RESUMEN

This field trial was conducted to evaluate two antibiotics at a close-up period in curing the existing IMI and to prevent new clinical and subclinical Intramammary infection (IMI). Two hundred and twelve Holstein cows were assigned to one of three treatment groups: TYLO, MARB and CONT. Cows in TYLO group received 10 mg/kg Tylosin for three days at the close-up period (21 days before calving), cows in MARB group received single SC injection of 8 mg/kg SC marbofloxacin at the close-up period and cows in CONT group remained untreated. Milk samples were collected for somatic cell count, microbial culture and Total oxidant/antioxidant capacity before drying-off, and 3 and 7days post calving. Antimicrobial susceptibility test and RAPT-PCR were performed on S.aureus isolates. No significant differences were detected in total cure rate within the groups, but S. aureus cure rates in TYLO and MARB were significantly higher than in CONT (74 and 73.5 % Vs 58.1 %). No significant differences in total new IMI were observed. Furthermore, the rate of new S. aureus IMI was higher in both treatment groups than in CONT group. Clinical mastitis rate in TYLO (3.8 %) and MARB (5.8 %) was significantly lower than CONT (11.3 %). Paired S. aureus isolates from dry-off and post-calving have been clustered into 9 different RAPD types (A-I). 8 paired strains collected at dry-off were identical to those at post-calving,and 35 strains had more than 60 % dissimilarity. Administration of Tylosin or Marbofloxacin is not useful in all cases;however, they have the potential to reduce the incidence of post-calving clinical mastitis and improve S.aureus cure rate if used selectively.

RESUMEN

Small extracellular vesicles called exosomes are released by almost all cell types and play a crucial role in both healthy and pathological circumstances. Exosomes, found in biological fluids (including plasma, urine, milk, semen, saliva, abdominal fluid and cervical vaginal fluid) and ranging in size from 50 to 150 nm, are critical for intercellular communication. Analysis of exosomal cargos, including micro RNAs (miRNAs), proteins and lipids, has been proposed as valuable diagnostic and prognostic biomarkers of disease. Exosomes can also be used as novel, cell-free, treatment strategies. In this review, we discuss the role, significance and application of exosomes and their cargos in diseases of animals.

Immunological and oxidative alterations have been reported around calving in dairy cattle. In addition, the levels of heavy metals rise in the blood around parturition, which might affect body systems. Therefore, in this Research Communication we evaluate the changes in whole blood lead (Pb), arsenic (As), and cadmium (Cd) around calving, in comparison with the beginning of the dry period, and assess the correlations of these elements with immunological factors and oxidative markers. Samples were collected from 30 clinically healthy dairy cows in the early dry period (-6 w), one week before expected calving (-1 w), and one week postpartum (+1 w). The highest concentrations of Pb, As, and Cd were observed at -1 w and all the three elements decreased after parturition leading to significantly lower As and Cd, compared to -1 w (P < 0.05). The lowest levels of tumor necrosis factor-alpha, immunoglobulin G, interleukin 4, interleukin 10 and haptoglobin were found at -1 w simultaneous with the highest measures of the heavy metals, with tumor necrosis factor-alpha being significantly lower at this time (P < 0.05). At -6 w, As concentration was significantly (P < 0.05) correlated negatively (r = -0.366) and positively (r = 0.417) with total antioxidant capacity and malondialdehyde, respectively. Furthermore, at -1 w Pb and As had significant (P < 0.05) negative correlations with interferon gamma (r = -0.502) and interleukin 4 (r = -0.483), respectively. After parturition, Pb was observed to be negatively correlated with total antioxidant capacity (r = -0.538, P < 0.05). The observed results revealed that the alterations in immunological factors and antioxidant capacity around parturition were correlated with Pb and As levels.

