RESUMEN

In response to increasingly complex social emergencies, this study realizes the optimization of logistics information flow and resource allocation by constructing the Emergency logistics information Traceability model (ELITM-CBT) based on alliance blockchain technology. Using the decentralized, data immutable and transparent characteristics of alliance blockchain technology, this research breaks through the limitations of traditional emergency logistics models and improves the accuracy and efficiency of information management. Combined with the hybrid genetic simulated Annealing algorithm (HGASA), the improved model shows significant advantages in emergency logistics scenarios, especially in terms of total transportation time, total cost, and fairness of resource allocation. The simulation results verify the high efficiency of the model in terms of timeliness of emergency response and accuracy of resource allocation, and provide innovative theoretical support and practical scheme for the field of emergency logistics. Future research will explore more efficient consensus mechanisms, and combine big data and artificial intelligence technology to further improve the performance and adaptability of emergency logistics systems.

RESUMEN

In interphase nuclei, chromatin forms dense domains of characteristic sizes, but the influence of transcription and histone modifications on domain size is not understood. We present a theoretical model exploring this relationship, considering chromatin-chromatin interactions, histone modifications, and chromatin extrusion. We predict that the size of heterochromatic domains is governed by a balance among the diffusive flux of methylated histones sustaining them and the acetylation reactions in the domains and the process of loop extrusion via supercoiling by RNAPII at their periphery, which contributes to size reduction. Super-resolution and nano-imaging of five distinct cell lines confirm the predictions indicating that the absence of transcription leads to larger heterochromatin domains. Furthermore, the model accurately reproduces the findings regarding how transcription-mediated supercoiling loss can mitigate the impacts of excessive cohesin loading. Our findings shed light on the role of transcription in genome organization, offering insights into chromatin dynamics and potential therapeutic targets.

RESUMEN

Optical frequency comb (OFC) technology can realize the rapid measurement of electric fields (E-fields) with large bandwidth. However, this technology suffers from the problem of high intensity noise, resulting in low sensitivity and a blind frequency region. In order to solve the above problems, a dual-path optical E-field sensor with a common reference arm based on a lithium niobate optical waveguide is proposed. The introduction of the reference arm improves the balance of optical paths and the degree of integration. A segmented electrode is also designed to ensure the generation of reverse electrical signals on two Mach-Zehnder interferometers (MZIs). After exiting from the differential photodetector (PD), the intensity noise can be removed, and the sensitivity of the sensor can be improved. After testing, the maximum intensity noise reduction is about 37 dB, the average noise reduction is about 22.3 dB, and the blind frequency region can be eliminated with the co-arm differential optical E-field (CDOE) sensor in the process of measuring the signal. This sensor can be used in the 1 MHz-12 GHz bandwidth with a sensitivity better than 10 mV/m·âˆšHz.

RESUMEN

The present work studied an acclimation method for phosphorus accumulating organisms (PAOs) with a high content of acetone in culture solutions to develop microbial-based enzyme sensors for highly hydrophobic organophosphorus (OP) pesticides. Through three steps of cultivation and acclimation, only rod-shaped bacteria survived among the various PAOs. The extracellular enzymes released from the acclimated PAOs were salted out by using ammonium sulfate, then purified by a dialysis membrane and a DEAE-Sepharose FF anion exchange column. Two enzyme components were successfully separated-both of which showed hydrolase activity on disodium p-nitrophenyl phosphate (enzyme I, 1.57 µmol/(min·µg); enzyme II, 0.88 µmol/(min·µg) at 45°C). Further, SDS-PAGE gel electrophoresis results showed that the molecular weights of enzymes I and II were about 15.11 and 11.98 kDa, respectively. On this basis, the applicability of the enzyme in hydrophobic OP biosensors was demonstrated.

RESUMEN

Nanochannel plastic membranes are excellent materials for electroosmotic pump (EOP) elements owing to their surface charge properties, flexibility and cost-effectiveness. However, the surface charge properties of plastics are inferior to those of silicate-based materials. This paper reports a performance-enhanced EOP equipped with a glassified track-etch polycarbonate membrane (PC), which has a nanochannel surface covered by allylhydridopolycarbosilane (AHPCS). The effects of applied voltage, pH and membrane pore size on the electroosmotic flow velocity, along with a comparative study of the EOP with coated and pure membranes were investigated. It was found that when low DC voltage (10 - 40 V) was applied to both ends of the pump, the magnitude of the electroosmotic flow was linearly proportional to the voltage when the pore size of the membrane was less than 600 nm. A higher flow rate was obtained with larger pore size membranes. Compared with the uncoated film, the coated one showed faster electroosmosis velocity, with higher stability under the same conditions. For pH 10.0 buffer solution, a flow rate of 89.13 µL/min was obtained in the modified membrane-based EOP with excellent repeatability and durability, while the flow rate was only 37.89 µL/min in the bare PC membrane under 20 V. In order to demonstrate the performance of the developed EOP, the EOP was used for microcomplexometric titration to determine actual tap water hardness. The measured results were highly consistent with the results of a conventional complexometric titration methed. The EOP with an AHPCS-coated plastic membrane expanded the application range to harsh condition solutions, such as high-concentration acids or bases.
