RESUMEN

The aged wine spirit is a beverage with great aromatic complexity. Their volatile compounds with odorant power coming from the distillate and from the wood used in its ageing, and the interactions that take place in the process, enhanced by oxygen, all contribute to this complexity. Due to time and cost inherent of ageing wine spirits in wooden barrels, research has sought to develop more sustainable alternatives to do it. In this context, the present study compares, the effect of traditional (wooden barrel) and alternative system (stainless steel tank with dipped staves and micro-oxygenation), on the odorant and sensory profile of a wine spirit, using Limousin oak and chestnut wood, after 12 months of ageing. The results suggest that the ageing process is accelerated by the alternative ageing technology and the chestnut wood, and the corresponding wine spirits presented characteristics of greater sensory evolution and strong wood compounds extraction.

PURPOSE: Targeted radiation therapy has seen an increased interest in the past decade. In vitro and in vivo experiments showed enhanced radiation doses due to gold nanoparticles (GNPs) to tumors in mice and demonstrated a high potential for clinical application. However, finding a functionalized molecular formulation for actively targeting GNPs in tumor cells is challenging. Furthermore, the enhanced energy deposition by secondary electrons around GNPs, particularly by short-ranged Auger electrons is difficult to measure. Computational models, such as Monte Carlo (MC) radiation transport codes, have been used to estimate the physical quantities and effects of GNPs. However, as these codes differ from one to another, the reliability of physical and dosimetric quantities needs to be established at cellular and molecular levels, so that the subsequent biological effects can be assessed quantitatively. METHODS: In this work, irradiation of single GNPs of 50 nm and 100 nm diameter by X-ray spectra generated by 50 and 100 peak kilovoltages was simulated for a defined geometry setup, by applying multiple MC codes in the EURADOS framework. RESULTS: The mean dose enhancement ratio of the first 10 nm-thick water shell around a 100 nm GNP ranges from 400 for 100 kVp X-rays to 600 for 50 kVp X-rays with large uncertainty factors up to 2.3. CONCLUSIONS: It is concluded that the absolute dose enhancement effects have large uncertainties and need an inter-code intercomparison for a high quality assurance; relative properties may be a better measure until more experimental data is available to constrain the models.

The goal of this study was to analyse the spatial pattern of tuberculosis (TB) mortality using different approaches, namely: mortality rates (MR), spatial relative risks (RR) and Bayesian rates (Global and Local) and their association with human development index (HDI), Global and its three dimensions: education, longevity and income. An ecological study was developed in Curitiba, Brazil based on data from Mortality Information System (2008-2014). Spatial scan statistics were used to compute RR and identify high-risk clusters. Bivariate Local Indicator of Spatial Associations was used to assess associations. MR ranged between 0 and 25.24/100.000 with a mean (standard deviation) of 1.07 (2.66). Corresponding values for spatial RR were 0-27.46, 1.2 (2.99) and for Bayesian rates (Global and Local) were 0.49-1.66, 0.90 (0.19) and 0-6.59, 0.98 (0.80). High-risk clusters were identified for all variables, except for HDI-income and Global Bayesian rate. Significant negative spatial relations were found between MR and income; between RR and HDI global, longevity and income; and Bayesian rates with all variables. Some areas presented different patterns: low social development/low risk and high risk/high development. These results demonstrate that social development variables should be considered, in mortality due TB.

