The purpose of this study was to investigate the effects of adding Pennisetum purpureum (P. purpureum, also known as Napier grass or elephant grass) to the diets of late gestation on the antioxidant indexes, immune indexes and faecal microbiota of sows. At the 90 days of gestation, 300 healthy sows were randomly divided into three groups, and they received the basic commercial diet or added 5% P. purpureum and 10% P. purpureum, respectively. The experiment started from 90 days of gestation to parturition. The results showed that the total antioxidant capacity, immunoglobulins and serum equol concentrations of sows on 100 days of gestation and at parturition increased linearly (p < .05) with the increase of the content of P. purpureum in the gestation diet. The 5% P. purpureum increased the relative abundance of Bacteroidetes (p = .027) and Actinobacteria (p < .001) at phylum level, Coriobacteriaceae (p < .001) at family level and Prevotellaceae_UCG_001 (p = .004) at genus level, and decreased the relative abundance of Escherichia_Shigella (p < .001) at genus level. In summary, this study shows that the additive of P. purpureum can increase the concentration of serum equol, improve the antioxidant capacity and immune function of sow in late gestation. In addition, the additive of 5% P. purpureum in the diet might change the composition of intestinal microbiota of sows, particularly the relative abundance of Coriobacteriaceae (p < .001) increased.

In this study there was evaluation of effects of dietary inulin during late gestation on sow physiology, farrowing duration and piglet performance. At day 80 of gestation sows were randomly assigned to four groups:basal diet (CON); or basal diet with 0.8 %; 1.6 %; or 2.4 % inulin. The feeding of the diet with 1.6 % inulin resulted in larger weights of the litter at birth a shorter duration of the farrowing period, lesser average birth interval between piglets, lesser number of piglets dead at birth, and fewer piglets/sow dead at birth (P < 0.05). When sows were fed 0.8 % and 1.6 % IN, there was a larger litter weight at weaning, sow average daily feed intake and piglet average daily gain during lactation compared with values for these variables in the CON group (P <  0.05). Additionally, there was an increase in serum concentration of free fatty acid, total cholesterol, and high-density lipoprotein cholesterol with increasing amounts of inulin in the diet (linear, P <  0.05). Sows fed 1.6 % IN had greater serum concentrations of glucose than those in the CON group (P <  0.05). Furthermore, there was a linear increase in serum activity of total antioxidant capacity, total superoxide dismutase and glutathione peroxidase with increasing amounts of inulin in the diet (P <  0.05). In conclusion, results of the present study indicated feeding inulin during late gestation improved reproductive performance of sows, thus, may be a novel additive for the pig industry in improving efficiency of pork production.

The early life period is crucial for the maturation of the intestinal barrier, its immune system, and a life-long beneficial host-microbiota interaction. The study aims to assess the impact of a beneficial dietary (short-chain fructooligosaccharides, scFOS) supplementation vs. a detrimental dietary environment (such as mycotoxin deoxynivalenol, DON) on offspring intestinal immune system developmental profiles. Sows were given scFOS-supplemented or DON-contaminated diets during the last 4 weeks of gestation, whereas force-feeding piglets with DON was performed during the first week of offspring life. Intestinal antigen-presenting cell (APC) subset frequency was analyzed by flow cytometry in the Peyer's patches and in lamina propria and the responsiveness of intestinal explants to toll-like receptor (TLR) ligands was performed using ELISA and qRT-PCR from post-natal day (PND) 10 until PND90. Perinatal exposure with scFOS did not affect the ontogenesis of APC. While it early induced inflammatory responses in piglets, scFOS further promoted the T regulatory response after TLR activation. Sow and piglet DON contamination decreased CD16+ MHCII+ APC at PND10 in lamina propria associated with IFNγ inflammation and impairment of Treg response. Our study demonstrated that maternal prebiotic supplementation and mycotoxin contamination can modulate the mucosal immune system responsiveness of offspring through different pathways.

