RESUMO

With the development of society, physical activity has come to be an effective means by which people pursue good health to improve the quality of life. However, with the increase of intensity and the passage of time, exercise injury has become a hazard that can no longer be ignored. It is imperative to find effective ways to inhibit or reduce the negative effects of exercise. Mitochondria are important organelles involved in exercise and play an important role in exercise injury and prevention. Studies have found that exercise preconditioning and increased mitochondrial nutrition can effectively decrease mitochondrial damage after exercise. Against this background, some of the newest developments in this important field are reviewed here. The results discussed indicate that exercise preconditioning and supplement mitochondrial nutrition need to be increased to prevent exercise-related injuries.

BACKGROUND: Cancer is a disease characterized by the invasion and uncontrolled growth of cells. One of the best ways to minimize the harmful effects of mutagens is through the use of natural antimutagens. In this regard, the search for new antimutagens that act in the chemoprevention could represent a promising field in this area. OBJECTIVE: In this study biological potential of 11 fractions from Coccoloba uvifera L. leaf hexane extract was evaluated by several in vitro tests. METHODS: Leaves were lyophilized and hexane extraction was performed. The extract was fractionated by column chromatography with hexane, ethyl acetate, and methanol. The antimutagenic (Ames test), antiproliferative (MTT test), and antioxidant capacity (DPPH, ABTS, and ferrous ion chelation) of the fractions were evaluated. RESULTS: Fractions 4, 6, 8, and 9 have antimutagenic activity (against sodium azide in strain TA100), fraction 11 showed antiproliferative capacity (IC50 of 24 ± 9 µg/mL in cells of HCT 116). The fractions with the highest activity were analyzed by HPLC-MS and lupeol, acacetin, and ß-sitosterol were identified. CONCLUSION: This study demonstrates, for the first time, the bioactivity of C. uvifera leaf as a new source of High Biological Value Compounds (HBVC), which can be of interest to the food and pharmaceutical industries.

A new pyrano coumarin, identified as excavatin A (1) together with two known compounds nordentatin (2) and binorpocitrin (3) was isolated from the 95% EtOH extract of Clausena excavata. All structures were elucidated by using spectroscopy methods such as extensive NMR and HR-FAB-MS spectrometry. All the isolated compounds were tested on antidiabetes activity by using α-glucosidase inhibition assay and the antioxidant activity by DPPH assay. Compounds 1-3 showed antioxidant activity with IC50 values 0.286, 0.02, 0.278 mM. Among them, 2 exhibited inhibition activity against maltase (IC50 5.45 µM) and sucrase (IC50 43.57 µM). However, compounds (1) and (3) displayed inhibition on yeast α-glucosidase with IC50 values 1.92 and 5.58 mM.[Figure: see text].

BACKGROUND: Chlorophytum comosum, popularly known as Spider Ivy, is used as a medicinal plant in traditional Chinese medicine, however, its detailed chemical composition and biological activity are yet unexplored. OBJECTIVE: To carry out the phytochemical investigation on different parts of Chlorophytum comosum using GCMS/ LC-ESI-MS and evaluation of its antioxidant, hemolytic and antiproliferative potential on breast cancer (MCF-7), lung cancer (A549, H1299) and normal lung (L-132) cell lines. METHODS: Chemical constituents from aqueous roots and leaves extracts were identified using LC-ESI-MS/GCMS. The identified compounds were annotated based on the match of mass spectra with the literature using NIST 14 and METLIN databases. Antioxidant activity was studied using DPPH, FRAP and TPC assays. The antiproliferative effects of ethanolic roots and leaves extracts of Chlorophytum comosum were measured by MTT assay on breast cancer (MCF-7), lung cancer (A549 & H1299) and normal lung (L-132) cell lines. The toxicity studies of the extracts were carried out using Hemolysis assay. RESULTS: GC-MS analysis identified 34 metabolites in roots and 17 from leaves, while 17 compounds from roots and 7 from leaves were detected by LC-ESI-MS. Significant antiproliferative effects were observed on the A549 and MCF-7 cancer cell lines with IC50 values ranging from 56.86 µg/ml to 68.68 µg/ml while no marked response was observed against normal cell line L-132. CONCLUSION: Our study represents the first report on the detailed chemical composition and antiproliferative potential of Chlorophytum comosum against lung and breast cancer cell lines.

