investigations coupled with laboratory incubations were employed to explore the surface sedimentary phosphorus (P) cycle in a mariculture area adjacent to the Yangma Island suffering from summer hypoxia in the North Yellow Sea. Five forms of P were fractionated, namely exchangeable P (Ex-P), iron-bound P (FeP), authigenic apatite (CaP), detrital P (De-P) and organic P (OP). Total P (TP) varied from 13.42 to 23.88 µmol g-1 with the main form of inorganic P (IP). The benthic phosphate (DIP) fluxes were calculated based on incubation experiments. The results show that the sediment was an important source of P in summer with ~39% of the bioavailable P (BioP) recycled back into the water column. However, the sediment acted a sink of P in autumn. The benthic DIP fluxes were mainly controlled by the remobilizing of FeP, Ex-P and OP under contrasting redox conditions. In August (hypoxia season), ~0.92 µmol g-1 of FeP and ~0.52 µmol g-1 of OP could be transformed to DIP and released into water, while ~0.36 µmol g-1 of DIP was adsorbed to clay minerals. In November (non-hypoxia season), however, ~0.54 µmol g-1 of OP was converted into DIP, while ~0.55 µmol g-1 and ~0.28 µmol g-1 of DIP was adsorbed to clay minerals and bind to iron oxides. Furthermore, scallop farming activities also affected the P mobilization through biological deposition and reduced hydrodynamic conditions. The burial fluxes of P varied from 11.67 to 20.78 µmol cm-2 yr-1 and its burial efficiency was 84.7-100%, which was consistent with that in most of the marginal seas worldwide. This study reveals that hypoxia and scallop farming activities can significantly promote sedimentary P mobility, thereby causing high benthic DIP flux in coastal waters.

Fourteen field cruises were carried out in a mariculture region of the northern Shandong Peninsula, North Yellow Sea, China from 2016 to 2017 for a better understanding of the biogeochemical behaviors, sources and export of dissolved inorganic nutrients. The spatial variations of nutrients were not obvious due to the influence of complex hydrological and biochemical conditions. Potential nutritional level was characterized in oligotrophy, and trophic status was rated at medium level. A preliminary estimation of nutrient budgets demonstrated that the dissolved inorganic nitrogen (DIN) load was mainly from atmospheric deposition and scallop excretion, accounting for 56.9% and 35.6% of its total influx. Scallop excretion and sediment release were the major source of phosphate (DIP), contributing to 25.2% and 44.3%, while dissolved silicon (DSi) was mainly from sediment release, accounting for 94.2%. In addition, about 136.7 × 103, 7.3 × 103 and 485.5 × 103 mol km-2 yr-1 of DIN, DIP and DSi could be converted into other forms, e.g. organic and particulate matter and gas species.

17ß-Hydroxysteroid dehydrogenases (17ß-HSDs) are multifunctional enzymes involved in the metabolism of steroids, fatty acids, retinoids and bile acid. In this study, two novel types of 17ß-HSDs (named as MgHsd17b10 and MgHsd17b12) were cloned from Mytilus galloprovincialis by using rapid amplification of cDNA ends (RACE) approaches. Sequence analysis showed that MgHsd17b10 and MgHsd17b12 encoded a polypeptide of 259 and 325 amino acids, respectively. Phylogenetic analysis revealed that MgHsd17b10 and MgHsd17b12 were evolutionarily clustered with other invertebrate 17ß-HSD type 10 and 17ß-HSD type 12 homologues. The MgHsd17b10 and MgHsd17b12 transcripts could be detected in all examined tissues with higher expression levels in digestive glands and gonad. After exposed to endocrine disrupting chemicals (Bisphenol A or 2,2',4,4'-tetrabromodiphenyl ether), the expression of MgHsd17b10 and MgHsd17b12 transcripts was both down-regulated in digestive glands. These findings suggest that MgHsd17b10 and MgHsd17b12 perhaps play an important role in the endocrine regulation of M. galloprovincialis.

