Touch sensitivity generally declines with age, contributing to loss of manual dexterity and tactile function. We investigated how touch changes over the lifespan, using different tests and on three body sites. We used a classical test of force detection sensitivity, where calibrated monofilaments were applied passively to the right index finger pad, forearm, and cheek. In addition, at the index, we used an active touch spatial discrimination task, developed by our group. Spatial discrimination was estimated through participants' ability to evaluate the distance between parallel bands printed on acrylic plates. Data were collected from 96 healthy women, aged 20-75 years. Force detection and tactile spatial discrimination on the index deteriorated significantly with age; however, no change was found for tactile detection on the forearm or cheek. Tactile detection on the cheek remained remarkably highly sensitive throughout life. There was a significant positive relationship between force detection and spatial discrimination on the index. Further, force detection on the forearm was significantly associated with detection on the index and cheek. Our results suggest a decrease in touch perception with age on the index finger pad, yet a preservation of tactile sensitivity in hairy skin. This opens discussion about the impact of daily activities upon the glabrous hand skin and on the function of hairs in tactile sensitivity. We highlight the need for new methods in evaluating tactile sensitivity on hairy skin.

The human body is encompassed by a thin layer of tissue, the skin, which is heterogenous and highly specialized to protect the body and encode interactions with the external world. There is a fundamental scientific drive to understand its function, coupled with the need to preserve skin as we age, which impacts on our physiological and psychological well-being. In the present study, we aimed to define differences in touch perception between age groups and with skin cream application. We investigated touch on the finger, the forearm and cheek in younger (20-28 years, n = 22) and older (65-75 years, n = 22) females. We measured skin hydration, touch detection, finger spatial discrimination, forearm tactile pleasantness together with electrodermal activity, and perceptual ratings about cream use, skin dryness, and cosmetic habits. Glabrous finger skin became drier and touch performance was impaired with age, but these aspects were preserved in hairy skin. Skin moisturization immediately increased hydration levels, but did not significantly change touch perception. We also found that touch appreciation increased with age. We conclude that reduced finger capacity may impact self-evaluation of the skin and that long-term skin care strategies should focus on hydrating the hand to preserve touch capacities.

The forces that are developed when manipulating objects generate sensory cues that inform the central nervous system about the qualities of the object's surface and the status of the hand/object interaction. Afferent responses to frictional transients or slips have been studied in the context of lifting/holding tasks. Here, we used microneurography and an innovative tactile stimulator, the Stimtac, to modulate both the friction level of a surface, without changing the surface or adding a lubricant, and, to generate the frictional transients in a pure and net fashion. In three protocols, we manipulated: the frictional transients, the friction levels, the rise times, the alternation of phases of decrease or increase in friction to emulate grating-like stimuli. Afferent responses were recorded in 2 FAIs, 1 FAII, 2 SAIs and 3 SAIIs from the median nerve of human participants. Independently of the unit type, we observed that: single spikes were generated time-locked to the frictional transients, and that reducing the friction level reduced the number of spikes during the stable phase of the stimulation. Our results suggest that those frictional cues are encoded in all the unit types and emphasize the possibility to use the Stimtac device to control mechanoreceptor firing with high temporal precision.

Many studies have compared active and passive touch to understand how motor action shapes touch perception. Current views emphasize the difficulties in making such a comparison and promote investigating how motor strategies enable the filtering out of sensory inputs to reshape touch perception. Cybulska-Klosowicz et al. (Cybulska-Klosowicz A, Tremblay F, Jiang W, Bourgeon S, Meftah E-M, Chapman CE. J Neurophysiol 123: 1072-1089, 2020) suggest that primary somatosensory (S1) cortical remodeling of digit representation occurs during active touch. Here, alternative interpretations are proposed, and the relevance of studying multidigit scanning is emphasized.

C-tactile (CT) afferents were long-believed to be lacking in humans, but these were subsequently shown to densely innervate the face and arm skin, and to a lesser extent the leg. Their firing frequency to stroking touch at different velocities has been correlated with ratings of tactile pleasantness. CT afferents were thought to be absent in human glabrous skin; however, tactile pleasantness can be perceived across the whole body, including glabrous hand skin. We used microneurography to investigate mechanoreceptive afferents in the glabrous skin of the human hand, during median and radial nerve recordings. We describe CTs found in the glabrous skin, with characteristics comparable with those in hairy arm skin, and detail recordings from three such afferents. CTs were infrequently encountered in the glabrous skin and we estimate that the ratio of recorded CTs relative to myelinated mechanoreceptors (1:80) corresponds to an absolute innervation density of around seven times lower than in hairy skin. This sparse innervation sheds light on discrepancies between psychophysical findings of touch perception on glabrous skin and hairy skin, although the role of these CT afferents in the glabrous skin remains subject to future work.NEW & NOTEWORTHY Human touch is encoded by low-threshold mechanoreceptors, including myelinated Aß afferents and unmyelinated C-tactile (CT) afferents. CTs are abundant in hairy skin and are thought to code gentle, stroking touch that signals positive affective interactions. CTs have never been described in human glabrous skin, yet we show evidence of their existence on the hand, albeit at a relatively low density. Glabrous skin CTs may provide modulatory reinforcement of gentle tactile interactions during touch using the hands.

