RESUMEN

This experiment was conducted to evaluate the effects of dietary supplementation with Clostridium butyricum on laying performance, egg quality, serum parameters, and cecal microflora of laying hens in the late phase of production. Jinghong-1 strain laying hens (n = 960; 48 wk of age) were randomly allocated to 5 treatment groups with 6 replicates of 32 hens. Hens were fed with basal diet (control) and basal diet supplemented with 2.5 × 104 (CB1), 5 × 104 (CB2), 1 × 105 (CB3), and 2 × 105 (CB4) cfu/g C. butyricum for 10 wk. The results showed that egg production, egg mass, and eggshell strength increased quadratically as supplemental C. butyricum increased, and these responses were maximized in the CB2 group (P < 0.05). Compared with the control group, the addition of C. butyricum resulted in quadratic effects on serum total protein, uric acid, calcium, complement component C3 and catalase concentrations, and these responses were maximized or minimized in the CB2 group (P < 0.05). Linear and quadratic increases were observed in serum IgM, total superoxide dismutase, and glutathione peroxidase concentrations, and these responses were maximized in CB2 or CB3 group (P < 0.05). The addition of C. butyricum in the CB2 group resulted in linearly increasing levels of serum IgG concentration as compared with the control group (P < 0.05). Spleen index increased (P < 0.05) in the CB2 group. Hens fed with C. butyricum reduced (P > 0.05) the population of E. coli, while Bifidobacterium counts increased quadratically and maximized in the CB2 group (P < 0.05). In conclusion, the results indicated that dietary supplementation with C. butyricum (5 × 104 or 1 × 105 cfu/g) could improve laying performance and egg quality by promoting immune function, enhancing antioxidative capacity, and benefiting the cecal microflora of laying hens in the late phase of production.

This study was to determine the effects of dietary Zn-methionine (Zn-Met) supplementation on the laying performance, egg quality, antioxidant capacity, and serum parameters of laying hens. Jingh ong-1 strain laying hens (n = 720, 49 wk of age) were randomly assigned to 6 treatments with 6 replications of 20 birds. The control was fed a basal diet supplemented with 80 mg of Zn/kg as Zn sulphate of diet and the 5 groups were fed a basal diet supplemented with 20, 40, 60, 80, and 100 mg of Zn/kg as Zn-Met of diet for 10 wk, respectively. At the terminal experiment, there were significant differences between control and 80 mg/kg Zn-Met group in feed intake (P < 0.05) and feed conversion ratio (FCR) (P < 0.01). Egg weight (P < 0.05) and albumen height (P < 0.01) reduced and were not significantly influenced by supplemental 80 mg/kg Zn-Met group until being stored 15 d as compared to the control. Zn-Met group in 100 mg/kg significantly increased haugh unit (P < 0.05) as compared to the control. The activity of MDA in serum had a linear decrease in 20 to 100 mg/kg Zn-Met. The activity of CAT in liver and GSH-Px in serum had quadratic effects in response to the Zn-Met treatments. Compared to the control, 60 mg/kg Zn-Met group increased the T-AOC, GSH-Px activity in serum (P < 0.01), and the T-AOC (P < 0.05), CuZnSOD (P < 0.01), GSH-Px (P < 0.01) activity in liver. Compared with the control, the concentration of serum ionic Ca in 80, 100 mg/kg Zn-Met treatments reduced (P < 0.01) significantly while the activity of serum alkaline phosphatase (AKP) increased in the Zn-Met groups of 40, 60, and 80 mg/kg (P < 0.01), and 100 mg/kg (P < 0.05). In conclusion, dietary Zn-Met supplementation at 60 to 80 mg/kg had more positive effects on performance, egg quality, and antioxidant capacity in laying hens as compared to 80 mg/kg ZnSO4.