The full-length cDNA sequences coding respectively for a manganese superoxide dismutase (Mg-MnSOD) and copper/zinc superoxide dismutase (Mg-CuZnSOD) were cloned from Mytilus galloprovincialis. Mg-MnSOD and Mg-CuZnSOD cDNAs encoded a polypeptide of 228 and 211 amino acids, respectively. Sequence analysis indicated Mg-MnSOD was a mitochondrial MnSOD and Mg-CuZnSOD was an intracellular CuZnSOD. Multiple alignment analysis showed that both Mg-MnSOD and Mg-CuZnSOD sequences had the common features conserved in MnSODs and CuZnSODs, respectively. Phylogenetic analysis revealed that Mg-MnSOD clustered together with MnSODs from other mollusks, whereas Mg-CuZnSOD clustered with other mollusk intracellular CuZnSODs with a wider phylogenetic distance. By quantitative real-time RT-PCR (qPCR) analysis, both Mg-MnSOD and Mg-CuZnSOD transcripts were detected in all tissues examined with the highest expression level in hepatopancreas. Following bacterial challenge, the expression level of Mg-MnSOD and Mg-CuZnSOD increased first and subsequently decreased to the original level in hemocytes. In hepatopancreas, Mg-CuZnSOD mRNA was up-regulated significantly at 72 h and 96 h post challenge, while the level of Mg-MnSOD transcript had no significant change. Therefore, Mg-MnSOD and Mg-CuZnSOD expressions were inducible and they were probably involved in the immune response against bacterial challenge. These results suggest that these SODs may play important roles in the immune defense system of M. galloprovincialis and perhaps contribute to the protective effects against oxidative stress in this mussel.

C-type lectins are a family of calcium-dependent carbohydrate-binding proteins. In the present study, a C-type lectin (designated as AiCTL5) was identified and characterized from Argopecten irradians. The full-length cDNA of AiCTL5 was of 673 bp, containing a 5' untranslated region (UTR) of 24 bp, a 3' UTR of 130 bp with a poly (A) tail, and an open reading frame (ORF) of 519 bp encoding a polypeptide of 172 amino acids with a putative signal peptide of 17 amino acids. A C-type lectin-like domain (CRD) containing 6 conserved cysteines and a putative glycosylation sites were identified in the deduced amino acid sequence of AiCTL5. AiCTL5 shared 11%-27.5% identity with the previous reported C-type lectin from A. irradians. The cDNA fragment encoding the mature peptide of AiCTL5 was recombined into pET-21a (+) with a C-terminal hexa-histidine tag fused in-frame, and expressed in Escherichia coli Origami (DE3). The recombinant AiCTL5 (rAiCTL5) agglutinated Gram-negative E. coli TOP10F' and Listonella anguillarum, but did not agglutinate Gram-positive bacteria Bacillus thuringiensis and Micrococcus luteus, and the agglutination could be inhibited by EDTA, indicating that AiCTL5 was a Ca(2+)-dependent lectin. rAiCTL5 exhibited a significantly strong activity to bind LPS from E. coli, which conformed to the agglutinating activity toward Gram-negative bacteria. Moreover, rAiCTL5 also agglutinated rabbit erythrocytes. These results indicated that AiCTL5 could function as a pattern recognition receptor to protect bay scallop from Gram-negative bacterial infection, and also provide evidence to understand the structural and functional diverse of lectin.