Free radicals can cause many diseases, such as cancer. Antioxidant is a compound that could scavenge free radicals. One of the natural antioxidants is guava. The goals of this research were to investigate the antioxidant activity of leaves and fruit of crystal guava by determining the value of the Antioxidant Activity Index (AAI) using DPPH, CUPRAC, and FRAP; evaluate the total phenolic content (TPC) and total flavonoid content (TFC); analyse the correlation between the TPC and TFC with AAI DPPH, CUPRAC, and FRAP, and analyse the correlation between the 3 methods. Extraction was performed by the reflux method using n-hexane, ethyl acetate, and ethanol. Determination of AAI DPPH, CUPRAC, FRAP, the TPC, and the TFC was performed by UV-visible spectrophotometry. The correlation of the TPC and TFC with AAI DPPH, CUPRAC, and FRAP and, also, the correlation of the 3 methods were investigated by Pearson's method. The antioxidant activity of leaves and fruit extracts of crystal guava showed AAI DPPH in the range of 0.33-56.46, CUPRAC 0.20-7.31, and FRAP 1.65-59.89. The highest TPC was given by ethanol leaf extracts (49.55 ± 1.45 g GAE/100 g), while the highest TFC was for n-hexane leaf extracts (9.68 ± 0.210 g QE/100 g). The TPC of leaves extract had a significantly positive correlation with AAI DPPH, CUPRAC, and FRAP. AAI DPPH, AAI CUPRAC, and AAI FRAP of leaves and fruit extract of crystal guava showed a significantly positive correlation. In general, leaves extract had strong antioxidant activity by the three methods. For the highest antioxidant activity, ethanol was the best solvent for extraction leaves and ethyl acetate for extraction fruit of crystal guava. The TPC in leaves extract contributed to the antioxidant activity by DPPH, CUPRAC, and FRAP methods. The Antioxidant activity of leaves and fruit extracts of crystal guava was linear by the three methods.

Chokecherry (Prunus virginiana L.) is rich in bioactive molecules as phenolics, which can act as antioxidants, anti-inflammatory, anticancer, among others; however, due to its high perishability, most of this fruit is wasted. Freezing and sun drying have been the most adopted techniques to avoid its postharvest deterioration. Nevertheless, both processes have presented some drawbacks as high storage costs and losses of bioactive molecules. Therefore, to preserve these molecules, this study compared the impact of convective airflow drying (CAD), freezing (FR), freeze drying (FD), and swell drying (SD). Total phenolics content (TPC), total flavonoids content (TFC), kuromanin concentration (KC), and antioxidant activity (antiradical activity (ARA) and Trolox equivalent antioxidant capacity assay (TEAC)) of chokecherries were measured. "Swell drying" is a drying process coupling convective airflow drying to the Instant Controlled Pressure Drop (DIC) expansion. A central composite rotatable design was applied to optimize the DIC variables and responses. Results showed that both freezing and swell drying effectively preserve the TPC, TFC, KC, and ARA. Moreover, SD samples also presented the highest TEAC. Contrary, in the case of CAD, it caused the highest losses of both antioxidant content and activity. Swell drying remedies the shrinkage and collapsing of dried food structure, which results in a better antioxidants extraction.

In this paper, polysaccharides from the seeds of three species of genus Glycyrrhiza were extracted to investigate the physicochemical properties, structural characteristics and antioxidant activities. The polysaccharides were composed of xylose, mannose, galactose, and glucose with different molar ratio. Fourier transform infrared spectroscopy showed the presence of key functional groups of polysaccharides whereas scanning electron microscopy analysis revealed the characteristic morphology of different polysaccharides, and thermogravimetric analysis exhibited good thermal stability of all samples. The antioxidant activities of polysaccharides were evaluated in vitro. All the three polysaccharides demonstrated strong reducing power, as well as scavenging activity against DPPH, ABTS, and hydroxyl free radicals. Antioxidant assays indicated that all the polysaccharides have obvious antioxidant activities and possess a potential development and application value in food, cosmetics as well as pharmaceutical industries.