AIM: Chicory fibre (CF) is rich in fructan, which always functions as a quality dietary fibre source during mammalian pregnancy; however, its effect on reproductive performance remains unclear. METHODS AND RESULTS: 40 pregnant SD rats were randomly allotted to receive one of four diets: basal diet (control group), basal diet + 5% CF, basal diet + 10% CF, and basal diet + 15% CF, respectively. We found that CF significantly increased the number born alive and total litter birth weight (P < 0·05), increased the expression of intestinal tight junction proteins, mucins and antimicrobial peptides, accompanied by the increase of villi height and the decrease of crypts depth of pregnant SD rats (P < 0·05). We also observed that CF markedly increased the acetic acid, propanoic acid, butyric acid and total SCFAs concentrations in caecum contents and promoted the expression of SCFAs-related receptors (P < 0·05). Notably, rats fed CF increased the relative abundance of Bacteroidetes (P < 0·001), decreased the relative abundance of Firmicutes and Proteobacteria, while markedly lowered the Firmicutes/ Bacteroidetes ratio (F/B ratio) (P < 0·05). Intriguingly, the number born alive and total litter birth weight were positively correlated with some probiotics and negatively correlated with other harmful bacteria by Pearson correlation analysis. CONCLUSION: Collectively, CF can enhance intestinal barrier function and maintain intestinal health, and may improve reproductive performance by altering intestinal microbiota composition. SIGNIFICANCE AND IMPACT OF THE STUDY: Adding suitable dietary fibre to the diet can improve the reproductive performance of sows. Indeed, there exist various problems in the application of traditional dietary fibres, including high insoluble fibre content and anti-nutritional factor level, and mycotoxin contamination. This study demonstrates that dietary CF supplementation improves reproductive performance and intestinal health. Thus, CF can be applied in pregnancy animals as a new dietary fibre additive in animal husbandry.

Progesterone (P4) plays a key role in pregnancy establishment and maintenance; during early pregnancy, P4 stimulates the production and release of uterine secretions necessary for conceptus growth prior to implantation; therefore, exogenous P4 supplementation may improve embryo development. This study evaluated the effects of supplementation during early pregnancy with long-acting injectable progesterone or altrenogest on embryonic characteristics of sows and gilts. Thus, a total of 32 sows and 16 gilts were used. On day 6 of pregnancy sows and gilts were allocated to one of the following groups: non-supplemented; supplemented with 20 mg of altrenogest, orally, from days 6 to 12 of pregnancy; supplemented with 2.15 mg/kg of long-acting injectable progesterone on day 6 of pregnancy. Animals were killed on day 28 of pregnancy, and ovulation rate, embryo survival, embryo weight, crown-to-rump length, uterine glandular epithelium and endometrial vascularization were assessed. Treatments had no effect on pregnancy rate, embryo survival or endometrial vascular density (P > 0.05). Non-supplemented gilts presented larger and heavier embryos compared to gilts from supplemented groups (P < 0.05). Sows in the altrenogest group presented larger and heavier embryos compared to non-supplemented sows and sows supplemented with long-acting injectable progesterone. In conclusion, supplementation of sows and gilts with progestagen from day 6 of pregnancy can be used as a means to improve embryo survival without deleterious effects.

Background: Nonruminant male and female offspring respond differently to gestational nutrition, with placenta contributing to the underlying mechanisms. However, similar data are lacking in large ruminants. Objectives: The aim of this study was to investigate the impact of methionine supply during late-gestation on metabolism and DNA methylation in placenta from cows carrying male or female calves. Methods: During the last 28 d of pregnancy, cows were individually fed a control diet (CON) or the control diet plus rumen-protected d,l-methionine (MET; 0.9 g/kg dry matter intake). Placentomes collected at term were classified according to cow dietary treatment and offspring sex as follows: Male CON (n = 7), Male MET (n = 7), Female CON (n = 8), and Female MET (n = 8). Calf growth was measured until 9 wk of age. Targeted metabolomics, RT-PCR, global DNA methylation, and activity of selected enzymes in one-carbon metabolism and transsulfuration pathways were performed. Statistics were conducted via ANOVA using MIXED models. Results: At birth, Male MET calves were heavier than Male CON calves (7.6%, P = 0.02), but body mass was similar at 9 wk of age. In contrast, compared with Female CON, Female MET calves had greater body mass at 9 wk of age (6.3%, P = 0.03). Compared with Male CON, placenta from Male MET calves had greater concentrations of tricarboxylic acid (TCA) cycle and transsulfuration intermediates (23-100%, P < 0.05), along with greater 5-methyltetrahydrofolatehomocysteine methyltransferase activity (67%, P = 0.03). Compared with Female CON, placenta from Female MET calves had greater concentrations of one-carbon metabolism intermediates (13-52%, P < 0.05). DNA methyltransferase 3A (DNMT3A) was upregulated (43%, P < 0.01) in placenta from Female MET compared with Female CON calves. Global DNA methylation was lower in placenta from Female MET compared with Female CON calves (45%, P = 0.06). Conclusions: Methionine supply affects placental metabolism, DNA methylation, and body mass of the calf in a sex-specific manner, underscoring its importance as dietary methyl-donor for pregnant cows.