An important factor in the course of daily medical diagnosis and treatment is understanding patients' emotional states by the caregiver physicians. However, patients usually avoid speaking out their emotions when expressing their somatic symptoms and complaints to their non-psychiatrist doctor. On the other hand, clinicians usually lack the required expertise (or time) and have a deficit in mining various verbal and non-verbal emotional signals of the patients. As a result, in many cases, there is an emotion recognition barrier between the clinician and the patients making all patients seem the same except for their different somatic symptoms. In particular, we aim to identify and combine three major disciplines (psychology, linguistics, and data science) approaches for detecting emotions from verbal communication and propose an integrated solution for emotion recognition support. Such a platform may give emotional guides and indices to the clinician based on verbal communication at the consultation time.

Herbal preparations have good medicinal value for potential use as therapeutic agents in cystic echinococcosis. The efficiency of crocin in the case of cystic echinococcosis was investigated, and compared with that of albendazole, one of the few licensed anti-echinococcosis drugs that served as a positive control. Five months after infecting BALB/C mice with E. granulosus sensu lato the experimental group was divided into 7 subgroups containing 10 animals each: 1- Crocin 80 (80 mg/kg), 2- Crocin 40 (40 mg/kg), 3- Crocin 10 (10 mg/kg), 4- Albendazole (200 mg/kg), 5- Crocin 10 (10 mg/kg) +Albendazole (100 mg/kg), 6- Crocin 20 (20 mg/kg) +Albendazole (50 mg/kg), 7- the control (infected and untreated). After two weeks of daily treatment, significant reductions of cysts' weights, sizes, and total numbers concerning the control group were achieved by treatment with crocin 80, crocin 40, crocin 10, crocin 10 + ABZ100, crocin 20+ ABZ50 and ABZ200 (p < 0.05). Moreover, there was no difference concentrations of crocin and those treated with albendazole,. The concentration of bilirubin was higher in the control group than all treated groups with crocin, significantly. However, the ALT activity showeda significant decrease in the crocin 10 group, compared to the crocin 80, crocin 40, crocin 10 + ABZ100, crocin 20 + ABZ50, control groups (p < 0.05). Based on our results, the administration of crocin used at 10 mg/kg concentrations seems a hopeful applicant for the treatment of cystic echinococcosis.

The protective effect of Biebersteinia Multifida on diazinon-induced toxicity in male Wistar rats was investigated over 8 weeks. Impacts of diazinon (10 mg/kg daily), Biebersteinia Multifida (500 mg/kg daily), and coadministration of them on oxidative stress parameters besides hematological and biochemical indices were assessed in various groups. The gas chromatography-mass spectrometry analysis was performed to identify the antioxidant components of plant extract by comparing the mass spectra and retention indices with those given in the literature. Pseudocholinesterase level demonstrated a significant attenuation in the Biebersteinia Multifida+diazinon-treated group in comparison to the diazinon group at the end of the 8th week. Statistical significant differences in hematological and biochemical indices were detectable when the diazinon group was compared to Biebersteinia Multifida+diazinon-treated rats. While diazinon destroyed hepatic and renal functions, Biebersteinia Multifida protected the liver and kidney from diazinon toxic effects by normalizing related function indices at the end of the 8th week. By diminishing malondialdehyde and enhancing the ferric-reducing power, Biebersteinia Multifida minimized the hazardous effect of diazinon-induced oxidative stress. Following these results, the beneficial effects of Biebersteinia Multifida in reducing the toxicity of diazinon should be taken into consideration.