The distribution of radiopharmaceuticals in tumor cells represents a fundamental aspect for a successful molecular targeted radiotherapy. It was largely demonstrated at microscopic level that only a fraction of cells in tumoral tissues incorporate the radiolabel. In addition, the distribution of the radionuclides at sub-cellular level, namely inside each nucleus, should also be investigated for accurate dosimetry estimation. The most used method to perform cellular dosimetry is the MIRD one, where S-values are able to estimate cellular absorbed doses for several electron energies, nucleus diameters, and considering homogeneous source distributions. However the radionuclide distribution inside nuclei can be also highly non-homogeneous. The aim of this study is to show in what extent a non-accurate cellular dosimetry could lead to misinterpretations of surviving cell fraction vs dose relationship; in this context, a dosimetric case study with 99mTc is also presented. METHODS: The state-of-art MCNP6 Monte Carlo simulation was used in order to model cell structures both in MIRD geometry (MG) and MIRD modified geometries (MMG), where also entire mitotic chromosome volumes were considered (each structure was modeled as liquid water material). In order to simulate a wide energy range of Auger emitting radionuclides, four mono energetic electron emissions were considered, namely 213eV, 6keV, 11keV and 20keV. A dosimetric calculation for 99mTc undergoing inhomogeneous nuclear internalization was also performed. RESULTS: After a successful validation step between MIRD and our computed S-values for three Auger-emitting radionuclides (99mTc, 125I and 64Cu), absorbed dose results showed that the standard MG could differ from the MMG from one to three orders of magnitude. These results were also confirmed by considering the 99mTc spectrum emission (Auger and internal conversion electrons). Moreover, considering an inhomogeneous radionuclide distribution, the average electron energy that maximizes the absorbed dose was found to be different for MG and MMG. CONCLUSIONS: The modeling of realistic radionuclide localization inside cells, including a inhomogeneous nuclear distribution, revealed that i) a strong bias in surviving cell fraction vs dose relationships (taking to different radiobiological models) can arise; ii) the alternative models might contribute to a more accurate prediction of the radiobiological effects inherent to more specific molecular targeted radiotherapy strategies.

The odorants compounds of aged wine brandies comprise compounds deriving from the wood, from the distillate and from the reactions that occur inside the barrel. The aim of this work was to study the kinetics of the odorant compounds of a wine brandy during two years of ageing in two ageing systems. The odorant compounds in the analysed brandies changed significantly over the time, but with different evolution patterns. The wood related compounds increased over time, with the highest increase in the first months of ageing. The kinetics of cis, trans-ß-methyl-γ-octalactone, acetovanillone and of seven volatile phenols are established for the first time in brandies. Moreover, a significant effect of the ageing system was found on the kinetics of the wood related compounds. These results pointed out the interest of these compounds as a tool to discriminate different ageing technologies.

In the image quality assessment for digital breast tomosynthesis (DBT), a breast phantom with an average percentage of 50 % glandular tissue is seldom used, which may not be representative of the breast tissue composition of the women undergoing such examination. This work aims at studying the effect of the glandular composition of the breast on the image quality taking into consideration different sizes of lesions. Monte Carlo simulations were performed using the state-of-the-art computer program PENELOPE to validate the image acquisition system of the DBT equipment as well as to calculate the mean glandular dose for each projection image and for different breast compositions. The integrated PENELOPE imaging tool (PenEasy) was used to calculate, in mammography, for each clinical detection task the X-ray energy that maximises the figure of merit. All the 2D cranial-caudal projections for DBT were simulated and then underwent the reconstruction process applying the Simultaneous Algebraic Reconstruction Technique. Finally, through signal-to-noise ratio analysis, the image quality in DBT was assessed.

Proton and ion beams are radiotherapy modalities of increasing importance and interest. Because of the different biological dose response of these radiations as compared with high-energy photon beams, the current approach of treatment prescription is based on the product of the absorbed dose to water and a biological weighting factor, but this is found to be insufficient for providing a generic method to quantify the biological outcome of radiation. It is therefore suggested to define new dosimetric quantities that allow a transparent separation of the physical processes from the biological ones. Given the complexity of the initiation and occurrence of biological processes on various time and length scales, and given that neither microdosimetry nor nanodosimetry on their own can fully describe the biological effects as a function of the distribution of energy deposition or ionization, a multiscale approach is needed to lay the foundation for the aforementioned new physical quantities relating track structure to relative biological effectiveness in proton and ion beam therapy. This article reviews the state-of-the-art microdosimetry, nanodosimetry, track structure simulations, quantification of reactive species, reference radiobiological data, cross-section data and multiscale models of biological response in the context of realizing the new quantities. It also introduces the European metrology project, Biologically Weighted Quantities in Radiotherapy, which aims to investigate the feasibility of establishing a multiscale model as the basis of the new quantities. A tentative generic expression of how the weighting of physical quantities at different length scales could be carried out is presented.

