RESUMEN

Objectives: To assess the impact of different concentrations TiO2-nt incorporated into a glass ionomer cement on the proliferation, mitochondrial metabolism, morphology, and pro- and anti-inflammatory cytokine production of cultured fibroblasts (NIH/3T3), whether or not stimulated by lipopolysaccharides (LPS-2 µg/mL, 24 h). Methods: TiO2-nt was added to KM (Ketac Molar EasyMix™, 3 %, 5 %, 7 % in weight); unblended KM was used as the control. The analyses included: Cell proliferation assay (n = 6; 24/48/72h); Mitochondrial metabolism assay (n = 6; 24/48/72h); Confocal laser microscopy (n = 3; 24/48/72h); Determination of biomarkers (IL-1ß/IL-6/IL-10/VEGF/TNF) by using both multiplex technology (n = 6; 12/18 h) and the quantitative real-time PCR assay (q-PCR) (n = 3, 24/72/120 h). The data underwent analysis using both the Shapiro-Wilk and Levene tests, and by generalized linear models (α = 0.05). Results: It demonstrated that cell proliferation increased over time, regardless of the presence of TiO2-nt or LPS, and displayed a significant increase at 72 h; mitochondrial metabolism increased (p < 0.05), irrespective of exposure to LPS (p = 0.937); no cell morphology changes were observed; TiO2-nt reverted the impact of KM on the secreted levels of the evaluated proteins and the gene expressions in the presence of LPS (p < 0.0001). Conclusions: TiO2-nt did not adversely affect the biological behavior of fibroblastic cells cultured on GIC discs.

Seagrass meadows provide several ecological functions that improve the overall ecological health of coastal systems and therefore, it is urgent to promote the restoration of such habitats. In Ria de Aveiro, a coastal lagoon in the Atlantic Coast of Portugal, a restoration initiative was responsible for transplanting the dwarf eelgrass Zostera noltei into a highly degraded area. This eelgrass was used as a nature-based solution (NbS) to mitigate some of the impacts of historical mercury contamination. Comparisons of key-species features (density and biomass), and some community-derived indicators (total density and biomass, species richness and Shannon-Wiener index) between the transplanted seagrass patch, their bare vicinities, and their counterpart habitats on the source area, provided signs of the effectiveness of the restoration action on the benthic communities' recovery. Indicators were higher within the restored meadow, and biomass derived indicators of the restored meadow were similar to the source meadow.

An innovative multiple regression analysis was used to evaluate metal/metalloid contamination in the surface sediments of a coastal lagoon. The concentrations of metals/metalloids were represented as a function of geochemical characteristics of the sediments (fine fraction, concentrations of organic carbon, Ca, Al, Mn) and distances between sampling points. The effect of distances on the concentrations were negligible for Li, Co, Ni, Ba, V, Cr, and only geochemical variables specific for each element explained its spatial variation. The concentrations of As, Cu, Zn and Pb were influenced by both geochemical and geographical distance variables, the latter representing the anthropogenic influence and the extent of transport of contaminants away from the upstream source. Enrichment of the sediment with Ba, As, Co, Cr and V was determined mainly by enrichment with Mn. The proposed approach is supplementary to the traditional utilization of enrichment factors, and is better suited for systems with anthropogenic influence.

The Paris Agreement signed by 195 countries in 2015 sets out a global action plan to avoid dangerous climate change by limiting global warming to remain below 2 °C. Under that premise, in situ experiments were run to test the effects of 2 °C temperature increase on the benthic communities in a seagrass bed and adjacent bare sediment, from a temperate European estuary. Temperature was artificially increased in situ and diversity and ecosystem functioning components measured after 10 and 30 days. Despite some warmness effects on the analysed components, significant impacts were not verified on macro and microfauna structure, bioturbation or in the fluxes of nutrients. The effect of site/habitat seemed more important than the effects of the warmness, with the seagrass habitat providing more homogenous results and being less impacted by warmness than the adjacent bare sediment. The results reinforce that most ecological responses to global changes are context dependent and that ecosystem stability depends not only on biological diversity but also on the availability of different habitats and niches, highlighting the role of coastal wetlands. In the context of the Paris Agreement it seems that estuarine benthic ecosystems will be able to cope if global warming remains below 2 °C.