This study was conducted to investigate the effects of dietary manganese-methionine (Mn-Met) supplementation on the egg quality of laying hens. A total of 480 Jinghong-1 strain layers aged 53 wk were divided into 5 groups with 6 replicates of 16 layers. Birds in the control group were fed a diet supplemented with 60 mg Mn/kg in the form of MnSO4; the birds in other 4 experimental groups were fed a diet supplemented with 20, 40, 60, and 80 mg Mn/kg as Mn-Met, respectively. Dietary Mn-Met treatments significantly affected (P < 0.05) the albumen height, yolk color, and Haugh unit compared to those of the control diet. The Mn contents in the eggshell increased (P < 0.01) significantly by increasing the Mn-Met supplementation, whereas Mn content in eggshell was triple that in the yolk or albumen. Compared with the 60 mg/kg Mn-Met group, the transverse surface in the control group had (P < 0.01) a greater width of mammillary cones, and there were obvious cracks on the outer surface in the control. There was no difference (P > 0.05) in the eggshell gland (ESG) in the expression of calbindin-D28k (CaBP-D28k) mRNA in response to any diet treatment. In conclusion, dietary Mn-Met supplementation increased internal egg quality and the ultrastructure of the eggshell. Compared to the control, 60 mg/kg Mn-Met treatment resulted in improving egg quality, and 20 mg/kg Mn-Met treatment had similar effects the control treatment had on the egg quality. This indicates that the inorganic Mn can be replaced by the lower concentration of Mn-Met.

OBJECTIVE: To study the effect of poly(ethylene glycol)methacrylate(PEG)-arginine-glycine-aspartic(RGD)polymer brushes graft on bacterial adhesion and MC3T3 osteoblast cell attachment on titanium, and to investigate if the modification of titanium will enable the implant to be anti-fouling and promot osteointegration. METHODS: PEG was tethered on titanium surface modified with 2-bromoisobutyryl bromide(denoted as Ti-Br)to form Ti-PEG brushes. Functionalization of the Ti-PEG surface with RGD was performed to form Ti-PEG-RGD brushes. The chemical composition of modified titanium surfaces was characterized by X-ray photoelectron spectroscopy(XPS). Changes in surface hydrophilicity and hydrophobicity were characterized by static water contact angle measurements. Streptococcus mutans(Sm), Actinomyces naeslundii(An)and osteoblast cell were cultured on pure titanium(Ti), Ti-PEG, Ti-PEG-RGD surfaces respectively. There were ten samples in each group. The bacterial adhesion ability and cell attachment were confirmed by fluorescence microscopy and scanning electron microscopy(SEM). RESULTS: The static water contact angle of Ti, Ti-Br, Ti-PEG, Ti-PEG-RGD was less than 10°, 80°, 45°, 55° respectively. XPS confirmed that PEG-RGD brushes were successfully tethered on titanium surfaces. Anti-bacterial test showed that on the pure-Ti, there were large amount of bacteria from both groups, however, in the Ti-PEG, Ti-PEG-RGD surfaces, both kind of bacteria were rare and distributed diffusely. Cell culture test showed that on the Ti-PEG-RGD surfaces, the number of cells was significantly more than that on the Ti and the Ti-PEG surfaces. CONCLUSIONS: PEG can inhibit both kind of bacteria adhesion and osteoblast cell attachment, and PEG-RGD brushes can not only inhibit bacterial adhesion but also promote osteoblast cell attachment.

OBJECTIVE: To investigate the association between breast tissue specified variants in p53 binding sites and the risk of BC in Chinese women. METHODS: ChIP-seq database on p53 binding sites in MCF-7 cell lines was extracted to identify the possible variants in p53 target genes. A hospital-based case-control study was then performed to investigate the association between variants in p53 binding sites and the risk of BC in a Chinese women population. RESULTS: Three variants were identified from the bioinformatics analysis. A total of 1 274 BC cases and 1 255 frequency-matched cancer-free controls were included in this case-control study. The average age was comparable between the case and the control groups, with the P value as 0.318. Meanwhile, distributions on menopausal status, smoking and alcohol intake between cases and controls were similar with the P values as 0.539, 0.258 and 0.131, respectively. The genotype distribution of rs1295925 was significantly different between the case and the control groups. Individuals that carrying rs1295925-CT and rs1295925-TT genotypes were significantly associated with an increased BC risk when compared with rs1295925-CC genotype after adjustment of age, menopausal status, smoking and alcohol intake (OR=1.32, 95%CI: 1.07-1.62 and OR=1.41, 95%CI: 1.13-1.78, respectively). Positive associations were also observed under the allelic, dominant and additive models. CONCLUSION: rs1295925 which located in VMP1 gene was associated with increased BC risk in the Chinese women population.

