Biotin (BI) and cobalamin (CA) are essential for rumen propionate production and hepatic gluconeogenesis. The study evaluated the influence of BI or/and coated CA (CCA) on milk performance and nutrient digestion in cows. Sixty Holstein dairy cows were assigned in a 2 × 2 factorial arrangement and randomised block design to four groups. The factors were BI at 0 or 20 mg/day and CCA at 0 or 9 mg CA/day. Dry matter intake increased with BI addition but was unchanged with CCA supply. Addition of BI or CCA increased fat-corrected milk, milk fat and milk protein yields and feed efficiency. Moreover, lactose yield was increased by CCA addition. Dry matter, organic matter, crude protein and acid detergent fibre total-tract digestibility increased for BI or CCA supply. When CCA was supplemented, positive response of neutral detergent fibre digestibility to BI addition was enhanced. Supplementing BI did not affect pH, propionate content and acetate to propionate ratio, but increased total volatile fatty acids (VFA) and acetate contents. Supplementing CCA decreased pH and acetate to propionate ratio, but increased total VFA, acetate and propionate contents. Rumen protease and carboxymethyl-cellulase activities and fungi, bacteria and Butyrivibrio fibrisolvens numbers increased for BI or CCA supply. In addition, protozoa increased for BI addition, and protease activity and Prevotella ruminicola increased for CCA supply. When CCA was supplemented, positive responses of R. albus and Ruminobacter amylophilus numbers to BI addition were enhanced. Blood glucose concentration was unchanged with BI supply, but increased for CCA supply. Blood nonesterified fatty acids and ß-hydroxybutyrate contents reduced with BI or CCA supply. Supplementation with BI or CCA increased blood BI or CA content. The results showed that supplementing BI or/and CCA improved lactation performance and nutrient digestion, and CCA supply did not enhance the lactation performance response to BI supply.

The microbiome has been linked to animal health and productivity, and thus, modulating animal microbiomes is becoming of increasing interest. Antimicrobial growth promoters (AGP) were once a common technology used to modulate the microbiome, but regulation and consumer pressure have decreased AGP use in food animals. One alternative to antimicrobial growth promoters are phytotherapeutics, compounds derived from plants. Capsaicin is a compound from the Capsicum genus, which includes chili peppers. Capsaicin has antimicrobial properties and could be used to manipulate the gastrointestinal microbiome of cattle. Both the rumen and fecal microbiomes are essential to cattle health and production, and modulation of either microbiome can affect both cattle health and productivity. We hypothesized that the addition of rumen-protected capsaicin to the diet of cattle would alter the composition of the fecal microbiome, but not the rumen microbiome. To determine the impact of rumen-protected capsaicin in cattle, four Holstein and four Angus steers were fed rumen-protected Capsicum oleoresin at 0 (Control), 5, 10, or 15 mg kg-1 diet dry matter. Cattle were fed in treatment groups in a 4 × 4 Latin Square design with a 21-d adaptation phase and a 7-d sample collection phase. Rumen samples were collected on day 22 at 0-, 2-, 6-, 12-, and 18-h post-feeding, and fecal swabs were collected on the last day of sample collection, day 28, within 1 h of feeding. Sequencing data of the 16s rRNA gene was analyzed using the dada2 pipeline and taxa were assigned using the SILVA database. No differences were observed in alpha diversity among fecal or rumen samples for either breed (P > 0.08) and no difference between groups was detected for either breed in rumen samples or for Angus steers in fecal samples (P > 0.42). There was a difference in beta diversity between treatments in fecal samples of Holstein steers (P < 0.01), however, a pairwise comparison of the treatment groups suggests no difference between treatments after adjusting for multiple comparisons. Therefore, we were unable to observe substantial overall variation in the rumen or fecal microbiomes of steers due to increasing concentrations of rumen-protected capsaicin. We do, however, see a trend toward increased concentrations of capsaicin influencing the fecal microbiome structure of Holstein steers despite this lack of significance.


The microbiome is the collection of microbes present in an animal's body and has been discovered to be directly connected to animal health and productivity. In production animals, such as feedlot cattle, the microbiome can be modulated by antimicrobials to promote growth, but increasing consumer pressure to reduce antimicrobial use has producers seeking alternatives. Capsaicin is a phytotherapeutic derived from chili peppers that can be used to modulate the microbiome due to its antimicrobial properties. Eight steers were fed rumen-protected Capsicum oleoresin to determine its effect on average daily gain. In addition, rumen and fecal samples were collected for microbiome testing. No differences were detected in the rumen microbiomes between cattle fed capsaicin (treatment) or those that received no capsaicin (control). While no overall effect was observed on the fecal microbiome of cattle fed different doses of capsaicin or control, we did observe changes in fecal beta diversity due to capsaicin treatment in Holstein steers fed greater doses. The fecal microbiome structure of Holsteins fed greater dosages of capsaicin differed from those fed control or low doses, as observed by the presence of two distinct clusters. This observation suggests an impact of greater doses of capsaicin treatment on microbiome structure.

