BACKGROUND: Sea lamprey (Petromyzon marinus) is a diadromous fish compromised by various stressors, which can lead to population decline and the urgency of stronger conservation regulation. In the absence of documentation of direct knowledge of local populations, a broader zoological and ecological understanding of sea lamprey fishing has become vital for the preservation of traditional practices and conservation of this migratory fish. To this purpose, we collected data from the P. marinus about the artisanal fisheries profile, folk taxonomy, habitat, reproduction, migration, and displacement using a low-cost methodology, through ethnobiology tools, in the four riverine fishing villages in Portugal. METHODS: A total of 40 semi-structured interviews were carried out during the winter of 2019 in crucial fishing villages in the Minho river. Fishers were selected by random sampling and the snowball technique when appropriate. Interviews applied contained four parts (fisher's profile, projective test, knowledge about fishing, and ethnozoological knowledge about the sea lamprey). Informal knowledge was analyzed following an emic-etic approach and the set-theoretical Union of all individual competences. The Code of Ethics of the International Society of Ethnobiology (ISE) was the main parameter for the conduction of this ethnozoological research and related activities in the Cooperminho project. RESULTS: This first ethnobiological study of the sea lamprey (Petromyzon marinus) in Portugal showed a sample of predominantly male fishers, averaging 57.13 years old, and average fishing experience of 37.18 years. The average income of fishers is about 688.28 Euros, and the level of education was predominantly basic. Data from artisanal fisheries showed the time and frequency of fishing, the characterization of fishing boats, and general information on catching lamprey in the Minho river. Three new folk names were attributed to P. marinus. Fishers mentioned sites with rock fragments and sandy bottoms and depth ranges ranging from 0 to 8 m as likely sea lamprey habitats. The villages of Monção and Melgaço are the last areas of the river where you could spot sea lamprey, as well as the last probable spawning grounds for this fish in the Minho river. The hydroelectric dams and predatory fisheries were considered the main obstacles to the migration of sea lamprey. Finally, local fishers also shared the lamprey migration season to feed and spawn. CONCLUSIONS: Fishers shared a vast informal knowledge of sea lamprey zoology and ecology typical of anadromous species of the Petromyzontidae family, in the central traditional Portuguese communities on the Minho river. This fisher's knowledge becomes essential to preserve cultural practices of the sea lamprey, which is currently highly susceptible to anthropogenic pressures. Given the real warning of population extinction in the Portuguese rivers (such as the Minho river) and a similar trend in Spanish territory, ethnozoological studies of sea lamprey in Spanish fishing communities may support our findings. Also, this study may assist in the adaptive participatory management of these anadromous fish, as well as in documentation of local ecological knowledge (LEK) and centuries-old fishing practices that are also vulnerable in modern times on the international frontier Minho river.

A central and long-standing tenet in the conceptualization of animal swimming is the idea that propulsive thrust is generated by pushing the surrounding water rearward. Inherent in this perspective is the assumption that locomotion involves the generation of locally elevated pressures in the fluid to achieve the expected downstream push of the surrounding water mass. Here we show that rather than pushing against the surrounding fluid, efficient swimming animals primarily pull themselves through the water via suction. This distinction is manifested in dominant low-pressure regions generated in the fluid surrounding the animal body, which are observed by using particle image velocimetry and a pressure calculation algorithm applied to freely swimming lampreys and jellyfish. These results suggest a rethinking of the evolutionary adaptations observed in swimming animals as well as the mechanistic basis for bio-inspired and biomimetic engineered vehicles.

Glutamate is the major excitatory neurotransmitter in vertebrates, and glutamatergic cells probably represent a majority of neurons in the brain. Physiological studies have demonstrated a wide presence of excitatory (glutamatergic) neurons in lampreys. The present in situ hybridization study with probes for the lamprey vesicular glutamate transporter (VGLUT) provides an anatomical basis for the general distribution and precise localization of glutamatergic neurons in the sea lamprey brainstem. Most glutamatergic neurons were found within the periventricular gray layer throughout the brainstem, with the following regions being of particular interest: the optic tectum, torus semicircularis, isthmus, dorsal and medial nuclei of the octavolateral area, dorsal column nucleus, solitary tract nucleus, motoneurons, and reticular formation. The reticular population revealed a high degree of cellular heterogeneity including small, medium-sized, large, and giant glutamatergic neurons. We also combined glutamate immunohistochemistry with neuronal tract-tracing methods or γ-aminobutyric acid (GABA) immunohistochemistry to better characterize the glutamatergic populations. Injection of Neurobiotin into the spinal cord revealed that retrogradely labeled small and medium-sized cells of some reticulospinal-projecting groups were often glutamate-immunoreactive, mostly in the hindbrain. In contrast, the large and giant glutamatergic reticulospinal perikarya mostly lacked glutamate immunoreactivity. These results indicate that glutamate immunoreactivity did not reveal the entire set of glutamatergic populations. Some spinal-projecting octaval populations lacked both VGLUT and glutamate. As regards GABA and glutamate, their distribution was largely complementary, but colocalization of glutamate and GABA was observed in some small neurons, suggesting that glutamate immunohistochemistry might also detect non-glutamatergic cells or neurons that co-release both GABA and glutamate.

We used Neurobiotin as a retrograde tract tracer in both larval and adult sea lampreys and observed a number of neuronal brainstem populations (mainly reticular and octaval populations and some diencephalic nuclei) that project to the spinal cord, in agreement with the results of previous tracer studies. We also observed small labeled neurons in the ventral hypothalamus, the mammillary region, and the paratubercular nucleus, nuclei that were not reported as spinal projecting. Notably, most of the labeled cells of the mammillary region and some of the ventral hypothalamus were cerebrospinal fluid-contacting (CSF-c) neurons. Combined tract tracing and immunocytochemistry showed that some of the labeled neurons of the mammillary and paratubercular nuclei were dopamine immunoreactive. In addition, some CSF-c cells were labeled in the caudal rhombencephalon and rostral spinal cord, and many were also dopamine immunoreactive. Results with other tracers (biotinylated dextran amines, horseradish peroxidase, and the carbocyanine dye DiI) also demonstrated that the molecular weight or the molecular nature of the tracer was determinant in revealing diencephalic cells with very thin axons. The results show that descending systems afferent to the spinal cord in lampreys are more varied than previously reported, and reveal a descending projection from CSF-c cells, which is unknown in vertebrates. The present results also reveal the existence of large differences between agnathans and gnathostomes in the organization of the dopaminergic cells that project to the spinal cord.