OBJECTIVE: To compare the consistency of angiography-guided and fractional flow reserve (FFR)-guided management strategy for intermediate coronary lesions. METHODS: The patients whose coronary intermediate lesions were assessed by measuring FFR from November 2012 to August 2014. The stenosis percentage and value of FFR during the procedure were collected. All the image data were collected and four experienced interventional cardiologists were invited to assess the target lesions and make a management strategy independently. The consistency of angiography-guided and fractional flow reserve-guided management strategy were analyzed. RESULTS: In the study, 151 patients were included, of whom, 70.2% were male, the average age was (62.7±9.6) years, 169 vessels were assessed by measuring FFR, 1 being left main, 116 left anterior descending, 27 left circumflex branch, and 25 right coronary artery. There were some correlationship between the stenosis percentage judged by four interventional cardiologists and the stenosis percentage judged during the procedure (r=0.29-0.38, P<0.001), but the difference was significant. When 0.80 was used as FFR threshold value, the consistency rates of angiography-guided management strategy decision made by the four cardiologists with fractional flow reserve-guided management strategy were 72.78%, 71.60%, 75.15%, and 72.78%, respectively. CONCLUSION: Angiography-guided management strategy decision is unreliable, FFR is recommended for management strategy decision for intermediate coronary lesions.

This study was conducted to determine the pharmacokinetic characteristics of bulleyaconitine A (BLA) after oral gavage and intravenous administration of BLA at a single dose of 0.04, 0.12, 0.36 mg/kg (oral) or 0.02 mg/kg (i.v.) in male Sprague-Dawley rats. Plasma concentration profiles were analysed using a non-compartmental pharmacokinetic method. Following i.v. 0.02 mg/kg and oral administration 0.04, 0.12 or 0.36 mg/kg, the geometric mean Cmax values were 19.97, 2.11, 5.11 and 11.47 ng/ml, respectively; the corresponding geometric mean AUC(0-t) values were 10.50, 3.19, 9.59 and 18.10 ng x h/ml, respectively. The median Tmax values were 0.033, 0.167, 0.167 and 0.167 h, respectively. The terminal elimination half-lives (t1/2) were 1.23, 2.48, 1.93 and 2.17h, respectively. The results showed that Cmax and AUC(0-t) increased with increasing doses of BLA. The increase in exposure with increasing dose was lower than expected under conditions of strict proportionality.

In this paper, the second and third order polarizabilities of small Ga(n)As(m) (n + m=4-10) clusters are systematically investigated using the time dependent density functional theory (TDDFT)6-311+G* combined with the sum-over-states method (SOSTDDFT6-311+G*). For the static second order polarizabilities, the two-level term (beta(vec.2)) makes a significant contribution to the beta(vec) for all considered Ga(n)As(m) clusters except for the Ga3As4 cluster. And, for the static third order polarizabilities, the positive channel (gamma(II)) makes a larger contribution to gamma(tot) than the negative channel (gamma(I)). Similar to the cubic GaAs bulk materials, the small Ga(n)As(m) cluster assembled materials exhibit large second order (1 x 10(-6) esu) and third order susceptibilities (5 x 10(-11) esu). The dynamic behavior of beta(-2omega; omega, omega) and gamma(-3omega; omega, omega, omega) show that the small Ga(n)As(m) cluster will be a good candidate of nonlinear optical materials due to the avoidance of linear resonance photoabsorption.

Configuration optimizations, excited state properties, and the frequency-dependent third-order nonlinear optical polarizabilities have been investigated on a series of methyl-silsesquioxane (MeT) cages [CH(3)SiO(1.5)](n) (n=4, 6, 8, and 10) using ab initio quantum mechanical methods coupled with the sum-over-states methods. The obtained electronic absorption spectra show a redshift as the cage size increases, and the absorption spectra are assigned as charge transfers from oxygen p type to silicon s type atomic orbitals. The calculated average third-order polarizabilities of in the three optical physical processes (third-harmonic generation, the electric-field-induced second-harmonic generation, and degenerate four-wave mixing) have wide nonresonance regions. For all the considered species, the values of gamma decrease in the order of [MeT](4)>[MeT](8)(C(2v))=[MeT](10)>[MeT](6)>[MeT](8)(O(h)).