Glutathione peroxidases (GPx) are key enzymes in the antioxidant systems of living organisms by catalyzing the reduction of peroxides to non-reactive products. In the present study, the full-length cDNA encoding a selenium-dependent GPx was identified from Venerupis philippinarum (designated as VpSe-GPx), and the spatial and temporal expression patterns post-Vibrio anguillarum, heavy metals and benzo[a]pyrene (B[a]P) challenge were also investigated. VpSe-GPx possessed all the conserved features critical for the fundamental structure and function of glutathione peroxidase. The VpSe-GPx mRNA was found to be most abundantly expressed in hepatopancreas. Vibrio challenge could significantly up-regulate the mRNA expression of VpSe-GPx, and the highest expression level was detected at 24 h post-infection with 6.5-fold increase compared with that in the control group. For heavy metals exposure, the expression of VpSe-GPx was significantly induced by 20, 40 µg L(-1) Cd and 10, 20 µg L(-1) Cu but depressed by 10 µg L(-1) Cd and 40 µg L(-1) Cu. With regards to B[a]P exposure, the expression of VpSe-GPx mRNA was significantly induced at 48 and 96 h post challenge. All these results suggested that VpSe-GPx was potentially involved in mediating the immune response and antioxidant defense in V. Philippinarum.

Thioredoxin (abbreviated as Trx) is an important ubiquitous disulfide reductase, which can protect organisms against various oxidative stresses. In the present study, thioredoxin 1 (named as VpTrx1) and thioredoxin-related protein (named as VpTrp14) were identified from Venerupis philippinarum, respectively. Similar to most Trx1s, VpTrx1 possessed all conserved features critical for the fundamental structure and function of Trx1s, such as the conserved catalytic residues (C-G-P-C), but lacked the other cysteine residues, while VpTrp14 contained the conserved motif (C-P-D-C). Quantitative Real-time PCR assay showed that VpTrx1 and VpTrp14 transcripts were distributed in a wide array of tissues most abundantly expressed in the hepatopancreas. The expression of VpTrp14 mRNA in the hepatopancreas was significantly up-regulated after exposure to 10 and 40µg/L Cd, while the VpTrx1 expression level was kept relatively constant. Both the expression levels of VpTrx1 and VpTrp14 in the hepatopancreas were induced after exposure to Cu, and increased to the peak value at 96h under the 40µg/L Cu exposure. These results showed that VpTrp14 transcripts responded to metal stress more acutely than VpTrx1, and both Trxs responded to Cu stress more sensitively than Cd. Together, it was suggested that VpTrx1 and VpTrp14 perhaps played important roles in the antioxidant responses against metal stress in V. philippinarum.

The glutathione peroxidases are essential enzymes of the cellular antioxidant defence system. In the present study, the full-length cDNA sequence encoding an extracellular glutathione peroxidase (designated CfGPx3) was isolated from Zhikong scallop Chlamys farreri. The complete cDNA was of 1194 bp, containing a 5' untranslated region (UTR) of 50 bp, a 3' UTR of 490 bp and an open reading frame (ORF) of 654 bp encoding a polypeptide of 217 amino acids. CfGPx3 possessed all the conserved features critical for the fundamental structure and function of glutathione peroxidase, such as the selenocysteine encoded by stop codon UGA, the GPx signature motif (96LGVPCNQF103) and the active site motif (179WNFEKF184). The high similarity of CfGPx3 with GPx from other organisms indicated that CfGPx3 should be a new member of the glutathione peroxidase family. By fluorescent quantitative real-time PCR, the CfGPx3 mRNA was universally detected in the tissues of haemocytes, gill, gonad, muscle and hepatopancreas with the highest expression in hepatopancreas. After scallops were challenged by Listonella anguillarum, the expression level of CfGPx3 transcript in haemocytes was significantly up-regulated (P<0.05) at 8h post challenge. These results suggested that CfGPx3 was potentially involved in the immune response of scallops and perhaps contributed to the protective effects against oxidative stress.