There is growing evidence that the radiation effects at low doses are not adequately described by a simple linear extrapolation from high doses, due, among others, to bystander effects. Though several studies have been published on this topic, the explanation of the mechanisms describing the bystander effects remains unclear. This study aims at understanding how the bystander signals are or can be propagated in the cell culture, namely if the number of irradiated cells influences the bystander response. An A549 cell line was exposed to several doses of α-particles, being the bystander response quantified in two non-irradiated areas. The radius of irradiated areas differs by a factor of 2, and the non-irradiated areas were optimally designed to have the same number of cells. Our results show evidence for bystander effects occurring in cells far away from the irradiated ones, meaning that bystander signals can easily spread throughout the cell culture. Additionally, our study highlights that the damage caused by radiation on the surrounding of irradiated areas could be different according to the number of irradiated cells, i.e., for the same dose value; the overall cellular damage could be different.

This study provides innovative information on the influence of new technologies of ageing (stainless steel tanks with wood staves or wood tablets of chestnut or Limousin oak), in comparison with traditional technology (oak wooden barrels), on the extraction/oxidation kinetics of low molecular weight compounds of wine brandy. The brandy was sampled and analysed by HPLC during the first year of ageing. The results show that most of the compounds tend to increase over the time, but their extraction/oxidation kinetics depend on the ageing technology. The wooden barrels promote greater enrichment in the majority of the compounds. However, gallic acid, ellagic acid and syringaldehyde, and vanillin and 5-methylfurfural, which are strong antioxidants and key-odourant compounds, respectively, present higher contents in the brandy aged with the alternative technologies. Chestnut proves to be a suitable alternative to Limousin oak for the ageing of brandy in all the studied technologies, inducing faster evolution and high quality.

Understanding the effects to human health resulting from exposure to low doses of ionizing radiation is a persisting challenge. No one questions the deleterious consequences for humans following exposure to high radiation doses; however, in the low dose range, the complex and to some extent unknown cellular responses raise important misgivings about the resulting protective or potentially detrimental effects. Bystander effects are involved in low dose exposures, being characterized by the appearance in unirradiated cells of a cellular damage associated with direct radiation exposure. The purpose of our work was to assess, by using clonogenic and micronuclei assays, the dose and time dependence of the bystander response after cells exposure to very low doses of α-particles and to evaluate its importance in the overall induced damage. The study includes an irradiated cells culture, a medium transfer culture with non-irradiated cells and a culture with irradiated cells after centrifugation. We observed a non-negligible contribution of the bystander effects in the overall cellular damage. Low-dose hyper-sensitivity was observed for medium transfer and irradiated cells after centrifugation cultures. Delayed and earlier cellular damage were similar in almost all experiments, suggesting an effectiveness of irradiated medium to induce a bystander response soon after irradiation.

Wooden barrels are used in the ageing or maturation of many alcoholic beverages, namely brandies and wines. However, the high costs related to ageing in wooden barrels have led to a search for alternative technologies. In this study we examined the application of wood fragments to the beverage in order to promote an accelerated ageing. We evaluated the sensory and chemical modifications in brandy aged in presence of two types of wood fragments, from two different woods (Limousin oak wood and Portuguese chestnut wood), and compared those with a brandy aged in wooden barrels. The results of the analysis of variance revealed more significant effects of wood botanical species than the ageing system on the sensory attributes. Concerning the ageing system, significant differences in brandy colour attributes were found, namely golden, topaz and greenish; olfactory attributes such as alcoholic, toasted and coffee; and the gustatory attribute, bitter. The brandies aged in the presence of wood tablets presented the highest intensities of topaz and greenish colour, toasted and coffee odours, while the brandies aged in wooden barrels presented the highest intensities of golden colour, alcohol odour and bitter taste. However, the overall quality of the brandies was similar. The analysis of odourant compounds showed a great discrimination of the brandies based on the ageing system. The brandies aged in wooden barrels presented the highest levels of several ethyl esters, acids, furanic aldehydes and the lowest levels of volatile phenols. Thus, considering the overall quality of the brandies, these results suggest the use of wood fragments to be an interesting alternative technology. On the other hand, the chemical analysis of the brandies showed the possibility of discriminating the ageing technologies based on odourant compound levels.

