Primiparous Angus × Simmental dams (n = 22) with an average body weight (BW) of 449 ±â€…32 kg of BW were divided based on two nutritional treatments: control (CTRL) and rumen-protected methionine (RPM). The control group received bermudagrass hay, corn gluten, and soybean hulls pellets supplementation (base diet); whereas the RPM group received the base diet in addition to 0.07% of DM of RPM at a fixed rate during the last trimester of gestation and the first ~80 d of lactation, in which calves (n = 17) were early weaned. Only male calves were included in this study. After weaning, calves born to RPM dams also received RPM from weaning (day 1) to day 100. Blood sampling and skeletal muscle biopsies for subsequent quantitative polymerase chain reaction (PCR) analysis were conducted on days 1, 25, 50, and 100 on calves. Quantitative PCR data were analyzed using GLIMMIX, and blood metabolites concentrations, BW, and body condition score (BCS) were analyzed using the MIXED procedure of SAS. There was no difference in maternal BW and BCS between treatments. Glucose and blood metabolites that served as biomarkers for liver health (e.g., aspartate transaminase, albumin, alkaline phosphatase, and alanine transaminase) were in the normal levels for all calves (P > 0.40). Calves in the RPM group had a greater expression of adipogenic genes (e.g., PPARG, LPL, and CEBPD) at day 100 compared with CTRL (P < 0.01). In addition, DNA methylation (DNMT1) and oxidative stress-related genes (SOD2 and NOS3) in the RPM group were upregulated at day 100 compared with CTRL (P < 0.01). These results may suggest that calves born to primiparous dams exposed to RPM supplementation are more prone to develop greater adipose tissue than CTRL calves. Furthermore, RPM supplementation may improve methylation processes, as shown by the upregulation of DNMT1. The results shown in our study aim at expanding the knowledge on fetal programming and early-life growth and development of beef cattle under supplementation with RPM.


Plane of nutrition plays a critical role in fetal and postnatal growth in beef cattle offspring. Methionine, a limiting amino acid in ruminants, is also involved in DNA methylation due to its role as S-adenosyl methionine precursor. A complete randomized design experiment was conducted to assess the fetal programming effect of rumen-protected methionine (RPM) supplementation on beef cattle. Calves born to primiparous beef heifers, that individually received RPM during the last trimester of gestation and lactation, had a greater expression of genes related to adipose tissue development, oxidative stress, and DNA methylation compared with those born to dams that did not receive rumen-protected supplementation. No difference in animal performance or blood parameters that serve as biomarkers for liver health status was detected. Our results suggest that maternal supplementation with RPM during the last trimester of gestation and lactation, and supplementation to the offspring after early weaning, could potentially increase adipose tissue development on male calves.

Research studies have modified traditional substances to seek fast-acting removal of phosphorus in constructed wetlands (CWs) and ecological dams, rather than develop a brand-new nano-adsorbent. This work synthesized FeCa-based layered double hydroxide (FeCa-LDH) with a chemical co-precipitation method, and the performance, mechanism and factors of phosphorus removal were investigated. FeCa-LDH showed a marked ability to adsorb phosphorus from waste water, with a removal efficiency of 94.4% and 98.2% in CWs and ecological dams, respectively. Both FTIR and XPS spectrum evidenced that FeCa-LDH removed phosphorus via electrostatic and hydrogen-bonding adsorption, as well as a coordination reaction and interlayer anion exchange. FeCa-LDH showed a higher capacity to remove phosphorus in alkaline and neutral waste water than in acid conditions. Co-occurrence anions, which influenced the efficiency of the phosphorus removal capacity are considered in the sequence below: CO32- ≈ HCO3- > SO42- > NO3-. Innovatively, FeCa-LDH was not affected by the low-temperature limitation for CWs, and phosphorus removal efficiency at 5 °C was almost equal to that at 25 °C. These results cast a new idea on the construction, application and phosphorus removal performance of CWs and ecological dams.