Sheep production systems in Brazilian caatinga rangelands require supplementation adapted to changes in floristic and chemical composition as dry seasons progress. Meeting sheep nutritional needs in extensive semiarid systems is challenging because of sheep dietary preferences and habits. The objective of this trial was to evaluate the substitutive effect of concentrate supplementation on grazing sheep in the Brazilian caatinga rangeland and its consequences on performance in different seasons. The trial was conducted from March to August 2013 at Embrapa Goat and Sheep in Sobral, Ceará State, Brazil. Thirty-two Brazilian Somali multiparous ewes were submitted to estrus synchronization and controlled breeding. At the start of the trial, ewes averaged 30.45 + 2.60 kg body weight (BW). Ewes were divided into four groups and individually offered 0, 200, 350, or 500 g supplement head-1 day-1. Intake prediction and digestibility trials were evaluated at three periods: rainy season (April), transition rainy-dry (June), and dry season (August). Sheep weights were taken every 14 days to measure their performance from late gestation until weaning. Ewe BW and body condition score changes were determined too. Lamb BW changes were also measured every 14 days from birth through weaning. A completely randomized design with split plot arrangement using eight replications was used for intake and digestibility measurements. The differences between supplement offered (0, 200, 350, and 500 g sheep-1) and season (rainy, transition rainy-dry, and dry) were submitted to analyses of variance and multiple means were separated, where differences were detected, using the Tukey's test. During lactation up through weaning, ewes supplemented at 500 g day-1 had greater BW than ewes without supplement. Ewes supplemented with 200 g concentrate head-1 day-1 had 9.1% greater (P ≤ 0.05) BW at weaning and their lambs had 19.7% greater birth and 16.6% heavier wean BW despite lower dry matter intake compared to unsupplemented animals. Supplementation with 200 g concentrate increased carrying capacity by 28.8% during the dry season and by 20.5% during the rainy season. This study confirmed that in the dry season, when quality of rangeland forages decreases, supplementation contributes to greater DMI, improves postpartum and lactation BW recovery of ewes, and contributes to greater lamb birth and wean weights. Moreover, supplementation leads to feed substitution effects that may increase rangeland resilience by mitigating overgrazing. Supplementation with 200 g concentrate can also prevent negative energy balance for grazing animals, improving longer postnatal recovery, longer intervals between parturitions, fewer double and is necessary to ensure a better BW at birth to lambs.

Our research group investigated the impacts of supplementing Ca salts of soybean oil (CSSO), a source of omega-6 fatty acids (FAs), on reproductive performance of beef cows. Initial studies were conducted with Nelore (Bos indicus) cows grazing tropical pastures. Cows were assigned to fixed-time artificial insemination (AI) and supplemented or not with 100 g/d (as-fed basis) of CSSO, and supplementation regimens ranged from days -11 to 28 relative to AI. Overall, CSSO supplementation during the 21 d after AI increased (P < 0.01) pregnancy rates from 38.1% (623/1,635 as pregnant/total nonsupplemented cows) to 49.0% (843/1,720 as pregnant/total CSSO-supplemented cows), and these outcomes were associated with enhanced early embryonic development and pregnancy establishment when omega-6 FA were supplemented. To verify this rationale, our group compared FA incorporation in grazing Nelore cows (n = 90) supplemented or not with CSSO (100 g/d; as-fed basis) beginning at fixed-time AI until slaughter at day 19 of gestation. Supplementing CSSO increased (P ≤ 0.05) incorporation of linoleic acid and its omega-6 derivatives in plasma, endometrium, corpus luteum, and conceptus, whereas the same responses were not observed (P ≥ 0.25) for omega-3 FA. Complementing these findings, grazing Nelore cows (n = 100) were supplemented or not with CSSO (100 g/d; as-fed basis) beginning at fixed-time AI, and assigned to transcervical uterine flush on day 15 of gestation. Supplementing CSSO increased (P ≤ 0.04) conceptus length (2.58 vs. 1.15 cm) and mRNA expression of interferon-tau (4.1-fold increase) and prostaglandin E synthase 2 (2.6-fold increase), which are critical regulators of pregnancy establishment. These outcomes were recently replicated in B. taurus beef cows consuming temperate forages. Pregnancy rates were greater (P = 0.01) in Angus cows receiving CSSO (100 g/d; as-fed basis) for 21 d after fixed-time AI (60.2%; 226/383 as pregnant/total cows) compared with nonsupplemented cows (51.7%; 193/388 as pregnant/total cows). Supplementing CSSO to Angus × Hereford cows (n = 96) beginning after AI also increased (P = 0.05) mRNA expression of interferon-tau in day 15 conceptuses (1.8-fold increase). Collectively, our research demonstrated that post-AI CSSO supplementation favors incorporation of omega-6 FA into maternal and embryonic tissues, which enhances interferon-tau synthesis by the conceptus and increases pregnancy rates to fixed-time AI in B. indicus and B. taurus beef cows.