The objective of this study was to compare the effects of anticoagulant and no anticoagulant on routine biochemical analytes in domestic pigeons (Columba livia domestica). Blood samples were obtained from 8 clinically healthy pigeons. The sample obtained from each bird was divided into 4 blood collection tubes containing either ethylenediaminetetraacetic acid (EDTA), lithium heparin, sodium citrate, or no anticoagulant. The concentrations of creatinine, uric acid, triglyceride, total cholesterol, glucose, phosphorus, calcium, magnesium, total protein, albumin, and iron, and the activities of alkaline phosphatase (ALP), alanine aminotransferase (ALT), and aspartate aminotransferase (AST), were measured in blood from each of the blood collection tubes. The values of the measured parameters, with the exception of iron, were significantly lower in the citrated plasma samples compared with the serum samples, even after correcting for dilution. In the lithium heparin plasma samples, significant decreases in albumin, triglyceride, calcium, total cholesterol, and ALP, and a significant increase in iron, were observed compared with the values in the serum samples. The concentrations of total protein, creatinine, glucose, total cholesterol, triglyceride, magnesium, calcium, and phosphorus, as well as the activities for AST and ALP, were significantly lower in the EDTA plasma samples compared with the serum samples. In conclusion, the anticoagulants had significant effects on most of the measured parameters compared with serum. The findings of the present study suggest that a lithium heparin sample is the most appropriate plasma sample for the measurement of blood biochemical parameters in the domestic pigeon.

BACKGROUND: Administration of antineoplastic drugs may cause azoospermia driving to subfertility. Production of animal azoospermia models is essential for evaluating new treatment methods before therapeutic interventions in human setup. This study aimed to investigate the toxic effects of Busulfan (an anticancer drug) on some vital organs and describe the best method and appropriate dose of Busulfan to induce an animal azoospermia model. METHODS: Rats were randomly assigned into four groups, treatment groups received 10 mg/kg, 40 mg/kg Busulfan intraperitoneally (IP), 5 mg/kg Busulfan intratesticular (IT), and control group. Blood, bone marrow, liver, renal, and testes samples were collected for histological (H&E staining), biochemical (serum levels of ALT, AST, ALP, creatinine, and urea), and hematological analyses. RESULTS: Results revealed severe anemia and leukopenia in rats that received Busulfan via IP. By contrast, injection of 5 mg/kg Busulfan via IT did not cause anemia except with a mild decrease in RBC count. Non-significant differences in the M/E ratio were observed in all groups. The administration of 40 mg/kg of Busulfan led to evacuation and destruction in the spermatogenesis process with thin-walled seminiferous epithelium in most tubules, but in rats treated with 10 mg/kg of Busulfan, the normal spermatogenesis process was notified. IT injection of Busulfan contributed to the complete degradation of spermatogenesis in which all spermatogenic cells degenerated. In the renal tissue, hyperemia, extensive tubular necrosis degeneration, and hyaline casts were found after IP injection of Busulfan. In hepatic tissue, focal hemorrhagic, chronic cholangitis, and hepatocyte degeneration, and swelling were noticed. Biochemical analysis revealed apparent Busulfan toxicity of both hepatic and renal tissues in IP Busulfan-treated rats. CONCLUSIONS: In summary, we found that the intratesticular injection of low doses of Busulfan (5 mg/kg) is a relatively non-invasive and safe method for producing the rat azoospermia model causing the least toxicity on vital organs.

Although serum protein electrophoresis is a diagnostic tool available through many veterinary laboratories, there currently are no reference intervals for protein fractions in healthy common mynahs (Acridotheres tristis). Therefore, electrophoretic patterns of proteins in serum and heparinized plasma of the common mynah were evaluated. Blood specimens were collected from 55 healthy adult common mynahs of unknown age (26 males and 29 females). The serum total protein and protein fractions were measured using the biuret method followed by cellulose acetate electrophoresis (CAE). The serum level of albumin was compared with bromocresol green (BCG) dye-binding and CAE methods. Four protein fractions, including albumin and α, ß, and γ globulins, were recorded in the electrophoretogram of serum specimens. Sex appeared to have no significant effect on the measured parameters. The serum BCG albumin fraction was significantly higher than the CAE albumin fraction (P = .01). Also, the comparison of total protein and protein fractions in serum and plasma specimens of 25 of the 55 birds sampled showed that total protein (Cohen index d = 0.66, P = .03), gamma globulin (d = 1.13, P = .00), and total globulin (d = 0.67, P = .00) in plasma samples were significantly higher than those in serum samples. The results of this study provide the specific reference intervals for total protein and protein fractions in common mynahs, which are essential for proper interpretation of laboratory results and also revealed that the albumin measurement by the BCG method yields unreliable results in common mynahs.