CONTEXTUALIZAÇÃO: As lesões nervosas periféricas podem comprometer atividades diárias de um indivíduo e resultam em perda da sensibilidade e motricidade do território inervado. OBJETIVO: Com o intuito de acelerar os processos regenerativos, objetivou-se analisar a influência da aplicação do laser de arsenieto de gálio e alumínio (AsGaAl, 660Nm) sobre a recuperação funcional do nervo ciático de ratos. MATERIAIS E MÉTODOS: O nervo ciático de 12 ratos Wistar foi submetido à lesão por neurotmese e anastomose epineural e divididos em dois grupos: controle e laserterapia. Após a lesão, utilizou-se o laser de GaAlAs, 660Nm, 4J/cm², 26,3mW, feixe de 0,63cm², em três pontos eqüidistantes sobre a lesão, por 20 dias. As impressões das pegadas dos animais foram obtidas antes e após (sete, 14 e 21 dias pós-operatórios) o procedimento cirúrgico e calculou-se o índice funcional do ciático (IFC). RESULTADOS: A comparação do IFC não resultou em diferença significante (p>0,05) entre os grupos. CONCLUSÕES: Conclui-se que os parâmetros e métodos empregados na laserterapia demonstram resultados nulos sobre o IFC no período avaliado.


CONTEXT: Peripheral nerve injuries result in sensory and motor losses in the innervated area and can hinder individuals' daily activities. Objective: The objective was to analyze the influence of applying gallium-aluminum-arsenide (GaAlAs) laser (660Nm) on the functional recovery of the sciatic nerve in rats. METHODS: The sciatic nerve of 12 Wistar rats was subjected to injury consisting of neurotmesis and epineural anastomosis. The rats were divided into two groups: control and laser therapy. After the injury, a GaAlAs laser was used (660Nm, 4J/cm², 26.3mW and 0.63cm² beam) at three equidistant points on the injury, for 20 days. Footprint impressions were obtained from the animals before and seven, 14 and 21 days after the surgical procedure and the sciatic functional index (SFI) was calculated. RESULTS: Comparison of the SFI did not show any significant difference (p>0.05) between the two groups. CONCLUSIONS: The parameters and methods used for the laser therapy did not produce any effect on the SFI over the period evaluated.

RESUMEN

The Monte Carlo simulation programs PENELOPE and MCNPX have been used for simulating the dose rate distribution in a (60)Co gamma irradiator. The simulated isodose curves obtained for each simulation code were validated comparing them to the dose measurements performed with a Fricke solution, which is a standard dosemeter widely used in radiation processing for calibration purposes. The agreement between the simulated values and the measurements indicates the effectiveness of both codes in performing the dose-mapping simulation for gamma irradiators.

A proposta deste estudo foi verificar a diferença entre o ângulo quadricipital em indivíduos sintomáticos e assintomáticos, em duas diferentes situações de exame, com o quadríceps relaxado e em contração isométrica voluntária máxima (CIVM) através da mensuração radiográfica para contribuir na avaliação e tratamento de pacientes com disfunção femoropatelar (DFP). Foram avaliadas 20 mulheres (40 joelhos), com idade média de 21 anos, através de método radiológico padronizado. Os indivíduos foram posicionados em decúbito dorsal com um estabilizador podálico em "U", com os membros inferiores relaxados, com a utilização de película de chumbo sobre a tuberosidade anterior da tíbia. Para a análise estatística foram utilizadas as médias dos grupos assintomático e sintomático, em estado de relaxamento e em CIVM, e o teste t de Student, com nível de significância de p < 0,05. Os valores médios do ângulo Q para os assintomáticos foram de 17,15º em relaxamento e de 14,5º em CIVM, enquanto os sintomáticos apresentaram 21,45º e 15,8º, respectivamente. Nos resultados para a análise da igualdade entre os grupos sintomáticos e assintomáticos no estado de relaxamento obteve-se p = 0,004, e para o estado de contração isométrica voluntária máxima, p = 0,29. Considerando os dados obtidos no presente estudo, pode-se verificar que em estado de relaxamento há diferença entre o valor do ângulo Q entre indivíduos sintomáticos e assintomáticos, sendo este maior nos portadores da DFP, enquanto que em estado de contração isométrica máxima do músculo quadricipital não houve diferença estatística, ocorrendo redução do ângulo em ambos os grupos.


