Triphyophyllum peltatum, a rare tropical African liana, is unique in its facultative carnivory. The trigger for carnivory is yet unknown, mainly because the plant is difficult to propagate and cultivate. This study aimed at identifying the conditions that result in the formation of carnivorous leaves. In vitro shoots were subjected to abiotic stressors in general and deficiencies of the major nutrients nitrogen, potassium and phosphorus in particular, to trigger carnivorous leaves' development. Adventitious root formation was improved to allow verification of the trigger in glasshouse-grown plants. Among all the stressors tested, only under phosphorus deficiency, the formation of carnivorous leaves was observed. These glandular leaves fully resembled those found under natural growing conditions including the secretion of sticky liquid by mature capture organs. To generate plants for glasshouse experiments, a pulse of 55.4 µM α-naphthaleneacetic acid was essential to achieve 90% in vitro rooting. This plant material facilitated the confirmation of phosphorus starvation to be essential and sufficient for carnivory induction, also under ex vitro conditions. Having established the cultivation of T. peltatum and the induction of carnivory, future gene expression profiles from phosphorus starvation-induced leaves will provide important insight to the molecular mechanism of carnivory on demand.

PREMISE: Carnivorous plants are often associated with nutrient-poor soils and fires. Fire can decrease available soil nitrogen (N) and increase light availability, thus potentially favoring carnivory if prey provide N. Prey can also be a source of phosphorus (P), however, and soil P-availability often increases and competition for prey can decrease following fire. Carnivory thus might be more advantageous before fire when prey and/or soil P are more limiting. METHODS: We examined nutrient limitation of growth in a carnivorous plant, Sarracenia alata, in a wet pine savanna in southeastern Mississippi, USA. We measured growth and N:P tissue concentration responses of adult plants to a factorial arrangement of prey capture, neighbor reduction, and addition of N, P, and ash to the soil. We tested two hypotheses: (1) Prey provide N, and neighbor reduction and ash addition increase light and soil P and thus the benefit of carnivory; and (2) Prey provide P, neighbor reduction increases prey and/or P, and prey exclusion reduces growth the most when neighbors are not reduced. RESULTS: The exclusion of prey reduced growth more when neighbors were not reduced, an effect that was ameliorated slightly by the addition of P to the soil (the P-limitation hypothesis). Prey exclusion caused a decrease in tissue P when N was added to the soil. CONCLUSIONS: The results of this study with adult plants differed from those of a previous study using small juvenile plants, suggesting a shift from light limitation to P and prey limitation with increasing size.

PREMISE: The association of carnivory (an adaptation to nutrient-poor soils) with fire has been described as a paradox, given increases in nutrient availability that often accompany fire. The nutrients that increase in availability following fire, however, may not be the same as those provided by prey and may not reduce nutrient limitation if accompanied by even greater increases in light. METHODS: Using a factorial experiment in the field, we examined how simulated fire (clipping plus nitrogen-free fertilizer addition) and prey-derived nutrient availability (prey exclusion) interacted to influence carnivorous potential in Sarracenia alata and belowground competition with its neighbors (manipulated via trenching). We hypothesized that simulated fire combined with prey exclusion would (1) increase the potential for prey capture relative to shade avoidance, hereafter, relative prey-capture potential (RPCP), and/or (2) increase belowground competition with neighboring plants. RESULTS: Sarracenia alata increased RPCP in response to the combination of simulated fire and prey exclusion, despite increases in phosphorus and other nutrients associated with the simulated fire treatment, suggesting that prey capture potential increases in response to increased nitrogen limitation resulting from increases in light and/or phosphorus after fire. We found no evidence of belowground competition. CONCLUSIONS: The potential importance of carnivory in Sarracenia alata increases following fire. This result helps to explain the paradoxical association of carnivorous plants with fire by demonstrating the potential for prey-derived nutrient limitation to increase rather than decrease in response to increases in light and nutrients other than nitrogen following fire.