Our objective was to evaluate relationships between milk components (acute phase proteins, enzymes, metabolic parameters and oxidative indices) and the spontaneous cure outcome of Staphylococcus aureus subclinical mastitis in dairy cows. The values of haptoglobin, serum amyloid A (SAA), malondialdehyde (MDA), total antioxidant capacity, milk urea nitrogen (MUN), lactate dehydrogenase (LDH), alkaline phosphatase (ALP), electrolytes (Cl and K), total protein, albumin, α-lactalbumin, ß-lactoglobulin, and immunoglobulin were measured in milk samples of S. aureus subclinical mastitis cows with spontaneous cure (n = 23), S. aureus subclinical mastitis cows without spontaneous cure (n = 29) and healthy cows (n = 23). The comparison of measured parameters revealed that subclinical mastitis cows with spontaneous cure had lower ALP and haptoglobin concentrations both at diagnosis and after cure (P < 0.05). In contrast, total antioxidant capacity and MDA concentration in subclinical mastitis cows without spontaneous cure significantly increased with time (P < 0.05). We can suggest that elevated haptoglobin concentration and higher ALP activity indicative of enhanced oxidative stress could potentially serve as early diagnostic indicators of chronic disease and the persistence of S. aureus subclinical mastitis in dairy cows.

AIMS: This study aimed to explore the therapeutic effects of amniotic fluid-derived extracellular vesicles including exosomes (AF-Exos) on the recovery of sperm production capacity in a rat model of azoospermia. MAIN METHODS: The non-obstructive azoospermia (NOA) was induced in rats using intratesticular administration of Busulfan. Azoospermia was confirmed by testis histology. AF-Exos samples containing 10 or 40 µg exosomal proteins were injected into testicular tissue of NOA rats. After two months, the recovery of spermatogenesis was monitored via histopathological staining, spermiogram, and hormonal analysis. Immunohistochemistry staining for OCT-3/4 was used to identify of spermatogonial progenitors. The expression of DAZL and VASA, was also measured. KEY FINDINGS: AF-Exos exhibited sphere-shaped morphology with the mean diameter and zeta potential of 50 ± 7.521 nm and -7.16 mV. Immunoblots revealed that isolated nanoparticles were CD63, CD9, and CD81 positive. Histopathological evaluation revealed that spermatogenesis was improved significantly in NOA rats after AF-Exos injection. Data showed that the sperm parameters and spermatogenesis index were significantly improved after AF-Exos injection compared to azoospermic groups. OCT-3/4+ cells were increased in NOA rats after AF-Exos injection, showing the restoration of spermatogenesis. In the present study, both doses of exosome (10 and 40 µg) restored the testicular function of NOA rats. DAZL and VASA were increased significantly in animals who received 40 µg exosomal protein compared to azoospermic rats. Except in a high dose of AF-Exos (40 µg) for Testosterone and FSH, no statistically significant differences were found regarding hormones post-exosome injection. SIGNIFICANCE: Our study demonstrated that AF-Exos regenerated spermatogenesis and improved sperm quality in NOA rats.