Most nutrition models and some nutritionists view ration formulation as accounting transactions to match nutrient supplies with nutrient requirements. However, diet and stage of lactation interact to alter the partitioning of nutrients toward milk and body reserves, which, in turn, alters requirements. Fermentation and digestion of diet components determine feeding behavior and the temporal pattern and profile of absorbed nutrients. The pattern and profile, in turn, alter hormonal signals, tissue responsiveness to hormones, and mammary metabolism to affect milk synthesis and energy partitioning differently depending on the physiological state of the cow. In the fresh period (first 2 to 3 wk postpartum), plasma insulin concentration and insulin sensitivity of tissues are low, so absorbed nutrients and body reserves are partitioned toward milk synthesis. As lactation progresses, insulin secretion and sensitivity increase, favoring deposition instead of mobilization of body reserves. High-starch diets increase ruminal propionate production, the flow of gluconeogenic precursors to the liver, and blood insulin concentrations. During early lactation, the glucose produced will preferentially be used by the mammary gland for milk production. As lactation progresses and milk yield decreases, glucose will increasingly stimulate repletion of body reserves. Diets with less starch and more digestible fiber increase ruminal production of acetate relative to propionate and, because acetate is less insulinogenic than propionate, these diets can minimize body weight gain. High dietary starch concentration and fermentability can also induce milk fat depression by increasing the production of biohydrogenation intermediates that inhibit milk fat synthesis and thus favor energy partitioning away from the mammary gland. Supplemental fatty acids also impact energy partitioning by affecting insulin concentration and insulin sensitivity of tissues. Depending on profile, physiological state, and interactions with other nutrients, supplemental fatty acids might increase milk yield at the expense of body reserves or partition energy to body reserves at the expense of milk yield. Supplemental protein or AA also can increase milk production but there is little evidence that dietary protein directly alters whole-body partitioning. Understanding the biology of these interactions can help nutritionists better formulate diets for cows at various stages of lactation.

The present study examined the effects of age at group housing and age at incremental hay supplementation in calf starters from 7.5 to 15% (dry matter, DM) and their interaction on growth performance, behavior, health of dairy calves, and development of heifers through first breeding. A total of 64 calves (n = 16 calves/treatment, 8 male and 8 female) were randomly assigned to 4 treatments in a 2 × 2 factorial arrangement, with age at group housing (early = d 28 ± 2, EG vs. late = d 70 ± 2, LG; 4 calves per group) and age at incremental hay supplementation of calf starters from 7.5 to 15% of DM (early = d 42 ± 2 d, EH vs. late = d 77 ± 2, LH) as the main factors. All calves (female and male) were weaned at 63 days of age and observed until 90 days of age. Heifer calves were managed uniformly from 90 days of age until first calving to evaluate the long-term effects of treatment. No interactions were observed between age at group housing and age at incremental hay to calves on starter feed intake, performance, calf health and behavior, and heifer development through first breeding, which was contrary to our hypothesis. The age at which incremental hay supplementation was administered had no effect on starter feed intake, growth performance, or heifer development until first calving. When EG calves were compared with LG calves, nutrient intake (starter, total dry matter, metabolizable energy, neutral detergent fiber, starch, and crude protein), average daily gain, and final body weight increased. In addition, frequency of standing decreased and time and frequency of eating increased in EG calves compared to LG calves. Overall, early group housing leads to improved growth performance in dairy calves with no negative effects on calf health compared to late group housing.

