RESUMEN

The pharmaceutical industry must comply with the requirements for good manufacturing practices to reduce inherent contamination risks in the production process. Bacillus and related genera are among the main bacterial isolated from clean areas, raw material, and products in the pharmaceutical industries, but the correct identification of these species is still a challenge. The aim of this study was to characterize by phenotyping, protein profiling, and 16S rRNA gene sequencing Sutcliffiellahorikoshii strains (n = 6) isolated from an immunobiological pharmaceutical facility, and to propose the reclassification of Bacillus tianshenii to the genus Sutcliffiella, and Sutcliffiella tianshenii sp. nov. The strains were characterized by VITEK®2, matrix-assisted laser desorption ionization-time of flight/mass spectrometry (MALDI-TOF/MS) using VITEK®MS, and 16S rRNA gene sequencing analysis. MALDI-TOF/MS did not identify any strains that were identified by 16S rRNA as S. horikoshii. VITEK®2 showed false-positive results, with misidentification as B. sporothermodurans (reclassified as Heyndrickxia sporothermodurans) and Geobacillus thermoleovorans. After MALDI-TOF/MS database expansion, with the creation of SuperSpectrum, the strains were correctly identified as S. horikoshii. This study is the first report of isolation of S. horikoshii strains from a pharmaceutical industry. More studies are necessary to better understand the ability of S. horikoshii to contaminate the environment and products.

BACKGROUND: Metabolic syndrome (MS) is defined as a low-grade proinflammatory state in which abnormal metabolic and cardiovascular factors increase the risk of developing cardiovascular disease and neuroinflammation. Events, such as the accumulation of visceral adipose tissue, increased plasma concentrations of free fatty acids, tissue hypoxia, and sympathetic hyperactivity in MS may contribute to the direct or indirect activation of Toll-like receptors (TLRs), specifically TLR4, which is thought to be a major component of this syndrome. Activation of the innate immune response via TLR4 may contribute to this state of chronic inflammation and may be related to the neuroinflammation and neurodegeneration observed in MS. In this study, we investigated the role of TLR4 in the brain microcirculation and in the cognitive performance of high-fat diet (HFD)-induced MS mice. METHODS: Wild-type (C3H/He) and TLR4 mutant (C3H/HeJ) mice were maintained under a normal diet (ND) or a HFD for 24 weeks. Intravital video-microscopy was used to investigate the functional capillary density, endothelial function, and endothelial-leukocyte interactions in the brain microcirculation. Plasma concentrations of monocyte chemoattractant protein-1 (MCP-1), adipokines and metabolic hormones were measured with a multiplex immunoassay. Brain postsynaptic density protein-95 and synaptophysin were evaluated by western blotting; astrocytic coverage of the vessels, microglial activation and structural capillary density were evaluated by immunohistochemistry. RESULTS: The HFD-induced MS model leads to metabolic, hemodynamic, and microcirculatory alterations, as evidenced by capillary rarefaction, increased rolling and leukocyte adhesion in postcapillary venules, endothelial dysfunction, and less coverage of astrocytes in the vessels, which are directly related to cognitive decline and neuroinflammation. The same model of MS reproduced in mice deficient for TLR4 because of a genetic mutation does not generate such changes. Furthermore, the comparison of wild-type mice fed a HFD and a normolipid diet revealed differences in inflammation in the cerebral microcirculation, possibly related to lower TLR4 activation. CONCLUSIONS: Our results demonstrate that TLR4 is involved in the microvascular dysfunction and neuroinflammation associated with HFD-induced MS and possibly has a causal role in the development of cognitive decline.

RESUMEN

This study investigates the extent to which aspects of the social integration of the elderly are linked to higher levels of fruit and vegetable consumption. It involved a cross-sectional study based on data from the National Study of Dependency in the Elderly, with a sample of 3278 elderly individuals. The variables of social integration considered were: frequency of meeting with close relatives, other relatives and neighbors and friends in the last 12 months; participation in recreational activities and in community groups, in addition to housing arrangements and marital status. The number of servings of fruit and vegetables consumed per day was the dependent variable. Logistic regression analysis was then conducted. In the adjusted model, more frequent encounters with siblings, in-laws and nephews, participation in community groups or organizations, are factors that increase the possibility of consuming two or more servings of fruit and vegetables a day, versus 1 serving or none; the opposite was observed when living alone. The variables of social integration that can increase the possibility of attaining the recommendation of consumption of fruit and vegetables daily (5 or more) in the adjusted model are frequency of encounters with siblings, in-laws and nephews and living alone.