Hypericum scabrum L. has been widely used in traditional medicine for the treatment of many diseases just as the other Hypericum species. In the present study, the antiradical, antimicrobial and cytotoxic activities of water and ethanol extracts of H. scabrum flowers were investigated. Their phytochemical contents and composition were also determined. The water and ethanol extracts are better scavenged ABTS (97.89 and 98.99%) and OH radicals (96.36 and 97.33%); the water extract is better scavenged DPPH radicals (91.66%) than the standard antioxidant BHA (94.33, 85.19, 90.16%, respectively). Flowers of H. scabrum contain flavonoids, phenolic acids, vitamins and phytosterols, dominated by catechin, vanillic acid, vitamin K and ergosterol. The extracts exhibit a strong cytotoxic activity against MCF-7, HCT-116, and LNCaP cancer cell lines. It is found that their antimicrobial activities are higher than the standard antibiotics. These results indicate that H. scabrum flowers have potent antiradical, antimicrobial and cytotoxic activities.

This study evaluates the antioxidant activity of cannabidiol (CBD), added to model systems of refined olive (ROO) and sunflower (SO) oils, by measuring the peroxide value, oxidative stability index (OSI), electron spin resonance (ESR) forced oxidation, and DPPH• assays. Free acidity, a parameter of hydrolytic rancidity, was also examined. CBD was compared using the same analytical scheme with α-tocopherol. CBD, compared to α-tocopherol, showed a higher scavenging capacity, measured by DPPH• assay, but not better oxidative stability (OSI) of the oily systems considered. In particular, α-tocopherol (0.5%) showed an antioxidant activity only in SO, registered by an increase of more than 30% of the OSI (from 4.15 ± 0.07 to 6.28 ± 0.11 h). By ESR-forced oxidation assay, the concentration of free radicals (µM) in ROO decreased from 83.33 ± 4.56 to 11.23 ± 0.28 and in SO from 19.21 ± 1.39 to 6.90 ± 0.53 by adding 0.5% α-tocopherol. On the contrary, the addition of 0.5% CBD caused a worsening of the oxidative stability of ROO (from 23.58 ± 0.32 to 17.28 ± 0.18 h) and SO (from 4.93 ± 0.04 to 3.98 ± 0.04 h). Furthermore, 0.5% of CBD did not lower dramatically the concentration of free radicals (µM) as for α-tocopherol, which passed from 76.94 ± 9.04 to 72.25 ± 4.13 in ROO and from 17.91 ± 0.95 to 16.84 ± 0.25 in SO.

This study investigated the phytochemical characteristics and antioxidant activity in leaves, roots, stem, flower, and seed parts of Datura alba (D. alba). The study also assessed the heavy metal (Cr, Mn, Zn, and Cu) accumulation in each part of the plant. Among the phytochemicals, alkaloids were found only in leaves while tannins, flavonoids, and phenols were present in all parts of the plant. For antioxidant activity, free radical scavenging assay for 2,2-diphenyl-1-picrylhydrazyl (DPPH) was performed using ascorbic acid as the standard. Higher activity was shown by stem extract in methanol and leaf extract in n-hexane, ethyl acetate, and chloroform. Furthermore, all the target heavy metals were detected in all plant sections with the highest concentration of Zn in leaves and Cu in stem, root, flower, and seed. Due to stronger antioxidant potential and phytochemical composition, D. alba could prove as valuable prospect in pharmaceutical formulations by taking part in the antioxidant defense system against generation of free radicals.