Increasing methionine availability in dairy cow diets during the first third of lactation may enhance their performance and health. The aim of this study was to determine the effect of supplementing rumen-protected methionine (Smartamine® M, SM) in a lactation diet with protein and energy levels calculated according to the literature. Seventy-six multiparous Holstein cows (39.1 ± 6.8 kg of milk/d and 65 ± 28 DIM) were assigned to 1 of 2 dietary treatments (38/treatment) according to a randomized complete block design with a 2-wk (covariate) and 10-wk experimental period. Treatments were a basal diet (CON; 3.77 Lys:1Met); and CON + 23 g SM (2.97 Lys:1 Met). Individual milk samples were taken every 2 weeks to determine milk composition. Blood was collected from 24 cows on d+30 d to measure plasma AA levels. Body weight and body condition score (BCS) were measured at the beginning and the end of the experiment. The SM diet promoted higher milk yield (41.7 vs. 40.1 kg/d; P = 0.03). Energy-corrected milk yield (41.0 vs. 38.0 kg/d), milk protein yield (1.30 vs. 1.18 kg/d), milk protein (3.14% vs. 2.97%) and casein (2.39% vs. 2.28%) were also different (P < 0.01) as well as milk fat yield (1.42 vs. 1.29 kg/d; P = 0.02). A trend (P = 0.06) for higher milk fat % (3.41% vs. 3.21%) was observed. Both diets resulted in similar body weight, but CON-fed cows tended (P = 0.08) to have higher BCS. Higher plasma methionine levels were determined with SM compared with CON (29.6 vs. 18.4 µM; P < 0.01), but lysine and histidine were not different. Dietary supplementation of RPM improved productive performance by increasing milk yield and milk components yields, suggesting better dietary AA utilization when Met levels are adjusted in Lys-adequate lactation diets.

Providing essential amounts of balanced nutrients is one of the most vital aspects of livestock production. Among nutrients, protein has an essential role in many physiological functions of animals. Amino acids in needs for both high and medium yielding ruminant animals are not fully covered by microbial degraded feed sources in the rumen of animals, and they must be met by protecting the proteins from being broken down in the rumen; hence, the dietary supplementation of rumen-protected proteins (RPP), including mainly rumen-protected methionine (RPM), became imperative. Many researchers are interested in studying the role of (RPM) in ruminant animals concerning its effect on milk yield, growth performance, digestibility, dry matter intake and nitrogen utilization efficiency. Unfortunately, results obtained from several investigations regarding RPM indicated great fluctuation between its useful and useless effects in ruminant nutrition particularly during early and late lactation period; therefore, this review article may be helpful for ruminant farm owners when they decide to supplement RPM in animal's diet. Conclusively, supplementation of RPM often has a balanced positive influence, without any reported negative impact on milk yield, growth performance and blood parameters especially in early lactating ruminant animals and when used with the low crude protein diet.

This experiment was carried out to study the effect of water extracted pomegranate peel extract (PE) on ruminal protein degradation and post-ruminal digestion in the dairy cow. PE was added at six levels of total phenolics (g/kg of the basal diet); 3.75 (PE1); 4.4 (PE2); 5.05 (PE3); 5.70 (PE4); and 6.35 (PE5). Rumen degradable crude protein (rdCP) decreased with PE addition (L < 0.0001), but total CP degradability (tdCP) was not affected. Compared to PE0, PE2, and PE3 diets showed higher (L = 0.054, Q = 0.029) digestibility of bypass CP (dBCP). Increasing levels of PE resulted in a decrease in proteolytic bacteria numbers (p < 0.0001). At PE4 and PE5 levels, total VFA and acetate concentrations linearly decreased compared to PE0. PE inclusion lowered the acetate:propionate ratio (L = 0.0001) and Ammonia-N production after 24 h (L = 0.0008) of incubation. The total number of protozoa, genera Dasytricha and Isotricha, and subfamilies Entodiniinae, Diplodiniinae, and Ophrioscolecinae decreased with increasing dietary PE concentration (p < 0.0001). The results suggest that all levels of PE addition reduce the protozoal population and Ammonia-N concentration. All PE levels slowed down protein degradation in the rumen but PE2 and PE3 showed the greatest effect.

The assay aimed to evaluate the effect of feeding a recently developed phytogenic feed additives mixture in diets of lactating Friesian cows (n = 30; 514 ± 10.1 kg body weight) for 3 months. Cows were stratified into three groups of 10 cows each and fed a control diet alone or the control diet supplemented with the additives mixture at 3 g (PHY3) or 6 g (PHY6)/cow daily. Menthol, levomenthol, ß-linaloolm, anethole, hexadecanoic acid and p-menthane were the principle compounds identified in the additives mixture. The PHY3 increased (p < 0.01) intake and nutrient digestibility. PHY3 and PHY6 increased (p < 0.01) ruminal pH, total volatile fatty acids, propionate and acetate. PHY3 and PHY6 improved serum total protein and antioxidant capacity and decreased the concentrations of serum urea-N, triglycerides, total lipids, cholesterol and malondialdehyde (p < 0.05). PHY3 increased milk production and milk content of total solids, protein, lactose and fat. Both PHY3 and PHY6 did not affect mineral concentrations in blood or milk. It is concluded that the inclusion of 3 g/cow/d of feed additives mixture in the lactating Friesian cows diet enhanced milk production and feed utilization, with negative effects observed with increasing the dose of additives mixture to 6 g/cow daily.