A 3 yr study evaluated the effects of three preweaning injections of bovine ST, administered 14 d apart, on growth and reproductive performance of Bos indicus-influenced beef heifers. On d 0 of each year, suckling Angus × Brangus heifers (n = 15 heifers/treatment/yr) were stratified by BW (147 ± 20 kg) and age (134 ± 11 d) and randomly assigned to receive an s.c. injection of saline (SAL; 5 mL; 0.9% NaCl) or 250 mg of sometribove zinc (BST; Posilac, Elanco, Greenfield, IN) on d 0, 14, and 28. Heifers and respective dams were managed as a single group on bahiagrass (Paspalum notatum) pastures from d 0 until weaning (d 127). From d 127 to 346, heifers were grouped by treatment, allocated to bahiagrass pastures (1 pasture/treatment/yr) and fed a molasses-based supplement (2.9 kg/heifer daily; DM basis) until d 346. Blood samples were collected on d 0, 14, 28, 42, and then every 9-10 d from d 179 to 346. In yr 3, liver biopsy samples were collected on d 0, 42, and 263. Heifers were exposed to mature Angus bulls from d 263 to 346. Growth performance and physiological parameters were analyzed using the MIXED procedure, whereas reproductive variables were analyzed using the GLIMMIX procedure of SAS. Effects of treatment × year and treatment × year × time were not detected for any variable measured in this study (P ≥ 0.14), except for calving percentage (P = 0.03). Heifers assigned to BST injections had greater overall plasma concentrations of IGF-1 and ADG from d 0 to 42 (P ≤ 0.05), less ADG from d 42 to 127 (P = 0.04), but had similar BW at weaning and postweaning ADG (P ≥ 0.25) compared to SAL heifers. Heifers assigned to BST tended to achieve puberty 26 d earlier (P = 0.10), had greater percentage of pubertal heifers on d 244, 263, 284, and 296 (P ≤ 0.04), tended to have greater overall pregnancy percentage (P = 0.10), and had greater (P ≤ 0.05) calving percentages in yr 1 and 2 (but not yr 3; P = 0.68) compared to SAL heifers. Liver mRNA expression of GHR-1B and IGF-1 on d 0 and 42 did not differ between treatments (P ≥ 0.15), but was greater for BST vs. SAL heifers on d 263 (P ≤ 0.02). Hence, administering three injections containing 250 mg of sometribove zinc at 14 d intervals before weaning (between 135 and 163 d of age) induced long-term impacts on liver gene expression and may be a feasible management practice to enhance puberty and pregnancy attainment in B. indicus-influenced replacement beef heifers.

Halphabarol, the active principle of Proximol, is the most potent of the four antispasmodics present in the national desert weed Cymbopogon proximus or ''Halfa Bar''. Halphabarol is of great value for the management of renal colic and in the expulsion of ureteric calculi as it causes dilation of the ureter below the site of calculus while active propulsion is maintained. Evaluation the congenital malformation of proximol in pregnant albino rats during gestation period. The virgin female rats were mated with male rats and the pregnant rats were orally administered a human equivalent dose (0.05 mg/kg) of Proximol from 5th-20th gestation day. At day 20 of pregnancy, all rats were anesthetized to obtained maternal and fetal data. The treatment group displayed some disorders, which can be summarized as growth retardation, external anomalies, embryonic resorption, and skeletal malformation. We concluded that the oral administration of Proximol resulted in embryonic abnormalities and skeletal malformations.


