The fouling phenomenon grabbed global attention and caused huge economic losses specifically in marine-related industries. Sessile behavior exposed the sponge to the risk of fouling. However, their bodies remained free from foulers, which were attributed to the chemical defense system. The objectives of this study were to determine the antibiofilm activity of the marine sponge, Stylissa carteri, and to characterize the isolated compound involved. The antibiofilm activity of S. carteri methanolic crude extract (MCE) and fractions was tested against biofilm-producing bacteria, Pseudomonas aeruginosa, using two different modes of crystal violet biofilm assays: preventive and detachment. Besides that, the disc-diffusion test was conducted to screen the antibacterial activity against gram-positive and gram-negative bacteria while a cytotoxicity assay was conducted on the HepG2 cell line. Bioassay-guided fractionation was carried out using vacuum liquid chromatography (VLC) and solid phase extraction using a C18 Sep-Pak Cartridge. The crystal compound was isolated and characterized through thin-layer chromatography (TLC), Fourier transform infrared (FTIR) spectroscopy, liquid chromatography-mass spectrometry (LCMS), and nuclear magnetic resonance (NMR) spectroscopy. The S. carteri MCE showed a promising result with a half-maximal inhibitory concentration (IC50) of 20.22 µg/mL in the preventive assay, while no IC50 was determined in the detachment assay since all inhibitions < 50%. The S. carteri MCE exhibited broad-spectrum antibacterial activity and displayed a non-cytotoxic effect. Fraction 4 from MCE of S. carteri (IC50 = 2.40 µg/mL) reduced the biofilm in the preventive assay at all concentrations and exhibited no antibacterial activity indicating the independence of antibiofilm from antibacterial properties. Based on the data obtained, an alkaloid named debromohymenialdisine (DBH) was identified from Fraction 4 of S. carteri MCE. In conclusion, S. carteri was able to reduce the establishment of the biofilm formed by P. aeruginosa and could serve as a prominent source of natural antifouling agents.

This present study aimed to investigate the effects of Gracilaria changii (Red seaweed) extract and its fraction on scavenger receptor class B type I (SR-BI) gene expressions. In vitro studies of the activity of G. changii extract and its fraction against the therapeutic target for hypocholesterolemia were conducted using high-density lipoprotein (HDL) receptor SR-BI via luciferase assay in HepG2 cells. In the current study, methanol crude extract (MCE) and Fraction-2 of G. changii showed the highest expression levels of the SR-BI gene via luciferase assay and increased SR-BI mRNA expression compared to the negative control. Metabolite profiling of the MCE of G. changii by GC-MS revealed nine major compounds, comprising various fatty acids, particularly hexadecenoic acid. Therefore, the MCE and Fraction-2 of G. changii have the potential to be explored in the treatment of hypercholesterolaemia to prevent or reduce the severity of atherosclerosis development.

Texture is an important sensory attribute for overall quality and consumer acceptance of prawns. However, texture is affected during cold storage due to the proteolytic activity of endogenous proteases, resulting in poor quality and a short shelf life. The objective of this study is to determine the inhibitory effects of Annona muricata leaves extract (AMLE) (0, 3, 10 and 20%) on the trypsin, cathepsin B and collagenase activities extracted from the cephalothorax of Macrobrachium rosenbergii. In addition, the textural changes in M. rosenbergii during 20 days of cold storage (4 °C) were also determined. M. rosenbergii were soaked in four different treatments: 0, 3, 10 and 20% AMLE and 1.25% sodium metabisulphate for 10 min at 4 °C. Protease activity was significantly (p < 0.05) reduced at 10 and 20% AMLE. Similarly, cathepsin B showed a significant (p < 0.05) low after treatment at 20% AMLE. The maximum inhibitory activity of trypsin was achieved at 20% AMLE and the standard inhibitor (Tosyl-L-lysyl-chloromethane hydrochloride (TLCK)) compared to the control. Whereas, the lowest collagenase activity was obtained at 20% AMLE compared to the control. These inhibitory effects further maintain the firmness of M. rosenbergii coated with 20% AMLE up to the eighth day of storage when compared to the control. Meanwhile, the highest penetration work was found in the M. rosenbergii coated with 20% AMLE at the twentieth day of storage. In conclusion, treatment at 20% AMLE could be used as a natural preservative to inhibit protease, trypsin and collagenase activity of M. rosenbergii and thus can maintain firmness for up to 8 days of storage.