This study aimed to evaluate the effects of the addition of Parkia platycephala pod meal (PP) and urea on the chemical composition, fermentation characteristics, and in situ ruminal degradability of elephant grass silage. A completely randomized design with a 4 × 2 factorial arrangement was adopted, with four levels of pod meal (0, 10, 20, and 30%) and two levels of urea (0 and 1.5%) on as fed basis. The produced silages were evaluated in terms of dry matter (DM), crude protein (CP), mineral matter (MM), neutral detergent fiber (NDF), acid detergent fiber (ADF), hemicellulose, ammonia nitrogen, pH, short chain fatty acids, and in situ ruminal degradability. For the study of degradation, four Santa Ines rumen fistulated sheep, with average weight of 45 ± 2.5 kg were used. The association of the additives increased (p < 0.05) DM, CP, DM degradability, fractions a, b, and effective degradability at all passage rates (2, 5, and 8%/h) and reduced NDF and ADF contents. DM disappearance increased (p < 0.05) during the incubation time, especially for the silages containing the two additives. The interaction in the rumen environment is essential for microbial multiplication. Thus, the use of additives such as PP and urea contributed to the availability of digestible fractions of the feed and greater use by ruminal microorganisms. The association of Parkia platycephala with urea improves the fermentation characteristics, chemical composition, and degradability of elephant grass silage.

The aim of this experiment was to test the effect of yeast-fermented de-hulled rice (YDR) levels of protein-rich feed with different kinds of roughages on in vitro gas production, nutrient degradability, and rumen fermentation. The treatments were randomly assigned according to a 2 × 4 factorial arrangement in a completely randomized design (CRD). The two experimental factors were comprised of two roughages (R) (untreated rice straw (RS) and sweet grass hay (SGH)) and four ratios of roughage to yeast-fermented de-hulled rice (R:YDR) (100:0, 75:25, 50:50, and 25:75). Thus, there were 8 treatment combinations. The results revealed that the interaction between R and R:YDR ratios influenced on the gas production rate constant for the insoluble fraction ratio (c) (P < 0.01). The in vitro dry mater degradability (IVDMD) was improved by SGH and R:YDR ratios (P < 0.05). Supplementation of YDR with both of roughage sources (RS and SGH) increased propionate (C3) (P < 0.05) and total VFA production (P < 0.01); both factors showed interactive effects on rumen methane production (P < 0.01). Moreover, bacterial population was significantly increased by the SGH:YDR ratios (P < 0.05). Therefore, it could be summarized that supplementing YDR, an enriched protein source with SGH:YDR ratio at 50-75:50-25 ratio significantly enhanced nutrient degradability and in vitro rumen fermentation efficiency.

Six steers were used to study the effects of dietary supplementation with sodium sulfate (Na2SO4) on rumen fermentation, nutrient digestion, rumen microbiota, and plasma metabolites. The animals were fed a basal ration with Na2SO4 added at 0 g/day (sulfur [S] content of 0.115% dry matter [DM]), 20 g/day (S at 0.185% DM), or 40 g/day (S at 0.255% DM) in a replicate 3-by-3 Latin square design. The results indicated that supplementing with Na2SO4 increased the ruminal concentration of total volatile fatty acids, the molar proportions of acetate and butyrate, the ruminal concentrations of microbial protein, SO42--S, and S2--S, and the digestibility of fiber, while it decreased the molar proportion of propionate and the ruminal concentration of ammonia nitrogen. Supplementing with Na2SO4 increased the diversity and the richness of rumen microbiota and the relative abundances of the phylum Firmicutes and genera Ruminococcus 2, Rikenellaceae RC9 gut group, and Desulfovibrio, whereas it decreased the relative abundances of the phylum Bacteroidetes and genera Prevotella 1, Prevotellaceae UCG-001, and Treponema 2 Supplementing with Na2SO4 also increased the plasma concentrations of amino acids (l-arginine, l-methionine, l-cysteine, and l-lysine), purine derivatives (xanthine and hypoxanthine), vitamins (thiamine and biotin), and lipids (acetylcarnitine and l-carnitine). It was concluded that supplementing the steer ration with Na2SO4 was beneficial for improving the rumen fermentation, fiber digestibility, and nutrient metabolism through modulating the rumen microbial community.IMPORTANCE Essential elements like nitrogen and sulfur greatly affect rumen fermentation and metabolism in ruminants. However, little knowledge is available on the effects of sulfur on the rumen microbiota and plasma metabolome. The results of the present trial demonstrated that supplementing the steer ration with sodium sulfate markedly improved rumen fermentation, fiber digestibility, and metabolism of amino acids, purine derivatives, and vitamins through effects on the ruminal microbiome. The facts obtained from the present trial clarified the possible mechanisms of the positive effects of sulfur on rumen fermentation and nutrient utilization.