The objective of the present study was to evaluate the alterations in selected indicators of immune responses and oxidative stress of broilers fed with nano-manganese. One hundred-sixty 1-day-old broiler chicks were randomly assigned into four groups with three replicates. Birds were fed the same basal diet supplemented with nano-manganese oxide, as 0.00 (control group), 50.00, 100, or 150 mg kg-1 of diet. The birds were vaccinated against avian influenza (AI), Newcastle disease (ND), infectious bronchitis (IB) and infectious bursal disease (IBD) as the standard vaccination schedule. Blood sample was taken from the brachial vein of birds on 42th day. A significant decrease in antibody titer against sheep RBC was revealed in the nano-manganese 100 and 150 groups compared to the control group. In addition, the antibody titers against IB and ND were significantly lower in the all nano-manganese groups compared to the control group. No significant difference was observed for the antibody titer against AI and oxidative stress indices among the experimental groups. The findings in the present study suggested that nano-manganese at 50.00, 100 and 150 mg kg-1 levels might suppress humoral immune response in broilers which should be taken into consideration in supplementation.

Along with the decline in oocyte quality, numerous defects such as mitochondrial insufficiency and the increase of mutation and deletion have been reported in oocyte mitochondrial DNA (mtDNA) following aging. Any impairments in oocyte mitochondrial function have negative effects on the reproduction and pregnancy outcome. It has been stated that infertility problems caused by poor quality oocytes in women with in vitro fertilization (IVF) and repeated pregnancy failures are associated with aging and could be overcome by transferring large amounts of healthy mitochondria. Hence, researches on biology, disease, and the therapeutic use of mitochondria continue to introduce some clinical approaches such as autologous mitochondrial transfer techniques. Following mitochondrial transfer, the amount of ATP required for aged-oocyte during fertilization, blastocyst formation, and subsequent embryonic development could be an alternative modality. These modulations improve the pregnancy outcome in women of high reproductive aging as well. In addition to overview the clinical studies using mitochondrial microinjection, this study provides a framework for future approaches to develop effective treatments and preventions of congenital transmission of mitochondrial DNA mutations/diseases to offspring. Mitochondrial transfer from ovarian cells and healthy oocytes could lead to improved fertility outcome in low-quality oocytes. The modulation of mitochondrial bioactivity seems to regulate basal metabolism inside target oocytes and thereby potentiate physiological activity of these cells while overcoming age-related infertility in female germ cells.

BACKGROUND: Exosomes may have critical roles in the maternal-embryo cross-talk for the recognition and maintenance of pregnancy during maternal aging. Exosomes have the capability to carry developmental signaling molecules with the ability to modulate gene expressions and affect growth and regulation of embryo during pregnancy. Systematic review aims to evaluate age-related alterations in the exosomal content and functions that can influence the reproductive performance in human and animal models as conveyors of senescence signals. METHODS: A literature search of electronic databases including MEDLINE (PubMed), Embase, ProQuest, Scopus, Google Scholar, WHO, SID, MAGIRAN, and Barakat will be conducted. Following the online search, articles will be screened by two independent reviewers according to inclusion and exclusion criteria. Eligible studies will be critically appraised by reviewers at the study level for methodological quality using Joanna Briggs Institute's standardized critical appraisal tools. The extracted data from selected studies will cover the study populations, methods, current evidence about the physiological role of extracellular vesicles and exosomes in reproductive system, relevant outcomes, and possible conclusions about the effectiveness of exposure. DISCUSSION: Regarding the role of exosomes and their cargoes in the function of reproductive tract, the possible beneficial or adverse effects following exosomal administration from younger women to older women will be evaluated in the systematic review. As a result, exosome therapy could be suggested as a novel therapeutic agent if the favorable results are identified.

An alternative mechanism of cell-to-cell communication via extracellular vesicles (EVs) has recently raised increasing attention. EVs are spherical structures comprising exosomes and microvesicles, capable of transferring regulatory molecules and genetic information from one cell to another. EVs act as modulators which can alter a wide spectrum of functions at the cellular level in the recipient cells, taking part in a variety of biological processes in both physiological and pathological conditions. Alteration in EVs content, notably exosomes, was reported during cellular senescence and in patients with age-related diseases. Most studies reported regulating the impacts of exosomes on fertility and pregnancy outcomes via their capability in carrying developmental signaling molecules like proteins, RNA cargos, influencing gene expressions, affecting growth, and development of embryos during aging. Alterations in the exosomal content and functions can influence the reproductive performance in human and animals as conveyors of senescence signals from outside of the cells. This review aimed to summarize evidence on the role of EVs on modulating fertility, early embryonic development, maternal-embryo crosstalk for the recognition, and maintenance of pregnancy during maternal aging. Advanced clinical studies are required to strengthen the findings that the benefit of exosomes can be extended to subjects undergoing reproductive aging. © 2019 BioFactors, 45(3):293-303, 2019.