Halphabarol, el principio activo de Proximol, es el más potente de los cuatro antiespasmódicos presentes en la maleza desértica nacional "Cymbopogon proximus" o "Halfa Bar". Halphabarol es de gran utilidad para el manejo de cólicos renales y para la expulsión de cálculos ureterales, ya que causa la dilatación del uréter por debajo del sitio de cálculo mientras se mantiene el mecanismo de propulsión activa. Se realizó una evaluación de la malformación congénita por Proximol en ratas albinas gestantes durante el período de gestación. Las ratas fueron apareadas y a las ratas gestantes se les administró oralmente, del 5 al 20 día de gestación, una dosis de Proximol (0,05 mg / kg), equivalente a la dosis humana. Al día 20 de gestación, todas las ratas fueron anestesiadas para obtener datos maternos y fetales. El grupo de tratamiento mostró algunos trastornos, que pueden resumirse como retraso del crecimiento, anomalías externas, resorción embrionaria y malformación esquelética. The objective of this experiment was to compare hormonal, uterine, and conceptus factors associated with pregnancy establishment in beef cows supplemented or not with Ca salts of soybean oil (CSSO) for 21 d beginning after timed AI. One hundred lactating multiparous Nelore cows were allocated to 20 groups of 5 cows/group and timed inseminated on d 0 of the experiment. After AI, groups were randomly assigned to receive (as-fed basis) 100 g of protein-mineral mix + 100 g of ground corn per cow per day, in addition to 1) 100 g/cow daily of CSSO ( = 10) or 2) 100 g/cow daily of kaolin (CON; rumen-inert indigestible substance; = 10). Groups were maintained in 4 pastures (5 groups from the same treatment within each pasture) with ad libitum access to forage. Groups were segregated daily and individually offered treatments from d 0 to 21. Blood samples were collected and transrectal ultrasonography was performed to verify ovulation and corpus luteum (CL) volume immediately before AI (d 0) and on d 7 and 15. After ultrasonography on d 15, 60 cows (30 cows/treatment and 3 cows/group) diagnosed without the presence of a CL on d 0 but with a CL greater than 0.38 cm3 in volume on d 7 and 15 were assigned to conceptus collection via transcervical flushing with PBS followed by endometrial biopsy in the uterine horn ipsilateral to the CL. Additional blood samples were collected for whole-blood RNA extraction (d 20), and pregnancy status was verified by transrectal ultrasonography (d 30) in cows not assigned to conceptus collection. Cows receiving CSSO had greater ( ≤ 0.04) mean plasma linoleic acid concentration, plasma linoleic:linolenic acid ratio, plasma progesterone (P4) concentration, and CL volume during the experiment compared with CON cows. Moreover, CSSO supplementation increased ( ≤ 0.04) length and mRNA expression of and by the conceptus as well as blood mRNA expression of interferon-stimulated genes on d 20 in gestating cows. No treatment differences were detected ( ≥ 0.30) for endometrial mRNA expression of and . In summary, post-AI CSSO supplementation to B. indicus beef cows increased plasma concentration of linoleic acid and enhanced pregnancy establishment factors, which included CL development and plasma P4 concentration, conceptus growth, and mRNA expression of as well as blood mRNA expression of interferon-stimulated genes.

The continued growth in biofuel production has led to a search for alternative value-added applications of its main by-product, crude glycerin. The surplus glycerin production and a higher cost of feedstuffs have increased the emphasis on evaluating its nutritive value for animal feeding. The aim of this research was to evaluate the effect of the dietary addition of crude glycerin on sow and litter performance, and to determine the serum concentrations of hormones related to energy metabolism and feed intake in sows during gestation and lactation. A total of 63 sows were assigned randomly to one of three dietary treatments, containing 0, 3 or 6% crude glycerin (G0, G3 and G6, respectively) added to a barley-soybean meal-based diet. During gestation, none of the dietary treatments had an effect on performance, while during lactation, glycerin-fed sows consumed less feed than those fed the control diet (3.8 v. 4.2kg DM/day; P=0.007). Although lactating sows fed the G3 diet had a higher BW loss than those fed the control diet (���20.6 v. ���8.7 kg; P=0.002), this difference was not reflected in litter performance. In gestation, the inclusion of glycerin did not affect blood concentrations of insulin or cortisol. However, pregnant sows fed diets supplemented with glycerin showed lower concentrations of acyl-ghrelin and higher concentrations of leptin (���55 and +68%, respectively; P<0.001). In lactating sows, there were no differences between dietary treatments for any of the hormones measured. Pre-prandial acyl-ghrelin concentrations were positively correlated with cortisol concentrations during gestation (r=0.81; P=0.001) and lactation (r=0.61; P=0.015). In conclusion, the inclusion of up to 6% crude glycerin did not affect the performance of sows during the gestation period; however it had a negative effect on the feed intake and weight loss of lactating sows. Moreover, further research is needed to elucidate the potential relationship between glycerin inclusion levels in the diet and the serum concentrations of hormones related to feed intake and energy balance control.

BACKGROUND: Prenatally stressed offspring exhibit increased susceptibility to inflammatory disorders due to in utero programming. Research into the effects of n-3 PUFAs shows promising results for the treatment and prevention of these disorders. The purpose of this study was to investigate whether maternal fishmeal supplementation during pregnancy and lactation protects against programming of the offspring's immune response following simulated maternal infection. METHODS: In order to accomplish this, 53 ewes were fed a diet supplemented with fishmeal (FM; rich in n-3 PUFA) or soybean meal (SM; rich in n-6 PUFAs) from day 100 of gestation (gd 100) through lactation. On gd135, half the ewes from each dietary group were challenged with either 1.2 µg/kg Escherichia coli lipopolysaccharide (LPS) endotoxin to simulate a bacterial infection, or saline as the control. At 4.5 months of age the offspring's dermal immune response was assessed by cutaneous hypersensitivity testing with ovalbumin (OVA) and candida albicans (CAA) 21 days after sensitization. Skinfold measurements were taken and serum blood samples were also collected to assess the primary and secondary antibody immune response. RESULTS: Offspring born to SM + LPS mothers had a significantly greater change in skinfold thickness in response to both antigens as well as a greater secondary antibody response to OVA compared to all treatments. CONCLUSIONS: Supplementation during pregnancy with FM appears to protect against adverse fetal programming that may occur during maternal infection and this may reduce the risk of atopic disease later in life.

