RESUMO

This study aimed to explore the mechanism of Tanreqing Injection in the treatment of acute lung injury(ALI) based on network pharmacology and molecular docking. The active components and action targets of Tanreqing Injection were retrieved from the Traditional Chinese Medicine Systems Pharmacology Database and Analysis Platform(TCMSP), PubChem, and SwissTargetPrediction databases, as well as available literature reports. The ALI-related targets were obtained from the GeneCards database and then mapped with Tanreqing Injection targets. Following the construction of "drug-component-potential target" network with Cytoscape 3.6.1, the potential targets were input into STRING to yield the protein-protein interaction(PPI) network, which was plotted using Cytoscape 3.6.1. Then the screened key targets were subjected to gene ontology(GO) and Kyoto encyclopedia of genes and genomes(KEGG) enrichment analysis based on DAVID database. The top three key targets RAC-alpha serine/threonine-protein kinase(AKT1), albumin(ALB) and interleukin-6(IL6) were docked to the top three key compounds by PyMOL and AutoDock vina. A total of 58 active components of Tanreqing Injection, 597 corresponding targets and 503 common targets shared by Tanreqing Injection and ALI were fi-gured out, with the key targets AKT1, ALB and IL6 involved. GO and KEGG enrichment analysis yielded 1 445 biological processes and 148 signaling pathways, respectively. Molecular docking verified a good binding ability of the top three key targets to the top three key compounds. The analysis based on network pharmacology and molecular docking uncovered that Tanreqing Injection directly or indirectly regulated the pulmonary capillary endothelial cells and alveolar epithelial cells via anti-inflammation, thus alleviating ALI.

In this study, the influence of vegetation type and environmental temperature on performance of constructed wetlands (CWs) was investigated. Results of vegetation types indicated that the removal of most nutrients in polyculture was greater than those in monoculture and unplanted control. The greatest removal percentages of NH4+-N, total nitrogen (TN) and total phosphorus (TP) in polyculture were 98.7%, 98.5%, and 92.6%, respectively. In experiments of different temperatures, the removal percentages of NH4+-N, NO3--N, TN and TP in all CWs tended to decrease with the decline of temperature. Especially, a sharp decline in the removal percentages of NO3--N (decreased by above 13.8%) and TN (decreased by above 7.9%) of all CWs was observed at low temperature (average temperature of 8.9 °C). Overall, the performance of CWs was obviously influenced by temperature, and the polyculture still showed best performance in the removal of nitrogen when the average temperature dropped to 19.8 °C. Additionally, the variations of urease activities in rhizosphere soil tended to decrease with the decreasing temperature. Overall, a substantial enhancement for nitrogen and TP removal in polyculture (Canna indica + Lythrum salicaria) was observed. In conclusion, CW cultivated with polyculture was a good strategy for enhancing nutrient removal when temperature was above 19.8 °C.

For long-term operation of highly loaded biotrickling filters (BTFs), the prevention of excess biomass accumulation was essential for avoiding BTF failure. In this study, we proposed low-dose ozonation as a biomass control strategy to maintain high removal efficiencies of volatile organic compounds (VOCs) over extended operation of BTFs. To obtain an optimized biomass control strategy, the relative performance of five parallel BTFs receiving different ozone doses was determined, and the affecting mechanism of ozonation on biofilm was elucidated. Experimental results showed that the decline in ozone-free BTF performance began from day 150, which was correlated with excess biomass accumulation, abundant excretion of extracellular polymeric substances (EPS) and a decline in metabolic activity of biofilm over extended operation. Ozone of 5-10 mg m(-3) was effective in preventing excessive growth and uneven distribution of biomass, and eventually maintaining long-term stable operations. Ozone of over 20 mg m(-3) possibly inhibited microorganism growth severely, thereby deteriorating the elimination performance instead. Comparison of the biofilm EPS indicated that the presence of ozone reduce EPS contents to different extents, which was possibly beneficial for mass transfer and metabolic activity. Comparative community analysis showed that ozonation resulted in different microbial communities in the BTFs. Dyella was found to be the most abundant bacterial genera in all BTFs regardless of ozonation, indicating strong resistance to ozonation. Chryseobacterium and Burkholderia members were markedly enriched in the ozone-added biofilm, implying good adaptation to ozone presence. These findings provided an improved understanding of low-dose ozonation in maintaining a stable long-term operation of BTF.