Conventional prosthetic arms suffer from poor controllability and lack of sensory feedback. Owing to the absence of tactile sensory information, prosthetic users must rely on incidental visual and auditory cues. In this study, we investigated the effect of providing tactile perception on motor coordination during routine grasping and grasping under uncertainty. Three transhumeral amputees were implanted with an osseointegrated percutaneous implant system for direct skeletal attachment and bidirectional communication with implanted neuromuscular electrodes. This neuromusculoskeletal prosthesis is a novel concept of artificial limb replacement that allows to extract control signals from electrodes implanted on viable muscle tissue, and to stimulate severed afferent nerve fibers to provide somatosensory feedback. Subjects received tactile feedback using three biologically inspired stimulation paradigms while performing a pick and lift test. The grasped object was instrumented to record grasping and lifting forces and its weight was either constant or unexpectedly changed in between trials. The results were also compared to the no-feedback control condition. Our findings confirm, in line with the neuroscientific literature, that somatosensory feedback is necessary for motor coordination during grasping. Our results also indicate that feedback is more relevant under uncertainty, and its effectiveness can be influenced by the selected neuromodulation paradigm and arguably also the prior experience of the prosthesis user.

Active sensing in biological system consists of emitting/receiving a periodic signal to explore the environment. The signal can be emitted toward distant objects, as in echolocation, or in direct contact with the object, for example, whisking in rodents. We explored the hypothesis that a similar mechanism exists in humans. Humans generate periodic signals at ~10 Hz during voluntary finger movements, which reflects a pulsatile motor command in the central nervous system. In the present study, we tested whether the ~10-Hz signal persists during the active exploration of textures and whether the textures' features can modulate the signal. Our results confirm our assumptions. The ~10-Hz signal persisted during active touch, and its amplitude increased with textures of higher friction. These findings support the idea that the ~10-Hz periodic signal generated during voluntary finger movements is part of an active sensing mechanism acting in a pulse-amplitude modulation fashion to convey relevant tactile information to the brain.NEW & NOTEWORTHY For the first time, we show that pulsatile motor output during voluntary movement of a finger persists during active exploration of a surface. We propose that this is part of an active sensing system in humans, with generation of an ~10-Hz signal during active touch that reinforces extraction of information about features of the touched surface.

Objective: Basic temporal dysfunctions have been described in patients with schizophrenia, which may impact their ability to connect and synchronize with the outer world. The present study was conducted with the aim to distinguish between interval timing and synchronization difficulties and more generally the spatial-temporal organization disturbances for voluntary actions. A new sensorimotor synchronization task was developed to test these abilities. Method: Twenty-four chronic schizophrenia patients matched with 27 controls performed a spatial-tapping task in which finger taps were to be produced in synchrony with a regular metronome to six visual targets presented around a virtual circle on a tactile screen. Isochronous (time intervals of 500 ms) and non-isochronous auditory sequences (alternated time intervals of 300/600 ms) were presented. The capacity to produce time intervals accurately versus the ability to synchronize own actions (tap) with external events (tone) were measured. Results: Patients with schizophrenia were able to produce the tapping patterns of both isochronous and non-isochronous auditory sequences as accurately as controls producing inter-response intervals close to the expected interval of 500 and 900 ms, respectively. However, the synchronization performances revealed significantly more positive asynchrony means (but similar variances) in the patient group than in the control group for both types of auditory sequences. Conclusion: The patterns of results suggest that patients with schizophrenia are able to perceive and produce both simple and complex sequences of time intervals but are impaired in the ability to synchronize their actions with external events. These findings suggest a specific deficit in predictive timing, which may be at the core of early symptoms previously described in schizophrenia.

The control of rhythmic action sequences may involve two distinct timing strategies, i.e., event-based and emergent timing, which are usually revealed through finger-tapping and circle-drawing tasks, respectively. There is a lively debate concerning the possibility of coexistence of the two modes of timing for the execution of a single task. If one considers emergent timing as simply an absence of explicit representation of a time interval, then by definition, the two modes of timing cannot coexist. However, if one considers that emergent timing engages control of another motor parameter, e.g., a control of movement through space rather than time, then the possibility of coexistence needs to be reassessed. In the present study, we designed a hybrid of finger-tapping and circle-drawing tasks for which the demands for space and time control were present at the same time in order to reassess the coexistence hypothesis. Seventy-eight participants performed a spatial-tapping task in which finger taps were to be produced in synchrony with a regular metronome to 6 visual targets presented around a virtual circle. The metronome set ten distinct tempi (1100-300 ms). Using autocorrelation analyses on timing variables, we show that motor timing was event-based at slow tempi and emergent at faster tempi. Through an analysis of the trajectory, we confirm that an increase in the spatial control of movement took place congruently with a switch from event-based to emergent timing modes. At these fast tempi, timing and spatial errors were correlated but only at the specific target location for which a dynamical anchor point was revealed. Hence, we conclude that the coding of emergent timing has a spatial nature from which emerge timing regularities. This spatio-temporal strategy insures the performance of sequential motor actions when cognitive effort is too high for the use of pure event-based timing strategies.