L-Arginine is an essential amino acid in many species that has been shown to influence reproduction. However, in horses a dose of 1% L-arginine of total dietary intake impaired absorption of other amino acids, whereas a dose of 0.5% did not. The objectives of this experiment were to evaluate postpartum parameters on mares supplemented with 0.5% L-arginine through the last 90d of gestation and 14d postpartum. Sixteen light-horse mares were randomly divided in two groups: 8 mares supplemented with 0.5% L-arginine and 8 mares fed an isonitrogenous equivalent. Gestation length, days to uterine clearance and days to first ovulation were compared. Uterine body depth, diameter of uterine horns, and length of largest pocket of uterine fluid were recorded daily via transrectal ultrasound. Measurements of foal weight, height, and cannon bone circumference were recorded for 9 weeks. Arginine treatment had no effect on gestation length (P=0.58). Supplemented mares cleared fluid quicker postpartum (6.8±0.53d; P=0.026) compared to control (9.0±0.38d). Mares supplemented with L-arginine had smaller diameter of fluid present in the postpartum uterus (P≤0.05). Days to first postpartum ovulation were not affected by treatment nor any influence on uterine involution. Finally, treatment had no effect on any foal's measured parameters. L-Arginine supplementation fed at 0.5% of daily intake during the last 90d of gestation and early postpartum in mares decreased uterine fluid accumulation, yet did not appear to have any effect on any other parameters measured.

Two experiments were designed to investigate the effects of feeding monensin and/or slow release urea with a fibrolytic feed enzyme (Optimase; Alltech, Inc., Nicholasville, KY) on performance, milk production, calf growth performance, and blood metabolites in beef cows. Spring-calving cows and heifers were used in a completely randomized design in Exp. 1 (N = 84; 534 ± 68 kg initial BW) and Exp. 2 (N = 107; 508 ± 72 kg initial BW). Exp. 1 supplements were formulated to meet cow protein requirements and fed daily and included 1) cottonseed meal with no monensin (control); or 2) monensin added to control to supply 200 mg per head per d (MON). In Exp. 2, experimental supplements included 1) cottonseed meal/wheat middlings (CS) fed at a rate to provide adequate DIP and CP according to , 2) the CS plus soybean hulls and 61 g per cow per d Optimase (OPT), 3) the CS plus monensin to supply 200 mg per cow per d (MON2), and 4) OPT plus MON2 (Combo). Cows were fed in last trimester through early lactation in Exp. 1 and during 2nd trimester in Exp. 2. Data were analyzed using the Mixed procedure in SAS with animal as the experimental unit. In Exp. 1, treatment did not affect cow BW or BCS change (P > 0.19). Calf birth BW was not affected by dam treatment (P = 0.24); however, calves from dams consuming MON weighed more (P < 0.04) at d 45 and at trial end. Calves also had greater (P = 0.04) ADG from birth to trial end. Milk production did not significantly differ among treatments (P > 0.41). In Exp. 2, mean cow BW and BCS were similar (P > 0.35) among treatments on d 90. However, from d 0 to 54, cows assigned to the OPT supplement gained less BCS (P = 0.02) compared with cows assigned to the CS supplement. Cumulative BCS gain was greater (P < 0.01) for CS-fed cows than for cows fed the OPT and MON2 supplements, although it was not significantly different for cows fed the Combo supplement. These studies indicate that the influence of monensin on cow BW and BCS change is inconsistent. The potential for monensin supplementation to positively impact calf performance during early lactation seems to be clearer. Replacing a portion of oilseed N in the supplement with Optimase may marginally reduce cow performance. Further research is needed to determine both the effects of monensin and the implications of combining monensin with Optimase on forage intake and cow performance at various stages of production.