The equilibrium geometries of three isomeric hexapyrrolidine C(60) adducts with T(h), D(3), and S(6) symmetries are optimized by means of the B3LYP method at the 6-31G basis sets in this paper. On the basis of the optimized structures, the excited state and third-order nonlinear optical properties, such as third-harmonic generation (THG), electric-field-induced second-harmonic generation (EFISHG), and degenerate four-wave mixing (DFWM), and two-photon absorption (TPA) cross sections, delta, are calculated by using the TDB3LYP model based on the 6-31G level coupled with the sum-over-states (SOS) method. The computational results show that the transition energies from S(0) to S(1) of the T(h) hexaadduct and the D(3) hexaadduct have a remarkable blue shift by comparison with that of the C(60) parent. These results are in agreement with experimental ones. However, the first singlet excitation energy of the S(6) hexaadduct has a red shift compared with that of the C(60) parent. Accordingly, we predict that different positions located by six addends may result in the different spectrum properties. Finally, the two-photon absorption cross sections indicate that the largest average value of resonant TPA, delta, of the D(3) hexaadduct has a red shift compared with those of the T(h) and S(6) hexaadducts.

Using the ab initio method, the geometrical structures of C(36) and the X (B,N)-doped isomers C(34)X(2) have been optimized. On the basis of the optimized structures, then, the third-order nonlinear optical polarizabilities gamma in the different optical processes of the third-harmonic generation, electric-field induced second-harmonic generation and degenerate four-wave mixing, and two-photon absorption (TPA) cross sections delta are calculated by using TDB3LYP method coupled with the sum-over-states method. The calculated results show that the one-photon allowed excitation process dominate the two-photon excitation process for C(36)-D(6h), whereas the two-photon allowed excitation process dominate the one-photon excitation process for C(36)-D(2d) and C(34)X(2) (B,N). It is found that the largest resonant TPA peaks of dopant fullerenes have a blueshift and the TPA cross sections have an enhancement compared with those of the parent fullerenes of isomers C(36)-D(6h) and C(36)-D(2d).

OBJECTIVE: To further develop Gaultheria leucocarpa var. yunnanensis, anti-bacteria constituents in it were screened. METHOD: The constituents were extracted by chromatographic process. The anti-bacteria test was made with regulatory method of analysis. RESULT AND CONCLUSION: Anti-bacteria test with extracts of water, acetic ester and n-butanol showed that 3 extracts from 22 samples had anti-Staphylococcus aureus action, and the extracts from root and stem showed the same result. 2 extracts could kill Escherichia coli and Pseudomonas aeruginosa. The lower the concentrate, the less the anti-bacteria action was. These results suggested that not only essential oil but other ingredients from G. leucocarpa var.yunnanensis have anti-bacteria activity. Anti-fungi test of the same extracts didn't indicate remarkable action.

RESUMEN

A computer model of radiometric scene simulation for simulated spectra for pollution clouds in complicated environment is proposed. The model is used to introduce the effects of an actual hazardous pollution clouds, such as (CH3)2CHO(CH3)FPO or (CLCH2CH2)2S, into exiting measured background spectra by passive Fourier transform infrared spectrometer (FTIS). The simulated results agree well with the experimental results.