The aim of this study is to perform a literature review about non-surgical facial lifting with PDS threads, addressing its variety of types, indications, organism degradation, insertion plan, contrain dications, advantages and possible complications. The researches were conducted in the Pubmed, Bi reme, Scielo, Sumários de Revistas Brasileiras and BVS data bases, selecting relevant articles available between 2005 and 2020. After applying the inclusion and exclusion criteria, of the 44 articles found, 16 were selected for this study. Being used the following descriptors in Portuguese and English for research purposes: "Facial Harmonization", "Supporti Threads", "Face Lift". The use of facial support threads consists in a minimally invasive, painless, outpatient method, local anesthesia and imme diate effect. Faced with the possible risks that accompany the surgical facelift, people are looking for less invasive alternatives, such as the support wires. Although they cannot be considered as an alternative to conventional reparative surgery, patients often end up opting for a facelift because they feel safer, even with more modest results. It was observed that the use of PDO threads has been shown to be effective in facial rejuvenation when well indicated, despite the possibility of adverse effects, usually small and transient... RESUMEN

Introduction: Crustaceans have economic and ecological importance and the role of caridean crustaceans in lentic environments is crucial to maintain the trophic structure. The species Atya scabra is occasionally associated with fisheries, especially in Northeast and Northern Brazil. Objective: To determine the population structure, reproductive period, patterns of growth and distribution of size classes among sexes in a population of the prawn A. scabra in the De Contas River, Bahia, Brazil. Methods: Individuals were sampled in a period of one year by passing sieves through marginal vegetation and using dragnets. In the laboratory, we recorded the sex of the individuals and measured them using a pachymeter with 0.001 mm precision. The dimensions measured were CL (carapace length), TL (total length) and CW (carapace width). In order to analyze data, linear models for the relationships CL × CW were constructed and the type of allometry for each sex was determined. Individuals were grouped into size classes of 5 mm intervals. A chi-square test (χ2) was used to test deviation from the 1:1 sex ratio. The proportion of ovigerous females was registered for each month to determine the breeding period. Results: A total of 517 individuals were collected, 328 were males and 186 females of which 169 were ovigerous. Males showed a polymodal distribution in size classes and were significantly larger than females, which presented a unimodal distribution in size classes. The observed growth patterns demonstrated that both sexes present negative allometry. However, sexual dimorphism was detected when comparing the growth parameters. Ovigerous females were sampled the entire year and size-frequency data demonstrated that recruitment also occurs throughout the year, which allowed us to classify their reproduction as continuous. Conclusions: Our data is essential to draw sustainable fishery management strategies. We suggest a minimum capture size of 70 mm and capture restrictions from May to August.

This study investigates the color correspondence of resin cements and try-in pastes, and the color stability of bonded lithium disilicate ceramic disks. Resin composite disks were fabricated (n = 36) to serve as the background for lithium disilicate disks prepared in two thicknesses (0.5 and 1.0 mm, n = 18 each). Two brands were used for try-in and cement procedures: Variolink Veneer and AllCem Veneer. For baseline, water was applied between the ceramic disks and their respective backgrounds to achieve the control group. This set was subjected to color measurement using an intraoral measurement device (T0). The try-in was inserted between background and ceramic, and this set was subjected to color measurement (T1). After adhesive procedures, the ceramic disk was placed under cement, and color measurement was performed with uncured cement (T2) and 24 h after light-curing (T3). Each set was immersed in distilled water and thermal-cycled, with color measurement being performed after 10,000 (T4) and 20,000 (T5) cycles. Color differences were calculated by CIELab (rEab) and CIEDE2000 (rE00). Data were analyzed by two-way ANOVA for repeated measurements and Tukey's test (α=5%). There was color correspondence of try-in and resin cement for the Variolink system, regardless of the ceramic thickness (p > 0.05). For the AllCem system, the thickness significantly influenced the color measurement (p < 0.001). The Variolink system also demonstrated color stability after 20,000 thermal cycles with rEab < 3.46 and rE00 < 2.25. It was concluded that the color correspondence between a try-in and its respective cement may vary according to resin cement composition.