The effects of source of corn silage and trace mineral on rumen fermentation, turnover, and particle passage rates were evaluated with 8 ruminally cannulated Holstein cows averaging 83 (standard error = 5) days in milk in a replicated 4 × 4 Latin square design with a 2 × 2 factorial arrangement of treatments and 28-d periods. The diets consisted (dry basis) of 55% conventional (CON) or brown midrib-3 (BM3) corn silage, 2% chopped wheat straw, and 43% grain mix with either sulfate (STM) or hydroxy (HTM) source of Cu, Zn, and Mn trace minerals. The targeted supplemental amount of Cu, Zn, and Mn was 194, 1,657, and 687 mg/d, respectively. The dietary treatments were (1) CON-STM, (2) CON-HTM, (3) BM3-STM, and (4) BM3-HTM. Dietary nutrient composition of BM3 diets averaged 32.1% amylase neutral detergent fiber on an organic matter basis (aNDFom) and 6.9% undigested neutral detergent fiber at 240 h of in vitro fermentation (uNDF240om; % of dry matter), and CON diets averaged 36.2% aNDFom and 8.6% uNDF240om (% of dry matter). Data were summarized by period and analyzed as a replicated Latin square design with fixed model effects for corn silage, trace mineral, corn silage and trace mineral interaction, period within replicated square, and replicated square using the MIXED procedure of SAS (version 9.4, SAS Institute Inc., Cary, NC). Cow within replicate was a random effect. Daily mean, standard deviation, minimum, and maximum for rumen pH were unaffected by corn silage or trace mineral source. Cows fed the CON diets had greater rumen acetate percentage than cows fed the BM3 diets (65.7 vs. 64.7 molar %). In contrast, cows fed the BM3 diets had greater rumen propionate percentage than cows fed the CON diets (21.4 vs. 20.4 molar %). Total volatile fatty acid concentration was lower for cows fed STM versus HTM in BM3 diets, but not for the cows fed the CON diets. Cows fed the BM3 diets had faster turnover rate and shorter turnover time for uNDF240om than cows fed the CON diets (3.12 vs. 2.86%/h and 33.3 vs. 36.5 h, respectively). Cows fed the BM3 diets had a faster passage rate of small and medium corn silage neutral detergent fiber particles than cows fed the CON diets (5.73 vs. 5.37%/h and 4.74 vs. 4.31%/h, respectively). We observed a corn silage by source of trace mineral interaction on organic matter and uNDF240om rumen pool size and organic matter turnover. Overall, source of corn silage had a pronounced influence on rumen dynamics presumably related to greater in vitro neutral detergent fiber digestibility and lower uNDF240om content of BM3 corn silage that allowed for faster turnover of indigestible neutral detergent fiber and greater passage rate of corn silage particles. In contrast, the source of trace mineral had much less significant effects on rumen fermentation, turnover, and particle passage rates. Corn silage-based diets intended to enhance rumen fiber fermentation, turnover, and passage are more affected by source and digestibility of neutral detergent fiber than source of dietary trace minerals.