The upper Yangtze River (UYR) plays an important role in water supply, hydropower generation, environmental and ecological protection. Constructions of Mega cascade reservoirs have significantly affected the transport of sediment and P, but the evolution of sediment and P in the mega cascade reservoirs of the UYR is unclear. This study investigated the variations in sediment load and total P (TP) flux based on the flow and sediment data from 1990 to 2019 and TP concentrations from 2005 to 2019. In addition, the proportion of sediment load and TP flux from tributaries, variations in the concentrations of particulate P (PP) and dissolved P, trapping effect of dams, and statistical uncertainties were analyzed and discussed. The main results are as follows: (1) the sediment load and TP flux evidently decreased after the impoundment of reservoirs in the UYR and Jinsha River, and the contribution rate of TP flux from main tributaries (except Wu River) to mainstream increased by 3.82-24 %; (2) the error of TP flux calculated by daily and monthly data is within 30 %, which shows that the uncertainty range is clear at some degree attributed to the different monitoring frequency, and the concentration of PP in flood season is greater than that in non-flood season; (3) the total retention rates of sediment and TP in the Three Gorges Reservoir and Gezhouba were 86.78 %, and 49.83 % (2009-2012), respectively, but decreased to 82.85 % and 15.26 % (2013-2019), and the values in Xiangjiaba and Xiluodu were 97.83 % and 60.27 % (2013-2019). The retention rates of newly built reservoirs (Wudongde and Baihetan) were predicted using an empirical method, and the results revealed that new dams would facilitate long-term reduction in sediment downstream.

Urban river pollution is considered a 'necessary evil' consequence of disproportionate developmental expansion in metropolises. Unprecedented expansion and anthropic activities lead to the deterioration of urban rivers with municipal and industrial sewage. The construction of sluices is one of the irrefutable parts of the process. In order to prevent floods and drought, many cities build sluices and dams in rivers to balance water quantity in different seasons. To explore the change characteristics of the water quality in urban rivers after the construction of sluices and dams, the change in the total phosphorus (TP) and total nitrogen (TN) concentrations upstream and downstream of rivers was investigated under the condition of sluices closure in Wuxi. According to the results, when the sluices were closed, the pollutants of TP and TN would accumulate upstream in rivers, which caused the water quality in the upper reaches to be worse than that in the lower reaches. Specifically, the TN and TP concentrations downstream of urban rivers in Wuxi were approximately 14.42% and 13.80% lower than those upstream when the sluices were closed. Additionally, the water quality in urban rivers was usually better in summer and autumn than in the other seasons, showing obvious seasonality after the construction of the sluices. The research will provide a theoretical basis for future sluice operation and the water resources management of urban rivers.

The Developmental Origins of Health and Disease (DOHaD) concept has been proposed to explain the influence of environmental conditions during critical developmental stages on the risk of diseases in adulthood. The aim of this study was to compare the impact of the prenatal vs. postnatal environment on the gut microbiota in dams during the preconception, gestation and lactation periods and their consequences on metabolic outcomes in offspring. Here we used the cross-fostering technique, e.g. the exchange of pups following birth to a foster dam, to decipher the metabolic effects of the intrauterine versus postnatal environmental exposures to a polyphenol-rich cranberry extract (CE). CE administration to high-fat high-sucrose (HFHS)-fed dams improved glucose homeostasis and reduced liver steatosis in association with a shift in the maternal gut microbiota composition. Unexpectedly, we observed that the postnatal environment contributed to metabolic outcomes in female offspring, as revealed by adverse effects on adiposity and glucose metabolism, while no effect was observed in male offspring. In addition to the strong sexual dimorphism, we found a significant influence of the nursing mother on the community structure of the gut microbiota based on α-diversity and ß-diversity indices in offspring. Gut microbiota transplantation (GMT) experiments partly reproduced the observed phenotype in female offspring. Our data support the concept that the postnatal environment represents a critical window to influence future sex-dependent metabolic outcomes in offspring that are causally but partly linked with gut microbiome alterations.