Explanations for the Upper Pleistocene extinction of megafauna from Sahul (Australia and New Guinea) remain unresolved. Extinction hypotheses have advanced climate or human-driven scenarios, in spite of over three quarters of Sahul lacking reliable biogeographic or chronologic data. Here we present new megafauna from north-eastern Australia that suffered extinction sometime after 40,100 (±1700) years ago. Megafauna fossils preserved alongside leaves, seeds, pollen and insects, indicate a sclerophyllous forest with heathy understorey that was home to aquatic and terrestrial carnivorous reptiles and megaherbivores, including the world's largest kangaroo. Megafauna species diversity is greater compared to southern sites of similar age, which is contrary to expectations if extinctions followed proposed migration routes for people across Sahul. Our results do not support rapid or synchronous human-mediated continental-wide extinction, or the proposed timing of peak extinction events. Instead, megafauna extinctions coincide with regionally staggered spatio-temporal deterioration in hydroclimate coupled with sustained environmental change.

The present study was conducted to explore the influence of dietary carbohydrate on antioxidant capacity and non-specific immunity of hybrid grouper, which would contribute to determine the tolerable dietary carbohydrate content. Seven diets with grade levels of carbohydrate (5.27, 8.95, 11.49, 14.37, 17.78, 20.82 and 23.65%) were fed to triplicate groups of fish for 10 weeks. Results showed that the inclusion of carbohydrate above 11.49% produced significant increased content of hydrogen peroxide (H2O2) in liver and malondialdehyde (MDA) in both serum and liver. The specific activity of catalase (CAT), superoxide dismutase (SOD), glutathione peroxidase (Gpx) and total antioxidative capacity (T-AOC) were significantly elevated with the increase of dietary carbohydrate from 8.95 to 23.65%, which may be associated with the reduced hepatic soluble protein content. However, opposite variation was observed in the expression of antioxidant related genes (SOD1 and Gpx), which was partly caused by the activation of NF-E2-related factor 2 (Nrf2) and inhibition of Kelch-like-ECH-associated protein 1 (Keap1) at the transcriptional level. The immunoglobulin M (lgM) content and activity of lysozyme and CCP in serum significantly depressed when dietary carbohydrate was above 11.49%. The expression of pro-inflammatory cytokines (TNF-α, IL-1ß and IL-8) was significantly increased with the increase of dietary carbohydrate from 5.27 to 8.95% and thereafter significantly reduced, which was consistent with the changed expression of toll-like receptor 2 (TLR2) and nuclear factor κΒ (NF-κΒ). In above, high dietary carbohydrate significantly impaired the antioxidant capacity and reduced the non-specific immunity of hybrid grouper, and the tolerable dietary carbohydrate content should not exceed 11.49%.

We used three complementary methods to assess the diet of two insectivorous bat species: one an obligate aerial hunter, Miniopterus natalensis, and the other Myotis tricolor whose morphology and taxonomic affiliation to other trawling bats suggests it may be a trawler (capturing insects from the water surface with its feet and tail). We used visual inspection, stable isotope values and fatty acid profiles of insect fragments in bat faeces sampled across five sites to determine the contribution of aquatic and terrestrial arthropods to the diets of the two species. The niche widths of M. tricolor were generally wider than those of Miniopterus natalensis but with much overlap, both taking aquatic and terrestrial insects, albeit in different proportions. The diet of M. tricolor had high proportions of fatty acids (20:5ω3 and 22:6ω3) that are only obtainable from aquatic insects. Furthermore, the diet of M. tricolor had higher proportions of water striders (Gerridae) and whirligig beetles (Gyrinidae), insects obtainable via trawling, than Miniopterus natalensis. These results suggest both species are flexible in their consumption of prey but that M. tricolor may use both aerial hawking and trawling, or at least gleaning, to take insects from water surfaces. The resultant spatial segregation may sufficiently differentiate the niches of the two species, allowing them to co-exist. Furthermore, our results emphasize that using a combination of methods to analyse diets of cryptic animals yields greater insights into animal foraging ecology than any of them on their own.