The objective of this study was to determine the effect of dry medicinal plants (wormwood, chamomile, fumitory and mallow) and dietary substrates containing a mix of the plants on the end products of in vitro ruminal and intestinal fermentation, rumen protozoan population and ruminal antioxidant capacity of sheep. The experiment consisted of fermentations with the four plants used individually as the sole substrate and fermentation of a mix of medicinal plants (Plants): meadow hay:barley grain (MH:B), 700/300 w/w and Plants:MH:B, 100/600/300 w/w/w. The experiment was conducted using the in vitro gas production technique (IVGPT) with 35 ml of buffered inocula and approximately 250 mg (DM basis) of substrate incubated for 24 hr at 39°C in anaerobic conditions. Quantitative analyses of the bioactive compounds by ultra-high-resolution mass spectrometry in Plants identified three main groups: flavonoids (22 mg/g DM), phenolic acids (15 mg/g DM) and alkaloids (3 mg/g DM). The total antioxidant capacity (TAC) of the plant extracts and rumen fluid was analysed using a ferric reducing antioxidant power assay. The values of total and individual short-chain fatty acids, acetate:propionate ratio, pH and total gas production were significantly affected by the single plant substrates and inocula (p < .001). Apart from these parameters, the values of ammonia N, methane production and total gas produced were decreased in Plants:MH:B in comparison with MH:B (p < .001). A positive correlation was recorded between total polyphenols content and TAC of plant extracts (R2  = .778, p < .001). The counts of the total ciliate protozoan population in rumen fluid after 24 hr of fermentation were not significantly different (p > .05). Results suggest that the dietary substrate containing the medicinal plant mix possessed strong ruminal antioxidant capacity, had the potential to reduce methane emission and ammonia concentration and caused desirable changes in the gastrointestinal ecosystem.

Evidence suggests that lipopolysaccharide (LPS) absorbed from the large intestine may contribute to the inflammatory response to high starch feeding in dairy cows. This work evaluated the impact of buffers or alkalinizing agents with expected large intestinal activity on faecal indicators of intestinal fermentation and LPS. Ten late-lactation cows were used in a replicated 5 × 5 Latin square design with 7-day periods. Cows were fed a diet containing 265 g/kg dry matter of starch and were abomasally infused with 1 g/kg body weight cornstarch daily. Treatments were control (CON), ration supplementation with 200 g/day sodium bicarbonate (FSB), 200 g/day calcium carbonate (FCC) or 125 g/day calcium carbonate plus 75 g/day of magnesium oxide (FCCM), or abomasal infusion of a lipid encapsulate providing 200 g/day sodium bicarbonate (ISB). The FCC, FCCM and ISB treatments were hypothesized to have large intestinal buffering effects, and FSB was included as a secondary control. Milk, feed, rumen and faecal samples were collected on day 7 of each period. Treatment did not affect intake, milk yield or milk composition. There were no effects of treatment on ruminal measures except that ISB tended to reduce and the post-ruminal treatments as a whole (FCC, FCCM and ISB) reduced rumen butyrate compared with CON. Faecal pH was greater for FCCM compared with all other treatments. Total faecal VFA tended to increase with FCC and FCCM compared with CON and was increased by the post-ruminal treatments as a whole compared with CON. Treatment did not affect faecal dry matter, lactate or LPS or apparent total tract nutrient digestibility. Although some treatments altered fermentation as evidenced by the change in faecal VFA, this was not accompanied by a decrease in faecal LPS. The strategies employed in this study had limited effects on large intestinal fermentation.

Exogenous fibrolytic enzymes (EFE) and yeast are feed supplements that improve forage digestion in rumen, but their influences on physical reticulorumen parameters are not well studied. This study was designed to evaluate the effect of the EFE:endo-ß-xylanase (37x104 U/cow/day), endocellulase (45x104 U/cow/day), endo-ß-glucanase (12x104U/cow/day), and active yeast - Saccharomyces cerevisiae CNCM-1077 (10x109CFU/cow/day) supplements on reticulorumen pH (RpH) and temperature (RT) in dairy cows. Nine Lithuanian Red cows were allocated into three groups (3 cows/group): control group (C) - farm diet without supplementation, enzyme group (E) - farm diet supplemented with EFE, enzyme and active yeast group (EY) - farm diet supplemented with EFE and active yeast. The feeding trial lasted for 60 d. All cows were equipped with reticuloruminal telemetric pH and temperature sensor device. Data provided by the device were used to calculate the mean RpH (RpH/24h), the mean minimal RpH (minRpH/24h) and mean of the time that RpH was below the threshold value of 6.0 (RpH⟨6.0/24h, min.). The highest RpH/24h (6.37±0.22) was observed in group EY and it was by 1.62% (p⟨0.05) and 1.27% (p⟨0.001) higher as compared with groups E and C, respectively. Also minRpH/24h (6.24±0.24) was highest in group EY and values were by 0.63% (p⟨0.001) and 0.65% (p⟨0.001) higher as compared with groups C and E, respectively. The shortest duration of RpH⟨6.0/24h, was recorded in group EY, and it was by 57.76% (p⟨0.05) and 47.87% shorter as compared with groups C and E, respectively. In conclusion, feed supplementation with EFE and Saccharomyces cerevisiae CNCM-1077 had beneficial effect on RpH.

