The use of hair samples to assess the mineral intake of horses under practical conditions has recently been attracting increased interest. The aims of this study were to compare mineral analysis results in equine mane hair between three commercial laboratories and to determine the relationship between the mineral intake and mineral content in mane hair. Four warmblood horses (14 ± 3 years) were included in the study. Horses were fed hay ad libitum and a commercial mineral supplement to meet nutrient requirements under maintenance conditions. Mane hair from the hairline was collected with a sample length of 2 to 3 cm according to the respective laboratory guidelines to monitor the feeding period of the last 1 to 2 months. Minerals in mane hair were analyzed by three commercial laboratories using inductively coupled plasma mass spectrometry (ICP-MS) or inductively coupled plasma optical emission spectroscopy and in feedstuffs using ICP-MS to calculate mineral intake. Hair mineral analysis showed that the results and their corresponding reference ranges widely varied among the three laboratories. The mean coefficient of variance ranged from a minimum of 10% for selenium (Se) to a maximum of 58% for iron (Fe). For example, Se supply (requirement, 1.17-1.28 mg/d; intake, 1.83 mg/d) was considered insufficient in all horses by two laboratories, whereas it was considered adequate by one laboratory. Intervariation of the same sample, as well as differences in reference ranges, was considerably high among the three laboratories. These results indicate that hair mineral analysis is not reliable for assessing mineral intake in horses.

Analysis of hair to gain insight into the trace mineral status and exposure to toxic heavy metals of horses is attractive because hair is an easily accessible sample material. To investigate the potential value of hair analysis in horses for determination of trace mineral and heavy metal concentrations, we analyzed mane hair and liver samples from 62 horses presented for slaughter at a facility in the Netherlands that receives horses from all regions of the country. Hair samples were cleaned in warm water. After acid digestion of hair and liver specimens, we quantified, with inductively coupled plasma-mass spectrometry, arsenic, cadmium, chromium, cobalt, copper, iron, lead, manganese, molybdenum, nickel, selenium, vanadium, and zinc in the digests. Based on Pearson product moment correlations, we found no statistically significant correlations between concentrations of trace minerals in liver and hair, with the exception of a slight correlation for copper that was too weak to be of clinical relevance. Our results do not support the use of hair to determine trace mineral status and exposure to toxic heavy metals in the horse under field conditions.

Objective: Endogenous hormones regulate numerous physiological processes in humans. Some of them are routinely measured in blood, saliva and/or urine for the diagnosis of disorders. The analysis of fluids may, however, require multiple samples collected at different time points to avoid the high variability in the concentration of some hormones. In contrast, hair analysis has been proposed as an interesting alternative to reveal average hormone levels over a longer period. In this work, we developed and validated an analytical method for analyzing 36 endogenous steroid and thyroid hormones and one pineal hormone in human hair using ultra-performance liquid chromatography (UPLC)-tandem mass spectrometry (MS/MS). Methods: Sample preparation involved hair decontamination, pulverization, methanol extraction, and purification with C18-solid phase extraction. Extracts were then divided into two portions, respectively injected into an UPLC-MS/MS system, and analyzed using two different instrumental methods. The method was applied to a healthy female population aged 25-45 years. Results: The method was validated on supplemented hair samples for the 37 targeted hormones, and its application to the population under study allowed to detect 32 compounds in 2-100% of the samples. Complete reference intervals (2.5-97.5th percentiles) were established for estrone, 17ß-estradiol, androstenedione, dehydroepiandrosterone, progesterone, 17α-hydroxyprogesterone, cortisone, cortisol and 3,3',5-triiodo-L-thyronine. Hair cortisone, cortisol, tetrahydrocortisone and tetrahydrocortisol concentrations were highly correlated with each other, with Kendall's τ correlation coefficients ranging from 0.52 to 0.68. Conclusion: Allowing the detection of 32 hormones from different chemical classes, the present method will allow to broaden hormonal profiling for better identifying endocrine disorders.

RESUMO

A previously published method for single hair analysis has been applied to a doping case for further clarification. Amphetamine could be detected in multiple micro segments resulting in two distinct concentration peaks in several hairs. The consumption of a contaminated food supplement as possible source for the amphetamine is discussed.

Due to its environmental persistence and bioaccumulative properties, mercury is considered highly toxic to humans, ecosystems, and wildlife. The present study aimed to investigate the total mercury (Hg) concentrations in hair samples of 40 wild cats belonging to eight different species collected from various provinces of Iran and to characterize their risk of Hg exposure. Total Hg levels in Iranian wild cats ranged from 62 to 3670 ng/g dw hair, with a median value of 488 ng/g dw hair. The lowest median Hg concentration was found in west Iran (251 ng/g dw hair), while the highest median level was measured in the north-east of the country (736 ng/g dw hair), likely related to its past use in agriculture and potential contamination of industrial effluents. The overall contamination of Iranian wild cats with mercury can be considered generally low, but 20% of the samples reached levels above 1100 ng/g dw hair, set as indicative of an environmental Hg concern according to the U.S. Fish and Wildlife Service. Our data are comparable with other investigations on wildlife worldwide, confirming the suitability of museum collection specimens to assess the environmental levels of Hg. We showed that, while diet and habitat can influence the mercury accumulation in wild cats, complementary factors, such as age, size, and interspecific metabolic differences, should be considered as well in future investigations.