Crown shape variation of the first lower molar in the arctic (Vulpes lagopus) and red foxes (Vulpes vulpes) was analyzed using five groups of morphotypes. Carnassial morphologies were compared between the species and between spatially and temporally distant populations: one Late Pleistocene (n = 45) and seven modern populations of the arctic fox (n = 259), and one Late Pleistocene (n = 35) and eight modern populations of the red fox (n = 606). The dentition of Holocene red foxes had larger morphotype variability than that of arctic foxes. The lower carnassials of the red fox kept have some primitive characters (additional cusps and stylids, complex shape of transverse cristid), whereas the first lower molars of the arctic fox have undergone crown shape simplification, with the occlusal part of the tooth undergoing a more pronounced adaptation to a more carnivorous diet. From the Late Pleistocene of Belgium to the present days, the arctic fox's crown shape has been simplified and some primitive characters have disappeared. In the red fox chronological changes in the morphology of the lower carnassials were not clearly identified. The phyletic tree based on morphotype carnassial characteristics indicated the distinctiveness of both foxes: in the arctic fox line, the ancient population from Belgium and recent Greenland made separate branches, whereas in the red foxes the ancient population from Belgium was most similar to modern red foxes from Belgium and Italy.

(E)-ß-Farnesene (EßF) is the predominant constituent of the alarm pheromone of most aphid pest species. Moreover, natural enemies of aphids use EßF to locate their aphid prey. Some plant species emit EßF, potentially as a defense against aphids, but field demonstrations are lacking. Here, we present field and laboratory studies of flower defense showing that ladybird beetles are predominantly attracted to young stage-2 pyrethrum flowers that emitted the highest and purest levels of EßF. By contrast, aphids were repelled by EßF emitted by S2 pyrethrum flowers. Although peach aphids can adapt to pyrethrum plants in the laboratory, aphids were not recorded in the field. Pyrethrum's (E)-ß-farnesene synthase (EbFS) gene is strongly expressed in inner cortex tissue surrounding the vascular system of the aphid-preferred flower receptacle and peduncle, leading to elongated cells filled with EßF. Aphids that probe these tissues during settlement encounter and ingest plant EßF, as evidenced by the release in honeydew. These EßF concentrations in honeydew induce aphid alarm responses, suggesting an extra layer of this defense. Collectively, our data elucidate a defensive mimicry in pyrethrum flowers: the developmentally regulated and tissue-specific EßF accumulation and emission both prevents attack by aphids and recruits aphid predators as bodyguards.

Identifying the presence of animals based on faecal deposits in modern and ancient environments is of primary importance to archaeologists, ecologists, forensic scientists, and watershed managers, but it has proven difficult to distinguish faecal material to the species level. Until now, four 5ß-stanols have been deployed as faecal biomarkers to distinguish between omnivores and herbivores, but they cannot distinguish between species. Here we present a database of faecal signatures from ten omnivore and herbivore species based on eleven 5ß-stanol compounds, which enables us to distinguish for the first time the faecal signatures of a wide range of animals. We validated this fingerprinting method by testing it on modern and ancient soil samples containing known faecal inputs and successfully distinguished the signatures of different omnivores and herbivores.

OBJECTIVES: The Early Neolithic involved an important social and economic shift that can be tested not only with the material culture, but also through biomolecular approaches. The Iberian Peninsula presents few Early Neolithic sites where fauna and humans can be analyzed together from an isotopic perspective. Here we present an isotopic study on the site of Cueva de Chaves as an example for understanding the dietary and economical changes that took place during Early Neolithic in Iberia. MATERIAL AND METHODS: Here we apply carbon and nitrogen stable isotope analysis to bone collagen from 4 humans and 64 faunal samples from 14 different species. The large dataset belongs to the same unique chrono-cultural context secured by 20 radiocarbon dates. Three direct new radiocarbon dates were carried out on the human remains analyzed. RESULTS: Faunal isotope values show no significant differences between wild and domestic herbivores, although the latter have more homogeneous values. Domestic pigs, potentially considered omnivorous, also show signatures of a herbivore diet. Human isotopic results show a diet mainly based on terrestrial C3 resources and possibly high meat consumption. The only individual found buried with a special funerary treatment presents a slightly different protein intake, when taking into account the long contemporaneous baseline analyzed. DISCUSSION: Similar values between wild and domestic species could be the result of common feeding resources and/or grazing on the same parts of the landscape. The herbivore diet seen amongst domestic pigs rules out feeding on household leftovers. High meat consumption by humans would support the hypothesis of the existence of a specialized animal husbandry management community in which agriculture was not intensively developed. Our results suggest that the development of agricultural practices and animal husbandry were not necessarily associated together in the early stages of the Western Mediterranean Neolithic.