Melatonin treatment in adult cashmere goats can increase cashmere yield and improve cashmere fibre quality by inducing cashmere growth during cashmere non-growth period, of which time cashmere goats are in the mid and late stages of lactation. However, whether melatonin treatment in adult cashmere goats affects their offspring's growth performance remains unknown. Therefore, the objectives of the current study were to determine the effects of melatonin treatment in adult cashmere goats on cashmere and milk production performance in dams and on hair follicle development and subsequent cashmere production in their offspring. Twenty-four lactating Inner Mongolian Cashmere goat dams (50 ± 2 days in milk, mean ± SD) and their single-born female offspring (50 ± 2 days old, mean ± SD) were randomly assigned to one of two groups supplemented with melatonin implants (MEL; n = 12) or without (CON; n = 12). The melatonin implants were subcutaneously implanted behind the ear at a dose of 2 mg/kg live weight on two occasions - 30 April and 30 June 2016. The results demonstrated that melatonin treatment in adult cashmere goats increased cashmere production and improved cashmere fibre quality as indicated by greater cashmere yield, longer cashmere fibre staple length, finer cashmere fibre diameter and thicker cashmere fibre density. The milk fat content was higher in MEL compared with CON cashmere goats. The daily yields of milk production, milk protein and milk lactose were lower in MEL compared with CON cashmere goats. Serum melatonin concentrations were greater, serum prolactin concentrations were lower and milk melatonin concentrations and yields were greater in MEL compared with CON cashmere goats. With regard to offspring, there were no differences in cashmere yield, fibre staple length, fibre diameter and fibre density at yearling combing, and the primary and secondary hair follicles population and maturation between treatments. In conclusion, melatonin treatment in adult cashmere goats during cashmere non-growth period is a practical and an effective way in cashmere industry as indicated by not only increasing cashmere yield and improving cashmere fibre quality in adult cashmere goat dams but also having no impairment in hair follicle development and the subsequent cashmere production in their single-born offspring.

Methylmercury (MeHg) is a well-known environmental pollutant associated with neurological and developmental deficits in animals and humans. However, epidemiological data showed that people living in the Amazon region although exposed to MeHg do not present these effects probably due to the protective effect of certain foods. We hypothesized here if guarana, a highly caffeinated fruit and consumed on a daily basis by Amazon people, could have some protective effect against MeHg toxicity using two complementary approaches. To assess locomotor impairment and sleep disruption, we used fruit fly (Drosophila melanogaster) model, and to evaluate neuroinflammation, we used human SH-SY5Y neural cells by measuring inflammatory cytokines levels. Results showed that guarana had a protective effect on the locomotor activity of male fruit flies reducing the excessive sleepiness caused by MeHg and increasing daily activity. Also, guarana increased the viability of flies and attenuated neural cells mortality. In addition, guarana reduced all pro-inflammatory cytokines levels increased by MeHg, along with caspase-1, caspase -3, caspase-8, and 8-dOHG levels, whereas increased the anti-inflammatory (IL-10) cytokine levels, which was decreased by MeHg. Our study provides new insights on the protective effects of guarana on the viability, locomotor activity, sleep, and activity patterns in vivo and the in vitro neuronal anti-inflammatory effect against MeHg toxicity.

Recent proliferation of hydro-dams was one of the nature-based solutions to meet the increasing demand for energy and food in the Lower Mekong River Basin (LMRB). While construction of these hydro-dams generated some hydropower and facilitated expansion of irrigated lands, it also significantly altered the basin-wide hydrology and subsequently impacted wetland ecosystems. Unintended adverse consequences of ecosystem services from lakes and wetlands offset the intended gains in hydroelectricity and irrigated agriculture. The trade-offs between gains in energy and food production and losses in aquatic ecosystem services were perceived to be significant but knowledge of the magnitude, spatial extent, and type of ecosystem services change is lacking and, therefore, the question whether the hydro-dam is an optimized solution or a potential ecological problem remains unanswered. In this study, as the first step to answer this question and using the Tonlé Sap Lake as an example, we quantified one of the impacts of hydro-dams on lake ecosystem's phenology in terms of open water area, a critical ecological characteristic that affects lake systems' fish production, biodiversity, and livelihoods of the local communities. We used the MODIS-NDVI time series, forecast function and the Mann-Kendall trend test method to first quantify the open water area, analyzed its changes over time, and then performed correlation analysis with climate variables to disentangle dam impacts. The results showed reduced hydro-periods, diminishing lake seasonality and a declining trend in Tonlé Sap Lake open water area over the past 15 years. These changes were insignificantly related to climatic influence during the same period. It is concluded that basin-wide hydro-dam construction and associated agricultural irrigation were deemed to be the primary cause of these ecological changes. Further analyses of changes in the lake's ecosystem services, including provision and cultural services, need to be carried out in order to have a holistic understanding of the trade-offs brought by the hydro-dam proliferation as a solution to the emerging energy and food demand in the LMRB.

