RESUMEN

Insects are a natural source of feed for fish and have received more attention as a potential source of sustainable high-quality protein. However, contrasting results in different feeding trials have been ascribed to the chitin contained in the exoskeleton of insects and highlighted the importance of developing reliable methods for the quantification of chitin to draw meaningful conclusions about its effect on fish health. A rapid method based on the hydrolysis of chitin into glucosamine and further quantification by liquid chromatography tandem mass spectrometry is evaluated. The method offers good selectivity, linearity, limit of detection (1.08 × 10-5 % w/v or 5.38 × 10-4 % w/w), limit of quantification (3.26 × 10-5 % w/v or 1.63 × 10-3 % w/w), trueness (88.39-109.29 %) and precision (2.24-10.72 %). The quantitative method was successfully applied to real samples of fish feed supplemented with chitin from black soldier fly (Hermetia illucens) larvae.

The level of perfluorinated alkyl substances (PFAS) was determined in North East Arctic cod (Gadus morhua) liver samples from 15 Norwegian fjords and harbors. Five harbors in the eastern part of Norway, six harbors in the western part and four harbours in the northern part. A total of 200 samples were analyzed for 16 PFAS. Determination of PFAS were carried out by LC-MS/MS following sample clean up by solid phase extraction and ultracentrifugation. The predominating PFAS was PFOS, which was found to be higher than the level of quantification (1.5 µg kg-1 wet weight) in 72% of the samples. The highest level of PFOS found was 21.8 µg kg-1 wet weight in a sample from Kragerø in the eastern part of Norway. A significantly higher level of PFOS was found in the eastern fjords and harbors compared to fjords and harbors in the western and northern part of Norway. Within the northern fjords and harbors elevated PFOS levels were found in Narvik, which may indicate a local source there. Variations in PFOS of the cod livers thus reflect differences in levels of pollution between the areas.

It is well-known that triacylglycerol (TAG) ions are suppressed by phospholipid (PL) ions in regiospecific analysis of TAG by mass spectrometry (MS). Hence, it is essential to remove the PL during sample preparation prior to MS analysis. The present article proposes a cost-effective liquid-liquid extraction (LLE) method to remove PL from TAG in different kinds of biological samples by using methanol, hexane and water. High performance thin layer chromatography confirmed the lack of PL in krill oil and salmon liver samples, submitted to the proposed LLE protocol, and liquid chromatography tandem MS confirmed that the identified TAG ions were highly enhanced after implementing the LLE procedure.

The content of triacylglycerols (TAG) in krill oil is generally omitted from the labels of commercial supplements and unacknowledged in studies aimed at proving its health benefits. The present study demonstrates that TAG compounds, in addition to phospholipids and lysophospholipids, are an important lipid class in pure krill oil. The fatty acid composition of TAG molecules from krill oil and their distribution on the backbone of TAG structures were determined by gas chromatography and liquid chromatography tandem mass spectrometric, respectively. The content of omega 3 polyunsaturated fatty acids (n-3 PUFA) was similar to those reported in the literature for fish oil. It was estimated that 21 % of n-3 PUFA were at the sn-2 position of TAG structures. To our knowledge, this is the first determination and structural characterization of TAG in pure krill oil supplements.
