RESUMO

Infectious Bursal Disease (IBD) is an economically important, immunosuppressive viral disease of chicken. Withania somnifera, a well-known Indian medicinal plant and functional food, finds extensive ethnomedicinal and ethnoveterinary use in the subcontinent. Root extracts of Withania somnifera have been shown to inhibit IBD virus (IBDV) in vitro. The effect of dietary supplementation with whole root powder of Withania somnifera was studied in chicken experimentally infected with IBDV. Dietary supplementation with the root powder improved erythrocytic indices, biochemical parameters, bursal weight index, and lymphocyte stimulation indices, and reduced histopathological insult in the infected birds. Viral load decreased to less than one-fourth in the birds receiving dietary supplementation with Withania somnifera root powder. It could be concluded that continued supplementation of IBDV-infected chicken with Withania somnifera root powder alleviated virus-induced stress and histological and immunological alterations and reduced viral persistence in the host.

Infectious bursal disease (IBD), caused by infectious bursal disease virus (IBDV), is an immunosuppressive infectious disease of global economic importance in poultry. This study was designed to evaluate the effect of oral administration of ginseng stem-leaf saponins (GSLS) on humoral and gut mucosal immunity in chickens vaccinated with live IBDV vaccine, and furthermore, to test its protective efficacy against virulent IBDV challenge following vaccination. In experiment 1, chickens were orally administered with GSLS at 5 mg/kg of BW for 7 d, and then immunized with live IBDV vaccine via the oral route. Serum was sampled on 0, 1, 2, 3, 4, and 5 wk postvaccination for detecting antibody titers by ELISA, and intestinal tissues were collected on 0, 1, 3, and 5 wk postvaccination for measurement of IgA-positive cells and intestinal intraepithelial lymphocytes by immunohistochemical and hematoxylin-eosin staining, respectively. Result showed that antibody titers, IgA-positive cells and intestinal intraepithelial lymphocytes were significantly higher in chickens drinking GSLS than the control, suggesting an enhanced effect of GSLS on humoral and gut mucosal immune responses. In experiment 2, chickens were delivered with GSLS and then vaccinated in the same way as in experiment 1. The birds were challenged with virulent IBDV at wk 3 postvaccination. Then the birds were weighed, bled, and necropsied at d 3 postchallenge and the bursae were sampled for gross and histopathological examination. Results demonstrated that GSLS provided a better protection against virulent IBDV challenge following vaccination than the control. In conclusion, oral administration of GSLS enhances both humoral and gut mucosal immune responses to IBDV and offers a better protection against virulent IBDV challenge. Considering its immunomodulatory properties to IBDV vaccine, GSLS might be a promising oral adjuvant for vaccination against infectious diseases in poultry.

This study was carried out to investigate the modulatory effects of dietary methionine and n-6/n-3 polyunsaturated fatty acids (PUFA) ratio on immune response and performance of infectious bursal disease (IBD)-challenged broiler chickens. In total, 350 one-day-old male broiler chicks were assigned to 1 of the 6 dietary treatment groups in a 3 × 2 factorial arrangement. There were 3 n-6/n-3 PUFA ratios (45, 5.5, and 1.5) and 2 levels of methionine (NRC recommendation and twice NRC recommendation). The results showed that birds fed with dietary n-6/n-3 PUFA ratio of 5.5 had higher BW, lower feed intake, and superior FCR than other groups. However, the highest antibody response was observed in birds with dietary n-6/n-3 PUFA ratio of 1.5. Lowering n-6/n-3 PUFA ratio reduced bursa lesion score equally in birds fed with n-6/n-3 PUFA ratio of 5.5 and 1.5. Supplementation of methionine by twice the recommendation also improved FCR and reduced feed intake and bursa lesion score. However, in this study, the optimum performance (as measured by BW, feed intake, and FCR) did not coincide with the optimum immune response (as measured by antibody titer). It seems that dietary n-3 PUFA modulates the broiler chicken performance and immune response in a dose-dependent but nonlinear manner. Therefore, it can be suggested that a balance of moderate level of dietary n-6/n-3 PUFA ratio (5.5) and methionine level (twice recommendation) might enhance immune response together with performance in IBD-challenged broiler chickens.

BACKGROUND: Infectious bursal disease (IBD) results in economic loss due to mortality, reduction in production efficiency and increasing the usage of antibiotics. This study was carried out to investigate the modulatory roles of dietary n-3 polyunsaturated fatty acids (PUFA) enrichment in immune response and performance of IBD challenged broiler chickens. METHODS: A total of 300 day old male broiler chicks were assigned to four dietary n-3 PUFA ascending levels as the treatment groups (T1: 0.5; T2: 8.0; T3: 11.5; T4: 16.5) using combinations of tuna oil and sunflower oil. All diets were isocaloric and isonitrogenous. On day 28, all birds were challenged with IBD virus. Antibody titer, cytokine production, bursa lesion pre and post-challenge and lymphoid organ weight were recorded. RESULTS: On d 42 the highest body weight was observed in the T2 and T3 and the lowest in T4 chickens. Feed conversion ratio of the T2 broilers was significantly better than the other groups. Although productive parameters were not responded to the dietary n-3 PUFA in a dose-dependent manner, spleen weight, IBD and Newcastle disease antibody titers and IL-2 and IFN-γ concentrations were constantly elevated by n-3 PUFA enrichment. CONCLUSIONS: Dietary n-3 PUFA enrichment may improve the immune response and IBD resistance, but the optimum performance does not coincide with the optimum immune response. It seems that dietary n-3 PUFA modulates the broiler chicken performance and immune response in a dose-dependent manner. Thus, a moderate level of dietary n-3 PUFA enrichment may help to put together the efficiency of performance and relative immune response enhancement in broiler chickens.