This work proposes a new method for the in vitro evaluation of the effect of UV irradiation on the production of free radicals and other reactive species during the photodecomposition of drugs. The method was based on the UV irradiation of antibiotics molecules to generate excited states that undergo to homolytic bond cleavages. These reactive species can be detected by their ability to oxidize the luminol, producing the electronically excited aminophtalate, which decays to the ground state releasing electromagnetic radiation in the visible zone of the spectrum. This method was applied to penicillin G, nafcillin, azlocillin and neomycin dissolved in water. It was found that the intensity of the luminol chemiluminescence emission (CL) was proportional to the concentration and dependent on the molecular structure of these drugs. Under the optimized conditions, it was found that penicillin and azlocillin were the most susceptible to photodegradation, while neomycin sulfate was the less affected by the UV light. It was observed that the addition to the antibiotics dissolutions of a hydro-alcoholic extract of petals of calyxes of Roselle reduced the CL intensity, indicating that the extract was able to scavenge the free radicals in the irradiated drugs. This result suggest that its addition to the antibiotics can help in the protection against the radicals formed during the exposition to solar light of patients treated with topic similar antibiotics.

OBJECTIVE: Currently, the cosmetic and medical industries are paying considerable attention to solve or prevent skin damage or diseases, such as hyperpigmentation and oxidation and free radical damage. In this study, the effective compounds in Myrica rubra fruit were extracted and studied the biological effects of these M. rubra fruit extracts. METHODS: In this study, we extracted M. rubra fruit using solutions with various ratios of water to ethanol (100:0, 50:50, 5:95) and studied the anti-melanogenesis, anti-oxidation and radical scavenging effects of these M. rubra fruit extracts on two melanoma cell lines: mouse melanoma (B16-F0) and human melanoma (A2058). The cytotoxicity, melanin synthesis, mushroom and cellular tyrosinase activities, enzyme kinetics, melanogenesis-related gene expression, melanogenesis-related protein secretion, radical DPPH scavenging activity and ROS inhibition after treatment with M. rubra fruit extracts were determined. RESULTS: The results showed that the water extract of M. rubra fruit was less cytotoxic to the melanoma cell lines, effectively inhibited melanin synthesis and tyrosinase activity and down-regulated the gene expression and protein secretion of MITF and TRP-1. In addition, the M. rubra fruit extracts also showed the abilities to scavenge DPPH free radicals and suppress ROS production. Finally, the effective compounds in the water extract were Myricetin-O-deoxyhexoside, Quercetin-O-deoxyhexoside, and Kaempferol-O-hexoside determined by LC/MS/MS assay. CONCLUSION: Overall, the water extract of M. rubra fruit is a safe and effective melanin inhibitor and anti-oxidant and can be applied widely in the fields of cosmetics and medicine.

Free radicals are partially reduced form of metabolites of Nitrogen and Oxygen. These are highly reactive and potentially toxic compounds which are contributing factors in different chronic disease. The present study was aimed to determine antioxidant capability and reducing ability of coded polyherbal capsules (Arthitec 1 & Arthitec 2). DPPH (2,2'-diphenyl-1-picryl hydrazyl) assay is most commonly used method for gauging antioxidant capability of natural compounds. In this assay DPPH act as stable free radical which react with an antioxidant. For measuring reducing ability suspected antioxidant react with ferric tripyridyltriazine (Fe3± TPTZ) complex and convert ferric into ferrous. Results are evident that both capsule formulations Arthitec 1 & Arthitec 2 have promising antioxidant activity and reducing potential. Antioxidant potential of both coded capsules with varied concentrations (10, 50 and 100 µg/ml) were compared and in both cases scavenging activity and as well as reducing ability raised in a dose dependent manner just like standard Butylated hydroxyanisole (BHA).