OBJECTIVE: The study aims to evaluate the efficacy and safety of two Chinese herbal formulae for the treatment of stable COPD. METHODS: A multicenter, double-blind, double-dummy, and randomized controlled trial (RCT) was conducted. All groups were treated with additional conventional medicines. There were a 6-month treatment and a 12-month follow-up for 5 times. Primary outcomes included lung function test, exacerbation frequency, score of SGRQ. Second outcomes consisted of 6MWD, BODE index, psychological field score, inflammatory factors and cortisol. RESULTS: A total of 331 patients were randomly divided into two active treatment groups (Bushen Yiqi (BY) granule group, n = 109; Bushen Fangchuan (BF) tablet group, n = 109) and a placebo group (n = 113). Finally 262 patients completed the study. BY granule & BF tablet increased the values of VC, FEV1 (%) and FEV1/FVC (%), compared with placebo. BY granule improved PEF. Both treatments reduced acute exacerbation frequency (P = 0.067), BODE index and psychological field score, while improved 6MWD. In terms of descent rang of SGRQ score, both treatments increased (P = 0.01). Both treatments decreased inflammatory cytokines, such as IL-8, and IL-17(P = 0.0219). BY granule obviously descended IL-17(P<0.05), IL-1ß (P = 0.05), IL-6, compared with placebo. They improved the level of IL-10 and cortisol. BY granule raised cortisol (P = 0.07) and decreased TNF-α. Both treatments slightly descended TGF-ß1. In terms of safety, subject compliance and drug combination, there were no differences (P>0.05) among three groups. CONCLUSIONS: BY granule and BF tablet were positively effective for the treatment of COPD, and the former performed better in general. Three bacterial strains utilizing paracetamol as the sole carbon, nitrogen, and energy source were isolated from a paracetamol-degrading aerobic aggregate, and assigned to species of the genera Stenotrophomonas and Pseudomonas. The Stenotrophomonas species have not included any known paracetamol degraders until now. In batch cultures, the organisms f1, f2, and fg-2 could perform complete degradation of paracetamol at concentrations of 400, 2,500, and 2,000 mg/L or below, respectively. A combination of three microbial strains resulted in significantly improved degradation and mineralization of paracetamol. The co-culture was able to use paracetamol up to concentrations of 4,000 mg/L, and mineralized 87.1 % of the added paracetamol at the initial of 2,000 mg/L. Two key metabolites of the biodegradation pathway of paracetamol, 4-aminophenol, and hydroquinone were detected. Paracetamol was degraded predominantly via 4-aminophenol to hydroquinone with subsequent ring fission, suggesting new pathways for paracetamol-degrading bacteria. The degradation of paracetamol could thus be performed by the single isolates, but is stimulated by a synergistic interaction of the three-member consortium, suggesting a possible complementary interaction among the various isolates. The exact roles of each of the strains in the consortium need to be further elucidated.

OBJECTIVE: To evaluate the efficacy and safety of treatment based on syndrome differentiation of Traditional Chinese Medicine (TCM) for community-acquired pneumonia (CAP). METHODS: A total of 240 CAP patients were randomly divided into the following two groups: the control group was treated by anti-infection plus conventional medicine treatment; and the trial group was treated by TCM plus the above-mentioned treatment given to the controls. The course of treatment was 14 days, and the patients were followed up for 7 days. RESULTS: Of the 240 patients, 235 accomplished the whole process of treatment. The five patients who withdrew from the study were brought into an intent-to-treat analysis. The therapeutic effects of the trial group were superior to those of the control group (P < 0.01). The trial group took less time to become clinically stable, with a higher score in the quality of life (P < 0.01). There were no significant differences in mortality rate (P > 0.05), white blood cell count (P > 0.05), bacterial clearance rate (P > 0.05), and adverse reactions between the two integrated groups. CONCLUSION: Treatment based on TCM syndrome differentiation for CAP has the advantages of resulting in less time to achieve a stable clinical condition, improvement of clinical symptoms and quality of life, and is comparatively safe.