The immunoregulatory actions of conjugated linoleic acid (CLA) of relevance to viral disease pathogenesis and immune responses were investigated. To test the hypothesis that CLA ameliorates immunosuppression, we developed a viral challenge model by infecting chickens with infectious bursal disease virus (IBDV). After 14 d of dietary supplementation with either soybean oil or CLA, half of the chickens in each group were challenged with IBDV. We examined the effect of CLA on the development of lesions (i.e., lymphoid depletion and necrosis) and observed the immune responses against IBDV. The IBDV infection depleted lymphocytes in the medullary area and significantly stimulated interferon (IFN)-γ and IL-6 mRNA relative expression of bursa (P < 0.05) compared with the uninfected bursa. Compared with the CLA diet, lymphocytes depletion was more accentuated in chickens fed the control diet, whereas IFN-γ and IL-6 mRNA relative expression were upregulated (P < 0.05). Additionally, histopathological examination of the bursa revealed that the pathological changes tended to be more severe in infected chickens fed the control diet, which also significantly decreased (P < 0.05) on lymphocyte proliferation. Significant interactions were found between infection and diets for lymphocyte proliferation, antibody titers, and IFN-γ mRNA relative expression (P < 0.05). The results of this study indicate that dietary CLA enhanced immune function in chickens, particularly those of the IBDV-immunosuppressive status. Furthermore, at the molecular level, the immunoregulatory functions of CLA on chickens are attributable mainly to the antiinflammatory properties of CLA and are mediated, at least in part, through suppressing IBDV-specific proinflammatory cytokines mRNA relative expression.

BACKGROUND: A large number of plants and their isolates have been shown to potentiate immunity. Some plants exert anti-inflammatory and anti-stress effects, others hepatoprotective activity. In this study, 320 1-day-old broiler chicks were randomly divided into four major groups A, B, C and D and fed rations supplemented with 0, 15, 20 and 22.5 g Berberis lycium kg⁻¹ ration respectively. Each group was further divided into two subgroups, one vaccinated against Newcastle disease (ND) and infectious bursal disease (IBD), the other non-vaccinated. Antibody titre against IBD and ND, relative weight of lymphoid organs, post-challenge morbidity and mortality, serum hepatic enzymes and total serum protein were observed. RESULTS: Group C had higher anti-IBD and anti-ND antibody titres. Relative bursa weight in groups C and D was higher until day 28, but birds in group C performed better at later stages of examination. Relative spleen weight was highest in group C. During initial stages there was no effect on relative thymus weight, but at later stages the effect was significant. Groups C and D performed similarly in terms of relative thymus weight. The birds were challenged to field IBD through intramuscular injection at a dose rate of 0.5 mL per bird. Post-challenge morbidity was lowest in groups C and D, while treatment significantly (P < 0.001) affected mortality amongst affected (morbid) birds. Levels of serum alanine aminotransferase and alkaline phosphatase were lowest in group C. Serum protein was similar in all groups and in both vaccinated and non-vaccinated broiler chicks. CONCLUSION: Berberis lycium added to feed at 20 g kg⁻¹ is effective in improving immunity against ND and IBD as well as liver function in broiler chicks.

Chicken embryo fibroblast (CEF) is a primary cellular material to research the infectious bursal disease virus (IBDV). Constructing the cDNA expression library of CEF is the foundation to research cell tropism and find cell receptors of IBDV from CEF. In order to achieve that purpose, a high-quality cDNA expression library of CEF was constructed by Gateway technology, which could avoid using the restriction enzyme for cloning to solve technical limitation of roution method. The mRNA was extracted from chicken embryonic fibroblast. Moreover, single-strand cDNA and double-strand cDNA were synthesized by using biotin-conjugated Oligo (dT) primer in turn. The double-strand cDNA was ligated Adapter and then purified by the cDNA Size Fractionation Columns. After BP recombination reaction, a cDNA entry library was constructed with a titer of 1 x 10(6) cfu/mL, total clones of 1.2 x 10(7) cfu and an average insertion size of about 2243 bp. After LR recombination reaction, the cDNA entry library was transformed into expression library which took on a titer of 5 x 10(5) cfu/mL, total clones of 5.5 x 10(6) cfu and an average insertion size of about 2411bp. The results indicate that the constructed cDNA expression library performs a remarkable high value in both recombination rate and library coverage. As a result, the cDNA expression library, with its good quality, may facilitate to identify the receptors associated with the resistance against IBDV in chicken embryonic fibroblast and to cast new light on the mechanism of cellular tropism. Moreover, it may also provide data of chicken embryonic fibroblast in transcription level and may be helpful to study its biological functions.

Addition of 5% freeze-dried IBD virus (IBDV)-immune bovine colostrum to the diet of chickens prevented infection when housed in an IBDV-contaminated environment. Four of five chickens receiving 2.5%, and all chickens receiving 0.5% or no IBDV-immune colostrum were infected. No interference with development of an active immunity was found in chickens receiving 5% IBDV-immune colostrum in their diet and vaccinated intramuscularly with modified IBDV.