Recent selective stimulation and ablation of galanin neurons in the preoptic area of the hypothalamus established their critical role in control of maternal behaviors. Here, we identified a group of galanin neurons in the anterior commissural nucleus (ACN), and a distinct group in the medial preoptic area (MPA). Galanin neurons in ACN but not the MPA co-expressed oxytocin. We used immunodetection of phosphorylated STAT5 (pSTAT5), involved in prolactin receptor signal transduction, to evaluate the effects of suckling-induced prolactin release and found that 76 % of galanin cells in ACN, but only 12 % in MPA were prolactin responsive. Nerve terminals containing tuberoinfundibular peptide 39 (TIP39), a neuropeptide that mediates effects of suckling on maternal motivation, were abundant around galanin neurons in both preoptic regions. In the ACN and MPA, 89 and 82 % of galanin neurons received close somatic appositions, with an average of 2.9 and 2.6 per cell, respectively. We observed perisomatic innervation of galanin neurons using correlated light and electron microscopy. The connection was excitatory based on the glutamate content of TIP39 terminals demonstrated by post-embedding immunogold electron microscopy. Injection of the anterograde tracer biotinylated dextran amine into the TIP39-expressing posterior intralaminar complex of the thalamus (PIL) demonstrated that preoptic TIP39 fibers originate in the PIL, which is activated by suckling. Thus, galanin neurons in the preoptic area of mother rats are innervated by an excitatory neuronal pathway that conveys suckling-related information. In turn, they can be topographically and neurochemically divided into two distinct cell groups, of which only one is affected by prolactin.

Creep feeding has been used to reduce calves' nutritional dependence on the cow, but research results under tropical conditions have not been conclusive about the effects on the cow. Therefore, this study was conducted to evaluate the effects of high and low supplementation levels for Nellore heifer calves on performance, milk production, and metabolic profile of their mothers. Fifty multiparous Nellore cows and their respective calves were used. The following treatments were evaluated: 0-control, no supplement was fed to calves; 3-calves received supplement in the amount of 3 g/kg of body weight (BW); 6-calves received supplement in the amount of 6 g/kg of BW. There was no significant effect of level of supplementation offered to offspring on cow BW, body condition score (BCS) and subcutaneous fat thickness (P > 0.05). Level of supplementation of heifer calves did not significantly affect milk production corrected to 4% of fat (P > 0.05). Fat, protein, lactose, and total solids of the milk also did not differ among supplementation strategies (P > 0.05). Level of supplement fed to calves had no effect on cows' glucose, total cholesterol, HDL, LDL, triglycerides, total protein, and albumin levels (P > 0.05), but cows nursing calves that did not receive supplement had lower level of serum urea N (SUN; P < 0.05). We conclude that creep feeding in the amounts of 3 or 6 g/kg of BW daily has no major impact on dams' performance and metabolism.

Fish oil is commonly taken by pregnant women, and supplements sold at retail are often oxidized. Using a rat model, we aimed to assess the effects of supplementation with oxidized fish oil during pregnancy in mothers and offspring, focusing on newborn viability and maternal insulin sensitivity. Female rats were allocated to a control or high-fat diet and then mated. These rats were subsequently randomized to receive a daily gavage treatment of 1 ml of unoxidized fish oil, a highly oxidized fish oil, or control (water) throughout pregnancy. At birth, the gavage treatment was stopped, but the same maternal diets were fed ad libitum throughout lactation. Supplementation with oxidized fish oil during pregnancy had a marked adverse effect on newborn survival at day 2, leading to much greater odds of mortality than in the control (odds ratio 8.26) and unoxidized fish oil (odds ratio 13.70) groups. In addition, maternal intake of oxidized fish oil during pregnancy led to increased insulin resistance at the time of weaning (3 wks after exposure) compared with control dams (HOMA-IR 2.64 vs. 1.42; P = 0.044). These data show that the consumption of oxidized fish oil is harmful in rat pregnancy, with deleterious effects in both mothers and offspring.