Recent genomic data have revealed multiple interactions between Neanderthals and modern humans, but there is currently little genetic evidence regarding Neanderthal behaviour, diet, or disease. Here we describe the shotgun-sequencing of ancient DNA from five specimens of Neanderthal calcified dental plaque (calculus) and the characterization of regional differences in Neanderthal ecology. At Spy cave, Belgium, Neanderthal diet was heavily meat based and included woolly rhinoceros and wild sheep (mouflon), characteristic of a steppe environment. In contrast, no meat was detected in the diet of Neanderthals from El Sidrón cave, Spain, and dietary components of mushrooms, pine nuts, and moss reflected forest gathering. Differences in diet were also linked to an overall shift in the oral bacterial community (microbiota) and suggested that meat consumption contributed to substantial variation within Neanderthal microbiota. Evidence for self-medication was detected in an El Sidrón Neanderthal with a dental abscess and a chronic gastrointestinal pathogen (Enterocytozoon bieneusi). Metagenomic data from this individual also contained a nearly complete genome of the archaeal commensal Methanobrevibacter oralis (10.2× depth of coverage)-the oldest draft microbial genome generated to date, at around 48,000 years old. DNA preserved within dental calculus represents a notable source of information about the behaviour and health of ancient hominin specimens, as well as a unique system that is useful for the study of long-term microbial evolution.

Diet is an important modifiable risk factor for cancer. Adequate diet modification may play a key role in reducing the incidence of some cancers. A growing body of epidemiological evidence suggested links of some nutritional exposures with individual cancers. This review updates and summarises the existing data on diet related factors for cancer prevention, evaluated in 2007 by World Cancer Research Fund/American Institute for Cancer Research and identifies the areas where more research is needed. Mechanisms of action of nutrients are discussed. For cancer prevention, more apparent association pertains to the role of foods from plant origin, processed meat products and alcohol. There is a lack of evidence to clarify the relationship of dairy and cereal products, different types of carbohydrates, micronutrients naturally found in foods vs supplements, industrial trans-fats, food preparation and handling techniques and dietary patterns and cancer, in order to implement safe cancer prevention strategies.

Resumen La dieta es un factor de riesgo modificable importante para el cáncer. Una modificación adecuada puede jugar un papel clave en la reducción de la incidencia de algunos cánceres. La evidencia epidemiológica sugiere enlaces de algunas exposiciones nutricionales con cánceres específicos. Esta revisión actualiza y resume los datos existentes sobre factores de la dieta que se relacionan con la prevención del cáncer, que fueron evaluados en 2007 por el World Cancer Research Fund/American Institute for Cancer Research, e identifica áreas para profundizar en investigación. Se discuten mecanismos de acción de los nutrientes. Para la prevención del cáncer, la evidencia epidemiológica se relaciona con los alimentos de origen vegetal, carnes procesadas y alcohol. Se necesita más investigación para aclarar la relación que tienen con el cáncer ciertos alimentos como lácteos, granos, diferentes tipos de carbohidratos, y otros factores que podrían intervenir: micronutrientes presentes en los alimentos contra suplementos, grasas industriales trans, preparación de alimentos y hábitos alimentarios. Esto facilitaría la creación de estrategias seguras de prevención de cáncer.