A 3-yr study was conducted to determine the effect of supplemental protein source on ADG, feed intake, calf birth BW, and subsequent pregnancy rate in pregnant beef heifers. Crossbred, Angus-based, AI-pregnant heifers (yr 1, n = 38; yr 2, n = 40; and yr 3, n = 36) were stratified by BW (450 ± 10 kg) and placed in a Calan Broadbent individual feeding system at approximately d 142 of gestation. Following a 25-d adaptation period, an 84-d feeding trial was conducted. Heifers were offered ad libitum grass hay (8 to 11% CP, DM basis) and no supplement (CON), 0.83 kg/d distillers-based supplement (HI), or 0.83 kg/d dried corn gluten-based supplement (LO). Supplements were formulated to be isocaloric, isonitrogenous (28% CP, DM basis), and equal in lipid content but differed in RUP, with HI (59% RUP) having greater levels of RUP than LO (34% RUP). Dry matter intake was also calculated based on feed NE values to account for different energy levels of the supplement compared with the control diet. Control heifers tended (P = 0.09) to consume less total DM than either supplement treatment. However, forage-only DMI was greater (P < 0.01) for CON heifers (9.94 ± 0.12 kg) compared with HI or LO heifers (8.50 and 8.34 ± 0.12 kg, respectively). Net energy DMI was less (P < 0.01) for CON heifers (4.98 ± 0.23 kg) compared with HI or LO heifers (5.43 and 5.35 ± 0.23 kg, respectively). Control heifers gained less (P < 0.01; 0.59 ± 0.14 kg/d) than either HI (0.82 ± 0.14 kg/d) or LO heifers (0.78 ± 0.14 kg/d), resulting in lower (501 ± 9 kg) BW (P < 0.01) than HI (519 ± 9 kg) heifers at the end of the feeding period. Calf birth BW was similar (P = 0.99) among treatments. At prebreeding, CON heifers weighed less (P < 0.03) than LO heifers. Cow BW was similar (P = 0.48) among treatments at pregnancy diagnosis, and final pregnancy rate was also similar (87%; P = 0.22). Protein supplementation increased ADG in pregnant heifers; however, calf birth BW and subsequent pregnancy rates were similar.

A 3-yr study using primiparous crossbred beef heifers (n = 114) was conducted to determine the effects of protein supplement during late gestation on progeny performance and carcass characteristics. Pregnant heifers were stratified by heifer development system, initial BW, and AI service sire and placed in an individual feeding system. Heifers were offered meadow hay (8 to 11% CP) from early November to mid-February and provided no supplement (CON; n = 37), 0.83 kg/d (DM basis) of a dried distillers grains with solubles-based supplement (HI; n = 39), or 0.83 kg/d (DM basis) of a dried corn gluten feed-based supplement (LO; n = 38). Supplements were designed to be isonitrogenous (28% CP) and isocaloric but to differ in RUP with HI (59% RUP) having greater levels of RUP than LO (34% RUP). After the individual feeding period, heifers were placed in a drylot for calving. All heifers were bred using a fixed-timed AI protocol and pairs were moved to a commercial ranch in the Nebraska Sandhills for summer grazing. Calf weaning BW did not differ (P = 0.14) based on maternal diet. However, feedlot entry BW was greater (P = 0.03) for HI compared with CON calves. Average daily gain during the initial feedlot phase tended (P = 0.10) to be greatest for calves born to CON dams and lowest for calves born to LO dams. However, overall ADG was similar (P = 0.50) for the entire feedlot period. Residual feed intake during the reimplant and total feeding period was improved in calves born to supplemented dams in yr 2 and 3 compared with calves born to CON dams. There was no difference in final BW among treatments (P = 0.71). Hot carcass weight was similar (P = 0.72) among treatments; however, steers had greater (P < 0.01) HCW than heifers. Furthermore, percent empty body fat and 12th rib fat thickness were lowest (P = 0.05 and P = 0.04) for calves born to LO dams. Tenderness measured by Warner-Bratzler shear force was increased (P = 0.03) in longissimus samples from calves from CON dams compared to calves from LO dams. Similarly, crude fat levels tended to be greater (P = 0.07) for calves from CON dams compared with calves from LO dams. Based on these data, providing RUP supplements, similar to those used in this study, to primiparous heifers in late gestation consuming ad libitum grass hay resulted in increased initial feedlot BW for HI compared to CON calves, improved feed efficiency, and altered carcass characteristics in calves born to supplemented compared with CON dams.