In the present research, the essential oil composition, bioactive compounds, anatomical and antioxidant activity of Achillea aucheri were investigated at four phenological stages. The yield of essential oil ranged from 0.1 % (five leaves appearance stage) to 0.7 % (full flowering stage). So, the oils from the plants harvested at the full flowering stage provided higher essential oil contents than those at other stages. The analysis of the essential oils by GC/MS revealed the presence of a large number of components represented mainly by oxygenated monoterpenes (24.36-77.81 %). The compounds revealed high variation at different phenological stages. The major constituents of A. aucheri were carvacrol (1.07-19.17 %), camphor (5.44-9.99 %), δ-cadinene (0.20-13.88 %), thymol (2.15-7.80 %), germacrene D (4.50-7.24 %), 1,8-cineole (1.62-5.54 %) and α-terpineol (1.71-3.42 %). The total phenolic (51.63 mg tannic acid/g DW) and flavonoid (10.88 mg quercetin/g W) contents were more accumulated at the 50 % flowering stage than others. The results revealed that the accumulation of essential oil in A. aucheri increased as the plant grew from the five leaves appearance stage to the 100 % flowering stage with increasing the size and number of secretory structures. Finally, the extracts collected at the 50 % flowering stage showed the highest antioxidant activity as measured based on FTC and DPPH method with the IC50 values ranging from 719.65 µg/mL to 1039.17 µg/mL for the samples.

The phytochemical screening and antioxidant potential of bark and stem of Parthenocissus quinquefolia (L.) planch was assessed in order to verify its ethnopharmacological significance. All major secondary metabolites e.g. alkaloids, flavonoids, saponins, terpenoids, tannins, reducing sugars, cardiac glycosides and anthraquinones were present. Antioxidant activity was analysed by using five techniques which included DPPH Free Radical Scavenging Activity, FRAP (Ferric Reducing Antioxidant Power), TAA (Total Antioxidant Activity, TPC (Total Phenolic Content and MC (Metal Chelating) Activity. Ethanolic extract of bark showed the highest scavenging effects of 90.01±0.01%, with IC50 value of 24.32mg/ml. Aqueous stem extract showed best activity with IC50 value of 13.6±0.34mg/ml. The significance antioxidant potential indicates the effectiveness of bark and stem of P. quinquefolia in treatment of many diseases.

Arthrospira platensis, better known as Spirulina, is one of the most important microalgae species. This cyanobacterium possesses a rich metabolite pattern, including high amounts of natural pigments. In this study, we applied a combined strategy based on Fourier Transform Ion Cyclotron Resonance Mass Spectrometry (FT-ICR-MS) and Ultra High-Performance Liquid Chromatography (UHPLC) for the qualitative/quantitative characterization of Spirulina pigments in three different commercial dietary supplements. FT-ICR was employed to elucidate the qualitative profile of Spirulina pigments, in both direct infusion mode (DIMS) and coupled to UHPLC. DIMS showed to be a very fast (4 min) and accurate (mass accuracy ≤ 0.01 ppm) tool. 51 pigments were tentatively identified. The profile revealed different classes, such as carotenes, xanthophylls and chlorophylls. Moreover, the antioxidant evaluation of the major compounds was assessed by pre-column reaction with the DPPH radical followed by fast UHPLC-PDA separation, highlighting the contribution of single analytes to the antioxidant potential of the entire pigment fraction. ß-carotene, diadinoxanthin and diatoxanthin showed the highest scavenging activity. The method took 40 min per sample, comprising reaction. This strategy could represent a valid tool for the fast and comprehensive characterization of Spirulina pigments in dietary supplements, as well as in other microalgae-based products.

The synthesis of ethyl 4-oxo-1,4-dihydroquinoline-3-carboxylates (4, 5) was performed via the reaction of corresponding anilines with diethyl ethoxymethylenemalonate under conventional and also microwave promoted conditions. The treatment of 4 and 5 afforded the corresponding hydrazides (6 and 7). These hydrazides were converted to the corresponding carbo(thio)amides (9a-f and 10a-e) which were then subjected to an intramolecular cyclisation leading to the formation of quinolone-triazole hybrids (11a-f and 12a-e). The newly synthesized compounds were screened for their biological activities such as antioxidant capacity (AC) and acetylcholinesterase Activity. Inhibition of cholinesterases is an effective method to curb Alzheimer's disease, a progressive and fatal neurological disorder. A series of some novel quinolonederivatives were designed, synthesized, and their inhibitory effects on AChE were evaluated. We obtained our compounds and determined their anticholinesterase activities according to the Ellman's method. 9b and 10c showed the best AChE inhibition with 0.48 ±â€¯0.02 and 0.52 ±â€¯0.07, respectively. Docking studies were performed for the most active compounds (9b, 10c) and interaction modes with enzyme active sites were determined. As a result of these studies, a strong interaction between these compounds and the active sites of AChE enzyme was revealed.