Biofouling is defined as the excessive colonization process of epibiotic organisms, ranging from microfoulers to macrofoulers, on any submerged surface in water. Previous research has attempted to explore the antifouling activity of bacterial isolates due to the biofouling problems occurring worldwide. One solution is to inhibit the early stage of fouling using secondary metabolites produced by marine bacteria. This study aims to determine the antifouling activities of the marine microorganism P. aeruginosa and to characterize the bacteria isolated as a potential anti-biofouling agent. The bacterial isolate was cultured and isolated on a media culture. The bacteria culture extract was extracted using ethyl acetate and concentrated prior to the bioassay method. It was screened for antibacterial activities against Gram-positive and Gram-negative bacteria, such as Bacillus cereus, Streptococcus uberis, Pseudomonas sp., and Vibrio parahaemolyticus, using the disk diffusion technique. The extract was investigated to verify its bioactivity in the prevention of biofilm formation following the crystal violet assay and aquarium test. The results indicated the inhibition of activity through biofilm formation, with the highest percentage at 83% of biofilm inhibition at a concentration of 0.1563 mg/mL. The bacterial isolate at a concentration of 5% showed the highest reduction in bacteria colonies in the aquarium test (161.8 × 103 CFU/mL compared to 722.5 × 103 CFU/mL for the blank sample). The bacterial isolate was characterized through phenotypic and genotypic tests for species identification. It was identified as a Gram-stain-negative, aerobic, and long-rod-shaped bacteria, designated as RLimb. Based on the 16S rDNA gene sequencing analysis, RLimb was identified as Pseudomonas aeruginosa (accession number: OP522351), exhibiting a similarity of 100% to the described neighbor P. aeruginosa strain DSM 50071. These results indicated that these isolated bacteria can potentially be used as a substitute for toxic antifoulants to prevent the formation of microfoulers.

Giant freshwater prawn (Macrobrachium rosenbergii) is one of the important aquaculture species and quickly expanding in many countries. High demand and mass commercialization on M. rosenbergii regulating 18% of the international seafood business. Seafood products contend with various level across the supply chains and time to reach the consumers depending upon the marketing and delivery channels after harvesting. Therefore, these may cause biodeterioration such as melanosis (dark pigmentation) and microbial changes that limit the shelf life. This studies reveal the antioxidant properties from Annona muricata leaves extract and their effectiveness in inhibiting the polyphenoloxidase (PPO) activity and delaying the bacterial accumulation during 20 days of chilled storage. Five metabolites including coumarins, flavonoid, glycoside, terpenoids and steroid compound were found in A. muricata leaves extract. Total phenolic content and total flavonoid content of A. muricata were recorded at 191.24 ± 0.03 mgGAEg-1 and 1777.48 ± 1.08 mgQEg-1, respectively. Sixteen percent (16%) of A. muricata leaf extract effectively inhibit 82.41% PPO. Furthermore, 15% of A. muricata leaves extracts showed a significant reduced (p < 0.05) in total bacteria count during 20 days of chilled storage of M. rosenbergii. These conclude that the present of listed secondary metabolites and at approximately ~ 15-16% of A. muricata leaves extracts were effectively inhibiting the melanosis and prolong the shelf life for up to 8 days of M. rosenbergii stored at chilled condition. Therefore, A. muricata leaves extract is potential used as natural preservative agent in obtaining high quality seafood products.