The objective of the present study was to investigate the effects of dietary supplementation with glycitein, a major soy isoflavone, during late pregnancy (starting on d 85) and lactation on antioxidative indices and performance in primiparous sows. A total of 227 gilts (Yorkshire × Landrace) were used, and after parturition, piglets were cross-fostered within treatment so that each sow suckled 10 piglets. Gilts were randomly divided into 4 groups on d 85 of pregnancy and thereafter fed the basal diets of gestation or lactation (controls) or those supplemented with 15, 30, or 45 mg/kg diet glycitein. Reproductive performance of the sows, growth of litters, concentration of plasma glycitein, milk composition, and antioxidative indices in sows' plasma and milk, such as the activities of superoxide dismutase (SOD), catalase (CAT) and glutathione peroxidase (GSH-Px), total antioxidant capacity (T-AOC), and the content of malondialdehyde (MDA), were measured. Supplementation of the dam's diets with glycitein increased ADG of piglets (linear, P = 0.003) and weaned BW of litters (linear, P = 0.01) and both variables were approximately 10% greater than controls with the 45 mg/kg treatment. The percentage of milk protein was linearly (P < 0.05) increased on d 1 and 7 of lactation, and milk fat content increased on d 7 and 14 (linear, P< 0.05). All other measured indices of performance were unaffected by supplementation with glycitein (P > 0.05). The glycitein concentration in sow's plasma was linearly (P < 0.0001) elevated. During lactation, linear increases occurred in plasma activities of SOD (P < 0.001) and T-AOC (P < 0.05 to P< 0.001), CAT (d 7 to 18 of lactation, P < 0.05) and GSH-Px (d 7 of lactation, P < 0.05), whereas the content of plasma MDA decreased (linear, P < 0.05) throughout lactation with glycitein supplementation. Activities of CAT and T-AOC in milk were not affected by maternal supplementation with glycitein, but increases in SOD on d 18 of lactation (P< 0.001) and GSH-Px in colostrum (P < 0.05) were observed. The content of MDA in milk was decreased (linear, P< 0.05) at all stages of lactation except d 14. Collectively, the results of this study indicate that dietary supplementation with glycitein in sows during late pregnancy and lactation elevated the antioxidative indices, decreased the content of MDA in sow's plasma and milk, improved milk composition, and enhanced the growth performance of the sucking piglets.

Previously we reported increased umbilical artery blood flow in ewes supplemented with melatonin from mid- to late-pregnancy, while maternal nutrient restriction decreased uterine artery blood flow. To further unravel these responses, this study was designed to assess placental cell proliferation and vascularity following supplementation with melatonin or maternal nutrient restriction. For the first experiment, 31 primiparous ewes were supplemented with 5mg of melatonin per day (MEL) or no melatonin (CON) and allocated to receive 100% (adequate fed; ADQ) or 60% (restricted; RES) of their nutrient requirements from day 50 to 130 of gestation. To examine melatonin receptor dependent effects, a second experiment was designed utilizing 14 primiparous ewes infused with vehicle, melatonin, or luzindole (melatonin receptor 1 and 2 antagonist) from day 62 to 90 of gestation. For experiment 1, caruncle concentrations of RNA were increased in MEL-RES compared to CON-RES. Caruncle capillary area density and average capillary cross-sectional area were decreased in MEL-RES compared to CON-RES. Cotyledon vascularity was not different across dietary treatments. For experiment 2, placental cellular proliferation and vascularity were not affected by infusion treatment. In summary, melatonin interacted with nutrient restriction to alter caruncle vascularity and RNA concentrations during late pregnancy. Although melatonin receptor antagonism alters feto-placental blood flow, these receptor dependent responses were not observed in placental vascularity. Moreover, placental vascularity measures do not fully explain the alterations in uteroplacental blood flow.

BACKGROUND: Drugs used in preterm labor (PTL) have side effects. Research into new tocolytic agents is recommended. The plant Bryophyllum pinnatum (Lam.) Oken (Kalanchoe pinnata (Lam.) Pers) is used in Anthroposophic Medicine for PTL, insomnia, and emotional disturbances. The aim of this study was to evaluate the effects of B. pinnatum mother tincture (MT) on Wistar rats and their fetuses throughout pregnancy. METHOD: Sixty animals were divided into 6 equal groups: controls C1 and C2 received 1 and 25 × the maximum daily dose of a 30% ethanol / water solution, serving as vehicle of B. pinnatum MT; B1 and B2 received 1 and 25 × the maximum daily dose of B. pinnatum MT; and B3 and B4 received 50 and 100 × B. pinnatum concentrate. The following parameters were assessed: weight gain; maternal and fetal mortality; implantations and resorptions; number and weight of fetuses and placentas; major external fetal malformations. RESULTS: Rat weight gain (excluding fetal and placental weight) was higher in group B4 and lower in group B2. There were no maternal or fetal deaths and no group differences in implantations and resorptions or number and weight of fetuses and placentas. No macroscopic fetal abnormalities were observed at the 4 dosage levels investigated. CONCLUSION: Daily administration of B. pinnatum MT at high doses to pregnant Wistar rats interfered with maternal weight gain and did not interfere with fetal compartment.