Recent work has indicated that stoichiometric food quality in terms of the carbon:phosphorus (C:P) ratio affects consumers whether the imbalance involves a deficit or an excess of nutrients; hence, organisms exist on a "stoichiometric knife edge". While previous studies have focused primarily on autotroph-herbivore trophic transfer, nutritional imbalances may also affect the interactions between species at higher trophic levels. Since the foods of carnivores are normally stoichiometrically similar to the body compositions of those carnivores, they may be more severely affected than herbivores if imbalances become pronounced. We analysed the response of the predatory copepod Parabroteas sarsi to monospecific diet treatments consisting of high and low C:P prey items. These dietary treatments strongly affected the predator's elemental composition and growth, although prey selection, excretion, egestion, and respiration rates were not affected. We suggest that, due to their low threshold elemental ratio and a narrow C:P stoichiometric knife edge, these predators are highly vulnerable to stoichiometric imbalances, whether an excess or a deficit of nutrients is involved. Our results demonstrating this high sensitivity to prey C:P ratio show that the stoichiometric knife edge may apply to not only herbivores but also higher trophic levels. Thus, predators such as P. sarsi, with a much narrower range of food quality, may also be strongly affected by fluctuations in the quality of their prey, with negative consequences for their secondary production.

The endogenous production of long-chain polyunsaturated fatty acids (LC-PUFA) in carnivorous teleost species inhabiting freshwater environments is poorly understood. Although a predatory lifestyle could potentially supply sufficient LC-PUFA to satisfy the requirements of these species, the nutrient-poor characteristics of the freshwater food web could impede this advantage. In this study, we report the cloning and functional characterisation of an elongase enzyme in the LC-PUFA biosynthesis pathway from striped snakehead (Channa striata), which is a strict freshwater piscivore that shows high deposition of LC-PUFA in its flesh. We also functionally characterised a previously isolated fatty acyl desaturase cDNA from this species. Results showed that the striped snakehead desaturase is capable of Δ4 and Δ5 desaturation activities, while the elongase showed the characteristics of Elovl5 elongases. Collectively, these findings reveal that striped snakehead exhibits the genetic resources to synthesise docosahexaenoic acid (DHA; 22:6n-3) from eicosapentaenoic acid (EPA; 20:5n-3). Both genes are expressed at considerable levels in the brain and the liver. In liver, both genes were up-regulated by dietary C18 PUFA, although this increase did not correspond to a significant rise in the deposition of muscle LC-PUFA. Brain tissue of fish fed with plant oil diets showed higher expression of fads2 gene compared to fish fed with fish oil-based diet, which could ensure DHA levels remain constant under limited dietary DHA intake. This suggests the importance of DHA production from EPA via the ∆4 desaturation step in order to maintain an optimal reserve of DHA in the neuronal tissues of carnivores.

RESUMO

Indigenous participation in land-based practices such as hunting, fishing, ceremony, and land care has a long history. In recent years, researchers and policy makers have advocated the benefits of these practices for both Indigenous people and the places they live. However, there have also been documented risks associated with participation in these activities. Environmental change brought about by shifts in land use, climate changes, and the accumulation of contaminants in the food chain sit alongside equally rapid shifts in social, economic and cultural circumstances, preferences and practices. To date, the literature has not offered a wide-ranging review of the available cross-disciplinary or cross-ecozone evidence for these intersecting benefits and risks, for both human and environmental health and wellbeing. By utilising hunting as a case study, this paper seeks to fill part of that gap through a transdisciplinary meta-analysis of the international literature exploring the ways in which Indigenous participation in land-based practices and human-environmental health have been studied, where the current gaps are, and how these findings could be used to inform research and policy. The result is an intriguing summary of disparate research that highlights the patchwork of contradictory understandings, and uneven regional emphasis, that have been documented. A new model was subsequently developed that facilitates a more in-depth consideration of these complex issues within local-global scale considerations. These findings challenge the bounded disciplinary and geographic spaces in which much of this work has occurred to date, and opens a dialogue to consider the importance of approaching these issues holistically.