The aim of this study was to elucidate the protective effect of glycyrrhizin on diazinon-induced changes in body and organ weights, blood hematology, lipid profile, biochemistry parameters and tissue markers of oxidative stress in male Wistar rats over a 7-week period. Rats were orally given sublethal dose of diazinon (10 mg/kg daily; 0.008 LD50), while glycyrrhizin (25 mg kg-1 daily) was given alone or in combination with diazinon. At the end of 7th week, statistically significant decrease of pseudocholinesterase activity was detected when diazinon- and glycyrrhizin + diazinon-treated groups were compared to the control group. Diazinon treated rats showed weight loss and organ weight changes which were comparable to other groups. There was a statistically significance in hematological indices except mean corpuscular hemoglobin (MCH) when diazinon treated group was compared to glycyrrhizin + diazinon treated rats. Glycyrrhizin protected the liver and kidney from diazinon toxic effects with significantly decrease in serum aspartate aminotransferase, alanine aminotransferase, alkaline phosphatase and lactate dehydrogenase activities as well as ameliorated hepatic and renal function indices (such as bilirubin, total protein, albumin, BUN, creatinine glucose). In addition, glycyrrhizin minimized the hazardous effect of diazinon on plasma lipids and lipoproteins. The protective effects of glycyrrhizin were confirmed by tissue markers of oxidative stress analysis as glycyrrhizin in combination diminished malondialdehyde and glycyrrhizin alone or in combination enhanced thiol group and the ferric reducing power. In accordance to these results, our observations demonstrated the beneficial effects of glycyrrhizin in reducing the toxicity of diazinon.

BACKGROUND: Diazinon (DZN) is an organophosphate pesticide commonly used for pest control in agriculture. It may engender a variety of negative effects in non-target species, including humans and animals. The objective of the present study was to evaluate the ameliorative properties of captopril (CAP), as a thiol containing an angiotensin-converting enzyme inhibitor, against DZN-induced oxidative stress. METHODS: Twenty-eight male Wistar rats were divided randomly into 4 groups. All the rats were treated orally via gavage once a day for 7 weeks: control (corn oil), CAP (10 mg/kg), DZN (10 mg/kg), and CAP+DZN combination (as mentioned above). Oxidative stress indices in blood serum, liver and kidney homogenates (malondialdehyde [MDA], total thiol groups, and total antioxidant capacity), and erythrocyte hemolysis (superoxide dismutase [SOD] and glutathione peroxidase) were evaluated. Statistical analysis was performed using GraphPad Prism software, version 6.0 (GraphPad, San Diego, CA, USA), by ANOVA, followed by the Tukey post hoc analysis. RESULTS: The MDA content and SOD activity increased significantly in the DZN group compared with those in the control group. Treatment with CAP in the DZN-exposed group significantly decreased (P<0.05) the MDA concentration and the SOD activity. The total thiol groups were decreased in the DZN group and elevated again by CAP treatment. CONCLUSION: The co-administration of CAP and DZN was able to attenuate lipid peroxidation and enzyme changes caused by DZN.