There is a risk of iron overload in grazing livestock. However, the effects on nutrient absorption and rumen function induced by excessive iron have not been well understood. Therefore, the purpose of present study was to investigate the impact of over-load iron on growth performance, nutrient digestibility, blood biochemistry, rumen fermentation and bacterial communities in sheep. Twenty-four German Mutton Merino cross-bred sheep with weight (42.66 ± 2.34 kg BW) were randomly divided into 4 groups, each with 6 replicates and 1 sheep per replicate. The basal diet consisted of 60% Leymus chinensis hay and 40% concentrate. The sheep in 4 groups were fed the basal diets supplemented with 50 (Control), 500 (T1), 1,000 (T2) and 1,500 (T3) mg Fe/kg as ferrous sulphate monohydrate (FeSO4 ·H2 O) respectively. And the actual contents of iron in the diet were determined to be 457.68 (control), 816.42 (T1), 1,256.78 (T2) and 1,725.63 (T3) mg/kg respectively. The experiment lasted 62 days including a 7-day metabolism trial. During the whole experiment, the digestibility of dry matter, organic matter, neutral detergent fibre and acid detergent fibre showed a quadratic increase with increasing over-load iron levels (p < .05), and maximum responses were found with 500 mg/kg supplementation. However, the response of total VFA concentration showed a quadratic decrease, as did the concentrations of propionate, butyrate and valerate (p < .05). Serum total iron-binding capacity on day 30 showed a quadratic decrease with the increase in high-dose iron, while the serum iron content increased linearly at day 60 (p < .05). Excessive iron resulted in the change in bacterial communities. An increase in over-load iron linearly decreased the abundance of bacteria in the phylum Bacteroidetes (p < .05), but linearly increased the Firmicutes (p = .037) and Proteobacteria (p = .018). In addition, there was a quadratic effect (p = .003) on the Fibrobacteres, which was higher in the 500 and 1,000 mg/kg Fe-supplemented groups. At the genus level, there were quadratic effects on the abundances of Selenomonas_1 (p = .023) and Ruminococcaceae_UCG-014 (p = .016). Furthermore, feeding of iron linearly increased the relative abundances of Succiniclasticum and Succinivibrionaceae_UCG-002 (p < .05). These results indicate that increasing ferrous sulphate monohydrate in diets had no negative impact on the growth performance, but it changed nutrient digestibility, blood iron parameters, rumen fermentation and bacterial communities in sheep.

Researches on sodium selenite (SS) mainly focus on production performance and rumen fermentation in ruminants, and the influence of dietary Se addition on ruminal microbial population and enzyme activity in dairy bulls is scarce. This study mainly evaluated the effects of SS on ruminal fermentation, microflora and urinary excretion of purine derivatives (PD) in dairy bulls. Eight ruminally cannulated dairy bulls were used in a replicated 4 × 4 Latin square design. Treatments were control, low SS (LSS), medium SS (MSS) and high SS (HSS) with 0, 0.1, 0.3 and 0.5 mg/kg of selenium (Se) from SS in dietary dry matter (DM), respectively. The supplement of SS (1.0 g/kg of Se) was mixed into the first third of the daily ration. Bulls were fed a total mixed ration with corn silage to concentrate ratio of 50:50 on a DM basis. Dry matter intake was not affected, average daily gain linearly increased, while feed conversion ratio quadratically decreased with increasing Se addition. The linearly increased digestibility of DM, organic matter, crude protein, ether extract, neutral detergent fibre and acid detergent fibre was observed. Both ruminal pH and ammonia-N concentration linearly decreased, whereas total volatile fatty acid concentration linearly increased. A lower acetate to propionate ratio was observed due to the unchanged acetate proportion and increased propionate proportion. Activity of cellobiase, xylanase, pectinase, α-amylase and protease, populations of total bacteria, fungi, protozoa, Ruminococcus (R.) albus, R. flavefaciens, Fibrobacter succinogenes, Butyrivibrio fibrisolvens and Ruminobacter amylophilus as well as urinary total PD excretion linearly increased, whereas populations of total methanogens and Prevotella ruminicola linearly decreased. The data indicated that dietary Se addition stimulated ruminal microbial growth and enzyme activity, and resulting in the increased nutrient digestion and growth performance, and the optimum supplementary dose of Se was 0.3 mg/kg dietary DM from SS in dairy bulls.

