RESUMO

Targeted green light photostimulation during the last stage of broiler incubation increases expression of the somatotropic axis. The purpose of this study was to further shorten the in ovo green light photostimulation and determine the critical age for photostimulation in broilers embryos, as a future strategy for broiler incubation. Fertile broilers eggs (n = 420) were divided into 5 treatment groups. The first group was incubated under standard conditions (in the dark) as the negative control group. The second was incubated under intermittent monochromatic green light using light-emitting diode lamps with an intensity of 0.1 W/m2 at shell level from embryonic day (ED) 0 of incubation until hatch, as a positive control. The third, fourth, and fifth groups were incubated under intermittent monochromatic green light from ED 15, 16, and 18 of incubation, respectively, until hatch. All treatment groups showed elevated somatotropic axis expression compared with the negative control, with the group incubated under monochromatic green light from ED 18 until hatch showing results closest to the positive control. This suggests that broiler embryos can be exposed to in ovo green light photostimulation from a late stage of incubation (when transferring the eggs to the hatchery) and exhibit essentially the same outcome as obtained by photostimulation during the entire incubation period.

Purpose: Gamma radiation is mainly used for disinfesting insect pests as an alternative for harmful fumigants. The specific dose of radiation is known to affect different developmental stages of insect pests. The study was conducted to determine the effective irradiation doses for inhibition of developmental stages and adult longevity of the coffee berry borer, Hypothenemus hampei (Ferrari). Materials and methods: Irradiation was carried with the following doses: five levels between 0.01 and 0.16 kGy for eggs, seven levels between 0.10 and 2.00 kGy for larva and prepupa, six levels between 0.10 and 1.60 kGy for pupa and ten levels between 0.10 and 3.20 kGy for adults. Results: Egg development was completely arrested at 0.160 kGy. A dose of 2.00 kGy caused 100% mortality in the first and second instar larva and 98.99% mortality in prepupa. The dose of 1.60 kGy prevented adult eclosion from the irradiated pupa. The adult mortality was 100% at 3.20 kGy. Conclusion: A dose of 3.20 kGy could successfully provide complete security from all developmental stages of H. hampei and prevent yield loss in green coffee as well as the spread of the pest.

It is well known that, under exposure to bright light, eggs tend to hatch earlier than control, without any damage to the birds. This report aims to systematically show the effect and establishes a proposal for a possible application to accelerate chicken egg formation, which could be extrapolated or adapted as a great advance in premature human newborns. Comparing several protocols, the experiments show that lower doses of light slowly delivered for 24 h promote higher efficiency in embryo development, increasing on average 25% of its size and more than 70% in weight when compared to the control. This weight difference shows promising results compared to rates of up to 17% found in the literature. These results can be a first step to reduce the stay of premature human infants in hospitals because light, when applied in very low doses, can accelerate the natural biological processes without risks.

Previous studies demonstrated that in ovo photostimulation with monochromatic green light increases body weight and accelerates muscle development in broilers. The mechanism in which in ovo photostimulation accelerates growth and muscle development is not clearly understood. The objective of the current study was to define development of the somatotropic axis in the broiler embryo associated with in ovo green light photostimulation. Two-hundred-forty fertile broiler eggs were divided into 2 groups. The first group was incubated under intermittent monochromatic green light using light-emitting diode (LED) lamps with an intensity of 0.1 W\m2 at shell level, and the second group was incubated under dark conditions and served as control. In ovo green light photostimulation increased plasma growth hormone (GH) and prolactin (PRL) levels, as well as hypothalamic growth hormone releasing hormone (GHRH), liver growth hormone receptor (GHR), and insulin-like growth factor-1 (IGF-1) mRNA levels. The in ovo photostimulation did not, however, increase embryo's body weight, breast muscle weight, or liver weight. The results of this study suggest that stimulation with monochromatic green light during incubation increases somatotropic axis expression, as well as plasma prolactin levels, during embryonic development.

Exposure to metal-contaminated water has been shown to result in a number of reproductive abnormalities in adult and larvae fish, such as failure of oocyte maturation and teratogenic effects. Recently, dietary uptake of metals by fish has been recognized as a critical route of exposure, however, the mechanisms of metal uptake and toxicity are poorly understood and in need of further investigation. The objectives of the present study are to quantify uranium (U dietary transfers from spiked artificial diets) in Danio rerio tissues and embryos, as well as establish its effect on reproduction and embryonic development. Uranium's environmental prominence is currently increasing because of new mining and milling activities. Uranium concentrations range from 0.02 µg/L in natural waters to 2 mg/L. The focus of this study was to examine the trophic transfer and effects of U following exposure modalities (dose, exposure duration 1 to 20 d). Two different isotopes were used to distinguish between chemical and radioactivity toxicity of U. Results showed that U trophic transfer was low (0.52%). Uranium tissue distributions showed that accumulation occurred in digestive organs (liver, digestive tract) following dietary exposure. High levels of U were measured in the gonads (female in particular, >20% of relative burden). High U accumulation levels in eggs indicated maternal transfer of the contaminant. Moreover, U trophic exposure led to a reduction in reproduction success as a function of U accumulated levels. High U exposure conditions strongly reduced the total number of eggs (50%) and their viability at 10 d (reduction of the clutch number, low quality of eggs).