Euonymus laxiflorus Champ., a medicinal herb collected in Vietnam, has been reported to show several potent bioactivities, including anti-NO, enzyme inhibition, hypoglycemic and antidiabetic effects. Recently, the antioxidant activity of Euonymus laxiflorus Champ. trunk bark (ELCTB) has also been reported. However, the active antioxidant and anti-NO constituents existing in ELCTB have not been reported in the literature. The objective of this study was to purify the active antioxidants from ELCTB and investigate the anti-NO effect of the major constituents. Twenty-two phenolics isolated from ELCTB, including 12 compounds newly isolated in this study (1⁻12) and 10 constituents obtained from our previous work, were evaluated for their antioxidant activity. Of these, 12 compounds (4⁻6, 9, 13⁻15, 18⁻22) showed a potent antioxidant capacity (FRS50 = 7.8⁻58.11 µg/mL), in comparison to α-tocopherol (FRS50 = 23 µg/mL). In the anti-NO activity tests, Walterolactone A (1a) and B (1b) ß-d-glucopyranoside (13) demonstrated the most effective and comparable activity to that of quercetin with max inhibition and IC50 values of 100%, 1.3 µg/mL, and 100%, 1.21 µg/mL, respectively. The crude extract and its major compounds showed no cytotoxicity on normal cells. Notably, three constituents (9, 11, and 12) were identified as new compounds, another three constituents, including 1, 7, and 8, were found to be new natural products, constituents 9 and 13 were determined to be new antioxidants, and compound 13 was reported to have novel potent anti-NO activity for the first time. The results of this study contribute to the enrichment of new natural products and compounds, as well as the novel biological activity of constituents isolated from Euonymus laxiflorus Champ. The current study also indicates ELCTB as a rich natural source of active phenolics. It is suggested that ELCTB could be developed as a health food with promising antioxidant and anti-NO effects, as well as other beneficial biological activities.

This study aimed to investigate antioxidant activities of a polysaccharide from Fallopia multiflora (Thunb.) Harald. F. multiflora polysaccharide (FMP) was prepared and partially characterized by high performance liquid chromatography, fourier transform infrared spectrum and anion chromatography, and its antioxidant activities both in vitro and in vivo were investigated. FMP, with a molecular weight of 17,758Da consisted of glucose. In vivo, FMP had high hydroxyl radical scavenging activity and reducing capacity. In vivo, FMP increased serum superoxide dismutase and glutathione peroxidise activities and decreased the level of malondialdehyde. These results indicated that FMP exhibits high antioxidant activity both in vitro and in vivo and might retard human ageing associated with free radicals.

AIM: The present study was carried out to assess a new series of triazoloquinazolines 1-40 for their antioxidant activities using 1,1-diphenyl-2-picryl hydrazyl radical scavenging, ferric reduction antioxidant power and reducing power capability assays. RESULTS: All triazoloquinazolines 1-40 exhibited antioxidant activity ranged from weak to moderate and high. The obtained findings revealed that the triazoloquinazolines 30, 36 and 38-40 have superiority among all compounds, demonstrating the highest capacity to deplete 1,1-diphenyl-2-picryl hydrazyl and free radicals, in relation to butylated hydroxyl toluene, as a synthetic antioxidant agent. CONCLUSION: Chemical modifications together with density functional theory study on the targets supplied us with some valuable clarifications about the required properties needed for the target compounds to be more active against free radicals.