The aim of this study was to investigate the relationships between oxidative stress, haematology and iron profile in neonatal dairy calves. Serum and haemolysate malondialdehyde (MDA), serum total antioxidant capacity, thiol groups, iron, total iron binding capacity, transferrin saturation and red blood cell (RBC) parameters were assessed in two groups: anaemic calves (n=14) and non-anaemic calves (n=16). Blood samples were collected from all of the calves within 24-48 hours after birth and at 7, 14, 21 and 28 days of age. A significant decrease in serum iron amount and transferrin saturation value (P<0.05) and a significant increase in haemolysate MDA concentration (P<0.05) in the anaemic calves were observed, when compared with non-anaemic calves. Total antioxidant capacity and thiol groups showed a significant positive correlation with iron profile and RBC parameters (haematocrit and haemoglobin) in the anaemic calves at day 21 (P<0.05). On the other hand, the concentration of haemolysate MDA was inversely correlated with the value of serum total antioxidant capacity (P<0.05). The results of the present study revealed that anaemic calves showed more severe oxidative stress than non-anaemic calves. In addition, iron insufficiency may be linked to the impairment of antioxidant defence system and oxidative damage of erythrocytes in the neonatal calves.

BACKGROUND: The aim of this study was to investigate the role of oxidative stress in the pathology of ovine dicrocoeliasis. METHODS: During Dec 2013 - Oct 2014, seventy-two sheep (1-3 years) with liver dicrocoeliasis along with 47 healthy sheep were selected from animals admitted for slaughtering at slaughterhouse located in Neyshabour, Razavi Khorasan Province, Iran. RESULTS: In comparison to healthy control, the malondialdehyde (MDA) level and serum total antioxidant capacity were significantly higher in the parasitized group (P<0.05). A significant increase in liver MDA concentration (P<0.05) of parasitized group was also observed. Packed cell volume (PCV), zinc, iron, total bilirubin and albumin sera levels were significantly lower in the parasitized group (P<0.05). In parasitized sheep, a significant positive correlation was seen between serum MDA concentration and the activity of aspartate aminotransferase (AST). On the other hand, the concentration of serum MDA was inversely correlated with the value of PCV. No significant differences were observed for MDA concentration and total antioxidant capacity between normal and abnormal hepatic lobes in the parasitized animals. Oxidative stress markers (MDA and total antioxidant capacity in serum and liver samples) showed no significant correlations with the extent of pathological lesions and serum variables of liver injury in the parasitized sheep. No significant correlation was observed between oxidative stress markers and the fluke's number in the parasitized animals. CONCLUSION: Oxidative stress may play an important role in the erythrocyte destruction in sheep naturally infected with D. dendriticum. However, no clear relationships were observed between the oxidative stress, hepatic damage and parasite burden.

BACKGROUND: We investigated the variations of heamathologic and biochemical parameters with different parasitic burdens and the correlation between fecal egg counts and fluke number in livers of sheep naturally infected with Dicrocoelium dendriticum. METHODS: In order to detect excretion of D. dendriticum eggs, fecal samples from 120 sheep of different age groups were collected individually at slaughterhouse in Neyshabur County in Razavi Khorasan Province during December 2013 - October 2014. Hematologic and blood biochemical values in 120 sheep naturally infected with D. dendriticum were compared with equivalent values in 120 parasite-free sheep from an organically farmed flock. Investigated animals were kept in outdoor system, on pastures covered with swamps, which remain flooded after rainy season. RESULTS: Total numbers of flukes in livers obtained from infected sheep at autopsy varied between 10 and 18,500. A positive relationship was noted (r= 0.94, P < 0.001) between the number of eggs excreted by each sheep and that of total fluke counts in the liver. Significantly, lower red blood cell, packed cell volume, neutrophil and albumin were observed in parasitized sheep, when compared to the healthy animals. White blood cells,, lymphocyte and aspartate aminotransferase values were significantly higher in parasitized sheep. No significant correlation was detected between the hematologic and biochemical parameters and the number of D. dendriticum in the liver. CONCLUSION: Infection with > 400 D. dendriticum in the liver could be diagnosed by egg per gram. However, there was no relationship between the parasite burden and hematologic and biochemical parameters.