Inadequate availability of feed ingredients significantly contributes to reduced animal production. Nutritional evaluation of feed ingredients for potential use in feed ration is key. Triplicate samples of oil extraction by-products viz.: amarula (Sclerocarya birrea), macadamia (integrifolia) and baobab (Adansonia digitata L.) oilcakes were analysed for proximate, mineral and amino acids content. To determine the in situ rumen degradability of dry matter (DM) and crude protein (CP) from the oilcakes, polyester bags were used to incubate triplicate subsamples of each oilcake in the ventral rumen of each of the three cannulated mid-lactating Holstein cows for 2, 4, 8, 16, 24, or 48 h. Amarula oilcake (AOC) had higher (P < 0.05) CP, ether extract and essential amino acid content than macadamia (MOC) and baobab (BOC) oilcakes. Fibre fractions were higher (P < 0.05) in MOC and BOC than in AOC. While calcium (Ca) and potassium (K) concentrations were high (P < 0.05) in MOC and BOC, phosphorus (P) concentration was high (P < 0.05) in AOC. The effective degradability of DM and water-soluble, rapidly degradable fractions of DM and CP were high (P < 0.05) in AOC. Insoluble but degradable fraction of CP was high (P < 0.00) in BOC and effective degradation of CP was high (P < 0.05) in AOC and BOC. The AOC with its high CP, EE and essential AA can be used as both protein and energy source for ruminants. Seemingly, MOC and BOC can be used as supplements for fibre as well as Ca and K in ruminant diets. Hence, these non-conventional oilcakes have the potential to partially replace conventional oilcakes in ruminant diets. Evaluation of growth performance, nitrogen digestion and carcass characteristics in ruminants fed diets containing the AOC, BOC and MOC are recommended.

We investigated the influence of rumen-protected conjugated linoleic acid (rpCLA) on growth performances, and carcass and meat quality traits in beef. Twenty-four young bulls and 30 heifers obtained from double-muscled beef sires and dairy cows were fed a low-protein ration (110 g/kg DM of crude protein) supplemented with 0, 8 or 80 g/d of a commercial rpCLA product. The animals were monthly weighed and scored for body muscularity and fatness. Blood samples were collected after 140 days on feed. Animals were slaughtered when they reached average in vivo fatness scores of around 2.5 (heifers) and 2.0 (bulls) points respectively. At slaughter, carcasses, various organs and parts of the gastrointestinal tract were weighed; the 5th rib was dissected and its tissue and muscle chemical composition was determined. The rpCLA had little influence on growth performance but decreased the blood urea content by 28% (p < 0.01). The rpCLA × sex interactions for daily gain (p < 0.05), conformation scores (p < 0.01), and blood creatinine content (p < 0.05) suggest that males were more responsive to rpCLA than females when fed a low-protein ration, probably because of the metabolic protein-sparing effect of CLA. Only slight differences were observed in carcass weight and quality at slaughter. The results indicate that the response of beef cattle to rpCLA is dependent on sex or on their propensity for lean and fat accretion. It is also possible that counteracting feedback mechanisms compensate for the influence of rpCLA administration over the course of growth.

This study evaluated the effects of rumen-protected folic acid (RPFA) supplementation and dietary protein level on growth performance, ruminal fermentation, nutrient digestibility and hepatic gene expression in calves. Forty Holstein male calves (161 ± 5.7 days of age and 192 ± 5.4 kg of body weight) were assigned to one of four groups in a randomized experimental design with a 2 × 2 factorial arrangement. Moderate crude protein (130.1 g CP/kg [MCP] or high crude protein (150.2 g CP/kg [HCP]) diets were fed without (RPFA-) or with 3.6 mg FA (RPFA+) as RPFA per kg dietary dry matter (DM). Calves were fed a total mixed ration with a corn silage to concentrate ratio of 50:50 on a DM basis. The CP×RPFA interaction was not significant for any of the studied variables. The unchanged DM intake, higher average daily gain and lower feed conversion ratio were observed for HCP or RPFA+. Ruminal pH was lower, and total volatile fatty acids (VFA) concentration was higher for HCP or RPFA+. Acetate proportion was higher, and propionate proportion was lower for HCP or RPFA+. As a result, the higher acetate to propionate ratio was observed. Ruminal ammonia N was higher for HCP, but was lower with RPFA supplementation. The higher digestibility of DM, OM, CP and NDF was observed. Blood glucose and insulin were unchanged, but albumin, total protein, GH and IGF-1 were higher. Similarly, the higher hepatic expression of GH, IGF-1, GHR, IGF-1R, PI3K, mTOR and P70S6K was observed for HCP or RPFA+. The results indicated that increasing dietary CP content or supplementation with RPFA promoted growth performance of calves by improving nutrient utilization and up-regulating hepatic expression of gene related to protein synthesis.