The purpose of this study was to analyze the difference between the angle of the quadriceps in symptomatic and asymptomatic individuals in two different examination situations, having the quadriceps relaxed and in a maximal voluntary isometric contraction (MVIC) through radiographic measurement, aiming to contribute to the assessment and treatment of patients with patelofemoral disorder (PFD). Through the standard radiological method twenty 21 years old mean women (40 knees) were assessed. All individuals were positioned supine using a U-podalic stabilizer, having their lower limbs relaxed, using a plumb film on the anterior tuberosity of the tibia. For the statistical analysis, the averages for the asymptomatic and symptomatic groups in a relaxed and MVIC status, as well as the Student's t-test with p < 0.05 significance level were used. The mean values to the Q angle compared to the asymptomatic group were 17.15º on relaxation, and 14.5º on MVIC, while the asymptomatic group presented 21.45º, and 15.8º, respectively. The results in the equality analysis between the symptomatic and asymptomatic groups on the relaxed status attained a p = 0.004, and to the maximal voluntary isometric contraction, p = 0.29. Considering the data attained in the present study, it can be verified that in a relaxing status, there is a difference between the value of the Q angle among symptomatic and asymptomatic individuals, being found a higher value in the FPD bearers, while in a maximal isometric contraction of the quadriceps muscle no statistical difference was found in the present study, with a reduction in the angle in both groups.


La propuesta de este estudio era verificar la diferencia entre el ángulo cuadricipital en los individuos sintomáticos y asintomáticos, en dos situaciones diferentes del examen físico; con el cuadriceps relajado y en el máximo de la reducción isométrico voluntario (CIVM) a través de la medida radiográfica para contribuir en la evaluación y el tratamiento de pacientes con trastorno fémoro-patelar (DFP). Se estimaron 20 mujeres (40 rodillas), con la edad media de 21 años, a través de método radiológico estandardizado. Los individuos se pusieron en decúbito con un estabilizador podálico en "U", con los miembros inferiores relajados, con el uso de película de plomo en la tuberosidad anterior de la espinilla. Para el análisis estadístico se usaron las medias del grupo asintomático y sintomático, en estado de relajación y en CIVM, y la prueba del test t, con un nivel de significancia de p < 0,05. Los valores medios del ángulo Q para los asintomáticos fueron de 17,15º en la relajación y de 14,5º en CIVM, mientras los sintomáticos presentaron 21,45º y 15,8º, respectivamente. En los resultados para el análisis de la igualdad entre los grupos sintomático y asintomático en el estado de relajación se obtuvo p = 0,004, y para el estado de reducción isométrica voluntaria máximo, p = 0,29. Considerando los datos obtenidos en el estudio presente, puede verificarse que en el estado de relajación hay diferencia entre el valor del ángulo Q entre los individuos sintomáticos y asintomáticos, siendo este más grande en los portadores de DFP, mientras en el estado de contracción isométrica máxima del músculo cuadriceps no presentó diferencia estadística, habiendo reducción del ángulo en ambos los grupos.

Oak and chestnut species have been largely used for the aging of brandies, but nowadays chestnut is rarely used. There have been no previous studies regarding the cooperage utilization of chestnut wood. This study provides, for the first time, specific information about the characterization of the northern Portuguese Castanea sativa wood and examines the influence of this wood and its heat treatment on the chemical composition of two-year-aged brandies, by the quantitative determination (HPLC) of low molecular weight phenolic compounds. The predominance of gallic acid among the analyzed extractable compounds both in chestnut wood and in the corresponding aged brandies was remarkable.