AIM: To compare the effects of water-soluble polysaccharides, FI0-b, and its formic acid-modified derivative, FI0-b-H, on production of human proinflammatory cytokines. METHODS: The polysaccharides were modified by formic acid. Cytokine production was quantitated by radioimmunoassay. mRNA for cytokines was measured by semi-quantitative RT-PCR. RESULTS: FI0-b and FI0-b-H 4, 40, and 400 mg/L significantly downregulated interleukin-1 alpha (IL-1 alpha) production by THP-1 cells induced by lypopolysaccharide (LPS) 1 or 10 mg/L and phorbol myristate acetate (PMA) 200 nmol/L. At lower stimulation with LPS 10 mg/L and PMA 200 nmol/L, both polysaccharides significantly upregulated tumor necrosis factor alpha (TNF alpha) production by THP-1 cells. However, at higher stimulation with LPS 100 mg/L and PMA 200 nmol/L, they downregulated TNF alpha production. FI0-b-H downregulated interleukin-8 (IL-8) production by THP-1 cells at a lower-dose of LPS 1 mg/L and PMA 200 nmol/L, but upregulated IL-8 production at a higher-dose of LPS 10 mg/L and PMA 200 nmol/L. Production of cytokines (IL-1 alpha and TNF alpha) was transcriptionally or post-transcriptionally regulated by FI0-b and FI0-b-H. CONCLUSION: The water-soluble polysaccharides of Ganoderma tsugae mycelium have bidirectional immunomodulatory effects on cytokine production in different stimulatory conditions in a dose-dependent manner. Compared with FI0-b, FI0-b-H has more marked effects on human proinflammatory cytokine production.

METHODS: The herbal polysaccharides were modified by chlorosulfornic acid in dimethyl sulfoxide (Me2SO). Cytokine production was measured by radioimmunoassay, mRNA for the cytokines was measured by semi-quantitative RT-PCR. RESULTS: FI0-c 4 mg/L itself induced IL-1 alpha production by THP-1 cells without stimulants, such as lipopolysaccharides (LPS) and phorbol myristate acetate (PMA). On the other hand, FI0-c and FI0-c-S inhibited the IL-1 alpha production by THP-1 cells with these stimulants. FI0-c and FI0-c-S significantly upregulated TNF alpha production by THP-1 cells without stimulants or at a low dose of LPS 10 mg/L and PMA 200 nmol/L, whereas these polysaccharides markedly downregulated the TNF alpha production by a high dose of LPS 100 mg/L and PMA. Human peripheral blood mononuclear cells (PBMC) responded to FI0-c and FI0-c-S in IL-1 alpha and TNF alpha production in a fashion similar to THP-1 cell responses. FI0-c 4 mg/L downregulated high-dose LPS- and PMA-induced IL-1 alpha or TNF alpha mRNA and their protein production by THP-1 cells. CONCLUSION: The water-soluble polysaccharides of Ganoderma tsugae mycelium have bidirectional immunomodulatory effects on cytokine production in different cell stimulatory conditions. Chemical modification of this polysaccharide changed the intensity of regulatory effect on cytokine production.

RESUMEN

To estimate the appropriate tolerance limit of fluoride in food in China, fluoride-related endemic diseases, background levels of fluoride in foods, and daily total intake of fluoride per capita were studied in addition to the subchronic toxicity test of fluoride in rats. In the general population, the daily total intake of fluoride from food, water, and air is 1.45-3.15 mg per capita. On the basis of these results and other information, it is suggested that the ADI of fluoride in the Chinese population should be 3.5 mg per capita, or 0.058 mg/kg body wt, and the tolerance limit of fluoride should be 1.0 ppm in rice, wheat flour, vegetables, and freshwater fish.

Using monoclonal antibodies with different specificity against the major allergenic components of ragweed pollen, we analyzed their cross-reactivity with two tree pollens, two grass pollens and five other weed pollens which are common in Taiwan by the immunoblot method. It was found that besides reacting with AgE and AgK of the ragweed pollen, the monoclonal antibody 48-5 also reacted with antigens of yellow dock pollen with molecular weights of 40K, 38K, 24K, and 21K. In a preliminary study, sera of two patients containing IgE antibodies to ragweed pollen antigens also reacted to the 40K component of the yellow dock pollen. Furthermore, from the results of allergenic skin testings on 109 patients with bronchial asthma, we found that of 22 patients who had a positive reaction to a crude extract of ragweed pollen, 18(81.8%) also reacted to the crude extract of yellow dock pollen. In conclusion, our results suggest that there exists a common allergenic determinant between pollens of ragweed and yellow dock. It may play an important role in the expression of the sensitivity of patients to these two kinds of pollens.