In this study, rat models of Parkinson's disease induced by substantia nigra injection of 6-hydroxy-dopamine were intragastrically administered Zhichan powder daily for 50 days. Reverse transcription PCR results showed that tyrosine hydroxylase mRNA expression in the rat substantia nigra was significantly increased, while monoamine oxidase B mRNA expression was significantly decreased in the Zhichan powder group, compared with the model group. In addition, the levels of striatal dopamine and homovanillic acid, the ratio of dopamine to homovanillic acid, and the activity of blood superoxide dismutase were all higher in the Zhichan powder group than in the model group, but the content of malondialdehyde in blood was lower. Our experimental findings indicate that Zhichan powder has an antioxidant effect, it can regulate the expression of monoamine oxidase B and tyrosine hydroxylase in the substantia nigra of Parkinson's disease rats, and it can facilitate the secretion of striatal dopamine and its metabolite homovanillic acid.

Previous studies have shown that Zhichan powder elevated immunity and suppressed oxidation in mice. Rat models of Parkinson's disease were induced by stereotaxically injecting 6-hydroxydopamine into the substantia nigra. The rat models were intragastrically treated with Zhichan powder, which is composed of milkvetch root, ginseng, bunge swallowwort root, himalayan teasel root, Magnolia officinalis, Ligustrum lucidum Ait. and szechwan lovage rhizome. Immunohistochemistry and reverse transcription-PCR results demonstrated that mRNA and protein expression of tumor necrosis factor receptor 1, Fas, caspase-8, cytochrome C, Bax, caspase-3, and p53 significantly increased, but Bcl-2 expression significantly decreased in the substantia nigra of rats with Parkinson's disease. Following Zhichan powder administration, mRNA and protein expression of tumor necrosis factor receptor 1, Fas, caspase-8, cytochrome C, Bax, caspase-3, and p53 diminished, but Bcl-2 expression increased in the rat substantia nigra. These results indicate that Zhichan powder regulates signal transduction protein expression, inhibits apoptosis, and exerts therapeutic effects on Parkinson's disease.

OBJECTIVE: To observe the difference between the therapeutic effect of Du-moxibustion (Moxibustion on the Governor Vessel) combined with western medicine and that of simple western medication for the remission stage of chronic obstructive pulmonary disease. METHODS: Two hundred and ten cases were randomly divided into an observation group (108 cases)and a control group (102 cases). The observation group was treated by routine treatment of western medicine combined with herb-partitioned spread moxibustion on the Governor Vessel between Dazhui (GV 14)and Yaoshu (GV 2). Simple western medicine was used in control group. The therapeutic effects of two groups were compared with the changes of symptom scores and pulmonary function before and after treatment. RESULTS: The total effective rate of observation group (90.7%, 98/108) was higher than that of control group (74.5%, 76/102) (P < 0.01). The symptom scores and some pulmonary function indices such as forced vital capacity (FVC), forced expiratory volume in one second (FEV1), the percentage of force expiratory volume in one second in predicted value(FEV1%) and maximal expiratory flow(PEF) after treatment were improved obviously than those before treatment in observation group (P < 0.05, P < 0.01), and the improvement degree was better than that of control group (all P < 0.01). CONCLUSION: Moxibustion on the Governor Vessel combined with western medicine can improve the clinical symptoms and pulmonary function of chronic obstructive pulmonary disease of lung and kidney qi deficiency type effectively,and the effect is better than that of simple western medication.

In this study, biotrickling filter (BTF) inoculated with acclimated sludge was established to treat waste gas containing chlorobenzene. The BTF performance, average well color development (AWCD) values and microbial community were examined in steady state. Results revealed BTF achieved removal efficiency more than 80% of chlorobenzene under the conditions of < 0.6 g x m(-3) inlet concentration and > 45 s EBRT. Therefore, BTF have an advantage in treating low-concentration waste gas containing chlorobenzene (< or = 0.6 g x m(-3)). The overall chlorobenzene elimination capacity reached a maximum of 70 g x (m3 x h)(-1) at an inlet load of 80 g x (m3 x h)(-1). The mass ratio of carbon dioxide produced to the BTo-X removed was approximately 1.92, which confirms complete degradation of chlorobenzene, given that some of the organic carbon consumed is also used for the microbial growth. The degradation of chlorobenzene in the BTF followed Michaelis-Menten kinetic model, the maximum specific degradation rate (r(max)) was 35.6 g x (m3 x h)(-1). The AWCD values indicated that the microorganisms in the BTF showed high the microbial metabolic activity. The PCR-DGGE fingerprinting analysis on biofilm samples in the BTF indicated that the microbial community had a relative stability and complexity during the steady-state phase. The stability and complexity of microbial community could contribute to the degradation and mineralization of chlorobenzene in BTF.