OBJECTIVE: To detect serum selenium level of people in non-selenium-deficiency region. METHODS: A total of 793 subjects were recruited by random sampling from people engaged in service sectors in Taign county, Shanxi province. Information about gender, age, occupation and educational level were collected and the content of serum selenium was measured by fluorospectrophotometry. All data were statistically analyzed with SPSS software version 13.0. RESULTS: The age distribution for these samples ranged from 20 to 67 years old. Their average serum selenium level and standard deviation (SD) were (0.0913 +/- 0.0186) mcirog/ml, (0.0947 +/- 0.0184) microg/ml and (0.0887 +/- 0.0184) microg/ml for total, male and female, respectively, and the level of male was significantly higher than that of female. There were no significant differences of serum selenium in each group of different ages, occupation, and educational level. Serum selenium in male were significantly higher than in female in both 20-29 years old group and 30-39 years old group, but there were no significant differences in groups older than 40. In 30-39 years old group, male had the highest serum selenium, but female occurred just the opposite. There was a decrease of serum selenium in male older than 40, but a great increase in female. CONCLUSION: The serum selenium level of adults in this area was adequate, but compared with progressed areas in the world, it could be categorized as low-middle level, serum selenium in male of 20-40 years old was significantly higher than in female, but the differences turn to be narrow in people older than 40 years old.

C-type lectins are a superfamily of carbohydrate-recognition proteins which play crucial roles in the innate immunity. In this study, the gene of a C-type lectin with multiple carbohydrate-recognition domains (CRDs) from scallop Chlamys farreri (designated as Cflec-3) was cloned by rapid amplification of cDNA ends (RACE) approach based on expression sequence tag (EST) analysis. The full-length cDNA of Cflec-3 was of 2256 bp. The open reading frame encoded a polypeptide of 516 amino acids, including a signal sequence and three CRDs. The deduced amino acid sequence of Cflec-3 showed high similarity to members of C-type lectin superfamily. By fluorescent quantitative real-time PCR, the Cflec-3 mRNA was mainly detected in hepatopancreas, adductor, mantle, and marginally in gill, gonad and hemocytes of healthy scallops. After scallops were challenged by Listonella anguillarum, the mRNA level of Cflec-3 in hemocytes was up-regulated and was significantly higher than that of blank at 8 h and 12 h post-challenge. The function of Cflec-3 was investigated by recombination and expression of the cDNA fragment encoding its mature peptide in Escherichia coli BL21 (DE3)-pLysS. The recombined Cflec-3 (rCflec-3) agglutinated Gram-negative bacteria Pseudomonas stutzeri. The agglutinating activity was calcium-dependent and could be inhibited by D-mannose. These results collectively suggested that Cflec-3 was involved in the immune response against microbe infection and contributed to nonself-recognition and clearance of bacterial pathogens in scallop.

Prophenoloxidase (proPO) is a conserved copper-containing enzyme that plays important roles in immune response of crustaceans and insects. In the present study, the full-length cDNA of a prophenoloxidase (designated EsproPO) was cloned from haemocytes of Chinese mitten crab Eriocheir sinensis by expressed sequence tag (EST) and PCR techniques. The isolated 3549bp full-length cDNA of EsproPO contained a 2040bp open reading frame (ORF) encoding a putative proPO protein of 679 amino acids, a 5'-untranslated region (UTR) of 68bp, and a long 3'-UTR of 1441bp. Two putative copper-binding sites, a proteolytic activation site, and a complement-like motif (GCGWPQHM) were identified in the deduced amino acid sequence of EsproPO. Homology analysis revealed that EsproPO was highly similar to other proPOs from crustaceans with identities from 52% to 68%. The conserved domains and motifs, and higher similarity with other proPOs suggested that EsproPO was a member of the proPO family. The mRNA expression of EsproPO and PO specific activities in the tissues of hepatopancreas, gill, gonad, muscle, heart, eye and haemocytes were measured by quantitative real-time PCR and colorimetric assay, respectively. The mRNA transcripts of EsproPO and PO specific activities could be detected in all the examined tissues with the highest level both in hepatopancreas. Three peaks of EsproPO mRNA expression were recorded at 2h, 12h and 48h in haemocytes of Chinese mitten crab post Vibrio anguillarum challenge, which was consistent with the temporal profile of PO specific activity. The mRNA expression pattern and the activity fluctuation of EsproPO post V. anguillarum stimulation indicated that it was potentially involved in the acute response against invading bacteria in Chinese mitten crab.