UNLABELLED: BACKROUND AND AIMS: It has been suggested that the rate of net photosynthesis (AN) of carnivorous plants increases in response to prey capture and nutrient uptake; however, data confirming the benefit from carnivory in terms of increased AN are scarce and unclear. The principal aim of our study was to investigate the photosynthetic benefit from prey capture in the carnivorous sundew Drosera capensis. METHODS: Prey attraction experiments were performed, with measurements and visualization of enzyme activities, elemental analysis and pigment quantification together with simultaneous measurements of gas exchange and chlorophyll a fluorescence in D. capensis in response to feeding with fruit flies (Drosophila melanogaster). KEY RESULTS: Red coloration of tentacles did not act as a signal to attract fruit flies onto the traps. Phosphatase, phophodiesterase and protease activities were induced 24 h after prey capture. These activities are consistent with the depletion of phosphorus and nitrogen from digested prey and a significant increase in their content in leaf tissue after 10 weeks. Mechanical stimulation of tentacle glands alone was not sufficient to induce proteolytic activity. Activities of ß-D-glucosidases and N-acetyl-ß-D-glucosaminidases in the tentacle mucilage were not detected. The uptake of phosphorus from prey was more efficient than that of nitrogen and caused the foliar N:P ratio to decrease; the contents of other elements (K, Ca, Mg) decreased slightly in fed plants. Increased foliar N and P contents resulted in a significant increase in the aboveground plant biomass, the number of leaves and chlorophyll content as well as AN, maximum quantum yield (Fv/Fm) and effective photochemical quantum yield of photosystem II (ΦPSII). CONCLUSIONS: According to the stoichiometric relationships among different nutrients, the growth of unfed D. capensis plants was P-limited. This P-limitation was markedly alleviated by feeding on fruit flies and resulted in improved plant nutrient status and photosynthetic performance. This study supports the original cost/benefit model proposed by T. Givnish almost 30 years ago and underlines the importance of plant carnivory for increasing phosphorus, and thereby photosynthesis.

We evaluated the levels of selenium and mercury in five fish species commonly eaten by local populations of the Brazilian Amazon. Fish specimens were sampled in two lotic and three lentic areas at two different phases of the hydrological cycle. Analyses of Carbon and Nitrogen stable isotopes allowed us to confirm the trophic levels of the fish species (one herbivorous, two omnivorous and two piscivorous) and verify that these levels remained unchanged with the habitats and the season. The levels of selenium and mercury in fish varied from 50ng/g to 1006ng/g and from 17ng/g to 3502ng/g respectively. For both seasons, fish from lotic ecosystems presented higher selenium concentrations. An inverse pattern was observed between selenium and mercury concentrations within the trophic chain, and this in both seasons. Indeed, the highest mean concentrations of selenium and lowest mean concentrations of mercury were measured in the herbivorous species and the opposite in the piscivorous species. Our results unequivocally demonstrate that local riverside populations will maximize the selenium health benefits of eating fish while minimizing their risk of being chronically exposed to mercury by preferentially consuming herbivorous species and to some extent omnivorous species, while avoiding piscivorous species.

Prior to hibernation, mammals accumulate large amounts of fat in their bodies. In temperate mammalian species, hibernation is improved by increasing the levels of poly-unsaturated fatty acids (PUFA) in the body. The saturation of fatty acids (FA) in both white adipose tissue (WAT) and membrane phospholipids of mammals often reflects their diet composition. We found that the greater mouse-tailed bat (Rhinopoma microphyllum) accumulates large amounts of fat at the end of summer by gradually shifting to a fat-rich diet (queen carpenter ants, Camponotus felah). PUFA are almost absent in this diet (<1 % of total FA), which contains a high fraction of saturated (SFA) and mono-unsaturated (MUFA) fatty acids. We found similar low levels of PUFA in mouse-tailed bat WAT, but not in their heart total lipids. The expression of two appetite-stimulating (orexigenic) hypothalamic neuropeptides, AgRP and NPY, increased in parallel to the shift in diet and with fat gain in these bats. To the best of our knowledge, this is the only documented example of specific pre-hibernation diet in bats, and one which reveals the most saturated FA composition ever documented in a mammal. We suggest that the increase in expression levels of NPY and AgRP may contribute to the observed diet shift and mass gain, and that the FA composition of the bat's specialized diet is adaptive in the relatively high temperatures we recorded in both their winter and summer roosts.