A batch of lab-based adsorption experiments were performed to investigate the arsenic (As) removal efficacy by activated alumina. Four factors including contact time, pH, initial As concentration and different coexisting ions were examined. The adsorbent made of activated alumina (AA) with particles of 2-4 mm diameter showed a high As removal efficiency and the As concentrations of the samples were below 0.05 mg/L when the hydraulic retention time (HRT) was operated above 5 min. The As concentrations of the samples could remain below 0.05 mg/L for 30 days. A series of AA adsorption dams coupled with several other supporting adsorption techniques were employed for As-contaminated river restoration. The engineering project functioned well, and the effluent As concentration was below 0.05 mg/L when the influent was between 0.2 and 0.7 mg/L, which met the discharge requirement of the Surface Water Quality Standards criteria III in China. The results demonstrated that AA adsorption dams could be applied for emergency treatments of small- or medium-sized rivers contaminated with As.

Nursing has important physiological and psychological consequences on mothers during the postpartum period. Tuberoinfundibular peptide of 39 residues (TIP39) may contribute to its effects on prolactin release and maternal motivation. Since TIP39-containing fibers and the receptor for TIP39, the parathyroid hormone 2 receptor (PTH2 receptor) are abundant in the arcuate nucleus and the medial preoptic area, we antagonized TIP39 action locally to reveal its actions. Mediobasal hypothalamic injection of a virus encoding an antagonist of the PTH2 receptor markedly decreased basal serum prolactin levels and the suckling-induced prolactin release. In contrast, injecting this virus into the preoptic area had no effect on prolactin levels, but did dampen maternal motivation, judged by reduced time in a pup-associated cage during a place preference test. In support of an effect of TIP39 on maternal motivation, we observed that TIP39 containing fibers and terminals had the same distribution within the preoptic area as neurons expressing Fos in response to suckling. Furthermore, TIP39 terminals closely apposed the plasma membrane of 82% of Fos-ir neurons. Retrograde tracer injected into the arcuate nucleus and the medial preoptic area labeled TIP39 neurons in the posterior intralaminar complex of the thalamus (PIL), indicating that these cells but not other groups of TIP39 neurons project to these hypothalamic regions. We also found that TIP39 mRNA levels in the PIL markedly increased around parturition and remained elevated throughout the lactation period, demonstrating the availability of the peptide in postpartum mothers. Furthermore, suckling, but not pup exposure without physical contact, increased Fos expression by PIL TIP39 neurons. These results indicate that suckling activates TIP39 neurons in the PIL that affect prolactin release and maternal motivation via projections to the arcuate nucleus and the preoptic area, respectively.

In addition to intellectual disability, individuals with Down syndrome (DS) exhibit dementia by the third or fourth decade of life, due to the early onset of neuropathological changes typical of Alzheimer's disease (AD). Deficient ontogenetic neurogenesis contributes to the brain hypoplasia and hypocellularity evident in fetuses and children with DS. A murine model of DS and AD (the Ts65Dn mouse) exhibits key features of these disorders, notably deficient ontogenetic neurogenesis, degeneration of basal forebrain cholinergic neurons (BFCNs), and cognitive deficits. Adult hippocampal (HP) neurogenesis is also deficient in Ts65Dn mice and may contribute to the observed cognitive dysfunction. Herein, we demonstrate that supplementing the maternal diet with additional choline (approximately 4.5 times the amount in normal rodent chow) dramatically improved the performance of the adult trisomic offspring in a radial arm water maze task. Ts65Dn offspring of choline-supplemented dams performed significantly better than unsupplemented Ts65Dn mice. Furthermore, adult hippocampal neurogenesis was partially normalized in the maternal choline supplemented (MCS) trisomic offspring relative to their unsupplemented counterparts. A significant correlation was observed between adult hippocampal neurogenesis and performance in the water maze, suggesting that the increased neurogenesis seen in the supplemented trisomic mice contributed functionally to their improved spatial cognition. These findings suggest that supplementing the maternal diet with additional choline has significant translational potential for DS.