This study aimed to assess the impact of trace element concentrations in fish assemblages of a recovering coastal lagoon. Fish, water and sediment were sampled in winter and summer in the Ria de Aveiro (Portugal). Multivariate analyses were used to examine the relationship between fish assemblages and environmental variables (physical-chemical parameters, contaminants and sediment grain size). In winter, fish density and biomass were mainly affected by the water turbidity, while Li concentration in the water column was found to be significant for fish biomass. During summer, a significant relationship was found between fish density and temperature, Hg, Li and Zn concentration in the sediment. These contaminants were mainly associated with the historically contaminated area, were Liza spp. and Dicentrarchus labrax appeared as dominant species. Environmental variables were not significant for fish biomass. The historical contamination in the Ria de Aveiro still seems to exert some influence on fish community structure.

Given their central role in mercury (Hg) excretion and suitability as reservoirs, bird feathers are useful Hg biomonitors. Nevertheless, the interpretation of Hg concentrations is still questioned as a result of a poor knowledge of feather physiology and mechanisms affecting Hg deposition. Given the constraints of feather availability to ecotoxicological studies, we tested the effect of intra-individual differences in Hg concentrations according to feather type (body vs. flight feathers), position in the wing and size (mass and length) in order to understand how these factors could affect Hg estimates. We measured Hg concentration of 154 feathers from 28 un-moulted barn owls (Tyto alba), collected dead on roadsides. Median Hg concentration was 0.45 (0.076-4.5) mg kg(-1) in body feathers, 0.44 (0.040-4.9) mg kg(-1) in primary and 0.60 (0.042-4.7) mg kg(-1) in secondary feathers, and we found a poor effect of feather type on intra-individual Hg levels. We also found a negative effect of wing feather mass on Hg concentration but not of feather length and of its position in the wing. We hypothesize that differences in feather growth rate may be the main driver of between-feather differences in Hg concentrations, which can have implications in the interpretation of Hg concentrations in feathers. Finally, we recommend that, whenever possible, several feathers from the same individual should be analysed. The five innermost primaries have lowest mean deviations to both between-feather and intra-individual mean Hg concentration and thus should be selected under restrictive sampling scenarios.

A mercury-cell chlor-alkali plant operated in Estarreja (North-western Portugal) for 50 years causing widespread environmental contamination. Although production by this process ceased in 2002, mercury contamination from the plant remains significant. The main objective of this study was to investigate mercury impact on the nearby environment and potential risks to local population. To assess the level of contamination soil samples were collected from agricultural fields in the vicinity of the plant, extending the study by taking samples of the predominant vegetation suitable for animal and human consumption, water samples, and fish species from a nearby coastal lagoon, to gain a preliminary insight into the potential for contamination of the terrestrial and aquatic food web. To determine population exposure to mercury, hair samples were collected from local residents. Total mercury concentration in the 0-15 cm layer of soil was found to be highly variable, ranging between 0.010 and 91 mg kg(-1), although mercury contamination of soils was found to be restricted to a confined area. Lolium perenne roots contained between 0.0070 and 2.0 mg kg(-1), and there is evidence that root systems uptake mercury from the soil. Levels of mercury in the aerial parts of plants ranged between 0.018 and 0.98 mg kg(-1). It appears that plants with higher mercury concentration in soils and roots also display higher mercury concentration in leaves. Total mercury concentration in water samples ranged between 12 and 846 ng L(-1), all samples presenting concentrations below the maximum level allowable for drinking water defined in the Portuguese law (1.0 microg L(-1)). Mercury levels in fish samples were below the maximum limit defined in the Portuguese law (0.5 mg kg(-1)), ranging from 0.0040 to 0.24 mg kg(-1). Vegetables collected presented maximum mercury concentration of 0.17 mg kg(-1). In general, food is not contaminated and should not be responsible for major human exposure to the metal. Mercury determined in human hair samples (0.090-4.2 mg kg(-1); mean 1.5 mg kg(-1)) can be considered within normal limits, according to WHO guidelines suggesting that it is not affecting the local population. Despite being subject to decades of mercury emissions, nowadays this pollutant is only found in limited small areas and must not constitute a risk for human health, should these areas be restricted and monitored. Considering the present data, it appears that the population from Estarreja is currently not being affected by mercury levels that still remain in the environment.