Technology is evolving rapidly around the world, and the use of mobile devices is increasing every day. Today, everyone owns a mobile device, including young children. Parents provide and allow young children to use mobile devices for various purposes. Due to the fact of these circumstances, children begin to become comfortable with the use of mobile devices, and they are prone to excessive use. Therefore, the purpose of this study was to examine the influence of sociodemographic factors on excessive mobile device use among young children. Sociodemographic variables, including the child's gender, the child's age when starting to use a mobile device, the parent's educational level, household income, type of application used, and the purpose of giving a mobile device to the child, were selected as predictive factors. A cross-sectional survey study design with a quantitative approach was conducted. A simple random sampling technique was employed, and a total of 364 parents completed the adapted questionnaire, namely, the Problematic Mobile Phone Use Scale (PMPUS). Data were statistically analyzed using descriptive and binary logistic regression analysis. The findings revealed that gender, age of the child when starting to use mobile devices, and purpose of parents providing mobile devices significantly contributed to 77.7% of the variance to make children users with a problem. However, the parent's educational level, household income, and type of application did not significantly contribute to the problem of mobile device use. Later, this study discusses the research implication, limitation, and recommendation for future research based on the finding.

Cancer is an uncontrolled multiplication of cells. The desire efficacy and severe toxicity of current anticancer drugs urge exploring and investigating a better alternative to existing chemotherapeutics. Natural products of marine origin are excellent sources of potential new drugs of enhanced biological activities. OBJECTIVES: Thus, the cytotoxic effects along with investigating the mode of cell death exerted by fractions, AP-9, AP-THR, DS-8 and DS-9 fraction of Acanthaster planci, Diadema setosum sp., on the human cervical cancer cell line, HeLa. METHODS: The cytotoxicity of fractions has determined by using an MTS assay. The early and late apoptosis was studied by using the High content Screening (HCS) instrument. RESULTS: The four fractions produced effective cytotoxicity effects with IC50 values at 72hr of less than 20 µg/ml in the order of AP-9 > DS-9 > APTHR-9 > DS-8. The fraction s exhibited cytotoxicity via mediating apoptotic mode of cell death. The early apoptosis by exposure of phosphatidylserine to the outer leaflet of the plasma membrane and late apoptosis due to the presence of green stain (DNA fragmentation) in treated cells. CONCLUSION: The potent bioactive compounds might be responsible for inducing apoptosis in cancer cells and, thus, the potential to be a successful candidate for exploring upcoming chemotherapeutic drugs.

Mangrove plants are endowed with various biologically active compounds which have potent antibacterial and antioxidant properties. In present study, a bioactivity-guided fractionation for antibacterial and antioxidant active metabolites from the twigs of Avicennia officinalis collected from Kuala Selangor Nature Park, Selangor, Malaysia gave 13 major fractions. The antibacterial activity of A. officinalis fractions using well-diffusion showed strong selectivity on the Gram-positive bacteria (Staphylococcus epidermidis, S. aureus and Bacillus subtilis) with minimum inhibition concentration (MIC) values of 0.156-5.00 mg/mL. However, no antibacterial activities were observed on the Gram-negative bacteria (Vibrio cholera, Enterobacter cloacae and Escherichia coli). The active antibacterial fractions were further isolated using several chromatographic techniques to give two naphthofuranquinones, namely, avicenol C (1) and stenocarpoquinone B (2). Meanwhile, the antioxidant activity of A. officinalis fractions were evaluated using DPPH radical scavenging assay exhibited low antioxidant activities. Molecular structure of the naphthofuranquinones was elucidated using 1 D and 2 D NMR spectroscopy.

Because students' ability to use statistics, which is mathematical in nature, is one of the concerns of educators, embedding within an e-learning system the pedagogical characteristics of learning is 'value added' because it facilitates the conventional method of learning mathematics. Many researchers emphasize the effectiveness of cognitive apprenticeship in learning and problem solving in the workplace. In a cognitive apprenticeship learning model, skills are learned within a community of practitioners through observation of modelling and then practice plus coaching. This study utilized an internet-based Cognitive Apprenticeship Model (i-CAM) in three phases and evaluated its effectiveness for improving statistics problem-solving performance among postgraduate students. The results showed that, when compared to the conventional mathematics learning model, the i-CAM could significantly promote students' problem-solving performance at the end of each phase. In addition, the combination of the differences in students' test scores were considered to be statistically significant after controlling for the pre-test scores. The findings conveyed in this paper confirmed the considerable value of i-CAM in the improvement of statistics learning for non-specialized postgraduate students.