Irradiation has been recognized and endorsed as a potential phytosanitary measure that could be an alternative to current quarantine treatments. Dosages of 50, 100, 150, 200, and 250 Gy were used to irradiate three different life stages (eggs, immatures, and adults) of Planococcus minor (Maskell) (Hemiptera: Pseudococcidae), focusing on females due to its parthenogenesis ability, with an aim to find the most tolerant stage and the most optimal dose to control P. minor. Cobalt 60 was the source of irradiation used. Irradiation of 150-250 Gy has a significant effect on all life stages of P. minor, decreasing its survival rate, percentage of adult reproduction, oviposition, and fertility rate. The adult was the most tolerant life stage in both mortality and fertility rate. All the different irradiated target life stage groups oviposited eggs, but none of the F2 eggs hatched at the most optimal dosage of 150-250 Gy.

This study reports on the effect of microwave radiation for inactivation of Ascaris lumbricoides eggs in 25 g of soil compared to ultraviolet irradiation and ozone expose. Microwave radiation at 700 W with 14% water content (w/w) achieved approximately 2.5 log inactivation of eggs in soil within 60s. On the other hand, UV irradiation at 3 mW cm(-2) with and without shaking soil for 3600 s achieved approximately 0.32 and 0.01 log inactivation of eggs, respectively. In ozone treatment, 0.13 log inactivation of eggs was achieved with 5.8+/-0.7 mg L(-1) of dissolved ozone dose for 30 min in a continuous diffusion reactor. In addition, the inactivation of eggs by three disinfection techniques was conducted in water in order to compare the inactivation efficiency of eggs in soil. The inactivation efficiency of microwave radiation was found to be no significant difference between in soil and water. However, the inactivation efficiency of UV irradiation was significantly increased in water while in ozone expose there was no significant difference between in soil and water. Microwave treatment thus proved to be the most efficient method in controlling A. lumbricoides eggs in soil.

The European corn borer, Ostrinia nubilalis (Hübner) (Lepidoptera: Crambidae), is a quarantine pest for several fresh commodities, including corn-on-the-cob, bell peppers, and green beans. Methyl bromide fumigation is the usual phytosanitary treatment, but the fumigant is under increasing regulation as a stratospheric ozone-depleting substance. Ionizing radiation is a relatively new commercial alternative that is currently used in several countries. The present research explored radiation doses that would provide quarantine security for commodities at risk of being infested by O. nubilalis. Radiotolerance of late pupae (the most tolerant stage infesting commodities) as determined by hatch of F1 eggs was not affected by host (meridic diet versus ear corn) or temperature (1 versus 13 degrees C) but was positively affected by low oxygen. Longevity was shorter for adults of irradiated than nonirradiated pupae. The minimum absorbed dose for phytosanitary irradiation against O. nubilalis could vary from 233 Gy for prevention of F1 pupation to 343 Gy for prevention of F1 egg hatch. Lower doses might be possible if greater risk of treatment failure was acceptable.

The photoenhanced toxicity of weathered Alaska North Slope crude oil (ANS) was investigated in the eggs and larvae of Pacific herring (Clupea pallasi) with and without the chemical dispersant Corexit 9527. Oil alone was acutely toxic to larvae at aqueous concentrations below 50 microg/L total polycyclic aromatic hydrocarbons (tPAH), and median lethal (LC50s) and effective concentrations (EC50s) decreased with time after initial oil exposure. Brief exposure to sunlight (approximately 2.5 h/d for 2 d) significantly increased toxicity 1.5- to 48-fold over control lighting. Photoenhanced toxicity only occurred when oil was present in larval tissue and increased with increasing tPAH concentration in tissue. Ultraviolet radiation A (UVA) treatments were less potent than natural sunlight, and UVA + sunlight caused greater toxicity than sunlight alone. The toxicity of chemically dispersed oil was similar to oil alone in control and UVA treatments, but oil + dispersant was significantly more toxic in the sunlight treatments. The chemical dispersant appeared to accelerate PAH dissolution into the aqueous phase, resulting in more rapid toxicity. In oil + dispersant exposures, the 96-h no-observed-effect concentrations in the UVA + sunlight treatment were 0.2 microg/L tPAH and 0.01 microg/g tPAH. Exposure of herring eggs to oil caused yolk sac edema, but eggs were not exposed to sun and UVA treatment did not cause phototoxicity. These results are consistent with the hypothesis that weathered ANS is phototoxic and that UV can be a significant and causative factor in the mortality of early life stages of herring exposed to oil and chemically dispersed oil.

A study was made of the effect of alpha-radiation of 238U solutions and enriched uranium (9.9 X 10(7) Bq/g) on developing Misgurnus fossilis eggs. The incidence of abnormal larvae and the death rate of eggs increased when the latter were incubated in 138 mg/l solution of uranium 238: this was not accompanied by the increase in the yield of the aberrant ana- and telophases in the epithelium cells of 1-day larvae.
